In recent years the concept of genomic resource banks has grown as a way of maintaining the genetic variability of populations, while the quality of cryopreservation of the gametes determines the effectiveness of such banks. However, the absence of basic knowledge regarding the physiology of species and their semen characteristics hampers the establishment of reproductive biotechnologies. Thus, this paper aimed to determine certain physicochemical (volume, colour, appearance, pH and osmolarity) and microscopic characteristics (mass movement, motility, vigour, concentration, and sperm morphology and morphometry) of semen of the species Mazama americana. To achieve this, five males of the species were used, and three semen samples per buck (electroejaculation) were collected at intervals of 2 weeks. The volume, pH and osmolarity of the ejaculate were 0.39 ± 0.14 mL, 6.90 ± 0.74 and 297.74 ± 19.10 mOsm/kg, respectively, while the values obtained for mass movement, motility, vigour and concentration were 3.33 ± 0.82; 69.6 ± 8.92%; 3.53 ± 0.50, and 244.07 ± 98.65 × 107/mL, respectively. Regarding the colour of the ejaculate, five samples were classified as ivory, two as yellowish, two as whitish and six as white. Regarding appearance, seven samples were considered creamy and eight, milky. Morphology was analysed in a humid chamber under phase contrast microscopy and 73.50 ± 5.57% of cells presented normal morphology, 8.37 ± 3.15% presented major defects and 18.13 ± 6.46% presented minor defects. To determine sperm morphometry, an optical microscope (Leica DM 5000B) and the Leica Qwin image analyser program were used, resulting in 8.09 ± 0.40, 4.65 ± 0.30, 2.81 ± 0.44 and 30.25 ± 3.02 m for length, largest width, smallest width and area, respectively. In this article, the authors aim to present a critical review of recent MRI studies addressing white matter (WM) abnormalities in Alzheimer's disease (AD) and mild cognitive impairment (MCI), by searching PubMed and reviewing MRI studies evaluating subjects with AD or MCI using WM volumetric methods, diffusion tensor imaging and assessment of WM hyperintensities. Studies have found that, compared with healthy controls, AD and MCI samples display WM volumetric reductions and diffusion tensor imaging findings suggestive of reduced WM integrity. These changes affect complex networks relevant to episodic memory and other cognitive processes, including fiber connections that directly link medial temporal structures and the corpus callosum. Abnormalities in cortico-cortical and cortico-subcortical WM interconnections are associated with an increased risk of progression from MCI to dementia. It can be concluded that WM abnormalities are detectable in early stages of AD and MCI. Degeneration of WM networks causes disconnection among neural cells and the degree of such changes is related to cognitive decline. © 2013 2013 Expert Reviews Ltd.

Considering the lack of detailed information about the morphology of paca, which is the second largest rodent of Brazilian's fauna, with excellent meat quality and, due to the importance of the common integument, we described the morphology, ultrastructure and morphometry of the skin of eight pacas (Cuniculus paca), males and females, through comparative analysis of skin's segments of the cervical, dorsal and medial carpal portions. Macroscopic characteristics of the coat were studied. Part of the segments of cutaneous regions was analyzed by light microscopy, and the other part by scanning electron microscopy. We measured the thickness of dermis, epidermis, stratum corneum, the profiles of the collagen fibers of reticular dermis and the area of filled sebaceous gland cells. The results were analyzed by descriptive statistics and T test (p<0.001). The color of the coat is reddish brown with bristles arranged in groups. The architecture of the skin and skin appendages resembles those of mammals in general, although there are no sweat glands. According to the morphometric analysis, this study shows differences of the skin architecture between male and female pacas and also between various body regions in the same animal.

The paca (Cuniculus paca) is the second largest rodent of the Brazilian fauna. The excellent meat quality of this specie encourages the development of their commercial production. Moreover, this animal can become a viable alternative for animal experimentation although there exists scarce detailed information concerning their morphology. Therefore the purpose of this study is to describe the morphology, morphometry and ultrastructure in segments of the cranial and caudal portions of vena cava in four adult males and females of Cuniculus paca from the squad of Wild Animals Sector of Animal Science Department of FCAV-Unesp. Parts of the segments were examined by light microscopy and part by scanning electron microscopy. Thickness measures of the tunica intima and media complex and tunica adventitia of the vena cava were taken and analyzed using T test (p<0.05). In vena cava the thickness values of the intima, media and adventitia, for all animals, were significantly higher in the cranial segment. The layers of the vessel walls showed variations in structure and thickness, presumably due to an adaptation to functional demand.

The goal of this study was to evaluate the quality of Cotesia flavipes from different bio-factories as biological control agents. We evaluated biological characteristics of the parasitoids throughout their lifespan, and measured the body length and width, abdomen width, thorax width and width and length of the right forewing of female and male parasitoids. Our results showed that the number of males and pupal viability were similar among the bio-factories; the number of emerged females was greater in bio-factories I and II; the egg-pupa period and the pupal period were shorter in bio-factory IV; and a greater longevity was found in bio-factories II and III. Sex ratio (at approximately 60% females) was satisfactory (in terms of suitability for release) across all bio-factories. For morphometric measurements, the body, abdomen and wing widths were similar in males; however, thorax width was greater in the males from bio-factory I; bio-factory III produced females with the highest body length; bio-factory I produced females with the greatest abdomen width; bio-factories III and IV produced females with the greatest wing length. Among the bio-factories studied, bio-factory IV produced the best quality C. flavipes, with respect to the greatest number of parasitoids per pupal mass (a mean of 57% more parasitoids) in a satisfactory sex ratio, and with the shortest developmental time, which facilitates faster rearing in the laboratory. Studies such as this, which assess the quality of a mass-produced C. flavipes, are crucial for the continued use of this parasitoid in controlling Diatraea saccharalis in the field. © 2013 Copyright Taylor and Francis Group, LLC.

This study aimed to evaluate the osteointegration and genotoxic potential of a bioactive scaffold, composed of alumina and coated with hydroxyapatite and bioglass, after their implantation in tibias of rats. For this purpose, Wistar rats underwent surgery to induce a tibial bone defect, which was filled with the bioactive scaffolds. Histology analysis (descriptive and morphometry) of the bone tissue and the single-cell gel assay (comet) in multiple organs (blood, liver, and kidney) were used to reach this aim after a period of 30, 60, 90, and 180 days of material implantation. The main findings showed that the incorporation of hydroxyapatite and bioglass in the alumina scaffolds produced a suitable environment for bone ingrowth in the tibial defects and did not demonstrate any genotoxicity in the organs evaluated in all experimental periods. These results clearly indicate that the bioactive scaffolds used in this study present osteogenic potential and still exhibit local and systemic biocompatibility. These findings are promising once they convey important information about the behavior of this novel biomaterial in biological system and highlight its possible clinical application. © 2013 Wiley Periodicals, Inc.

Background: Although skeletal muscle atrophy and changes in myosin heavy chain (MyHC) isoforms have often been observed during heart failure, their pathophysiological mechanisms are not completely defined. In this study we tested the hypothesis that skeletal muscle phenotype changes are related to myogenic regulatory factors and myostatin/follistatin expression in spontaneously hypertensive rats (SHR) with heart failure. Methods: After developing tachypnea, SHR were subjected to transthoracic echocardiogram. Pathological evidence of heart failure was assessed during euthanasia. Age-matched Wistar-Kyoto (WKY) rats were used as controls. Soleus muscle morphometry was analyzed in histological sections, and MyHC isoforms evaluated by electrophoresis. Protein levels were assessed by Western blotting. Statistical analysis: Student's t test and Pearson correlation. Results: All SHR presented right ventricular hypertrophy and seven had pleuropericardial effusion. Echocardiographic evaluation showed dilation in the left chambers and left ventricular hypertrophy with systolic and diastolic dysfunction in SHR. Soleus weight and fiber cross sectional areas were lower (WKY 3615±412; SHR 2035±224 μm2; P < 0.001), and collagen fractional volume was higher in SHR. The relative amount of type I MyHC isoform was increased in SHR. Myogenin, myostatin, and follistatin expression was lower and MRF4 levels higher in SHR. Myogenin and follistatin expression positively correlated with fiber cross sectional areas and MRF4 levels positively correlated with I MyHC isoform. Conclusion: Reduced myogenin and follistatin expression seems to participate in muscle atrophy while increased MRF4 protein levels can modulate myosin heavy chain isoform shift in skeletal muscle of spontaneously hypertensive rats with heart failure. © 2012 Elsevier B.V.

The aim of this study was to evaluate the survival rate, the intestinal microbiota, the mucosal integrity, and the carcass quality of juvenile Nile Tilapia, Oreochromis niloticus, after 80 days being fed on a diet containing probiotic additive (Bacillus cereus 4.0×108 CFUg-1 and Bacillus subtilis 4.0×108 CFUg-1), at the ratio of 4g/kg of pelleted feed. The completely randomized design with two treatments was used: one control group and one group fed on the mentioned diet. The evaluation of survival rate, the intestinal microbiota analysis by microbiological culture, histomorphometrical analysis of intestinal mucosa and chemical analysis of carcass was performed. The results showed that tilapias from the treated group had higher relative survival rate (P<0.05) than the control group, higher number of colony-forming units (P<0.05) regarding intestinal colonization by B. cereus and B. subtilis, and higher rates of intestinal mucosal integrity (P<0.05), evaluated by histomorphometry. As for the latter, the group being fed on feed with probiotic additive was observed to have higher and larger villi, besides having a higher number of goblet cells than the control group. Concerning the carcass quality, the results showed that there was positive interference (P<0.05) of the probiotic on the treated group in comparison to the control group as in regard to levels of protein and ether extract. These results allow the inference that the supplementation with probiotic, as tested in this experiment, led to the intestinal colonization by beneficial bacteria and resulted in higher relative survival rate, decreased the mucosal desquamation and helped in the increase of the number of goblet cells.

The relative growths of Persephona lichtensteinii, P. mediterranea, and P. punctata were investigated on the south-eastern Brazilian coast, focusing on differences in the growth rates between immature and mature phases, the onset of morphological sexual maturity, and the breeding seasons of these species. Crabs were collected every two months from January 1991 through to November 1992, from a shrimp fishing boat equipped with two otter-trawl nets. Significant differences in the patterns of body growth were observed between immature and mature phases of all three species. Changes in the growth rates of the chelipeds (males) and abdomen (females) observed for P. lichtensteinii, P. mediterranea, and P. punctata, seem to be related to the puberty moult for both sexes. Males of P. mediterranea and P. punctata reached larger mean sizes of carapace width than females, whereas no difference was recorded for P. lichtensteinii. The body size at which 50% of males attained sexual maturity was also larger in P. mediterranea and P. punctata, and smaller in P. lichtensteinii. The absence of a pronounced sexual dimorphism and the size at the onset of sexual maturity observed only for P. lichtensteinii might be explained by distinct reproductive strategies of males. The presence of ovigerous females during the entire sampling period suggests that all three species have a continuous reproduction pattern at the Ubatuba region. Future studies on the population structure, functional maturity, and mating system should improve the understanding of factors driving the biology and ecology of these species at a subtropical region. © Marine Biological Association of the United Kingdom 2013.

Soft tissue sarcomas (STSs) are a heterogeneous group of mesenchymal tumors of >50 subtypes. However, STSs represent <1% of types of cancer. Despite this low frequency, the disease is aggressive and treatment, when possible, is based on traditional chemotherapies. A number of cases of resistance to adjuvant therapies have been reported. Metastases are commonly identified in STS patients during diagnosis and the development of effective clinical parameters is crucial for correct management of the disease. The use of biological markers in cancer is a useful tool to determine patient prognosis. Ki--67 is a protein marker for proliferation of somatic cells and is widely used in prognostic studies of various types of tumor, including STSs. Cluster of differentiation 100 (CD100) is a member of the semaphorin family. The family was initially described as axon guidance molecules important for angiogenesis, organogenesis, apoptosis and neoplasia. CD100 was previously utilized as a prognostic factor in tumors and also in STSs. In the present study, protein expression of Ki--67 and CD100 was analyzed by immunohistochemistry in samples of STS patients of the Barretos Cancer Hospital (Barretos, Brazil) to establish prognostic criteria of the disease. Results demonstrate a correlation between CD100 expression and poor prognosis, consistent with a previous study. Moreover, the expression of Ki-67 was identified to correlate with presence of local or locoregional recurrence. To the best of our knowledge, no large casuistic study has revealed this correlation between Ki--67 and local recurrence in STSs. The use of Ki--67 and CD100 as markers in clinical pathological analysis may be suitable as a prognostic criterion in disease progression.
