Migration carriers act as the “Bridges” connecting source rock and traps and play important roles in petroleum migration and accumulation system. Among various types of carriers, sandstone carrier constitutes the basis of carrier system consisting of connected sandstone bodies, of sand-bodies connected with other carriers, such as faults and/or unconformities. How do we understand sandstone carrier beyond the traditional reservoirs concept? How could we characterize quantitatively this kind of carriers for petroleum migration? Such subjects are important and difficult contents in dynamic studies on hydrocarbon migration and accumulation. Sandstone carrier of Chang 8 member in Longdong area of Ordos Basin is selected as the research target in this thesis. Through conducting integrated reservoir analysis on many single wells, the correlation between single sandstone thickness and oil thickness seems good. Sketch sandstone is defined in this thesis as the principal part of carrier based on systematical analysis on lithology and sandstone thickness. Geometry connectivity of sandstone bodies was identified by the spatial superposition among them and was proved by the oil property features in oilfields. The connectivity between sandstone carriers is also hydrodynamically studied by observing and analyzed various diagenetic phenomena, especially the authigenic minerals and their forming sequence. The results were used to characterize transporting capability of sandstone carriers during the key petroleum migration periods. It was found that compaction and cementation are main causes to reduce pore space, and resolution may but not so importantly increases pore space after the occurrence of first migration. The cements of ferrocalcite and kiesel seem like the efficient index to demonstrate the hydraulic connection among sandy bodies. Diagenetic sequence and its relationship with petroleum migration phases are analyzed. Sandstone carrier of Chang 8 member was then characterized by studying their pore space and permeable properties. The results show an average porosity and permeability of Chang 8 carriers are respectively 8% and 0.50md, belongs to low porosity - low permeability reservoirs. Further, the physical properties of Chang 81 member are commonly better than those of Chang 82 member. Methods to reconstruct property of sandstone carrier during petroleum migration phase (late Jurassic) are built based on diagenetic sequence. Planal porosity, porosity and permeability of sandstone carrier in this period are statistically analyzed. One combining index - product of thickness and ancient porosity - is selected as the idea parameter to characterize sandstone carrier of late Jurassic after contrast with other parameters. Reservoirs of Chang 8 member in Longdong area are lithological reservoir controlled by sand body in which oil layers in middle part are clamped with dry layers in upper and lower parts, in a sandwich way. Based a newly proposed “migration-diagensis-remigration” model in low permeability sandstone of Chang 8 member in Longdong area, oil migration and accumulation processes during different periods are simulated with the reconstructed sandstone carriers system. Results match well with current reservoir distributions. Finally, suggestions for next favorable exploration areas are given based on all research achievements.

A large number of mantle-derived fluid activities occurred in the Dongying Sag. On the basis of the studies on the geochemical characteristics of these fluids in this sag, the spatial distribution of biomarkers in petroleum and their relationships with the parameters of mantle-derived fluids were studied, to reveal the influence of mantle-derived fluids on the biomarkers and to evaluate the reliability of these biomarkers when applied to oil-source correlation and maturity analysis. Most biomarkers used in oil correlation kept the characteristics of their sources during burial thermal evolution. Even some of them were not influenced by mantle derived fluids, such as the relative content of C27-C29 steroid(ααα20R)and C21/C23 tricyclic terpane. However, Pr/Ph and C35/C34 hopanes were sensitive to both heat energy and materiel input by the mantle-derived fluids. γ-waxnae/C30hopanes and C24 tetracyclic terpanes /C26 tricyclic terpanes responded only to thermal influence by mantle-derived fluids. They did not chemically reacted with the mantle-derived fluids. Fluorene series compounds reacted with hydrogen and / or carbon dioxide from the mantle. Mantle-derived fluids affected most maturity index. The huge thermal energy with mantle-devied fluids weakened the relationship between the maturity parameters and depth. Among them, pregnane/C27-29 steroid and Ts/(Ts+Tm) were more sensitive to the heat of the fluids. ααα20S/(20S+20R) took the second place. αββ/(ααα+αββ) and 22S/(22S+22R) were not thermally influenced by the mantle-derived fluid. Besides, the substance of mantle-derived fluids reacted with fragrants, hopanes or moretanoids and thus altered the values of MPI1, MPI2, MPR, C30 hopanes/(C30 hopanes + moretanoids) and alkyl-diben zothiophene/diben zothiophene. The thermal alernation of phenanthrene series and their spatial distribution show that the heat energy carried by mantle-derived fluids was not fierce but spread widely in Dongying Sag, which is favorable to hydrocarbon generation with little destroy. In sum, mantle-derived fluids affected biomarkers through thermal energy and chemical reactions and changed the values of oil-source correlation and maturity parameters in the deep-seated fault belts. Therefore, in the deep-seated fault belts, oil-source correlation should be restudied and the new parameters need to be explored.

The modeling of petroleum flow path (petroleum charging) and the detail of corresponding software development are presented in this paper, containing principle of petroleum charging, quantitative method, and practical modeling in two oil fields. The Modeling of Petroleum Flow Path is based on the result of basin modeling, according to the principle of petroleum migrating along the shortest path from the source to trap, Petroleum System Dynamics (Prof. Wu Chonglong, 1998), the concept of Petroleum Migration and Dynamic Accumulation (Zhou Donyan, Li Honhui, 2002), etc. The simulation is done combing with all parameters of basin, and considering the flow potential, non-uniformity of source and porous layer. It's the extending of basin modeling, but not belong to it. It is a powerful simulating tool of petroleum system, and can express quantitatively every kind of geology elements of a petroleum basin, and can recuperate dynamically the geology processes with 3D graphics. At result, we can give a result that the petroleum flow shows itself the phenomena of main path, and without using the special theory such as deflection flow in fractures(Tian Kaiming, 1989, 1994, Zhang Fawang, Hou Xingwei, 1998), and flow potential(England, 1987). The contour map of petroleum flow quantitative show clearly where the coteau - dividing slot is, and which convergence region are the main flow path of petroleum, and where is the favorable play of petroleum. The farsighted trap can be determined if there are enough information about structural diagram and can be evaluated, such as the entrapment extent, spill point, area, oil column thickness, etc. Making full use of the result of basin modeling with this new tool, the critical moment and scheme of the petroleum generation and expulsion can be showed clearly. It's powerful analysis tool for geologist.

Voice alarm plays an important role in emergency evacuation of public place, because it can provide information and instruct evacuation. This paper studied the optimization of acoustic and semantic parameters of voice alarms in emergency evacuation, so that alarm design can improve the evacuation performance. Both method of magnitude estimation and scale were implemented to investigate participants' perceived urgency of the alarms with different parameters. The results indicated that, participants evaluated the alarms with faster speech rate, with greater signal to noise ratio (SNR) and under louder noises more urgent. There was an interaction between noise level and content of voice alarm. Signals with speech rate below 4 characters / second were evaluated as non urgent at all. Intelligibility of the voice alarm was investigated by evaluating the key pointed recognition performance. The results showed that, speech rate’s effect was a marginal significance, and 7 characters / second has the highest intelligibility. It might because that the faster the signal spoken, the more attention was paid. Gender of speaker and SNR did not have a significant effect on the signals’ intelligibility. This paper also investigated impact of voice alarms' content on human behavior in emergency evacuation in a 3-D virtual reality environment. In condition of "telling the occupants what had happened and what to do", the number of participants who succeeded in evacuation was the largest. Further study, in which similar numbers of participants evacuate successfully in three conditions, indicated that the reaction time and evacuation time was the shortest in the aforesaid condition. Although one-way ANOVA shows that the difference was not significant, the results still provided some reference to the alarm design. In sum, parameters of voice alarm in emergency evacuation should be chosen to meet needs from both perceived urgency and intelligibility. Contents of the alarms should include "what had happened and what to do", and should vary according to noise levels in different public places.

RESUMO: O presente trabalho teve como objetivo avaliar o teor e a produtividade de genótipos de girassol semeados em segunda safra no ano de 2014 em Campo Novo do Parecis ? MT, no campo experimental do Instituto Federal de Educação Ciência e Tecnologia de Mato Grosso. O delineamento experimental utilizado foi o de blocos casualizados, com 16 tratamentos (16 genótipos) e quatro repetições. As parcelas experimentais foram constituídas de 4 linhas com 6,5 m de comprimento, com espaçamento entrelinhas de 0,45 m, contendo área de 11,7 m², totalizando uma área de 748 m². Foi utilizada a população de 45000 plantas por hectare. Os dados foram submetidos à análise de variância e ao teste Scott-Knott, a 5% de probabilidade. Os genótipos que se destacaram em relação à produtividade de aquênios foram o MG 360, AGUARÁ 06, MG 305, AGUARÁ 04, CF 101, SYN 045, GNZ NEON, HELIO 251 e SYN 3950HO. Para o teor de óleo nos aquênios e produtividade de óleo, o genótipo MG 360 apresentou o maior valor e se destacou em relação aos demais genótipos analisados. ABSTRACT: This study aimed to evaluate genotypes of sunflower seeded second harvest in the year 2014 in Campus Campo Novo do Parecis, in the experimental field of the Instituto Federal de Educação Ciência e Tecnologia de Mato Grosso. The experimental design was a randomized block design with treatments 16 (16 genotypes) and four replications. The experimental plots consisted of four rows 6.5 m long with row spacing of 0.45 m, containing area of 11.7 m², totaling an area of 748 m². The population of 45000 plants per hectare is used. Data were subjected to analysis of variance and the Scott-Knott test at 5 % probability. The genotypes that stood out in relation to achenes productivity were MG 360, AGUARÁ 06, MG 305, AGUARÁ 04, CF 101, SYN 045, GNZ NEON, HELIO 251 and SYN 3950HO. For oil content and oil productivity, MG 360 genotype showed the highest value and stood out in relation to other genotypes.

With web caching and cache-related services like CDNs and edge services playing an increasingly significant role in the modern internet, the problem of the weak consistency and coherence provisions in current web protocols is becoming increasingly significant and drawing the attention of the standards community [LCD01]. Toward this end, we present definitions of consistency and coherence for web-like environments, that is, distributed client-server information systems where the semantics of interactions with resource are more general than the read/write operations found in memory hierarchies and distributed file systems. We then present a brief review of proposed mechanisms which strengthen the consistency of caches in the web, focusing upon their conceptual contributions and their weaknesses in real-world practice. These insights motivate a new mechanism, which we call "Basis Token Consistency" or BTC; when implemented at the server, this mechanism allows any client (independent of the presence and conformity of any intermediaries) to maintain a self-consistent view of the server's state. This is accomplished by annotating responses with additional per-resource application information which allows client caches to recognize the obsolescence of currently cached entities and identify responses from other caches which are already stale in light of what has already been seen. The mechanism requires no deviation from the existing client-server communication model, and does not require servers to maintain any additional per-client state. We discuss how our mechanism could be integrated into a fragment-assembling Content Management System (CMS), and present a simulation-driven performance comparison between the BTC algorithm and the use of the Time-To-Live (TTL) heuristic.

Object detection can be challenging when the object class exhibits large variations. One commonly-used strategy is to first partition the space of possible object variations and then train separate classifiers for each portion. However, with continuous spaces the partitions tend to be arbitrary since there are no natural boundaries (for example, consider the continuous range of human body poses). In this paper, a new formulation is proposed, where the detectors themselves are associated with continuous parameters, and reside in a parameterized function space. There are two advantages of this strategy. First, a-priori partitioning of the parameter space is not needed; the detectors themselves are in a parameterized space. Second, the underlying parameters for object variations can be learned from training data in an unsupervised manner. In profile face detection experiments, at a fixed false alarm number of 90, our method attains a detection rate of 75% vs. 70% for the method of Viola-Jones. In hand shape detection, at a false positive rate of 0.1%, our method achieves a detection rate of 99.5% vs. 98% for partition based methods. In pedestrian detection, our method reduces the miss detection rate by a factor of three at a false positive rate of 1%, compared with the method of Dalal-Triggs.

Yersiniosis is an acute or chronic enteric zoonosis caused by enteropathogenic Yersinia species. Although yersiniosis is predominantly associated with gastroenteric forms of infection, extraintestinal forms are often reported from the elderly or patients with predisposing factors. Yersiniosis is often reported in countries with cold and mild climates (Northern and Central Europe, New Zealand and North of Russian Federation). The Irish Health Protection Surveillance Centre (HPSC) currently records only 3-7 notified cases of yersiniosis per year. At the same time pathogenic Yersinia enterocolitica is recovered from pigs (main source of pathogenic Y. enterocolitica) at the levels similar to that observed in Yersinia endemic countries. Introduction of Yersinia selective culture procedures may increase Yersinia isolation rates. To establish whether the small number of notifications of human disease was an underestimate due to lack of specific selective culture for Yersinia we carried out a prospective culture study of faecal samples from outpatients with diarrhoea, with additional culture of appendix and throat swabs. Higher levels of anti-Yersinia seroprevalence than yersiniosis notification rates in endemic countries suggests that most yersiniosis cases are unrecognised by culture. Subsequently, in addition to a prospective culture study of clinical specimens, we carried out serological screening of Irish blood donors and environmental screening of human sewage. Pathogenic Yersinia strains were not isolated from 1,189 faeces samples, nor from 297 throat swabs, or 23 appendix swabs. This suggested that current low notification rates in Ireland are not due to the lack of specific Yersinia culture procedures. Molecular screening detected a wider variety of Y. enterocolitica-specific targets in pig slurry than in human sewage. A serological survey for antibodies against Yersinia YOP (Yersinia Outer Proteins) proteins in Irish blood donors found antibodies in 25%, with an age-related trend to increased seropositivity, compatible with the hypothesis that yersiniosis may have been more prevalent in Ireland in the recent past. Y. enterocolitica is a heterogeneous group of microorganisms that comprises strains with different degree of pathogenicity. Although non-pathogenic Y. enterocolitica lack conventional virulence factors, these strains can be isolated from patients with diarrhoea. Insecticidal Toxin Complex (ITC) and Cytolethal Distending Toxins can potentially contribute to the virulence of non-pathogenic Y. enterocolitica in the absence of other virulence factors. We compared distribution of ITC and CDT loci among pathogenic and non-pathogenic Y. enterocolitica. Additionally, to demonstrate potential pathogenicity of non-pathogenic Y. enterocolitica we compared their virulence towards Galleria mellonella larvae (a non-mammalian model of human bacterial infections) with the virulence of highly and mildly pathogenic Y. enterocolitica strains. Surprisingly, virulence of pathogenic and non-pathogenic Y. enterocolitica in Galleria mellonella larvae observed at 37°C did not correlate with their pathogenic potential towards humans. Comparative phylogenomic analysis detects predicted coding sequences (CDSs) that define host-pathogen interactions and hence providing insights into molecular evolution of bacterial virulence. Comparative phylogenomic analysis of microarray data generated in Y. enterocolitica strains isolated in the Great Britain from humans with diarrhoea and domestic animals revealed high genetic heterogeneity of these species. Because of the extensive human, animal and food exchanges between the UK and Ireland the objective of this study was to gain further insight into genetic heterogeneity and relationships among clinical and non-clinical Y. enterocolitica strains of various pathogenic potential isolated in Ireland and Great Britain. No evidence of direct transfer of strains between the two countries was found.

Both the emission properties and the evolution of the radio jets of Active Galactic Nuclei are dependent on the magnetic (B) fields that thread them. A number of observations of AGN jets suggest that the B fields they carry have a significant helical component, at least on parsec scales. This thesis uses a model, first proposed by Laing and then developed by Papageorgiou, to explore how well the observed properties of AGN jets can be reproduced by assuming a helical B field with three parameters; pitch angle, viewing angle and degree of entanglement. This model has been applied to multifrequency Very Long Baseline Interferometry (VLBI) observations of the AGN jets of Markarian 501 and M87, making it possible to derive values for the helical pitch angle, the viewing angle and the degree of entanglement for these jets. Faraday rotation measurements are another important tool for investigating the B fields of AGN jets. A helical B field component should result in a systematic gradient in the observed Faraday rotation across the jet. Real observed radio images have finite resolution; typical beam sizes for cm-wavelength VLBI observations are often comparable to or larger than the intrinsic jet widths, raising questions about how well resolved a jet must be in the transverse direction in order to reliably detect transverse Faraday-rotation structure. This thesis presents results of Monte Carlo simulations of Faraday rotation images designed to directly investigate this question, together with a detailed investigation into the probabilities of observing spurious Faraday Rotation gradients as a result of random noise and finite resolution. These simulations clearly demonstrate the possibility of detecting transverse Faraday-rotation structures even when the intrinsic jet widths are appreciably smaller than the beam width.

RNA editing is a biological phenomena that alters nascent RNA transcripts by insertion, deletion and/or substitution of one or a few nucleotides. It is ubiquitous in all kingdoms of life and in viruses. The predominant editing event in organisms with a developed central nervous system is Adenosine to Inosine deamination. Inosine is recognized as Guanosine by the translational machinery and reverse-transcriptase. In primates, RNA editing occurs frequently in transcripts from repetitive regions of the genome. In humans, more than 500,000 editing instances have been identified, by applying computational pipelines on available ESTs and high-throughput sequencing data, and by using chemical methods. However, the functions of only a small number of cases have been studied thoroughly. RNA editing instances have been found to have roles in peptide variants synthesis by non-synonymous codon substitutions, transcript variants by alterations in splicing sites and gene silencing by miRNAs sequence modifications. We established the Database of RNA EDiting (DARNED) to accommo-date the reference genomic coordinates of substitution editing in human, mouse and fly transcripts from published literatures, with additional information on edited genomic coordinates collected from various databases e.g. UCSC, NCBI. DARNED contains mostly Adenosine to Inosine editing and allows searches based on genomic region, gene ID, and user provided sequence. The Database is accessible at http://darned.ucc.ie RNA editing instances in coding region are likely to result in recoding in protein synthesis. This encouraged me to focus my research on the occurrences of RNA editing specific CDS and non-Alu exonic regions. By applying various filters on discrepancies between available ESTs and their corresponding reference genomic sequences, putative RNA editing candidates were identified. High-throughput sequencing was used to validate these candidates. All predicted coordinates appeared to be either SNPs or unedited.

Mesenchymal stem cells (MSCs) are currently under investigation as repair agents in the preservation of cardiac function following myocardial infarction (MI). However concerns have emerged regarding the safety of acute intracoronary (IC) MSC delivery specifically related to mortality, micro-infarction and microvascular flow restriction post cell therapy in animal models. This thesis aimed to firstly identify an optimal dose of MSC that could be tolerated when delivered via the coronary artery in a porcine model of acute MI (AMI). Initial dosing studies identified 25x106 MSC to be a safe MSC cell dose, however, angiographic observations from these studies recognised that on delivery of MSC there was a significant adverse decrease in distal blood flow within the artery. This observation along with additional supportive data in the literature (published during the course of this thesis) suggested MSC may be contributing to such adverse events through the propagation of thrombosis. Therefore further studies aimed to investigate the innate prothrombotic activity of MSC. Expression of the initiator of the coagulation cascade initiator tissue factor (TF) on MSC was detected in high levels on the surface of these cells. MSC-derived TF antigen was catalytically active, capable of supporting thrombin generation in vitro and enhancing platelet-driven thrombus deposition on collagen under flow. Infusion of MSC via IC route was associated with a decreased coronary flow reserve when delivered but not when coadministered with an antithrombin agent heparin. Heparin also reduced MSC-associated in situ thrombosis incorporating platelets and VWF in the microvasculature. Heparin-assisted MSC delivery reduced acute apoptosis and significantly improved infarct size, left ventricular ejection fraction, LV volumes, wall motion and scar formation at 6 weeks post AMI. In addition, this thesis investigated the paracrine factors secreted by MSC, in particular focusing on the effect on cardiac repair of a novel MSC-paracrine factor SPARCL1. In summary this work provides new insight into the mechanism by which MSC may be deleterious when delivered by an IC route and a means of abrogating this effect. Moreover we present new data on the MSC secretome with elucidation of the challenges encountered using a single paracrine factor cardiac repair strategy.
