On Sunday, Switzerland votes on its federal parliament. The members of both chambers - 200 National Councillors and 46 Councillors of States - are directly elected, with the 26 Swiss cantons (or provinces) forming the districts. Seats in the National Council are distributed to the cantons based on their population size, while in the Council of States each full canton gets two seats and the six half-cantons get one each. Here are six things to look out for.

In this paper we consider a model with two industrialized countries that face a flow of immigration from the "rest of the world." The countries differ in three characteristics: the labor complementarity between the "native" population and immigrants, the population size, and the magnitude of the cultural friction between the natives and immigrants. We consider a non-cooperative game between two countries' when their strategic instrument is the choice of an immigration quota and the world immigrant wages introduce the spill-over effect between two countries. We first show that the quota game admits unique pure strategies Nash equilibrium. We then compare the equilibrium choices of two countries and show that even though the larger country attracts more immigrants, it chooses lower quota than its smaller counterpart. It also turns out that higher degree of labor complementarity between natives and immigrants and a lower degree of cultural friction between two groups yield higher immigration quota. We also examine the welfare implications of countries choices' and argue that coordinated and harmonized immigration policies may improve the welfare of both countries.

This paper proposes a general equilibrium model of a monocentric city based on Fujita and Krugman (1995). Two rates of transport costs per distance and for the same good are introduced. The model assumes that lower transport costs are available at a few points on a line. These lower costs represent new transport facilities, such as high-speed motorways and railways. Findings is that new transport facilities connecting the city and hinterlands strengthen the lock-in effects, which describes whether a city remains where it is forever after being created. Furthermore, the effect intensifies with better agricultural technologies and a larger population in the economy. The relationship between indirect utility and population size has an inverted U-shape, even if new transport facilities are used. However, the population size that maximizes indirect utility is smaller than that found in Fujita and Krugman (1995).

Probabilistic modeling is the de�ning characteristic of estimation of distribution algorithms (EDAs) which determines their behavior and performance in optimization. Regularization is a well-known statistical technique used for obtaining an improved model by reducing the generalization error of estimation, especially in high-dimensional problems. `1-regularization is a type of this technique with the appealing variable selection property which results in sparse model estimations. In this thesis, we study the use of regularization techniques for model learning in EDAs. Several methods for regularized model estimation in continuous domains based on a Gaussian distribution assumption are presented, and analyzed from di�erent aspects when used for optimization in a high-dimensional setting, where the population size of EDA has a logarithmic scale with respect to the number of variables. The optimization results obtained for a number of continuous problems with an increasing number of variables show that the proposed EDA based on regularized model estimation performs a more robust optimization, and is able to achieve signi�cantly better results for larger dimensions than other Gaussian-based EDAs. We also propose a method for learning a marginally factorized Gaussian Markov random �eld model using regularization techniques and a clustering algorithm. The experimental results show notable optimization performance on continuous additively decomposable problems when using this model estimation method. Our study also covers multi-objective optimization and we propose joint probabilistic modeling of variables and objectives in EDAs based on Bayesian networks, speci�cally models inspired from multi-dimensional Bayesian network classi�ers. It is shown that with this approach to modeling, two new types of relationships are encoded in the estimated models in addition to the variable relationships captured in other EDAs: objectivevariable and objective-objective relationships. An extensive experimental study shows the e�ectiveness of this approach for multi- and many-objective optimization. With the proposed joint variable-objective modeling, in addition to the Pareto set approximation, the algorithm is also able to obtain an estimation of the multi-objective problem structure. Finally, the study of multi-objective optimization based on joint probabilistic modeling is extended to noisy domains, where the noise in objective values is represented by intervals. A new version of the Pareto dominance relation for ordering the solutions in these problems, namely �-degree Pareto dominance, is introduced and its properties are analyzed. We show that the ranking methods based on this dominance relation can result in competitive performance of EDAs with respect to the quality of the approximated Pareto sets. This dominance relation is then used together with a method for joint probabilistic modeling based on `1-regularization for multi-objective feature subset selection in classi�cation, where six di�erent measures of accuracy are considered as objectives with interval values. The individual assessment of the proposed joint probabilistic modeling and solution ranking methods on datasets with small-medium dimensionality, when using two di�erent Bayesian classi�ers, shows that comparable or better Pareto sets of feature subsets are approximated in comparison to standard methods.

Esta tesis doctoral pretende profundizar en el conocimiento de la ecología de Ulmus laevis Pallas, especie autóctona en peligro de extinción en la Península Ibérica, con el fin de proponer medidas adecuadas para su conservación. Se ha estudiado la distribución natural de la especie atendiendo a aspectos edáficos. Los resultados muestran que U. laevis presenta menor capacidad de acidificación de la rizosfera, menor actividad de la reductasa férrica y menor homeostasis que U. minor Mill. cuando crecen en sustratos con una disponibilidad de hierro limitada. Estas diferencias ayudan a comprender la distribución de ambas especies en la Península Ibérica: U. laevis se ve restringido a suelos ácidos o moderadamente ácidos, mientras que U. minor es capaz de habitar tanto suelos ácidos como básicos. Se han analizado las propiedades hidráulicas y anatómicas de U. laevis, constatando que sus características son favorables en ambientes con gran disponibilidad hídrica y que se trata del olmo ibérico más vulnerable a la cavitación por estrés hídrico, por lo que la aridificación del clima y la pérdida de los freáticos supone un riesgo para sus poblaciones. Para evaluar la capacidad de recuperación de la especie se han estudiado la diversidad y estructura genética espacial de las dos mayores poblaciones españolas. Los resultados evidencian que estas poblaciones mantienen niveles de diversidad equiparables o ligeramente superiores a los europeos, pese a haber sufrido un cuello de botella prolongado durante las glaciaciones y a las reducciones poblacionales recientes. En la actualidad la endogamia no representa un riesgo para estas poblaciones. También se ha analizado la producción, dispersión y predación de semillas en Valdelatas (Madrid). Los resultados han mostrado que el viento dispersa las sámaras a corta distancia (<30 m) y que los años no veceros las probabilidades de establecimiento de regenerado son bajas. Además, la producción de sámaras vanas puede tratarse de un carácter adaptativo que aumenta la eficiencia biológica de la especie, ya que favorece la supervivencia de las semillas embrionadas disminuyendo sus tasas de predación pre- y post-dispersión. La modificación del hábitat de esta especie como consecuencia de las actividades humanas afecta de manera negativa al establecimiento del regenerado. La conservación de esta especie a largo plazo requiere la recuperación de los niveles freáticos y de regímenes hidrológicos que permitan avenidas, ya que estas crean las condiciones adecuadas para el establecimiento de regenerado al eliminar la vegetación preexistente y depositar barro. ABSTRACT Ulmus laevis Pallas is an endangered species in the Iberian Peninsula. Therefore, in order to be able to propose adequate management guidelines for its conservation, this PhD Thesis intends to advance the knowledge on the species ecology in the region. Firstly, the species natural distribution was studied in relation to soil nature. Results show that U. minor Mill. had a higher root ferric reductase activity and proton extrusion capability than U. laevis, and maintained a better nutrient homeostasis when grown under iron limiting conditions. These differences in root Fe acquisition efficiencies proved helpful to understand the distribution of these species in the Iberian Peninsula, where U. laevis is restricted to acid or moderately acid soils, whereas U. minor can grow both in acid and basic soils. Secondly, we studied Ulmus laevis’ xylem anatomy and hydraulic traits. These proved favourable for growing under high water availability, but highly susceptible to drought-stress cavitation. Therefore, this species is vulnerable to the Iberian Peninsula’s aridification. Spatial genetic structure and diversity were evaluated in two of the biggest U. laevis populations in Spain in order to evaluate their recovery capabilities. These populations maintain similar or slightly higher diversity levels than European populations, despite having undergone an ancestral genetic bottleneck and having suffered recent population size reductions. No inbreeding problems have been detected in these populations. Seed production, dispersal and predation were assessed in Valdelatas’ elm grove (Madrid). Despite U. laevis samaras being winged nuts, wind dispersed them short distances from the mother tree (<30 m). The seed shadow models show that non-mast years provide very few chances for the stand to regenerate due to their low full seed flux. Empty samaras deceive pre- and post-dispersal predators increasing full seed survival probabilities. Therefore, empty fruit production might be an adaptive trait that increases plant fitness. Finally, human-induced changes in water-table levels and river regulation may affect U. laevis seed dispersal and regeneration establishment negatively. The long-term conservation and expansion of this species in the Iberian Peninsula requires the recovery of water-tables and of natural hydrological regimes, as flooding eliminates vegetation, creating open microhabitats and deposits mud, creating the ideal conditions for seedling establishment.

En Ecuador el maíz es el cultivo más importante en superficie y es base de la alimentación para la población rural que vive en los Andes. A diferencia de lo que sucede en la Costa, en la región Sierra todavía se cultivan numerosas variedades tradicionales que se agrupan en veinticuatro razas. Mantener esta diversidad es, pues, de gran importancia no solo para la seguridad alimentaria, sino también como fuente de genes para tolerancia a factores abióticos que podrían ser incorporados a las variedades modernas. Si bien parte de esta diversidad fue recolectada a mediados del siglo pasado y está siendo conservada en distintos bancos de germoplasma, es deseable que su conservación in situ también esté asegurada, entre otras razones, porque de esta manera el cultivo puede seguir evolucionando. Para poder implementar un plan de conservación en finca que contribuya a preservar este patrimonio, resulta imprescindible identificar áreas idóneas donde concentrar los recursos y conocer las características y tipologías de los agricultores que manejan la diversidad actual. Generar esta información es el objetivo principal de esta investigación y para lograrlo se han llevado a cabo cuatro estudios: (1) Análisis de la diversidad a nivel de razas e identificación de áreas de alta riqueza de razas, alta diversidad morfológica y/o alta diversidad ecogeográfica en la Sierra de Ecuador, (2) Identificación del perfil y las características de los agricultores que conservan y manejan las variedades tradicionales de maíz en la Sierra de Ecuador, (3) Análisis del conocimiento local, manejo y usos de variedades tradicionales de maíz en la Sierra de Ecuador, y (4) Identificación de áreas de alta diversidad y bajo riesgo de pérdida para la conservación en finca de maíz en la Sierra de Ecuador. Para el primer estudio se visitaron 303 fincas distribuidas a lo largo de la Sierra y se recolectaron 636 muestras que fueron caracterizadas morfológicamente mediante 14 variables: 8 relacionadas con la mazorca (forma, longitud y diámetro de la mazorca, color y diámetro de olote y número y disposición de hileras) y 7 referidas el grano (número total de granos, color, forma, longitud, anchura y grosor de grano y tipo de endospermo). Adicionalmente, las fincas donde se tomaron las muestras fueron caracterizadas ecogeográficamente mediante 5 variables climáticas (temperatura media estacional, rango de temperatura media anual, temperatura mínima de diciembre, precipitación estacional y precipitación de octubre), 2 geofísicas (altitud y pendiente) y 5 edáficas (textura principal del suelo, profundidad a roca, pH, contenido en materia orgánica y fertilidad). A partir de esta información y mediante técnicas de sistemas de información geográfica (SIG), se generaron mapas de distribución por raza en formato vectorial y un mapa de riqueza de razas, un mapa de diversidad morfológica y un mapa de diversidad ecogeográfica en formato ráster con celdas de 10 km x 10 km. Los resultados permitieron constatar que, en los últimos 60 años, no se ha perdido ninguna raza. Sin embargo, Canguil, Chaucho y Clavito han dejado de cultivarse en algunas provincias con la consiguiente erosión genética del cultivo. La caracterización morfológica detectó diferencias en el grado de variabilidad intra-raza, siendo Patillo Ecuatoriano, Racimo de Uva y Uchima las razas más heterogéneas tanto para los caracteres cualitativos como cuantitativos. A nivel climático y geofísico, también se detectaron diferencias en el grado de variación intra-raza; Cuzco Ecuatoriano, Kcello Ecuatoriano y Montaña Ecuatoriana fueron las razas que en promedio presentaron mayores rangos y coeficientes de variación para estas variables ecogeográficas. En cuanto a las condiciones edáficas todas las razas, excepto Cónico Dentado, presentaron una gran heterogeneidad, pudiendo crecer tanto en suelos ricos como pobres, con valores de pH entre ácido y moderadamente alcalino. La comparación entre razas reveló diferencias significativas en los rangos ambientales de algunas razas como Cónico Dentado, que tiende a cultivarse a menor altitud y, por tanto, en ambientes menos fríos y de mayor precipitación que Blanco Blandito, Patillo Ecuatoriano, Sabanero Ecuatoriano, Uchima y Zhima. Para la mayoría de las razas se encontraron materiales potencialmente adaptados a condiciones de estrés (precipitación estacional inferior a 500 mm y suelos con pH entre 4.5 y 5.5). Finalmente, los mapas de riqueza, de diversidad morfológica y de diversidad ecogeográfica mostraron 36 celdas de alta diversidad repartidas en las 10 provincias de la Sierra: 11 celdas en las provincias del norte, 11 en las provincias del centro y 14 en las provincias del sur. Para la caracterización e identificación de las tipologías de los agricultores que cultivan maíz en la Sierra de Ecuador y el análisis de los posibles factores de riesgo de pérdida de diversidad, se realizaron entrevistas individuales y semiestructuradas a los agricultores dueños de las fincas donde se recolectaron las muestras para el estudio de diversidad (254 en total). Las preguntas que se formularon (11 abiertas y 5 cerradas) estuvieron organizadas en seis bloques: datos del agricultor, características de la finca, diversidad y conocimiento del cultivo, manejo del cultivo, usos y flujo de semillas. Los resultados indicaron que la diversidad de maíz que hay en la Sierra de Ecuador es manejada mayoritariamente por agricultores mestizos, de entre 30 y 55 años, que cultivan una o dos variedades tradicionales para autoconsumo, en parcelas de menos de 0.5 ha y en asocio con fréjol. El análisis de segmentación mediante el algoritmo Chi-square automatic interaction detection (CHAID) permitió identificar un pequeño grupo de agricultores indígenas con parcelas medianas (entre 0.5 ha y 1.5 ha) que conservan un mayor número de variedades tradicionales por finca que el agricultor promedio. Los análisis estadísticos no detectaron diferencias significativas entre etnias (mestizo vs. indígena), géneros (hombre vs. mujer) y grupos de edad (jóvenes menores de 30 años, adultos entre 30 y 55 años y adultos mayores de 55 años) en lo que respecta al conocimiento del cultivo (criterios de reconocimiento y razones de preferencia) y manejo (tipo de cultivo), pero sí detectaron diferencias entre regiones, principalmente en el modo de cultivar el maíz; mientras que en el norte y sur tienden a sembrarlo en asocio y con un mayor número de especies, en el centro acostumbran a cultivarlo preferentemente solo. En cuanto a los usos, se recopilaron hasta 39 modos diferentes de consumir maíz, siendo Kcello Ecuatoriano y Zhima las razas para las que se registró un mayor número de usos. La comparación del número medio de usos por variedad entre etnias evidenció que los agricultores mestizos utilizan sus variedades tradicionales de forma más variada que los indígenas. Entre los factores de riesgo que se analizaron, el bajo porcentaje de jóvenes agricultores que se ocupan de las fincas podría suponer una amenaza a medio plazo por falta de relevo generacional. Adicionalmente, las numerosas sinonimias y homonimias que se detectaron y el bajo intercambio de semillas también podrían ser causa de pérdida de diversidad, bien por reemplazo o por envejecimiento de la semilla. Finalmente, se concluyó que las razas Chaucho, Complejo Chillo-Huandango, Complejo Mishca-Huandango, Cónico Dentado, Montaña Ecuatoriana y Sabanero Ecuatoriano son particularmente vulnerables, no solo por su baja presencia, sino también por el color de grano que tienen (los mismos que la mayoría de las razas más comunes) y carecer de nombres y usos específicos. Finalmente, para la priorización de áreas de conservación en finca para maíz en la Sierra de Ecuador, se utilizaron 13 criterios de diferente naturaleza: 2 ecogeográficos (precipitación, diversidad ecogeográfica), 6 biológicos (grado de presencia del cultivo, riqueza de razas, diversidad morfológica, presencia de mezclas, presencia de razas locales y riesgo de erosión genética), 3 culturales (abundancia de variedades por finca, diversidad de usos y frecuencia de intercambio) y 2 demográficos (tamaño de la población y distancia a núcleos urbanos). Mediante técnicas SIG y de evaluación multicriterio, los valores originales de las capas-criterio fueron transformados a una escala de 0 a 100. Posteriormente, las capas-criterio normalizadas fueron sumadas utilizando tres métodos de ponderación: (1) mismo peso, (2) diferente peso según la puntuación otorgada por 72 expertos, y (3) diferente peso según el método de comparación entre pares de criterios. Los resultados permitieron identificar ocho celdas de 10 km x 10 km con alta puntuación (> 65): tres celdas en el norte (una en cada una de las provincias), una celda en el centro (en la provincia de Cotopaxi), y cuatro celdas en la región sur (dos en Azuay y otras dos en Loja). ABSTRACT In Ecuador, the maize is the most important cultivation in surface and it is a base of the feeding for the rural population who lives in the Andes. In contrast to what it happens on the Coast, in the Sierra region still there are cultivated numerous traditional varieties that are grouped into twenty-four races. Maintaining this diversity is, therefore, of great importance not only for food security, but also as a source of genes for tolerance to abiotic factors could be incorporated into modern varieties. Although part of this diversity was collected in the middle of the last century and is still preserved in various germplasm banks, it is desirable for the in situ conservation also is assured, among other reasons, because in this way the crop can continue to evolve. To be able to implement a conservation plan on farm that contribute to preserving this heritage, it is essential to identify suitable areas where to concentrate resources and know the characteristics and typology of farmer who managed the current diversity. To generate this information is the main target of this investigation and to achieve this, four studies have been carried out: (1) Analysis of the diversity at races and identification of areas of high richness of races, high morphological diversity and / or ecogeographical high diversity in the Sierra of Ecuador, (2) Identification of the profile and characteristics of farmers who conserve and manage traditional varieties of maize in the Sierra of Ecuador, (3) Analysis of local knowledge, management and use of traditional varieties of maize in the Sierra of Ecuador, and (4) Identification of areas of high diversity and low risk of loss for the conservation of maize in the Sierra of Ecuador. For the first study were visited 303 farms distributed along the Sierra and collected 636 samples that were characterized morphologically by 14 variables: 8 related to the ear (shape, length and diameter of the cob, colour, and diameter of cob and number and arrangement of rows) and 7 referred to the grain (total number of grain, colour, shape, length, width, and thickness and type of grain endosperm). In addition, the farms where the samples were taken were characterized ecogeographically through 5 climatic variables (seasonal average temperature, range of average annual temperature, minimum temperature for December, seasonal precipitation and precipitation of October), 2 geophysical (altitude and slope) and edaphic 5 (main texture of the soil, deep rock, pH, content of organic matter and fertility). From this information and techniques of geographic information systems (GIS), maps were generated for distribution by race in vector format and a map of richness of races, a map of morphological diversity and a map of ecogeographical diversity in raster format with cells of 10 km x 10 km. The results allowed observing that, over the past 60 years, it has not lost any race. Nevertheless, Canguil, Chaucho and Clavito have stopped being cultivated in some provinces with the consequent genetic erosion of the cultivation. The morphological characterization detected differences in the degree of variability intra-race, being Patillo Ecuatoriano, Racimo de Uva and Uchima races more heterogeneous both for the qualitative and quantitative characters. At climate and geophysical level, also detected differences in the degree of variation intra-race; Cuzco Ecuatoriano, Kcello Ecuatoriano and Montaña Ecuatoriana were races that, on average, showed higher ranges and coefficients of variation for these geographical characters. In terms of the edaphic conditions, all races, except Cónico Dentado, showed a great heterogeneity, and can grow both in rich and poor soils, with pH values between acid and moderately alkaline. The comparison between races revealed significant differences in the environmental ranges in some races as Cónico Dentado, which tends to be grown at lower elevations and, therefore, in environments less cold and greater precipitation than Blanco Blandito, Patillo Ecuatoriano, Sabanero Ecuatoriano, Uchima and Zhima. For most of the races were found materials potentially adapted to stress conditions (seasonal precipitation less than 500 mm and soil with a pH between 4.5 and 5.5). Finally, the maps of richness, morphologic diversity and ecogeographical diversity showed 36 cells high diversity distributed in 10 provinces of the Sierra: 11 cells in the northern provinces, 11 in the central provinces and 14 in the southern provinces. For the characterization and identification of the typology of the farmers who cultivate corn in the Sierra of Ecuador and the analysis of the possible factors of risk of loss of diversity, there were realized interviews individual and semistructured to the farmers’ owners of the farms where the samples were gathered for the study of diversity (254 in whole). The questions that were formulated (11 opened ones and 5 closed ones) were organized in six blocks: data of the farmer, characteristics of the farm, diversity and knowledge of the crop, crop management, uses and seed flow. The results indicated that the maize diversity that exist in the Sierra of Ecuador is managed mainly by mestizo farmers, aged between 30 and 55, who cultivate one or two traditional varieties for self-consumption, on plots of less than 0.5 has and in associated with beans. The segmentation analysis algorithm using the Chi-square automatic interaction detection (CHAID technique), allowed to identify a small group of indigenous farmers with medium-sized plots (between 0.5 there is and 1.5 it is) that a major number of traditional varieties preserves for farm that the average farmer. The statistical analysis did not detect significant differences between ethnic groups (mestizos vs. indigenous), genres (man vs. women) and age groups (young people under 30 years of age, adults between 30 and 55 years and adults over 55 years old) in regards to the knowledge of the cultivation (recognition criteria and reasons of preference) and management (type of crop), but if detected differences between regions, mainly on the mode of cultivating the maize; while in the north and south they tend to sow in associate and with a greater number of species, in the center accustomed to cultivate it preferably only. In regards to the uses, they were compiled up to 39 different ways of consuming maize, being Kcello Ecuatoriano and Zhima the races for which a major number of uses registered. The comparison of the average number of uses per variety between ethnic groups showed that the mestizo farmers used their traditional varieties of form more varied than the indigenous people. Between the factors of risk that were analyzed, the low percentage of young farmers who deal with the farms might suppose a medium-term threat for lack of generational relief. In addition, the numerous synonyms and homonyms that were detected and the low seed exchange could also be a cause of loss of diversity, either by replacement or by aging of the seed. Finally, it was concluded that the races Chaucho, Complex Chillo-Huandango, Complex Mishca-Huandango, Cónico Dentado, Montaña Ecuatoriana and Sabanero Ecuatoriano are particularly vulnerable, not only because of their low presence, but also by the grain color they have (the same as the majority of races more common) and lack of names and specific uses. Finally, for the prioritization of maize conservation areas on farm in the Sierra of Ecuador, used 13 criteria of different nature: 2 ecogeographic (precipitation, diversity ecogeographical), 6 biological (degree of presence of the crop, races richness, morphological diversity, the presence of mixtures, presence of local races and risk of genetic erosion), 3 cultural (abundance of varieties per farm, diversity of uses and frequency of exchange) and 2 demographic (population size and distance to urban centers). Using GIS techniques and multicriteria evaluation, the original values of the layers-criterion were transformed to a scale of 0 to 100. Later, the normalized layers - criteria were added using three weighting methods: (1) the same weight, (2) different weight according to the score given by 72 experts, and (3) different weight according to the method of comparison between pairs of criteria. The results allowed to identify eight 10 km cells x 10 km with high punctuation (> 65): three cells in the north (one in each of the provinces), a cell in the center (in the Cotopaxi province), and four cells in the south region (two in Azuay and other two in Loja).

Esta Tesis tiene como objetivo demostrar que los programas de preservación del patrimonio, durante su fase de implementación, deben someterse a un análisis multidisciplinar que haga un balance de su ejecución. Dicho análisis permitirá identificar resultados significativos, capaces de fundamentar la rectificación de las bases conceptuales de la política pública en cuestión. Este reajuste podrá darse, por lo tanto, durante su vigencia y, de forma más relevante, posteriormente, sus motivos y resultados permitirán elaborar nuevas estrategias que serán aplicadas en futuros programas de intervención. Por otro lado se indagó, además, si las ciudades participantes en un programa nacional de preservación del patrimonio, regidas por normas y metas comunes, podrían alcanzar resultados diferentes. Para atender a estos objetivos, la investigación se encuentra enfocada a la realidad brasileña, siendo seleccionado como objeto de estudio el Programa Monumenta. Este Programa formó parte de la política pública cultural del Ministerio de Cultura con una importante implicación del Banco Interamericano de Desarrollo. Implementado a partir de 1999, procuró promover un proceso de recuperación urbana sostenible y de preservación del patrimonio de 26 Sitios Históricos Urbanos o Conjuntos de Monumentos Urbanos protegidos por el Instituto del Patrimonio Histórico y Artístico Nacional (Iphan). El Programa contó con el apoyo de la Unesco y del Iphan, así como con la participación de las Administraciones Municipales y/o Estatales, sectores privados y la sociedad civil. Las hipótesis planteadas en esta Tesis Doctoral fueron: (a) el análisis de los resultados en la fase de implementación de un programa de preservación del patrimonio es imprescindible, porque permite extraer conclusiones preliminares y orientar sus reformas; (b) los objetivos a corto plazo establecidos por el Programa Monumenta fueron alcanzados de modo diferenciado en las distintas ciudades beneficiadas; (c) a pesar de las diferencias, el Programa Monumenta presentó resultados preliminares positivos y significativos en la preservación del patrimonio histórico urbano brasileño. Los procedimientos metodológicos se centraron en un análisis cuantitativo, cualitativo y comparativo de los resultados alcanzados por tres ciudades beneficiadas por el Programa Monumenta, seleccionadas según su tamaño poblacional: Pelotas, Porto Alegre (Estado de Rio Grande do Sul) y São Francisco do Sul (Estado de Santa Catarina). Estos procedimientos fueron aplicados en los siguientes indicadores: utilización de los equipamientos culturales, características de la población y de los domicilios, variación de las actividades económicas, financiación destinada al sector privado para la recuperación de inmuebles y el fomento de la seguridad urbana. La Tesis ha englobado discusiones y conceptos abordados en las disciplinas de la Sociología Urbana, Geografía Urbana, Historia, Economía y Estadística de modo que se atribuye al objeto de investigación una visión interdisciplinar que ayudará a la comprensión de la teoría y la práctica preservacionistas. El análisis de la varianza, la regresión lineal y el análisis factorial fueron las herramientas estadísticas aplicadas sobre los datos con el objetivo de constatar la significación de los resultados y la relación de correspondencia entre algunas variables. Esta Tesis contribuye a la elaboración de una metodología analítica que puede ser aplicada en el cálculo de la superficie ocupada por las actividades económicas, con base en el método estadístico del Diagrama de Caja y Bigotes, de John Wilder Tukey. Las conclusiones corroboran las hipótesis planteadas y pretenden contribuir al diseño de las nuevas políticas públicas de preservación de sitios históricos de carácter urbano, enfatizando, con ello, la necesidad de evaluaciones más profundas de los resultados durante su fase de implementación. ---------------------------------------------------------------------------------- RESUMO--------------------------------------------------------------------------- A presente Tese apresenta como objetivo principal demonstrar que os programas de preservação do patrimônio histórico, durante a sua fase de implementação, necessitam de uma análise multidisciplinar sobre a sua execução. Essa análise permite identificar resultados significativos, capazes de fundamentar a retificação das bases conceituais da política pública em questão. A correção poderá, portanto, ser realizada tanto durante a sua vigência como posteriormente, ao permitir a elaboração de novas estratégias a serem aplicadas nos futuros programas de intervenção. Por outro lado, indagou-se se cidades participantes de um mesmo programa nacional de preservação do patrimônio histórico, regidas por normas e metas comuns, poderiam alcançar resultados não similares. Para atender tais objetivos, a investigação enfoca a realidade brasileira, tendo sido selecionado o Programa Monumenta como objeto de estudo. Esse Programa fez parte de uma política pública cultural do Ministério da Cultura, que atuou em parceria com o Banco Interamericano de Desenvolvimento. Thesis is to demonstrate that a multidisciplinary analysis is needed during the implementation phase of preservation of heritage programmes. Such analysis allows the identification of significant results, which in turn can serve as the foundation for the conceptual bases of the public policy at hand. Thus, any corrections can be made both during the programme and afterward, by introducing new strategies to be applied in future intervention programmes. On the other hand, this project also asks whether cities participating in the same national preservation of heritage programme with common rules and goals can achieve distinct results. In order to meet these objectives, the project chose Brazil as its focus and Monumenta Programme for its object of study. This Programme is part of the Ministry of Culture’s public cultural policy, and was developed with cooperation by the Inter-American Development Bank. Implemented at the national level in 1999, the Programme aimed at promoting a process of sustainable urban renewal and the preservation of 26 urban historic sites or urban monumental ensembles, protected by the National Historic and Artistic Heritage Institute (IPHAN). UNESCO and IPHAN supported the Programme, and participants included municipal and state offices, private businesses, and local residents. The hypotheses established in this Doctoral Thesis were: (a) the analysis of the results in the implementation phase of a cultural public policy is imperative, because it enables preliminary conclusions to be drawn and orientate reforms; (b) the short-term objectives set out in the Monumenta Programme were achieved differently in the benefitted cities; (c) despite the differences, the Monumenta Programme displayed significant positive preliminary results in the conservation of the urban historic heritage in Brazil. Methodology procedures centered around quantitative, qualitative, and comparative analyses of the results achieved in three benefiting cities, selected according to population size: Pelotas, Porto Alegre (Rio Grande do Sul State) and São Francisco do Sul (Santa Catarina State). These procedures were applied to the following indicators: the use of the cultural facilities, characteristics of the population and the residential housing, variation of the economic activities, financing destined for the recovery of real estate, and promoting urban safety. The Thesis includes discussions and concepts addressed in urban sociology, urban geography, history, economics, and statistics, in order to examine the object of study with an interdisciplinary eye and an understanding between preservation theory and practice. Variance analysis, linear regression, and factorial analysis were the statistical techniques applied to the data, with the goal of defining the significance of the results and the correspondence ratio between some of the variables. This Thesis attempted to elaborate an applied methodology for calculating the space occupied by economic activities, using John Wilder Tukey's statistical method of Box-and-Whisker Plot. The conclusions corroborated the hypotheses established and are meant to contribute to the design of new public policies of historical site preservation in urban settings, emphasizing the need for deeper evaluations of the results during the implementation phase.

Maize (Zea mays ssp. mays) is genetically diverse, yet it is also morphologically distinct from its wild relatives. These two observations are somewhat contradictory: the first observation is consistent with a large historical population size for maize, but the latter observation is consistent with strong, diversity-limiting selection during maize domestication. In this study, we sampled sequence diversity, coupled with simulations of the coalescent process, to study the dynamics of a population bottleneck during the domestication of maize. To do this, we determined the DNA sequence of a 1,400-bp region of the Adh1 locus from 19 individuals representing maize, its presumed progenitor (Z. mays ssp. parviglumis), and a more distant relative (Zea luxurians). The sequence data were used to guide coalescent simulations of population bottlenecks associated with domestication. Our study confirms high genetic diversity in maize—maize contains 75% of the variation found in its progenitor and is more diverse than its wild relative, Z. luxurians—but it also suggests that sequence diversity in maize can be explained by a bottleneck of short duration and very small size. For example, the breadth of genetic diversity in maize is consistent with a founding population of only 20 individuals when the domestication event is 10 generations in length.

In populations that are small and asexual, mutations with slight negative effects on fitness will drift to fixation more often than in large or sexual populations in which they will be eliminated by selection. If such mutations occur in substantial numbers, the combined effects of long-term asexuality and small population size may result in substantial accumulation of mildly deleterious substitutions. Prokaryotic endosymbionts of animals that are transmitted maternally for very long periods are effectively asexual and experience smaller effective population size than their free-living relatives. The contrast between such endosymbionts and related free-living bacteria allows us to test whether a population structure imposing frequent bottlenecks and asexuality does lead to an accumulation of slightly deleterious substitutions. Here we show that several independently derived insect endosymbionts, each with a long history of maternal transmission, have accumulated destabilizing base substitutions in the highly conserved 16S rRNA. Stabilities of Domain I of this subunit are 15–25% lower in endosymbionts than in closely related free-living bacteria. By mapping destabilizing substitutions onto a reconstructed phylogeny, we show that decreased ribosomal stability has evolved separately in each endosymbiont lineage. Our phylogenetic approach allows us to demonstrate statistical significance for this pattern: becoming endosymbiotic predictably results in decreased stability of rRNA secondary structure.

Classical quorum-sensing (autoinduction) regulation, as exemplified by the lux system of Vibrio fischeri, requires N-acyl homoserine lactone (AHL) signals to stimulate cognate transcriptional activators for the cell density-dependent expression of specific target gene systems. For Pantoea stewartii subsp. stewartii, a bacterial pathogen of sweet corn and maize, the extracellular polysaccharide (EPS) stewartan is a major virulence factor, and its production is controlled by quorum sensing in a population density-dependent manner. Two genes, esaI and esaR, encode essential regulatory proteins for quorum sensing. EsaI is the AHL signal synthase, and EsaR is the cognate gene regulator. esaI, ΔesaR, and ΔesaI-esaR mutations were constructed to establish the regulatory role of EsaR. We report here that strains containing an esaR mutation produce high levels of EPS independently of cell density and in the absence of the AHL signal. Our data indicate that quorum-sensing regulation in P. s. subsp. stewartii, in contrast to most other described systems, uses EsaR to repress EPS synthesis at low cell density, and that derepression requires micromolar amounts of AHL. In addition, derepressed esaR strains, which synthesize EPS constitutively at low cell densities, were significantly less virulent than the wild-type parent. This finding suggests that quorum sensing in P. s. subsp. stewartii may be a mechanism to delay the expression of EPS during the early stages of infection so that it does not interfere with other mechanisms of pathogenesis.

Using systematic evolution of ligands by exponential enrichment (SELEX), an RNA molecule was isolated that displays a 1,000-fold higher affinity for guanosine residues that carry an N-7 methyl group than for nonmethylated guanosine residues. The methylated guanosine residue closely resembles the 5′ terminal cap structure present on all eukaryotic mRNA molecules. The cap-binding RNA specifically inhibited the translation of capped but not uncapped mRNA molecules in cell-free lysates prepared from either human HeLa cells or from Saccharomyces cerevisiae. These findings indicate that the cap-binding RNA will also be useful in studies of other cap-dependent processes such as pre-mRNA splicing and nucleocytoplasmic mRNA transport.

Phylogenetic trees for groups of closely related species often have different topologies, depending on the genes used. One explanation for the discordant topologies is the persistence of polymorphisms through the speciation phase, followed by differential fixation of alleles in the resulting species. The existence of transspecies polymorphisms has been documented for alleles maintained by balancing selection but not for neutral alleles. In the present study, transspecific persistence of neutral polymorphisms was tested in the endemic haplochromine species flock of Lake Victoria cichlid fish. Putative noncoding region polymorphisms were identified at four randomly selected nuclear loci and tested on a collection of 12 Lake Victoria species and their putative riverine ancestors. At all loci, the same polymorphism was found to be present in nearly all the tested species, both lacustrine and riverine. Different polymorphisms at these loci were found in cichlids of other East African lakes (Malawi and Tanganyika). The Lake Victoria polymorphisms must have therefore arisen after the flocks now inhabiting the three great lakes diverged from one another, but before the riverine ancestors of the Lake Victoria flock colonized the Lake. Calculations based on the mtDNA clock suggest that the polymorphisms have persisted for about 1.4 million years. To maintain neutral polymorphisms for such a long time, the population size must have remained large throughout the entire period.

Using a PCR approach we have isolated racF1, a novel member of the Rho family in Dictyostelium. The racF1 gene encodes a protein of 193 amino acids and is constitutively expressed throughout the Dictyostelium life cycle. Highest identity (94%) was found to a RacF2 isoform, to Dictyostelium Rac1A, Rac1B, and Rac1C (70%), and to Rac proteins of animal species (64–69%). To investigate the role of RacF1 in cytoskeleton-dependent processes, we have fused it at its amino-terminus with green fluorescent protein (GFP) and studied the dynamics of subcellular redistribution using a confocal laser scanning microscope and a double-view microscope system. GFP–RacF1 was homogeneously distributed in the cytosol and accumulated at the plasma membrane, especially at regions of transient intercellular contacts. GFP–RacF1 also localized transiently to macropinosomes and phagocytic cups and was gradually released within <1 min after formation of the endocytic vesicle or the phagosome, respectively. On stimulation with cAMP, no enrichment of GFP–RacF1 was observed in leading fronts, from which it was found to be initially excluded. Cell lines were obtained using homologous recombination that expressed a truncated racF1 gene lacking sequences encoding the carboxyl-terminal region responsible for membrane targeting. These cells displayed normal phagocytosis, endocytosis, and exocytosis rates. Our results suggest that RacF1 associates with dynamic structures that are formed during pinocytosis and phagocytosis. Although RacF1 appears not to be essential, it might act in concert and/or share functions with other members of the Rho family in the regulation of a subset of cytoskeletal rearrangements that are required for these processes.

Association of mRNA with the cytoskeleton represents a fundamental aspect of RNA physiology likely involved in mRNA transport, anchoring, translation, and turnover. We report the initial characterization of a protein complex that binds RNA in a sequence-independent but size-dependent manner in vitro. The complex includes a ∼160-kDa protein that is bound directly to mRNA and that appears to be either identical or highly related to a ∼1600-kDa protein that binds directly to mRNA in vivo. In addition, the microtubule-associated protein, MAP 1A, a cytoskeletal associated protein is a component of this complex. We suggest that the general attachment of mRNA to the cytoskeleton may be mediated, in part, through the formation of this ribonucleoprotein complex.
