The steroidal glycoalkaloid solamargine and its parent aglycone solasodine, isolated from Solanum palinacanthum, were studied in vitro for cytotoxicity and biotransformation by the hepatic S9 fraction as the metabolic activating system. The MTT uptake assay was used to determine viability after 24 h in RAW 264.7 mouse macrophage-like and SiHa cells exposed to various concentrations of the alkaloids in the presence and absence of the hepatic S9 microsomal fraction. The dose-response curves were established for solamargine and solasodine in the presence and absence of external metabolizing system. From these data, the cytotoxic index (CI50) was calculated with mean values of 7.2 and 13.6 μg/mL for Raw cells and 8.6 and 26.0 μg/mL for SiHa cells, respectively. Mass spectrometry was performed to compare the fragmentation patterns of the alkaloids to predict metabolism by the S9 fraction. The mass spectra demonstrated a distinct fragmentation patterns for solamargine and solasodine after the addition of the S9 fraction. In the present study, we demonstrate that the cytotoxic effect of solamargine and solasodine and their metabolites prepared in vitro by biotransformation with the S9 fraction are comparable. These findings suggest that the metabolic activation system S9 fraction may fail to suppress the cytotoxicity of these alkaloids. © 2013 Springer-Verlag Berlin Heidelberg.

Background: Helicobacter pylori infection is usually acquired in childhood and persists into adulthood if untreated. The bacterium induces a chronic inflammatory response, which is associated with epigenetic alterations in oncogenes, tumor-suppressor genes, cell-cycle regulators, and cell-adhesion molecules. Aim: The aim of this study was to analyze the effect of H. pylori infection on the methylation status of Thrombospondin-1 (THBS1), Hypermethylated in cancer 1 (HIC1) and Gata binding protein-4 (GATA-4) in gastric biopsy samples from children and adults infected or uninfected with the bacterium and in samples obtained from gastric cancer patients. Methods: The methylation pattern was analyzed with methylation-specific PCR. Results: Our results showed that H. pylori infection was associated with methylation of the promoter regions of the THBS1 and GATA-4 genes in pediatric and adult samples (p < 0.01). HIC1 showed the lowest level of methylation, which was not an early event during gastric carcinogenesis. Conclusions: The results from this study indicate that methylation of THBS1 and GATA-4 occurs in the early stages of chronic gastritis and gastric cancer in association with H. pylori infection; however, in gastric cancer samples, other mechanisms cooperate with the down-regulation of these genes. Methylation of HIC1 may not be the principal mechanism implicated in its down-regulation in gastric cancer samples. © 2013 Springer Science+Business Media New York.

We studied the distribution of NADPH-diaphorase activity in the visual cortex of normal adult New World monkeys (Saimiri sciureus) using the malic enzyme "indirect" method. NADPH-diaphorase neuropil activity had a heterogeneous distribution. In coronal sections, it had a clear laminar pattern that was coincident with Nissl-stained layers. In tangential sections, we observed blobs in supragranular layers of V1 and stripes throughout the entire V2. We quantified and compared the tangential distribution of NADPH-diaphorase and cytochrome oxidase blobs in adjacent sections of the supragranular layers of V1. Although their spatial distributions were rather similar, the two enzymes did not always overlap. The histochemical reaction also revealed two different types of stained cells: a slightly stained subpopulation and a subgroup of deeply stained neurons resembling a Golgi impregnation. These neurons were sparsely spined non-pyramidal cells. Their dendritic arbors were very well stained but their axons were not always evident. In the gray matter, heavily stained neurons showed different dendritic arbor morphologies. However, most of the strongly reactive cells lay in the subjacent white matter, where they presented a more homogenous morphology. Our results demonstrate that the pattern of NADPH-diaphorase activity is similar to that previously described in Old World monkeys.

Uma das áreas mais promissoras para o estudo das sociedades pré-colombianas complexas na bacia do rio Amazonas é a área do baixo curso dos rios Tapajós, Trombetas e Nhamundá. Há relatos escritos sobre os Konduri e os Tapajós, apresentando informações sobre a sua organização social regional, padrões de comércio, a abundância de alimentos, e o mundo material. Assim, evidências arqueológicas – sítios imensos, solos antrópicos, e belos artefatos – podem ser contrastadas com a informação escrita. Estes ricos depósitos culturais estão desaparecendo a um ritmo alarmante, à medida em que os centros urbanos crescem e se expande a agricultura na região. Apesar desta situação, pouca pesquisa arqueológica tem sido realizada na área, especialmente quando se trata de investigar os antigos padrões de assentamento e redes de trocas. Nos últimos anos, temos realizado pesquisas na bacia do baixo rio Tapajós. O registro arqueológico indica que a cerâmica mostrando forte semelhança estilística forte com a encontrada no grande sítio central está espalhada por pelo menos 90 quilômetros ao sul da presente cidade de Santarém. Este artigo apresenta os resultados de uma pesquisa regional realizada nas proximidades de Santarém, no planalto de Belterra e Alter do Chão, oferecendo uma análise preliminar do sistema de assentamentos na região.

Estudos parasitológicos em populações naturais de primatas neotropicais são relativamente raros, existindo poucos dados disponíveis sobre o guariba-de-mão-ruiva, Alouatta belzebul. No presente estudo, populações de A. belzebul foram amostradas em cinco locais na área do reservatório da Usina Hidrelétrica de Tucuruí no sudeste da Amazônia Brasileira, correspondendo à margem direita do Rio Tocantins. As áreas de coleta incluíram a floresta contínua e fragmentos de hábitats em ilhas, com tamanhos que variaram de 180 a 484 hectares. O principal objetivo deste estudo foi a avaliação dos efeitos da perturbação do hábitat sobre os padrões de infestação por endoparasitas. A densidade populacional foi estimada para cada ponto de coleta usando o método de transecção linear, que variou de 100-108 km percorridos por ponto. Amostras fecais foram coletadas de seis a quatorze grupos em cada local, com um total de 40-46 amostras por ponto (n = 212). As amostras fecais foram fixadas em MIF e observadas através de microscópio óptico, com aumentos de até 400x. A densidade populacional variou entre 66,4 e 191,5 indivíduos por km2. No total, 76,4% das amostras foram positivas para pelo menos uma espécie de endoparasita. Foram identificadas doze táxons de endoparasitas, oito de helmintos e cinco de protozoários. Amostras individuais apresentaram até cinco diferentes espécies de endoparasitas. Em cada local de coleta, o número de espécies identificadas variou entre seis e doze e as taxas de infecção ficaram entre 67,5% e 86%. Não foram encontrados padrões sistemáticos na diversidade de parasitas ou nas taxas de infecção em relação a variáveis como, tamanho de população, densidade ou fragmentação de hábitat. A diversidade e as taxas de infecção variaram mais entre os dois pontos de floresta contínua que nos locais fragmentados e, no geral, foram menores nos locais com menor densidade populacional. A única exceção foi o Trypanoxyuris mirtutus, um oxiurídeo bastante comum transmitido através do contato direto, para o qual foi encontrada uma correlação forte entre as taxas de infecção e a densidade populacional. No geral, foram encontradas poucas evidências capazes de sustentar a hipótese de que a fragmentação do hábitat tem um efeito sistemático nos padrões de infestação em A. belzebul. Contudo, recomenda-se a realização de mais estudos detalhados antes de se estabelecer conclusões definitivas.

The purpose of this study was to compare contrast sensitivity estimated from transient visual evoked potentials (VEPs) elicited by achromatic pattern-reversal and pattern-onset/offset modes. The stimuli were 2-cpd, achromatic horizontal gratings presented either as a 1 Hz pattern reversal or a 300 ms onset/700 ms offset stimulus. Contrast thresholds were estimated by linear regression to amplitudes of VEP components vs. the logarithm of the stimulus contrasts, and these regressions were extrapolated to the zero amplitude level. Contrast sensitivity was defined as the inverse of contrast threshold. For pattern reversal, the relation between the P100 amplitude and log of the stimulus contrast was best described by two separate linear regressions. For the N135 component, a single straight line was sufficient. In the case of pattern onset/offset for both the C1 and C2 components, single straight lines described their amplitude vs. log contrast relations in the medium-to-low contrast range. Some saturation was observed for C2 components. The contrast sensitivity estimated from the low-contrast limb of the P100, from the N135, and from the C2 were all similar but higher than those obtained from the high-contrast limb of the P100 and C1 data, which were also similar to each other. With 2 cpd stimuli, a mechanism possibly driven by the M pathway appeared to contribute to the P100 component at medium-to-low contrasts and to the N135 and C2 components at all contrast levels, whereas another mechanism, possibly driven by the P and M pathways, appeared to contribute to the P100 component at high contrast and C1 component at all contrast levels.

This study compared the effectiveness of the multifocal visual evoked cortical potentials (mfVEP) elicited by pattern pulse stimulation with that of pattern reversal in producing reliable responses (signal-to-noise ratio >1.359). Participants were 14 healthy subjects. Visual stimulation was obtained using a 60-sector dartboard display consisting of 6 concentric rings presented in either pulse or reversal mode. Each sector, consisting of 16 checks at 99% Michelson contrast and 80 cd/m2 mean luminance, was controlled by a binary m-sequence in the time domain. The signal-to-noise ratio was generally larger in the pattern reversal than in the pattern pulse mode. The number of reliable responses was similar in the central sectors for the two stimulation modes. At the periphery, pattern reversal showed a larger number of reliable responses. Pattern pulse stimuli performed similarly to pattern reversal stimuli to generate reliable waveforms in R1 and R2. The advantage of using both protocols to study mfVEP responses is their complementarity: in some patients, reliable waveforms in specific sectors may be obtained with only one of the two methods. The joint analysis of pattern reversal and pattern pulse stimuli increased the rate of reliability for central sectors by 7.14% in R1, 5.35% in R2, 4.76% in R3, 3.57% in R4, 2.97% in R5, and 1.78% in R6. From R1 to R4 the reliability to generate mfVEPs was above 70% when using both protocols. Thus, for a very high reliability and thorough examination of visual performance, it is recommended to use both stimulation protocols.
