Contents Fibroblast growth factor (FGF10) acts at the cumulus oocyte complex, increasing the expression of cumulus cell expansion-related genes and oocyte competency genes. We tested the hypothesis that addition of FGF10 to the maturation medium improves oocyte maturation, decreases the percentage of apoptotic oocytes and increases development to the blastocyst stage while increasing the relative abundance of developmentally important genes (COX2, CDX2 and PLAC8). In all experiments, oocytes were matured for 22h in TCM-199 supplemented with 0, 2.5, 10 or 50ng/ml FGF10. In Experiment 1, after maturation, oocytes were stained with Hoechst to evaluate meiosis progression (metaphase I, intermediary phases and extrusion of the first polar body) and submitted to the TUNEL assay to evaluate apoptosis. In Experiment 2, oocytes were fertilized and cultured to the blastocyst stage. Blastocysts were frozen for analysis of COX2, CDX2 and PLAC8 relative abundance. In Experiment 1, 2.5ng/ml FGF10 increased (p<0.05) the percentage of oocytes with extrusion of the first polar body (35%) compared to 0, 10 and 50ng/ml FGF10 (21, 14 and 12%, respectively) and FGF10 decreased the percentage of oocytes that were TUNEL positive in all doses studied. In Experiment 2, there was no difference in the percentage of oocytes becoming blastocysts between treatments and control. Real-time RT-PCR showed a tendency of 50ng/ml FGF10 to increase the relative abundance of COX2 and PLAC8 and of 10ng/ml FGF10 to increase CDX2. In conclusion, the addition of FGF10 to the oocyte maturation medium improves oocyte maturation in vitro, decreases the percentage of apoptotic oocytes and tends to increase the relative abundance of developmentally important genes.

A composição e estrutura da comunidade epifítica herbácea de fuste baixo, assim como sua distribuição vertical, foram estudadas. O DAP de hospedeiros arbóreos e o tipo de casca influenciam a riqueza e abundância dessas espécies em um trecho de floresta de terra firme na Amazônia Oriental (1º57’36"S 51º36’55"W). Foram identificadas, no total, 37 espécies herbáceas epifíticas, sendo 60% delas Araceae. A riqueza de espécies e a abundância de herbáceas epifíticas mostraram tendência de correlação positiva com o tamanho de hospedeiros arbóreos e nenhuma relação com o tipo de casca. Correlação positiva baixa pode ser um subproduto da predominância de árvores de menor diâmetro na amostragem em vez de refletir relação neutra. A ausência de relações com o tipo de casca deve ser parcialmente explicada pelo grande número de hemiepífitas secundárias, generalistas, e também refletir a ausência de substratos adequados em árvores de menor diâmetro.

1. The crabeater seal Lobodon carcinophaga is considered to be a key species in the krill-based food web of the Southern Ocean. Reliable estimates of the abundance of this species are necessary to allow the development of multispecies, predator–prey models as a basis for management of the krill fishery in the Southern Ocean. 2. A survey of crabeater seal abundance was undertaken in 1500 000 km2 of pack-ice off east Antarctica between longitudes 64–150° E during the austral summer of 1999/2000. Sighting surveys, using double observer line transect methods, were conducted from an icebreaker and two helicopters to estimate the density of seals hauled out on the ice in survey strips. Satellite-linked dive recorders were deployed on a sample of seals to estimate the probability of seals being hauled out on the ice at the times of day when sighting surveys were conducted. Model-based inference, involving fitting a density surface, was used to infer densities in the entire survey region from estimates in the surveyed areas. 3. Crabeater seal abundance was estimated to be between 0.7 and 1.4 million animals (with 95% confidence), with the most likely estimate slightly less than 1 million. 4. Synthesis and applications. The estimation of crabeater seal abundance in Convention for the Conservation of Antarctic Marine Living Resources (CCAMLR) management areas off east Antarctic where krill biomass has also been estimated recently provides the data necessary to begin extending from single-species to multispecies management of the krill fishery. Incorporation of all major sources of uncertainty allows a precautionary interpretation of crabeater abundance and demand for krill in keeping with CCAMLR’s precautionary approach to management. While this study focuses on the crabeater seal and management of living resources in the Southern Ocean, it has also led to technical and theoretical developments in survey methodology that have widespread potential application in ecological and resource management studies, and will contribute to a more fundamental understanding of the structure and function of the Southern Ocean ecosystem.

Killer whale (Orcinus orca Linnaeus, 1758) abundance in the North Pacific is known only for a few populations for which extensive longitudinal data are available, with little quantitative data from more remote regions. Line-transect ship surveys were conducted in July and August of 2001–2003 in coastal waters of the western Gulf of Alaska and the Aleutian Islands. Conventional and Multiple Covariate Distance Sampling methods were used to estimate the abundance of different killer whale ecotypes, which were distinguished based upon morphological and genetic data. Abundance was calculated separately for two data sets that differed in the method by which killer whale group size data were obtained. Initial group size (IGS) data corresponded to estimates of group size at the time of first sighting, and post-encounter group size (PEGS) corresponded to estimates made after closely approaching sighted groups.

We studied the temporal distribution and reproductive biology of marine podonids during two consecutive years off Ubatuba, southeast coast of Brazil. Podonid specimens and their eggs and embryos were counted, measured and classified into categories. Pseudevadne tergestina was the most abundant species, and was more abundant in surface layers, in warm seasons, when the water column was stratified because of bottom intrusions of the cold and nutrient-rich South Atlantic Central Water (SACW) onto the inner shelf. Evadne spinifera had a similar temporal and vertical distribution, but with lower abundance and frequency. Pleopis schmackeri did not show a clear seasonal distribution, but preferred bottom layers. Pleopis polyphemoides and Podon intermedius occurred in low abundances, and only under SACW influence. Parthenogenetic females were dominant among all podonid species. Gamogenetic females of P. polyphemoides and P. intermedius were observed, but males of neither species occurred. This suggests that in tropical and subtropical regions, P. tergestina, E. spinifera and P. schmackeri reproduce through parthenogenesis during most of the year.

This paper presents a survey of the insects that feed on fruits of Psittacanthus Martius (Santalales: Loranthaceae), a hemiparasitic mistletoe genus that infects trees in Brazil and other neotropical countries. The aim of the study was to identify candidate insects for biological control of Psittacanthus mistletoes. Unripe and mature fruits were collected in several localities of Cerrado, bordering South Pantanal, Southwestern Brazil, from 29 Apr 1998 to 30 Jul 2000. A total of 24,710 fruits (54 samples) of Psittacanthus acinarius infecting 15 species from 10 plant families were evaluated. Psittacanthus acinarius (Mart.) was the most abundant and frequent species of mistletoe parasitizing trees in the ecotonal Cerrado-Pantanal. From 24,710 fruits of Psittacanthus acinarius were obtained 1,812 insect larvae including 1,806 Neosilba McAlpine (Diptera: Lonchaeidae) species and 6 Thepytus echelta (Hewitson) (Lepidoptera: Lycaenidae). From these emerged 1,550 Neosilba spp. adults and 6 T. echelta. Neosilba pantanense Strikis was described from this research. Larvae of T. echelta occurred in fruits of P. acinarius parasitizing Cecropia pachystachya Trecul (Urticaceae) and Anadenanthera colubrina (Vellozo) Brenan (Fabaceae). Larvae of Neosilba caused no adverse effects on the germination of infected fruits of Psittacanthus, because they do not eat the embryo or viscin tissues. This differs from the larvae of T. echelta that interrupted the germination of seeds by feeding on those tissues. Thepytus echelta may be a promising insect for the biological control of P. acinarius in the ecotonal Cerrado-Pantanal, although its abundance and frequency were low throughout the sampling period.

The need for biodiversity conservation is increasing at a rate much faster than the acquisition of knowledge of biodiversity, such as descriptions of new species and mapping species distributions. As global changes are winning the race against the acquisition of knowledge, many researchers resort to the use of surrogate groups to aid in conservation decisions. Reductions in taxonomic and numerical resolution are also desirable, because they could allow more rapid the acquisition of knowledge while requiring less effort, if little important information is lost. In this study, we evaluated the congruence among 22 taxonomic groups sampled in a tropical forest in the Amazon basin. Our aim was to evaluate if any of these groups could be used as surrogates for the others in monitoring programs. We also evaluated if the taxonomic or numerical resolution of possible surrogates could be reduced without greatly reducing the overall congruence. Congruence among plant groups was high, whereas the congruence among most animal groups was very low, except for anurans in which congruence values were only slightly lower than for plants. Liana (Bignoniaceae) was the group with highest congruence, even using genera presence-absence data. The congruence among groups was related to environmental factors, specifically the clay and phosphorous contents of soil. Several groups showed strong spatial clumping, but this was unrelated to the congruence among groups. The high degree of congruence of lianas with the other groups suggests that it may be a reasonable surrogate group, mainly for the other plant groups analyzed, if soil data are not available. Although lianas are difficult to count and identify, the number of studies on the ecology of lianas is increasing. Most of these studies have concluded that lianas are increasing in abundance in tropical forests. In addition to the high congruence, lianas are worth monitoring in their own right because they are sensitive to global warming and the increasing frequency and severity of droughts in tropical regions. Our findings suggest that the use of data on surrogate groups with relatively low taxonomic and numerical resolutions can be a reliable shortcut for biodiversity assessments, especially in megadiverse areas with high rates of habitat conversion, where the lack of biodiversity knowledge is pervasive. We carried out 84 trawls in 41 seagrass meadows composed of the phanerogam Cymodocea nodosa at three islands of the Canarian Archipelago, during June to September 2003, in order to describe the associated ichthyofauna (composition, richness, and abundance), to analyze the role that this habitat can play in fish recruitment, and to determine the potential relationship between the spatial structure of the seagrass meadow and the patterns of richness and abundance of the fish assemblage. A total of 8298 individuals were captured. The five most relevant species, in terms of abundance and frequency, were Spondyliosoma cantharus, Diplodus annularis, Syngnathus typhle, Mullus surmuletus, and Pagellus erythrinus. Gran Canaria had the largest species richness (36 species) and mean number of species per sample (8.69 ± 0.49; mean ± SE). Lanzarote had the largest number of individuals (64.83% of the total registered) and mean total abundance per sample (168.39 ± 30.91). High densities of individuals were registered (95.86 ± 13.5) and 92.91% of fishes were juveniles. Our data showed that the physical configuration of the seagrass meadows did not significantly affect the patterns of richness and abundance of the associated fish assemblage. In conclusion, the C. nodosa meadows exhibited a singular ichthyofauna and they contribute to the maintenance of the diversity of the coastal fish assemblages in the Canarian Archipelago. This habitat constitutes, during spring and summer, a nursery habitat for juvenile fishes of many species, several of them commercially targeted.

Cadmium is a highly volatile element and its abundance in meteorites may help better understand volatility-controlled processes in the solar nebula and on meteorite parent bodies. The large thermal neutron capture cross section of 113Cd suggests that Cd isotopes might be well suited to quantify neutron fluences in extraterrestrial materials. The aims of this study were (1) to evaluate the range and magnitude of Cd concentrations in magmatic iron meteorites, and (2) to assess the potential of Cd isotopes as a neutron dosimeter for iron meteorites. Our new Cd concentration data determined by isotope dilution demonstrate that Cd concentrations in iron meteorites are significantly lower than in some previous studies. In contrast to large systematic variations in the concentration of moderately volatile elements like Ga and Ge, there is neither systematic variation in Cd concentration amongst troilites, nor amongst metal phases of different iron meteorite groups. Instead, Cd is strongly depleted in all iron meteorite groups, implying that the parent bodies accreted well above the condensation temperature of Cd (i.e., ≈650 K) and thus incorporated only minimal amounts of highly volatile elements. No Cd isotope anomalies were found, whereas Pt and W isotope anomalies for the same iron meteorite samples indicate a significant fluence of epithermal and higher energetic neutrons. This observation demonstrates that owing to the high Fe concentrations in iron meteorites, neutron capture mainly occurs at epithermal and higher energies. The combined Cd-Pt-W isotope results from this study thus demonstrate that the relative magnitude of neutron capture-induced isotope anomalies is strongly affected by the chemical composition of the irradiated material. The resulting low fluence of thermal neutrons in iron meteorites and their very low Cd concentrations make Cd isotopes unsuitable as a neutron dosimeter for iron meteorites.

Most empirical studies support a decline in speciation rates through time, although evidence for constant speciation rates also exists. Declining rates have been explained by invoking pre-existing niches, whereas constant rates have been attributed to non-adaptive processes such as sexual selection and mutation. Trends in speciation rate and the processes underlying it remain unclear, representing a critical information gap in understanding patterns of global diversity. Here we show that the temporal trend in the speciation rate can also be explained by frequency-dependent selection. We construct a frequency-dependent and DNA sequence-based model of speciation. We compare our model to empirical diversity patterns observed for cichlid fish and Darwin's finches, two classic systems for which speciation rates and richness data exist. Negative frequency-dependent selection predicts well both the declining speciation rate found in cichlid fish and explains their species richness. For groups like the Darwin's finches, in which speciation rates are constant and diversity is lower, speciation rate is better explained by a model without frequency-dependent selection. Our analysis shows that differences in diversity may be driven by incipient species abundance with frequency-dependent selection. Our results demonstrate that genetic-distance-based speciation and frequency-dependent selection are sufficient to explain the high diversity observed in natural systems and, importantly, predict decay through time in speciation rate in the absence of pre-existing niches.

Leptospiral pulmonary haemorrhage syndrome (LPHS) is a particularly severe form of leptospirosis. LPHS is increasingly recognized in both humans and animals and is characterized by rapidly progressive intra-alveolar haemorrhage leading to high mortality. The pathogenic mechanisms of LPHS are poorly understood which hampers the application of effective treatment regimes. In this study a 2-D guinea pig proteome lung map was created and used to investigate the pathogenic mechanisms of LPHS. Comparison of lung proteomes from infected and non-infected guinea pigs via differential in-gel electrophoresis revealed highly significant differences in abundance of proteins contained in 130 spots. Acute phase proteins were the largest functional group amongst proteins with increased abundance in LPHS lung tissue, and likely reflect a local and/or systemic host response to infection. The observed decrease in abundance of proteins involved in cytoskeletal and cellular organization in LPHS lung tissue further suggests that infection with pathogenic Leptospira induces changes in the abundance of host proteins involved in cellular architecture and adhesion contributing to the dramatically increased alveolar septal wall permeability seen in LPHS. BIOLOGICAL SIGNIFICANCE The recent completion of the complete genome sequence of the guinea pig (Cavia porcellus) provides innovative opportunities to apply proteomic technologies to an important animal model of disease. In this study, the comparative proteomic analysis of lung tissue from experimentally infected guinea pigs with leptospiral pulmonary haemorrhage syndrome (LPHS) revealed a decrease in abundance of proteins involved in cellular architecture and adhesion, suggesting that loss or down-regulation of cytoskeletal and adhesion molecules plays an important role in the pathogenesis of LPHS. A publically available guinea pig lung proteome map was constructed to facilitate future pulmonary proteomics in this species.
