Subtitle D of the Resource Conservation and Recovery Act (RCRA) requires a post closure period of 30 years for non hazardous wastes in landfills. Post closure care (PCC) activities under Subtitle D include leachate collection and treatment, groundwater monitoring, inspection and maintenance of the final cover, and monitoring to ensure that landfill gas does not migrate off site or into on site buildings. The decision to reduce PCC duration requires exploration of a performance based methodology to Florida landfills. PCC should be based on whether the landfill is a threat to human health or the environment. Historically no risk based procedure has been available to establish an early end to PCC. Landfill stability depends on a number of factors that include variables that relate to operations both before and after the closure of a landfill cell. Therefore, PCC decisions should be based on location specific factors, operational factors, design factors, post closure performance, end use, and risk analysis. The question of appropriate PCC period for Florida’s landfills requires in depth case studies focusing on the analysis of the performance data from closed landfills in Florida. Based on data availability, Davie Landfill was identified as case study site for a case by case analysis of landfill stability. The performance based PCC decision system developed by Geosyntec Consultants was used for the assessment of site conditions to project PCC needs. The available data for leachate and gas quantity and quality, ground water quality, and cap conditions were evaluated. The quality and quantity data for leachate and gas were analyzed to project the levels of pollutants in leachate and groundwater in reference to maximum contaminant level (MCL). In addition, the projected amount of gas quantity was estimated. A set of contaminants (including metals and organics) were identified as contaminants detected in groundwater for health risk assessment. These contaminants were selected based on their detection frequency and levels in leachate and ground water; and their historical and projected trends. During the evaluations a range of discrepancies and problems that related to the collection and documentation were encountered and possible solutions made. Based on the results of PCC performance integrated with risk assessment, projection of future PCC monitoring needs and sustainable waste management options were identified. According to these results, landfill gas monitoring can be terminated, leachate and groundwater monitoring for parameters above MCL and surveying of the cap integrity should be continued. The parameters which cause longer monitoring periods can be eliminated for the future sustainable landfills. As a conclusion, 30 year PCC period can be reduced for some of the landfill components based on their potential impacts to human health and environment (HH&E).

Characterizing engineered human lung tissue is an important step in developing a functional tissue replacement for lung tissue repair and in vitro analysis. Small tissue constructs were grown by seeding IMR-90 fetal lung fibroblasts and adult microvascular endothelial cells onto a Polyglycolic acid (PGA) polymer template. Introducing the constructs to dynamic culture conditions inside a bioreactor facilitated three-dimensional growth seen in scanning electron microscopy images (SEM). Characterization of the resultant tissue samples was done using SEM imagery, tensile tests, and biochemical assays to quantify extra-cellular matrix (ECM) composition. Tensile tests of the engineered samples indicated an increase in the mechanical properties when compared with blank constructs. Elastin and collagen content was found to average 3.19% and 15.49% respectively in relation to total mass of the tissue samples. The presence of elastin and collagen within the constructs most likely explains the mechanical differences that we noted. These findings suggest that the necessary ECM can be established in engineered tissue constructs and that optimization of this procedure has the capacity to generate the load bearing elements required for construction of a functional lung tissue equivalent.

The problem of solid waste that involves the management, the management and the allocation of these is an issue that permeates all spheres of society, which denotes environmental implications that contribute to the discussion of the environmental crisis in progress. From this perspective, this study aims to investigate and understand the role that consortia, especially the Regional Public Waste Consortium Solid Seridó/RN have waste management, in order to observe their strengths and weaknesses in order to conclude whether they are an appropriate solution to the problem of solid waste. In studying a region that is undergoing a process of institutional organization guided in the solid waste, the focus turned into an interesting academic research point. Since this is a qualitative research, readings were taken of relevant authors to the object and the following legal frameworks, namely: the National Basic Sanitation Policy (Law nº 11.445/2007), the National Policy on Solid Waste (Law nº 12.305/2010 ) and the Law on Public Consortia (Law nº 11.107/2005). The consortium region has about 290.000 inhabitants, generated 40.000 tons of waste a year. As for the final disposal of waste, all municipalities in the region deposit their waste in the open, or in garbage dumps. In the analysis of the Consortium Seridó, a list of issues that are hindering their implementation has been identified. The demands come mainly from the political sphere, but also financial, technical and logistical. It is expected that with the realization of the Consortium by building its supporting structures (overfill Station and Landfill), a new model of solid waste management is implemented.

This work was performing effluent degradation studies by electrochemical treatment. The electrochemical oxidation (EO) hydroquinone (H2Q) was carried out in acid medium, using PbO2 electrode by galvanostatic electrolysis, applying current densities of 10 and 30 mA/cm2 . The concentration of H2Q was monitored by differential pulse voltammetry (DPV). The experimental results showed that the galvanostatic electrolysis process performance significantly depends on the applied current density, achieving removal efficiencies of 100% and 80 % and 10 applying 30 mA/cm2 , respectively. Furthermore, the electroanalytical technique was effective in H2Q be used as a detection method. In order to test the efficiency of PbO2 electrode, the electrochemical treatment was conducted in an actual effluent, leachate from a landfill. The liquid waste leachate (600ml effluent) was treated in a batch electrochemical cell, with or without addition of NaCl by applying 7 mA/cm2 . The efficiency of EO was assessed against the removal of thermo-tolerant coliforms, total organic carbon (TOC), total phosphorus and metals (copper, cobalt, chromium, iron and nickel). These results showed that efficient removal of coliforms was obtained (100%), and was further decrease the concentration of heavy metals by the cathode processes. However, results were not satisfactory TOC, achieving low total removal of dissolved organic load. Because it is considered an effluent complex were developed other tests with this effluent to monitor a larger number of decontamination parameters (Turbidity, Total Solids, Color, Conductivity, Total Organic Carbon (TOC) and metals (barium, chromium, lithium, manganese and Zinc), comparing the efficiency of this type of electrochemical treatment (EO or electrocoagulation) using a flow cell. In this assay was compared to electro streaming. In the case of the OE, Ti/IrO2-TaO5 was used as the anode, however, the electrocoagulation process, aluminum electrodes were used; applying current densities of 10, 20 and 30 mA/cm2 in the presence and absence of NaCl as an electrolyte. The results showed that EO using Ti/IrO2–TaO5 was anode as efficient when Cl- was present in the effluent. In contrast, the electrocoagulation flow reduces the dissolved organic matter in the effluent, under certain experimental conditions.

Tissue-engineered blood vessels (TEBV) can serve as vascular grafts and may also play an important role in the development of organs-on-a-chip. Most TEBV construction involves scaffolding with biomaterials such as collagen gel or electrospun fibrous mesh. Hypothesizing that a scaffold-free TEBV may be advantageous, we constructed a tubular structure (1 mm i.d.) from aligned human mesenchymal cell sheets (hMSC) as the wall and human endothelial progenitor cell (hEPC) coating as the lumen. The burst pressure of the scaffold-free TEBV was above 200 mmHg after three weeks of sequential culture in a rotating wall bioreactor and perfusion at 6.8 dynes/cm(2). The interwoven organization of the cell layers and extensive extracellular matrix (ECM) formation of the hMSC-based TEBV resembled that of native blood vessels. The TEBV exhibited flow-mediated vasodilation, vasoconstriction after exposure to 1 μM phenylephrine and released nitric oxide in a manner similar to that of porcine femoral vein. HL-60 cells attached to the TEBV lumen after TNF-α activation to suggest a functional endothelium. This study demonstrates the potential of a hEPC endothelialized hMSC-based TEBV for drug screening.

Oil spills in marine environments often damage marine and coastal life if not remediated rapidly and efficiently. In spite of the strict enforcement of environmental legislations (i.e., Oil Pollution Act 1990) following the Exxon Valdez oil spill (June 1989; the second biggest oil spill in U.S. history), the Macondo well blowout disaster (April 2010) released 18 times more oil. Strikingly, the response methods used to contain and capture spilled oil after both accidents were nearly identical, note that more than two decades separate Exxon Valdez (1989) and Macondo well (2010) accidents.

The goal of this dissertation was to investigate new advanced materials (mechanically strong aerogel composite blankets-Cabot® Thermal Wrap™ (TW) and Aspen Aerogels® Spaceloft® (SL)), and their applications for oil capture and recovery to overcome the current material limitations in oil spill response methods. First, uptake of different solvents and oils were studied to answer the following question: do these blanket aerogel composites have competitive oil uptake compared to state-of-the-art oil sorbents (i.e., polyurethane foam-PUF)? In addition to their competitive mechanical strength (766, 380, 92 kPa for Spaceloft, Thermal Wrap, and PUF, respectively), our results showed that aerogel composites have three critical advantages over PUF: rapid (3-5 min.) and high (more than two times of PUF’s uptake) oil uptake, reusability (over 10 cycles), and oil recoverability (up to 60%) via mechanical extraction. Chemical-specific sorption experiments showed that the dominant uptake mechanism of aerogels is adsorption to the internal surface, with some contribution of absorption into the pore space.

Second, we investigated the potential environmental impacts (energy and chemical burdens) associated with manufacturing, use, and disposal of SL aerogel and PUF to remove the oil (i.e., 1 m3 oil) from a location (i.e., Macondo well). Different use (single and multiple use) and end of life (landfill, incinerator, and waste-to-energy) scenarios were assessed, and our results demonstrated that multiple use, and waste-to-energy choices minimize the energy and material use of SL aerogel. Nevertheless, using SL once and disposing via landfill still offers environmental and cost savings benefits relative to PUF, and so these benefits are preserved irrespective of the oil-spill-response operator choices.

To inform future aerogel manufacture, we investigated the different laboratory-scale aerogel fabrication technologies (rapid supercritical extraction (RSCE), CO2 supercritical extraction (CSCE), alcohol supercritical extraction (ASCE)). Our results from anticipatory LCA for laboratory-scaled aerogel fabrication demonstrated that RSCE method offers lower cumulative energy and ecotoxicity impacts compared to conventional aerogel fabrication methods (CSCE and ASCE).

The final objective of this study was to investigate different surface coating techniques to enhance oil recovery by modifying the existing aerogel surface chemistries to develop chemically responsive materials (switchable hydrophobicity in response to a CO2 stimulus). Our results showed that studied surface coating methods (drop casting, dip coating, and physical vapor deposition) were partially successful to modify surface with CO2 switchable chemical (tributylpentanamidine), likely because of the heterogeneous fiber structure of the aerogel blankets. A possible solution to these non-uniform coatings would be to include switchable chemical as a precursor during the gel preparation to chemically attach the switchable chemical to the pores of the aerogel.

Taken as a whole, the implications of this work are that mechanical deployment and recovery of aerogel composite blankets is a viable oil spill response strategy that can be deployed today. This will ultimately enable better oil uptake without the uptake of water, potential reuse of the collected oil, reduced material and energy burdens compared to competitive sorbents (e.g., PUF), and reduced occupational exposure to oiled sorbents. In addition, sorbent blankets and booms could be deployed in coastal and open-ocean settings, respectively, which was previously impossible.

Synthesis of Polyhydroxyalkanoates (PHAs) by Pseudomonas mendocina, using different vegetable oils such as, coconut oil, groundnut oil, corn oil and olive oil, as the sole carbon source was investigated for the first time. The PHA yield obtained was compared with that obtained during the production of PHAs using sodium octanoate as the sole carbon source. The fermentation profiles at shaken flask and bioreactor levels revealed that vegetable oils supported the growth of Pseudomonas mendocina and PHA accumulation in this organism. Moreover, when vegetable oil (coconut oil) was used as the sole carbon source, fermentation profiles showed better growth and polymer production as compared to conditions when sodium octanoate was used as the carbon source. In addition, comparison of PHA accumulation at shaken flask and fermenter level confirmed the higher PHA yield at shaken flask level production. The highest cell mass found using sodium octanoate was 1.8 g/L, whereas cell mass as high as 5.1 g/L was observed when coconut oil was used as the feedstock at flask level production. Moreover, the maximum PHA yield of 60.5% dry cell weight (dcw) was achieved at shaken flask level using coconut oil as compared to the PHA yield of 35.1% dcw obtained using sodium octanoate as the sole carbon source. Characterisations of the chemical, physical, mechanical, surface and biocompatibility properties of the polymers produced have been carried out by performing different analyses as described in the second chapter of this study. Chemical analysis using GC and FTIR investigations showed medium chain length (MCL) PHA production in all conditions. GC-MS analysis revealed a unique terpolymer production, containing 3-hydroxyoctanoic acid, 3-hydroxydecanoic acid and 3-hydroxydodecanoic acid when coconut oil, groundnut oil, olive oil, and corn oil were used as the carbon source. Whereas production of the homopolymer containing 3-hydroxyoctanoic acid was observed when sodium octanoate was used as the carbon source. MCL-PHAs produced in this study using sodium octanoate, coconut oil, and olive oil exhibited melting transitions, indicating that each of the PHA was crystalline or semi-crystalline polymer. In contrast, the thermal properties of PHAs produced from groundnut and corn oils showed no melting transition, indicating that they were completely amorphous or semi-crystalline, which was also confirmed by the X-Ray Diffraction (XRD) results obtained in this study. Mechanical analysis of the polymers produced showed higher stiffness of the polymer produced from coconut oil than the polymer from sodium octanoate. Surface characterisation of the polymers using Scanning Electron Microscopy (SEM) revealed a rough surface topography and surface contact angle measurement revealed their hydrophobic nature. Moreover, to investigate the potential applicability of the produced polymers as the scaffold materials for dental pulp regeneration, multipotent human Mesenchymal stem cells (hMSCs) were cultured onto the polymer films. Results indicated that these polymers are not cytotoxic towards the hMSCs and could support their attachment and proliferation. Highest cell growth was observed on the polymer samples produced from corn oil, followed by the polymer produced using coconut oil. In conclusion, this work established, for the first time, that vegetable oils are a good economical source of carbon for production of MCL-PHA copolymers effectively by Pseudomonas mendocina. Moreover, biocompatibility studies suggest that the produced polymers may have potential for dental tissue engineering application.

The new engine plant by General Motors (GM) in Joinville-SC, inaugurated on February 27th 2013, incorporates the most advanced automotive technology processes and broad compliance with environmental standards and energy efficiency. The initiatives implemented in this industrial plant include processes with 100% of recycled industrial waste (landfill free) and pioneer systems in energy efficiency and environmental protection, qualifying the plant to obtain the global certification of Leadership in Energy and Environmental Design (LEED). This industrial project reveals the strategic importance of the region and of Brazil in the growth of GM in the world, becoming a reference for studies and project evaluations of "green" factories in the automotive sector. The present study performs an exploratory research based on scientific publications, assessing the direct and indirect impacts on the business outcome, resulting from implementation of industrial serviceoriented sustainability of its operations, referred to in this article as "Green Factory”. We concluded that the adopted technologies focused on sustainability, study and development, represent a new step for the design of new plants and future expansions of the company in the region, combining low operating cost, low environmental impact and conservation of natural resources.

Un bon fonctionnement du coeur humain est primordial pour maintenir une bonne qualité de vie. Cependant, lorsque le coeur est défaillant, certaines interventions chirurgicales s’avèrent nécessaires pour prolonger l’espérance de vie. Dans le cadre d’un projet multidisciplinaire reliant le génie mécanique avec le domaine biomédical, notre équipe travaille sur la fabrication de valves cardiaques conçues entièrement par génie tissulaire. Pour y parvenir, il est important d’obtenir des propriétés mécaniques optimales pour les tissus biologiques. Afin d’obtenir ces propriétés mécaniques, un outil important a été fabriqué lors d’une étude antérieure : le bioréacteur cardiaque. Le bioréacteur cardiaque permet de reproduire l’environnement physiologique du coeur, notamment les conditions de débit et de pression. Il est crucial de bien contrôler ces conditions, car celles-ci jouent un rôle important lors du conditionnement des substituts valvulaires. Toutefois, il est complexe de contrôler simultanément ces deux conditions de manière efficace. C’est pourquoi notre équipe s’est concentrée sur le développement d’une nouvelle stratégie de contrôle afin que le bioréacteur puisse reproduire le plus fidèlement possible l’environnement physiologique. Plusieurs techniques de contrôle ont été essayés jusqu’à maintenant. Par contre, leur précision était généralement limitée. Une nouvelle approche a donc été envisagée et est présentée dans ce mémoire. Cette nouvelle approche pour le contrôle du bioréacteur est basée sur un type d’algorithme bien connu mais encore très peu utilisé en contrôle : les algorithmes génétiques. Cette approche prometteuse nous a permis de produire des résultats dépassant tous ceux obtenus jusqu’à maintenant pour l’une des deux conditions, soit le débit physiologique.

Les substituts valvulaires disponibles actuellement comportent encore plusieurs lacunes. La disponibilité restreinte des allogreffes, les risques de coagulation associés aux valves mécaniques et la durabilité limitée des bioprothèses en tissu animal sont toutes des problématiques que le génie tissulaire a le potentiel de surmonter. Avec la méthode d’auto-assemblage, le seul support des cellules consiste en leur propre matrice extracellulaire, permettant la fabrication d’un tissu entièrement libre de matériau exogène. Ce projet a été précédé par ceux des doctorantes Catherine Tremblay et Véronique Laterreur, ayant respectivement développé une méthode de fabrication de valves moulées par auto-assemblage et une nouvelle version de bioréacteur. Au cours de cette maîtrise, le nouveau bioréacteur a été adapté à une utilisation stérile avec des tissus vivants et la méthode de fabrication de valves moulées a été modifiée puis éprouvée avec la production de 4 prototypes. Ces derniers n’ont pas permis d’obtenir des performances satisfaisantes en bioréacteur, motivant la conception d’une nouvelle méthode. Plutôt que de tenter de répliquer la forme native des valves cardiaques, des études récentes ont suggéré une géométrie tubulaire. Cela permettrait une fabrication simplifiée, une implantation rapide, et un encombrement minimal en vue d’opérations percutanées. Cette approche minimaliste s’accorde bien avec la méthode d’auto-assemblage, qui a déjà été utilisée pour la production de vaisseaux de petits diamètres. Un total de 11 tubes ont été produits par l’enroulement de feuillets fibroblastiques auto-assemblés, puis ont été transférés sur des mandrins de diamètre inférieur, leur permettant de se contracter librement. La caractérisation de deux tubes contrôles a démontré que cette phase de précontraction était bénéfique pour les propriétés du tissu en plus de prévenir la contraction en bioréacteur. Les prototypes finaux pouvaient supporter un écoulement physiologique pulmonaire. Cette nouvelle méthode montre que le procédé d’auto-assemblage a le potentiel d’être utilisé pour fabriquer des valves cardiaques tubulaires.

O crescente consumo de energia, bem como a possibilidade de esgotamento dos recursos não renováveis, tem fomentado a busca de fontes de energia alternativas. O biodiesel é um biocombustível obtido a partir de fontes renováveis e a sua utilização permite reduzir as emissões de gases com efeito de estufa. Nos últimos anos tem-se produzido biodiesel a partir de óleos alimentares usados (OAU), sendo que com esta aplicação valoriza-se um resíduo e simultaneamente produz-se um combustível “verde”. O biodiesel é produzido através das reações de transesterificação e/ou esterificação entre triglicerídeos e/ou ácidos gordos livres e um álcool, na presença de um catalisador. O rendimento do processo está estritamente relacionado com o tipo de catalisador e as condições que este opera. O principal objetivo do presente trabalho consistiu na avaliação do efeito de alguns parâmetros operacionais no desempenho de uma lípase imobilizada (Novozyme® 435), nomeadamente: (i) índice de acidez do óleo, (ii) razão mássica de enzima/óleo e (iii) método regeneração da enzima com vista à sua reutilização. Também foi objeto de estudo do presente trabalho a produção em contínuo, num (bior)reator tubular de leito fixo, de ésteres metílicos de ácidos gordos (FAME) usando a referida enzima. Registou-se um aumento rendimento em com o incremento do índice de acidez do óleo usado, o que indicia que a enzima catalisa simultaneamente as reações de esterificação e transesterificação. Relativamente à razão mássica de enzima/óleo, dentro da gama testada verificou-se um aumento do rendimento em FAME com a concentração da enzima em meio reacional. Dos vários solventes testados, a aplicação de solvente tert-butanol na regeneração (com incubação) da enzima foi o que melhores resultados teve. Finalmente, os resultados obtidos no ensaio de produção de FAME num biorreator contínuo são motivadores, criando expectativas de uma possível aplicação industrial no futuro.

A produção de combustível derivado de resíduos (CDR) resultou de uma decisão política que permitiu a instalação em Portugal de um significativo conjunto de processos, cuja avaliação técnico-económica e ambiental pode já ser feita. Este trabalho faz uma avaliação técnica e ambiental da linha de produção de CDR da Recivalongo. A avaliação técnica consistiu na análise dos caudais de material que entraram na linha de produção, na quantidade de CDR produzido, contabilizando também os gastos de recursos da linha (energia e materiais auxiliares). A avaliação da qualidade de CDR produzido a partir do ensaio laboratorial representou também uma parte muito significativa do trabalho produzido. A avaliação ambiental foi efetuada com base na metodologia da Avaliação de Ciclo de Vida (ACV). Os resultados obtidos permitiram concluir que o processo da Recivalongo transforma 69% do material de entrada, recuperando 2% em metais ferrosos e rejeitando para aterro a restante fração de 29%. As análises efetuadas ao CDR mostraram que apresenta um PCI compreendido entre 17 e 20 MJ/kg; o teor em cloro está compreendido entre 0,2 e 0,8% Conclui-se que neste processo o parâmetro mais difícil de controlar é o teor em cloro no CDR, pois existe uma grande diversidade de resíduos com grandes quantidades de cloro na sua constituição e que por muitas vezes são difíceis de identificar e/ou separar na primeira fase do tratamento dos resíduos. Da análise ciclo de vida efetuada à produção de CDR pode-se afirmar que esta operação de gestão de resíduos apresenta uma mais-valia quando comparada com a deposição dos resíduos em aterro, não sendo a diferença entre destinos tão significativa quanto o esperado. Desta avaliação pode concluir-se que a instalação operou muito abaixo da sua capacidade, sendo esta considerada uma das melhores linhas de produção de CDR a nível nacional.

O bioetanol constitui uma alternativa renovável aos combustíveis fósseis. Contudo, o bioetanol de primeira geração, produzido a partir de matérias-primas alimentares, desencadeou sérios problemas económicos e sociais, pelo que é fundamental encontrar estratégias que permitam a viabilidade comercial do bioetanol de segunda geração, produzido a partir de matérias-primas lenho-celulósicas. O licor de cozimento ao sulfito ácido de árvores folhosas (HSSL) é um subproduto da indústria papeleira que, devido ao seu elevado conteúdo em açúcares, pode ser utilizado como substrato para a produção de bioetanol de segunda geração. No entanto, a maior fração dos açúcares do HSSL é composta por pentoses. Por isso, a fermentação do HSSL é realizada pela levedura Scheffersomyces stipitis, pois esta é capaz de fermentar tanto as hexoses como as pentoses. Todavia, a S. stipitis só produz etanol sob condições microaerófilas, pelo que o maior desafio da produção de bioetanol por S. stipitis reside no estabelecimento das condições ótimas de arejamento. Este trabalho teve assim por objetivo estabelecer uma estratégia de arejamento que permita a eficiente produção de bioetanol a partir de HSSL por S. stipitis C4, a qual é uma estirpe adaptada a este substrato. Deste modo, foram realizados ensaios em Erlenmeyer, de modo a caracterizar o crescimento da S. stipitis C4, e ensaios em biorreator, com vista a estudar a produção de etanol por S. stipitis C4 em duas estratégias de arejamento diferentes. Na primeira estratégia foi usado apenas um único estágio de arejamento, com controlo da tensão de oxigénio dissolvido, DOT (%), e na segunda estratégia foram usados dois estágios de arejamento, com controlo da DOT no primeiro estágio e com restrição de oxigénio no segundo estágio. Nos ensaios em Erlenmeyer com HSSL o crescimento da S. stipitis C4 foi completamente inibido. Por sua vez, nos ensaios em biorreator com um único estágio de arejamento o controlo da DOT não permitiu a produção de etanol. No entanto, nos ensaios com dois estágios de arejamento em meio sintético foi possível produzir etanol de forma eficiente. Nesta estratégia, a utilização de um maior valor de DOT no primeiro estágio de arejamento permitiu aumentar a taxa específica de crescimento máxima e o rendimento em biomassa do primeiro estágio. Para além disso, a utilização de um maior valor de DOT no primeiro estágio também permitiu aumentar a produtividade em etanol durante o segundo estágio de arejamento. Por sua vez, no segundo estágio de arejamento verificou-se que a restrição de oxigénio evitou a reassimilação de etanol pela S. stipitis C4. Deste modo, os melhores resultados para a produção de etanol foram obtidos com controlo da DOT a 50% durante o primeiro estágio e com 0 mLAr.min-1 e 250 rpm durante o segundo estágio de arejamento. A aplicação desta estratégia de arejamento a 60% HSSL/40% meio sintético permitiu obter, no primeiro estágio de arejamento, uma taxa específica de crescimento máxima de 0,17 h-1, o que demonstra que a elevada disponibilidade de oxigénio durante o primeiro estágio aumenta a tolerância da S. stipitis C4 aos inibidores. Para além disso, a taxa volumétrica de produção de etanol e o rendimento em etanol de toda a fermentação foi de respetivamente de 0,03 g.L-1.h-1 e 0,38 g.g-1. Assim, a elevada eficiência de conversão dos açúcares em etanol (74,4%) demostra que a fermentação com dois estágios de arejamento constitui uma estratégia promissora para a produção de bioetanol de segunda geração a partir de HSSL.

With the disorganized decentralization occurred in Brazil after the 1988 Constitution, municipalities have risen to the level of federal entities. This phenomenon became known as "municipalism" also brought some negative effects such as low capacity financial, economic and political of these entities. In the face of this reality , the municipalities sought in models of collaborative features to address public policy issues ultrarregionais, one of these models are the Public Consortia. Characterized as the organization of all federal entities that aim to solve public policy implementation alone that they could not, or spend great resources for such. This reality of the municipalities have an aggravating factor when looking at the situation in Metropolitan Regions (MRs). This is because the RMs has a historical process of formation that does not encourage cooperation, since that were created top-down during the military regime. Furthermore, the metropolitan municipalities have significant power asymmetries, localist vision, rigidity earmarked revenues, different scenarios conurbation, difficulty standardization of concepts and others that contribute to the vision of low cooperation of these metropolitan areas. Thus, the problem of this work is in the presence of collaborative arrangements, such as the Public Consortia in metropolitan areas, which are seen as areas of low cooperation. To elucidate this research was used for analysis the cases of CONDIAM/PB and Consórcio Grande Recife/PE, because they are apparently antagonistic, but with some points of similarity. The cases has as foundation the Theory of Common Resources, which provides the possibility of collective action through the initiative of individuals. This theory has as its methodology for analyzing the picture IAD Framework, which proposes its analysis based on three axes: external variables, the arena of action and results. The nature of the method of this research was classified as exploratory and descriptive. For the stage of date analysis, was used the method of document analysis and content, Further than of separation of the cases according to theur especificities. At the end of the study, noted that the CONDIAM/PB was a strategy of municipal government of Joao Pessoa to attract funds from the Federal Government for the purpose of to build a landfill, and over the years the ideology of cooperation was left aside, the prevailing view localist municipalities. In the case of Consórcio Grande Recife/PE, members act with some degree of cooperation, especially the collaborative aspect of the region, however, still prevails with greater strength the power of the state of Pernambuco in the decisions and paths of the consortium. Thus, was conclude that the Public Consortia analyzed are an experience of collaborative arrangement, from the initiative of members, as the theory of common resources says, but has not actually signed as a practice of collective action to overcome the dilemmas faced by metropolitan areas

Carbon capture and storage (CCS) in the oil and water industries is becoming common and a significant consumer of energy typically requiring 150–450 °C and or several hundred bar pressure [1] particularly in geological deposition. A biological carbon capture and conversion has been considered in conventional anaerobic digestion processes. The process has been utilised in biological mixed culture, where acetoclastic bacteria and hydrogenophilic methanogens play a major key role in the utilisation of carbon dioxide. However, the bio catalytic microorganisms, hydrogenophilic methanogens are reported to be unstable with acetoclastic bacteria. In this work the biochemical thermodynamic efficiency was investigated for the stabilisation of the microbial process in carbon capture and utilisation. The authors observed that a thermodynamic efficiency of biological carbon capture and utilisation (BCCU) had 32% of overall reduction in yield of carbon dioxide with complimentary increase of 30% in yield of methane, while the process was overall endothermic. Total consumption of energy (≈0.33 MJ l−1) was estimated for the carbonate solubility (0.1 mol l−1) in batched BCCU. This has a major influence on microbial composition in the bioreactor. This thermodynamic study is an essential tool to aid the understanding of the interactions between operating parameters and the mixed microbial culture.
