[EN] To determine whether conditions for O2 utilization and O2 off-loading from the hemoglobin are different in exercising arms and legs, six cross-country skiers participated in this study. Femoral and subclavian vein blood flow and gases were determined during skiing on a treadmill at approximately 76% maximal O2 uptake (V(O2)max) and at V(O2)max with different techniques: diagonal stride (combined arm and leg exercise), double poling (predominantly arm exercise), and leg skiing (predominantly leg exercise). The percentage of O2 extraction was always higher for the legs than for the arms. At maximal exercise (diagonal stride), the corresponding mean values were 93 and 85% (n = 3; P < 0.05). During exercise, mean arm O2 extraction correlated with the P(O2) value that causes hemoglobin to be 50% saturated (P50: r = 0.93, P < 0.05), but for a given value of P50, O2 extraction was always higher in the legs than in the arms. Mean capillary muscle O2 conductance of the arm during double poling was 14.5 (SD 2.6) ml.min(-1).mmHg(-1), and mean capillary P(O2) was 47.7 (SD 2.6) mmHg. Corresponding values for the legs during maximal exercise were 48.3 (SD 13.0) ml.min(-1).mmHg(-1) and 33.8 (SD 2.6) mmHg, respectively. Because conditions for O2 off-loading from the hemoglobin are similar in leg and arm muscles, the observed differences in maximal arm and leg O2 extraction should be attributed to other factors, such as a higher heterogeneity in blood flow distribution, shorter mean transit time, smaller diffusing area, and larger diffusing distance, in arms than in legs.

Age-related physiological changes in the gastrointestinal tract, as well as modification in lifestyle, nutritional behaviour, and functionality of the host immune system, inevitably affect the gut microbiota. The study presented here is focused on the application and comparison of two different microarray approaches for the characterization of the human gut microbiota, the HITChip and the HTF-Microb.Array, with particular attention to the effects of the aging process on the composition of this ecosystem. By using the Human Intestinal Tract Chip (HITChip), recently developed at the Wageningen University, The Netherland, we explored the age-related changes of gut microbiota during the whole adult lifespan, from young adults, through elderly to centenarians. We observed that the microbial composition and diversity of the gut ecosystem of young adults and seventy-years old people is highly similar but differs significantly from that of the centenarians. After 100 years of symbiotic association with the human host, the microbiota is characterized by a rearrangement in the Firmicutes population and an enrichment of facultative anaerobes. The presence of such a compromised microbiota in the centenarians is associated with an increased inflammation status, also known as inflamm-aging, as determined by a range of peripheral blood inflammatory markers. In parallel, we overtook the development of our own phylogenetic microarray with a lower number of targets, aiming the description of the human gut microbiota structure at high taxonomic level. The resulting chip was called High Taxonomic level Fingerprinting Microbiota Array (HTF-Microb.Array), and was based on the Ligase Detection Reaction (LDR) technology, which allowed us to develop a fast and sensitive tool for the fingerprint of the human gut microbiota in terms of presence/absence of the principal groups. The validation on artificial DNA mixes, as well as the pilot study involving eight healthy young adults, demonstrated that the HTF-Microb.Array can be used to successfully characterize the human gut microbiota, allowing us to obtain results which are in approximate accordance with the most recent characterizations. Conversely, the evaluation of the relative abundance of the target groups on the bases of the relative fluorescence intensity probes response still has some hindrances, as demonstrated by comparing the HTF.Microb.Array and HITChip high taxonomic level fingerprints of the same centenarians.

Weaning is a crucial period in the management of piglets. In modern piggeries economic interest make weaning age decrease more and more and the detrimental consequences of weaning have as much importance as earlier the weaning occurs. The risk of development of post-weaning diarrhea (PWD) in piglets is high and PWD is the cause of serious economic losses in pig herds. In the past the supplementation of the feed given after weaning with growth promoters antibiotics, in order to keep PWD under control, used to be a common practice, but their usage has been banned in EU since 2006. This measure led to the investigation of alternative suitable feed supplements that would be reasonably efficient in protecting and sustaining animal health and performance. Aim of this thesis was to evaluate the effect of some different alternatives to growth-promoters antibiotics on weaning piglets and to assess if some of them could be considered as valuables options to replace auxinic in animal feeding. The study is composed by four experimental trials. The first one aims to identify mechanisms involved in the auxinic effects of antibiotics in the diets; the following three evaluate the addition butyric acid, tryptophan, and nitrate as alternative to in-feed antimicrobials. Although some results are controversial, it appears from the data presented that the alternatives to in-feed antibiotics considered may exert positive effects on some zootechnical and health parameters on piglet in the post-weaning period. Anyway, the mechanism of action and the interaction with microbiota of such additives should be investigated inside out because many effects remains poorly understood.

Due to its high Curie temperature of 420K and band structure calculations predicting 100% spin polarisation, Sr2FeMoO6 is a potential candidate for spintronic devices. However, the preparation of good quality thin films has proven to be a non-trivial task. Epitaxial Sr2FeMoO6 thin films were prepared by pulsed laser deposition on different substrates. Differing from previous reports a post-deposition annealing step at low oxygen partial pressure (10-5 mbar) was introduced and enabled the fabrication of reproducible, high quality samples. According to the structural properties of the substrates the crystal structure and morphology of the thin films are modified. The close interrelation between the structural, magnetic and electronic properties of Sr2FeMoO6 was studied. A detailed evaluation of the results allowed to extract valuable information on the microscopic nature of magnetism and charge transport. Smooth films with a mean roughness of about 2 nm have been achieved, which is a pre-requisite for a possible inclusion of this material in future devices. In order to establish device-oriented sub-micron patterning as a standard technique, electron beam lithography and focussed ion beam etching facilities have been put into operation. A detailed characterisation of these systems has been performed. To determine the technological prospects of new spintronics materials, the verification of a high spin polarisation is of vital interest. A popular technique for this task is point contact Andreev reflection (PCAR). Commonly, the charge transport in a transparent metal-superconductor contact of nanometer dimensions is attributed solely to coherent transport. If this condition is not fulfilled, inelastic processes in the constriction have to be considered. PCAR has been applied to Sr2FeMoO6 and the Heusler compound Co2Cr0.6Fe0.4Al. Systematic deviations between measured spectra and the standard models of PCAR have been observed. Therefore existing approaches have been generalised, in order to include the influence of heating. With the extended model the measured data was successfully reproduced but the analysis has revealed grave implications for the determination of spin polarisation, which was found to break down completely in certain cases.

Das Time-of-Flight Aerosol Mass Spectrometer (ToF-AMS) der Firma Aerodyne ist eine Weiterentwicklung des Aerodyne Aerosolmassenspektrometers (Q-AMS). Dieses ist gut charakterisiert und kommt weltweit zum Einsatz. Beide Instrumente nutzen eine aerodynamische Linse, aerodynamische Partikelgrößenbestimmung, thermische Verdampfung und Elektronenstoß-Ionisation. Im Gegensatz zum Q-AMS, wo ein Quadrupolmassenspektrometer zur Analyse der Ionen verwendet wird, kommt beim ToF-AMS ein Flugzeit-Massenspektrometer zum Einsatz. In der vorliegenden Arbeit wird anhand von Laborexperimenten und Feldmesskampagnen gezeigt, dass das ToF-AMS zur quantitativen Messung der chemischen Zusammensetzung von Aerosolpartikeln mit hoher Zeit- und Größenauflösung geeignet ist. Zusätzlich wird ein vollständiges Schema zur ToF-AMS Datenanalyse vorgestellt, dass entwickelt wurde, um quantitative und sinnvolle Ergebnisse aus den aufgenommenen Rohdaten, sowohl von Messkampagnen als auch von Laborexperimenten, zu erhalten. Dieses Schema basiert auf den Charakterisierungsexperimenten, die im Rahmen dieser Arbeit durchgeführt wurden. Es beinhaltet Korrekturen, die angebracht werden müssen, und Kalibrationen, die durchgeführt werden müssen, um zuverlässige Ergebnisse aus den Rohdaten zu extrahieren. Beträchtliche Arbeit wurde außerdem in die Entwicklung eines zuverlässigen und benutzerfreundlichen Datenanalyseprogramms investiert. Dieses Programm kann zur automatischen und systematischen ToF-AMS Datenanalyse und –korrektur genutzt werden.

The only nuclear model independent method for the determination of nuclear charge radii of short-lived radioactive isotopes is the measurement of the isotope shift. For light elements (Z < 10) extremely high accuracy in experiment and theory is required and was only reached for He and Li so far. The nuclear charge radii of the lightest elements are of great interest because they have isotopes which exhibit so-called halo nuclei. Those nuclei are characterized by a a very exotic nuclear structure: They have a compact core and an area of less dense nuclear matter that extends far from this core. Examples for halo nuclei are 6^He, 8^He, 11^Li and 11^Be that is investigated in this thesis. Furthermore these isotopes are of interest because up to now only for such systems with a few nucleons the nuclear structure can be calculated ab-initio. In the Institut für Kernchemie at the Johannes Gutenberg-Universität Mainz two approaches with different accuracy were developed. The goal of these approaches was the measurement of the isotope shifts between (7,10,11)^Be^+ and 9^Be^+ in the D1 line. The …first approach is laser spectroscopy on laser cooled Be^+ ions that are trapped in a linear Paul trap. The accessible accuracy should be in the order of some 100 kHz. In this thesis two types of linear Paul traps were developed for this purpose. Moreover, the peripheral experimental setup was simulated and constructed. It allows the efficient deceleration of fast ions with an initial energy of 60 keV down to some eV and an effcient transport into the ion trap. For one of the Paul traps the ion trapping could already be demonstrated, while the optical detection of captured 9^Be^+ ions could not be completed, because the development work was delayed by the second approach. The second approach uses the technique of collinear laser spectroscopy that was already applied in the last 30 years for measuring isotope shifts of plenty of heavier isotopes. For light elements (Z < 10), it was so far not possible to reach the accuracy that is required to extract information about nuclear charge radii. The combination of collinear laser spectroscopy with the most modern methods of frequency metrology …finally permitted the …first-time determination of the nuclear charge radii of (7,10)^Be and the one neutron halo nucleus 11^Be at the COLLAPS experiment at ISOLDE/ CERN. In the course of the work reported in this thesis it was possible to measure the absolute transition frequencies and the isotope shifts in the D1 line for the Be isotopes mentioned above with an accuracy of better than 2 MHz. Combination with the most recent calculations of the mass effect allowed the extraction of the nuclear charge radii of (7,10,11)^Be with an relative accuracy better than 1%. The nuclear charge radius decreases from 7^Be continuously to 10^Be and increases again for 11^Be. This result is compared with predictions of ab-initio nuclear models which reproduce the observed trend. Particularly the "Greens Function Monte Carlo" and the "Fermionic Molecular Dynamic" model show very good agreement.

The ingestion of a meal evokes a series of digestive processes, which consist of the essential functions of the digestive system: food transport, secretory activity, absorption of nutrients and the expulsion of undigested residues do not absorbed. The gastrointestinal chemosensitivity is characterized by cellular elements of the endocrine gastrointestinal mucosa and nerve fibers, in particular of vagal nature. A wide range of mediators endocrine and/or paracrine can be released from various endocrine cells in response to nutrients in the diet. These hormones, in addition to their direct activity, act through specific receptors activating some of the most important functions in the control of energy intake and energy homeostasis in the body. For integration of this complex system of control of gastrointestinal chemosensitivity, recent evidence demonstrates the presence of taste receptors (TR) belonging to the family of G proteins coupled receptor expressed in the mucosa of the gastrointestinal tract of different mammals and human. This thesis is divided into several research projects that have been conceived in order to clarify the relationship between TR and nutrients. To define this relationship I have used various scientific approaches, which have gone on to evaluate changes in signal molecules of TR, in particular of the α-transducin in the fasting state and after refeeding with standard diet in the gastrointestinal tract of the pig, the mapping of the same molecule signal in the gastrointestinal tract of fish (Dicentrarchus labrax), the signaling pathway of bitter TR in the STC-1 endocrine cell line and finally the involvement of bitter TR in particular of T2R38 in patients with an excessive caloric intake. The results showed how there is a close correlation between nutrients, TR and hormonal release and how they are useful both in taste perception but also likely to be involved in chronic diseases such as obesity.

Co-evolving with the human host, gut microbiota establishes configurations, which vary under the pressure of inflammation, disease, ageing, diet and lifestyle. In order to describe the multi-stability of the microbiome-host relationship, we studied specific tracts of the bacterial trajectory during the human lifespan and we characterized peculiar deviations from the hypothetical development, caused by disease, using molecular techniques, such as phylogenetic microarray and next-generation sequencing. Firstly, we characterized the enterocyte-associated microbiota in breast-fed infants and adults, describing remarkable differences between the two groups of subjects. Successively, we investigated the impact of atopy on the development of the microbiome in Italian childrens, highlithing conspicuous deviations from the child-type microbiota of the Italian controls. To explore variation in the gut microbiota depending on geographical origins, which reflect different lifestyles, we compared the phylogenetic diversity of the intestinal microbiota of the Hadza hunter-gatherers of Tanzania and Italian adults. Additionally, we characterized the aged-type microbiome, describing the changes occurred in the metabolic potential of the gut microbiota of centenarians with respect to younger individuals, as a part of the pathophysiolology of the ageing process. Finally, we evaluated the impact of a probiotics intervention on the intestinal microbiota of elderly people, showing the repair of some age-related dysbioses. These studies contribute to elucidate several aspects of the intestinal microbiome development during the human lifespan, depicting the microbiota as an extremely plastic entity, capable of being reconfigured in response to different environmental factors and/or stressors of endogenous origin.

The gut microbiota (GM) is essential for human health and contributes to several diseases; indeed it can be considered an extension of the self and, together with the genetic makeup, determines the physiology of an organism. In this thesis has been studied the peripheral immune system reconstitution in pediatric patients undergoing allogeneic hematopoietic stem cell transplantation (aHSCT) in the early phase; in parallel, have been also explored the gut microbiota variations as one of the of primary factors in governing the fate of the immunological recovery, predisposing or protecting from complications such as the onset of acute graft-versus-host disease (GvHD). Has been demonstrated, to our knowledge for the first time, that aHSCT in pediatric patients is associated to a profound modification of the GM ecosystem with a disruption of its mutualistic asset. aGvHD and non-aGvHD subjects showed differences in the process of GM recovery, in members abundance of the phylum Bacteroidetes, and in propionate fecal concentration; the latter are higher in the pre-HSCT composition of non-GvHD subjects than GvHD ones. Short-chain fatty acids (SCFAs), such as acetate, butyrate and propionate, are end-products of microbial fermentation of macronutrients and distribute systemically from the gut to blood. For this reason, has been studied their effect in vitro on human DCs, the key regulators of our immune system and the main player of aGvHD onset. Has been observed that propionate and, particularly, butyrate show a strong and direct immunomodulatory activity on DCs reducing inflammatory markers such as chemokines and interleukins. This study, with the needed caution, suggests that the pre-existing GM structure can be protective against aGvHD onset, exerting its protective role through SCFAs. They, indeed, may regulate cell traffic within secondary lymphoid tissues, influence T cell development during antigen recognition, and, thus, directly shape the immune system.

Il microbiota intestinale riveste un ruolo importantissimo nell’influenzare la salute dell’ospite. È stato dimostrato come la composizione della dieta possa condizionare lo stato di benessere dell’animale, inducendo importanti cambiamenti tra le popolazioni batteriche che coabitano l’intestino; l’uso di prebiotici rappresenta una delle strategie maggiormente impiegate per modulare positivamente la composizione ed il metabolismo dell’ecosistema gastroenterico. Il presente progetto di dottorato si è proposto di indagare gli effetti sul microbiota intestinale del cane e del gatto di diete a diverso tenore proteico e contenenti proteine di diversa digeribilità in presenza o meno di sostanze prebiotiche. Inoltre, sono stati valutati gli effetti della presenza di un estratto di Yucca schidigera e di tannini sulla microflora intestinale del gatto. In ultima istanza, sono state valutate le conseguenze di dosi crescenti di lattosio sul benessere intestinale del cane. I risultati del presente studio hanno rilevato come le sostanze prebiotiche influiscono sulla composizione e sul metabolismo della microflora del cane e del gatto, e come l’impiego di diete ricche di proteine possa avere conseguenze negative sull’ambiente intestinale. Tuttavia, la presenza di oligosaccaridi non sembra contrastare gli effetti negativi che diete ad alto tenore proteico potrebbero avere sull’ecosistema intestinale dell’animale. Nella successiva prova è stato evidenziato come l’inclusione nella dieta di estratti di Yucca e tannini possa contribuire a mitigare l’emanazione di sostanze maleodoranti dalle deiezioni degli animali da compagnia. Nel corso dell’ultima prova, nonostante non siano state osservate differenze tra le popolazioni microbiche intestinali, la somministrazione di dosi crescenti di lattosio ha indotto una certa riduzione delle fermentazioni proteolitiche microbiche. Ulteriori studi sono necessari per stabilire in che misura la dieta e gli alimenti “funzionali” possano influire sul microbiota intestinale del cane e del gatto e come queste informazioni possono essere utilizzate per migliorare miratamente l’alimentazione e lo stato di salute degli animali da compagnia.

In this thesis two approaches were applied to achieve a double general objective. The first chapter was dedicated to the study of the distribution of the expression of genes of several bitter and fat receptor in several gastrointestinal tracts. A set of 7 genes for bitter taste and for 3 genes for fat taste was amplified with real-time PCR from mRNA extracted from 5 gastrointestinal segments of weaned pigs. The presence of gene expression for several chemosensing receptors for bitter and fat taste in different compartments of the stomach confirms that this organ should be considered a player for the early detection of bolus composition. In the second chapter we investigated in young pigs the distribution of butyrate-sensing olfactory receptor (OR51E1) receptor along the GIT, its relation with some endocrine markers, its variation with age, and after interventions affecting the gut environment and intestinal microbiota in piglets and in different tissues. Our results indicate that OR51E1 is strictly related to the normal GIT enteroendocrine activity. In the third chapter we investigated the differential gene expression between oxyntic and pyloric mucosa in seven starter pigs. The obtained data indicate that there is significant differential gene exression between oxintic of the young pig and pyloric mucosa and further functional studies are needed to confirm their physiological importance. In the last chapter, thymol, that has been proposed as an oral alternative to antibiotics in the feed of pigs and broilers, was introduced directly into the stomach of 8 weaned pigs and sampled for gastric oxyntic and pyloric mucosa. The analysis of the whole transcript expression shoes that the stimulation of gastric proliferative activity and the control of digestive activity by thymol can influence positively gastric maturation and function in the weaned pigs.

Der Ginkgo biloba-Extrakt EGb 761 besteht aus einer Reihe pharmakologisch wirksamer Substanzen, welche gut beschriebene Wirkungen auf verschiedene potentiell zytoprotektive Signalwege ausüben und u.a. antioxidative Wirksamkeit haben. Folglich wurde EGb 761 bisher als eine natürliche Behandlung bei neurodegenerativen Erkrankungen mit zellulärem oxidativen Stress angewendet, einschließlich der Alzheimer-Krankheit (AD). Aufgrund von vielen gemeinsamen Merkmalen zwischen der AD und der Huntington-Krankheit (HD) wurde vermutet, dass EGb 761 eventuell auch positive Wirksamkeit bei der HD aufweisen könnte. rnDie Neuropathologie der HD wird durch pathologische Verlängerung an Glutamin-Wiederholungen im Huntingtin-Protein (polyQ-Protein) verursacht, wodurch es zu Fehlfaltungen im Protein kommt und hierdurch der proteasomale Abbau aberranter Proteine erschwert wird. Somit sollten in der vorliegenden Arbeit die EGb 761-Wirkungen auf die Proteasom-Aktivität und die Proteinaggregation in zellulären Modellen der HD untersucht werden. rnWie die ersten Untersuchungen in nativen HEK293-Zellen ergaben, bewirkte die Behandlung der Zellen mit EGb 761 eine Steigerung der basalen Proteasom-Aktivität sowie des proteasomalen Proteinabbaus und erhöhte die Transkription proteasomaler Gene. Hieraus ergaben sich Untersuchungen in Zellen mit Expressionen pathologischer Varianten von polyQ-Proteinen als zelluläre Modelle der HD. Hierbei konnte festgestellt werden, dass die Expression aberranter polyQ-Proteine eine verminderte zelluläre Proteasom-Aktivität bewirkte. Interessanterweise verursachte EGb 761 eine Abmilderung der pathologisch-induzierten verminderten Proteasom-Aktivität, in dem die EGb 761-Behandlung der Zellen zu einer erhöhten Proteasom-Aktivität, einem verbesserten proteasomalen Proteinabbau, sowie zu einer erhöhten Transkription proteasomaler Gene führte. Da diese EGb 761-Effekte unabhängig von der Expression aberranter polyQ-Proteine waren, demonstrierten diese Ergebnisse eine allgemeine EGb 761-Wirkungen auf die Proteasom-Aktivität. Anhand dieser Ergebnisse sollten anschließend weitere Untersuchungen mit zellulären Modellen der HD die genau Wirkung von EGb 761 auf die Degradation von abnormal verlängerten polyQ-Proteinen sowie auf die Bildung von polyQ-Aggregaten klären. rnHier konnte gezeigt werden, dass die Expression aberranter polyQ-Proteinen zu einer Akkumulation von SDS-resistenten bzw. SDS-unlöslichen, aggregierten polyQ-Proteinen führte, sowie die Bildung von sichtbaren polyQ-Aggregaten in Zellen bewirkte. Hierbei verursachte eine EGb 761-Behandlung der Zellen eine signifikante Verminderung im Gehalt an SDS-resistenten polyQ-Proteinen sowie eine Reduzierung von Aggregat-tragenden Zellen. Zudem konnte gezeigt werden, dass eine pharmakologische Inhibition des Proteasoms in EGb 761-behandelten Zellen, den Gehalt an SDS-unlöslichen polyQ-Proteinaggregate wieder erhöhte und somit den Effekt von EGb 761 aufhob. Folglich zeigten diese Ergebnisse, dass die EGb 761-induzierte Reduzierung der polyQ-Proteinaggregate durch einen effizienteren proteasomalen Abbau von fehlgefalteten, aberranten polyQ-Proteinen bewirkte wurde. rnAufbauend auf diesen Ergebnissen wurde eine experimentell-therapeutische Anwendung von EGb 761 in Modellen der HD in vitro und in vivo überprüft und hierzu primäre humane Fibroblasten sowie transgene C. elegans Würmer mit Expressionen aberranter polyQ-Proteine untersucht. Interessanterweise konnte in vitro und in vivo gezeigt werden, dass die EGb 761-Behandlung auch hier eine Reduzierung von SDS-unlöslichen polyQ-Proteinen bewirkte und zudem eine Reduzierung des pathologisch erhöhten Gehalts an Polyubiquitin-Proteinen bewirkte. Folglich wurde auch hier vermutet, dass EGb 761 einen verbesserten proteasomalen Abbau von polyQ-Proteinen induzierte und dies eine Verminderung der polyQ-Proteinaggregate verursachte. Darüber hinaus führte die EGb 761-Behandlung von seneszenten Fibroblasten zur Reduzierung von altersabhängig erhöhten Mengen von polyQ-Aggregaten, wodurch ein therapeutischer Effekt auf den proteasomalen Abbau der polyQ-Proteine verdeutlicht wurde. Zusätzlich konnte in polyQ-transgenen C. elegans demonstriert werden, dass eine EGb 761-Behandlung die Abmilderung eines typischen pathologischen Phänotyps bewirkte, indem eine polyQ-induzierte verminderte Motilität der Nematoden verbessert wurde und hierdurch eine positive EGb 761-Wirkung auf die Pathologie der HD in vivo dargestellt wurde. rnZusammenfassend konnten in dieser Arbeit neue Wirkungen von EGb 761 in der HD demonstriert werden. Hierbei wurde gezeigt, dass EGb 761 die Aggregation von pathogenen aberranten polyQ-Proteinen in vitro und in vivo reduziert, indem eine effizientere Degradation von polyQ-Proteinen erfolgt. Somit könnte diese Wirkungen von EGb 761 eine potentiell therapeutische Anwendung in der HD und ähnliche neurodegenerativen Erkrankungen darstellen.

It was the aim of the study to evaluate the clinical and antibacterial effect of a dentifrice containing an anti-inflammatory plant extract (SB) versus a placebo (PLA) using an experimental gingivitis model. Forty subjects (20 per group) discontinued all oral hygiene measures for four teeth for a period of 21 days using a shield (to generate a possible gingivitis) while they could brush the other teeth normally. After brushing, the shield was removed and teeth were treated with the randomly assigned toothpaste slurry for 1 min. Löe and Silness gingival index (GI), Silness and Löe plaque index (PI), and biofilm vitality (VF%) were assessed at days 0, 14, and 21, respectively.
