Durant aquests darrers anys hem pogut veure com el concepte de qualitat apareixia amb molta força a casa nostra. En l'àmbit empresarial i possiblement degut a la influència que ha tingut la implicació de les grans empreses en tot aquest moviment, o potser només a la creixent competència de les empreses estrangeres, el petit i mitjà empresari del nostre país s'ha vist obligat a obrir les portes de casa seva al que podríem definir com a la "Cultura de la Qualitat". És evident que en aquesta entrada l'administració també hi ha pres part. Ara bé, sovint aquest missatge a favor de la qualitat ha estat "disfressat" de la tant coneguda normativa ISO 9000. Poc a poc, sota aquestes sigles la gent s'ha fet la distorsionada idea de que això vol dir qualitat i tot el que surti d'això és sinònim de mala qualitat. Així doncs, val realment la pena la implicació en el compliment d'aquestes normes? És realment per això pel que la majoria de les petites i mitjanes empreses del nostre entorn s'han implicat en l'assegurament de la qualitat segons la ISO 9000? I més encara, una vegada han aconseguit el certificat de qualitat n'estan realment satisfetes de les millores que han tingut? O la única millora important és la de la imatge externa de l'empresa? La recerca d'una resposta amb base científica és la que porta al desenvolupament del present treball, omplint el buit que hi ha actualment entre els treballs teòrics i la realitat. Aquesta tesi vol esbrinar quin ha estat l'impacte que hi ha hagut a les empreses catalanes degut a la implicació en la normativa d'assegurament de la qualitat ISO 9000. El mètode de recerca es basarà en una anàlisi de les dades que es recullin d'un treball empíric a realitzar en petites i mitjanes empreses de les comarques de Catalunya. Al primer capítol es defineix exactament quin és l'objectiu d'aquest treball de recerca. Al capítol dos es pot trobar una perspectiva del que ha estat la qualitat en la història i de la gent que ha fet possible que prengués la importància que ara té. La normativa d'assegurament de la qualitat ISO 9000, marc de treball d'aquesta tesi es mereix un capítol apart, el tres. A partir de les hipòtesis a contrastar, i la literatura sobre la gestió de la qualitat, al quart capítol es defineix com es portarà a terme es treball, quedant definit doncs quin serà el treball empíric que s'ha portat a terme. Al cinquè capítol es troben els resultats obtinguts del treball empíric, així com un anàlisi descriptiu d'aquests. Les hipòtesis plantejades es resolen al capítol sisè, mitjançant l'aplicació de la tècnica estadística denominada: "Anàlisi cluster". Aquesta tècnica permetrà veure per a quines agrupacions d'empreses es compleixen les hipòtesis i per quines no, realitzant-se així un estudi més concret de la situació. Finalment al capítol setè és on s'hi troben les conclusions d'aquest treball,analitzant-se també quines poden ser futures línies d'investigació en aquest camp.

La present tesi pretèn ser,tal com el seu mateix nom indica, un estudi de totes aquelles informacions que ens proporcionen les fonts escrites d'època tardoantiga (segles IV-VII)i que fan referència, directament o indirecta, a les terres del nord-est de Catalunya.Fou aquesta àrea geogràfica la que, d'ençà el segle IX, i un cop superada la invasió sarraïna que suposà, entre altres coses, la definitiva desaparició de la seu episcopal emporitana, acabà per convertir-se en el futur territori del bisbat de Girona, una demarcació que englovava els antics dominis eclesiàstics i territorials de les seus visigòtiques de Gerunda i Impurias. És una realitat que l'estudi de la tardoantiguitat al nord-est de Catalunya ha experimentat un important impuls en els darrers anys. Es en aquest marc, que la investigació que presentem, pretén ser una aportació a una millor comprensió d'aquest període, amb la sistematització i catalogació de tots aquells textos, la cronologia dels quals abraça el període comprés entre els segles IV-VII, on es fa menció expressa a l'àrea geogràfica que ens ocupa. Aquest estudi s'estructura en set parts clarament diferenciades encara que interdependents entre si, cadascuna de les quals amb una especificitat i una metodologia pròpia. A la primera d'elles (caps.2-3), i a mode d'introducció, es fixen els límits territorials (marc geogràfic) i cronològics (marc històric) de la investigació, elements del tot indispensables i necessaris per a comprendre millor l'escenari on tenen lloc els fets descrits. Un segon gran apartat està enterament dedicat a l'estudi i anàlisi de les principals aportacions realitzades per la historiografia del nostre país en els darrers segles, del XVII fins a l'actualitat, en l'estudi del tema que ens ocupa. En tercer lloc, es passa a analitzar individualment totes i cadascuna de les referències textuals recollides en aquest estudi. L'anàlisi l'hem divididida en cin grans apartats, atenent i respectant la diversa tipologia de les fonts consultades: 1) els itineraria romana; 2) els textos de la patrologia tardoantiga; 3) les actes conciliars; 4) els textos litúrgics; i 5) l'epigrafia. L'estudi de cada text es realitza d'una manera global i sitemàtica, analitzant l'autor, el text i l'època de composició, i tot això en el marc més ampli de la societat del seu temps. Un apartat certament important és enterament dedicat a l'estudi dels orígens i difusió del culte al màrtir Feliu de Girona durant l'antiguitat tardana, a partir sobretot de la informació proporcionada per les fonts escrites i epigràfiques de l'època. El culte al màrtir gironí del segle IV fou, sense cap mena de dubtes, un dels elements característics i definidors en la vida de la Gerunda tardoantiga i, al mateix temps, un signe del gran prestigi assolit per la seu episcopal gironina a la resta d'Hispània durant l'antiguitat tardana. Finalment, i després de presentar, a tall de síntesi, les principals conclusions d'aquesta investigació, un darrer gran apartat ve constituït per un apèndix documental, en el qual s'hi recullen, seguint un criteri cronològic, tots els textos estudiats, transcrits segons les principals edicions crítiques citades. Val a dir, que en aquesta investigació hem utilitzat en la majoria dels casos les millors edicions crítiques de cadascuna de les obres estudiades, un fet que ha permès, en ocasions, corregir antigues transcripcions molt menys acurades i crítiques que s'havien inclòs en moltes de les obres dels darrers dos-cents anys on s'estudiava el període històric que ens ocupa.

A dataset of 1,846,990 completed lactation record,; was created Using milk recording data from 8,967 commercial dairy farms in the United Kingdom over a five year period. Herd-specific lactation curves describing levels of milk, Cat and protein by lactation number and month of calving were generated for each farm. The actual yield of milk and protein proportion at the first milk recording of individual cow lactations were compared with the levels taken from the lactation curves. Logistic regression analysis showed that cows production milk with a lower percentage of protein than average had a significantly lower probability of being in-calf at 100 days post calving and it significantly higher probability of being culled at the end of lactation. The culling rates derived from the studied database demonstrate the current high wastage rate of commercial dairy cows. Well of this wastage is due to involuntary culling as a result of reproductive failure.

Accurate knowledge of lactation curves has an important relevance to management and research of dairy production systems. A number of equations have been proposed to describe the lactation curve, the most widely applied being the gamma equation. The objective of this work was to compare and evaluate candidate functions for their predictive ability in describing lactation curves from central Mexican dairy cows reared under 2 contrasting management systems. Five equations were considered: Gaines ( exponential decay), Wood ( gamma equation), Rook ( Michaelis-Menten x exponential), and 2 more mechanistic ones (Dijkstra and Pollott). A database consisting of 701 and 1283 records of cows in small-scale and intensive systems, respectively, was used in the analysis. Before analysis, the database was divided into 6 groups representing first, second, and third and higher parity cows in both systems. In all cases except second and above parity cows in small-scale systems, all models improved on the Gaines equation. The Wood equation explained much of the variation, but its parameters do not have direct biological interpretation. Although the Rook equation fitted the data well, some of the parameter estimates were not significant. The Dijkstra equation consistently gave better predictions, and its parameters were usually statistically significant and lend themselves to physiological interpretation. As such, the differences between systems and parity could be explained due to variations in theoretical initial milk production at parturition, specific rates of secretory cell proliferation and death, and rate of decay, all of which are parameters in the model. The Pollott equation, although containing the most biology, was found to be over-parameterized and resulted in nonsignificant parameter estimates. For central Mexican dairy cows, the Dijkstra equation was the best option to use in describing the lactation curve.

Milk supply from Mexican dairy farms does not meet demand and small-scale farms can contribute toward closing the gap. Two multi-criteria programming techniques, goal programming and compromise programming, were used in a study of small-scale dairy farms in central Mexico. To build the goal and compromise programming models, 4 ordinary linear programming models were also developed, which had objective functions to maximize metabolizable energy for milk production, to maximize margin of income over feed costs, to maximize metabolizable protein for milk production, and to minimize purchased feedstuffs. Neither multicriteria approach was significantly better than the other; however, by applying both models it was possible to perform a more comprehensive analysis of these small-scale dairy systems. The multi-criteria programming models affirm findings from previous work and suggest that a forage strategy based on alfalfa, rye-grass, and corn silage would meet nutrient requirements of the herd. Both models suggested that there is an economic advantage in rescheduling the calving season to the second and third calendar quarters to better synchronize higher demand for nutrients with the period of high forage availability.

This study sets out to find the best calving pattern for small-scale dairy systems in Michoacan State, central Mexico. Two models were built. First, a linear programming model was constructed to optimize calving pattern and herd structure according to metabolizable energy availability. Second, a Markov chain model was built to investigate three reproductive scenarios (good, average and poor) in order to suggest factors that maintain the calving pattern given by the linear programming model. Though it was not possible to maintain the optimal linear programming pattern, the Markov chain model suggested adopting different reproduction strategies according to period of the year that the cow is expected to calve. Comparing different scenarios, the Markov model indicated the effect of calving interval on calving pattern and herd structure.

A limitation of small-scale dairy systems in central Mexico is that traditional feeding strategies are less effective when nutrient availability varies through the year. In the present work, a linear programming (LP) model that maximizes income over feed cost was developed, and used to evaluate two strategies: the traditional one used by the small-scale dairy producers in Michoacan State, based on fresh lucerne, maize grain and maize straw; and an alternative strategy proposed by the LIP model, based on ryegrass hay, maize silage and maize grain. Biological and economic efficiency for both strategies were evaluated. Results obtained with the traditional strategy agree with previously published work. The alternative strategy did not improve upon the performance of the traditional strategy because of low metabolizable protein content of the maize silage considered by the model. However, the Study recommends improvement of forage quality to increase the efficiency of small-scale dairy systems, rather than looking for concentrate supplementation.

Small-scale dairy systems play an important role in the Mexican dairy sector and farm planning activities related to resource allocation have a significant impact on the profitability of such enterprises. Linear programming is a technique widely used for planning and ration formulation, and partial budgeting is a technique for assessing the impact of changes on the profitability of an enterprise. This study used both methods to optimise land use for forage production and nutrient availability, and to evaluate the economic impact of such changes in small-scale Mexican dairy systems. The model showed satisfactory performance when optimal solutions were compared with the traditional strategy. The strategy using fresh ryegrass, maize silage and oat hay, and the strategy using a combination of alfalfa hay, maize silage, fresh ryegrass and oat hay appeared attractive options for providing a better nutrient supply and maintaining a higher stocking rate throughout the year than the traditional strategy.

Statistical approaches have been applied to examine amino acid pairing preferences within parallel beta-sheets. The main chain hydrogen bonding pattern in parallel beta-sheets means that, for each residue pair, only one of the residues is involved in main chain hydrogen bonding with the strand containing the partner residue. We call this the hydrogen bonded (HB) residue and the partner residue the non-hydrogen bonded (nHB) residue, and differentiate between the favorability of a pair and that of its reverse pair, e.g. Asn(HB)-Thr(nHB)versus Thr(HB)-Asn(nHB). Significantly (p < or = 0.000001) favoured pairings were rationalised using stereochemical arguments. For instance, Asn(HB)-Thr(nHB) and Arg(HB)-Thr(nHB) were favoured pairs, where the residues adopted favoured chi1 rotamer positions that allowed side-chain interactions to occur. In contrast, Thr(HB)-Asn(nHB) and Thr(HB)-Arg(nHB) were not significantly favoured, and could only form side-chain interactions if the residues involved adopted less favourable chi1 conformations. The favourability of hydrophobic pairs e.g. Ile(HB)-Ile(nHB), Val(HB)-Val(nHB) and Leu(HB)-Ile(nHB) was explained by the residues adopting their most preferred chi1 and chi2 conformations, which enabled them to form nested arrangements. Cysteine-cysteine pairs are significantly favoured, although these do not form intrasheet disulphide bridges. Interactions between positively and negatively charged residues were asymmetrically preferred: those with the negatively charged residue at the HB position were more favoured. This trend was accounted for by the presence of general electrostatic interactions, which, based on analysis of distances between charged atoms, were likely to be stronger when the negatively charged residue is the HB partner. The Arg(HB)-Asp(nHB) interaction was an exception to this trend and its favorability was rationalised by the formation of specific side-chain interactions. This research provides rules that could be applied to protein structure prediction, comparative modelling and protein engineering and design. Objectives: Influenza A H3N2 viruses isolated recently have characteristic receptor binding properties that may decrease susceptibility to neuraminidase inhibitor drugs. A panel of clinical isolates and recombinant viruses generated by reverse genetics were characterized and tested for susceptibility to zanamivir. Methods: Plaque reduction assays and neuraminidase enzyme inhibition assays were used to assess susceptibility to zanamivir. Receptor binding properties of the viruses were characterized by differential agglutination of red blood cells (RBCs) from different species. Sequence analysis of the haemagglutinin (HA) and neuraminidase (NA) genes was carried out. Results: Characterization of a panel of H3N2 clinical isolates from 1968 to 2000 showed a gradual decrease in agglutination of chicken and guinea pig RBCs over time, although all isolates could agglutinate turkey RBCs equally. Sequence analysis of the HA and NA genes identified mutations in conserved residues of the HA1 receptor binding site, in particular Leu-226 --> Ile-226/Val-226, and modification of potential glycosylation site motifs. This may be indicative of changes in virus binding to sialic acid (SA) receptors in recent years. Although recent isolates had reduced susceptibility to zanamivir in MDCK cell based plaque reduction assays, no difference was found in an NA enzyme-inhibition assay. Assays with recombinant isogenic viruses showed that the recent HA, but not the NA, conferred reduced susceptibility to zanamivir. Conclusion: This study demonstrates that recent clinical isolates of influenza A H3N2 virus no longer agglutinate chicken RBCs, but despite significant receptor binding changes as a result of changes in HA, there was little variation in sensitivity of the NA to zanamivir.

Single crystal X-ray diffraction studies reveal that the incorporation of meta-amino benzoic acid in the middle of a helix forming hexapeptide sequence such as in peptide I Boc-Ile(1)-Aib(2)-Val(3)-m-ABA(4)-Ile(5)-Aib(6)-Leu(7)-OMe (Aib: alpha-amino isobutyric acid: m-ABA: meta-amino benzoic acid) breaks the helix propagation to produce a turn-linker-turn (T-L-T) foldamer in the solid state. In the crystalline state two conformational isomers of peptide I self-assemble in antiparallel fashion through intermolecular hydrogen bonds and aromatic pi-pi interactions to form a molecular duplex. The duplexes are further interconnected through intermolecular hydrogen bonds to form a layer of peptides. The layers are stacked one on top of the other through van der Waals interactions to form hydrophilic channels filled with solvent methanol. (C) 2009 Elsevier B.V. All rights reserved.

Single crystal X-ray diffraction studies and solvent dependent NMR titration reveal that the designed pepticles I and 11, Boc-Xx(1)-Aib(2)-Yy(3)-NH(CH2)(2)NH-Yy(3)-Aib(2)-Xx(1)-Boc, where Xx and Yy are lie and Leu in peptide I and Leu and Val in peptide 11, respectively, fold into a turn-linker-turn (T-L-T) conformation both in the solid state and in solution. In the crystalline state the T-L-T foldamers; of peptide I and II self-assemble to form a three-dimensional framework of channels. The insides of the channels are hydrophilic and found to contain solvent CHCl3 hydrogen bonded to exposed C=O of Aib located at the turn regions. (c) 2008 Elsevier B.V. All rights reserved.

A series of water-soluble synthetic dipeptides (1-3) with an N-terminally located beta-alanine residue, beta-alanyl-L-valine (1), beta-alanyl-L-isoleucine (2), and beta-alanyl-L-phenylalanine (3, form hydrogen-bonded supramolecular double helices with a pitch length of 1 nm, whereas the C-terminally positioned beta-alanine containing dipeptide (4), L-phenylalanyl-beta-alanine, does not form a supramolecular double helical structure. beta-Ala-Xaa (Xaa = Val/Ile/Phe) can be regarded as a new motif for the formation of supramolecular double helical structures in the solid state.

A terminally protected acyclic tetrapeptide Boc-Aib-Val-Aib-beta-Ala-OMe 1 (Aib: alpha-aminoisobutyric acid, beta-Ala: beta-Alanine) self-assembles into a continuous hydrogen-bonded supramolecular helix with an average diameter of 10Angstrom (1nm) starting from a double bend molecular conformation in crystals and further self-assembly of this supramolecular architecture leads to the formation of polydisperse nanorods of diameters 10-40 nm.
