Glycosyl hydrolases are enzymes capable of breaking the glycosidic linkage of polysaccharides and have considerable industrial and biotechnological applications. Driven by the later applications, it is frequently desirable that glycosyl hydrolases display stability and activity under extreme environment conditions, such as high temperatures and extreme pHs. Here, we present X-ray structure of the hyperthermophilic laminarinase from Rhodothermus marinus (RmLamR) determined at 1.95 angstrom resolution and molecular dynamics simulation studies aimed to comprehend the molecular basis, for the thermal stability of this class of enzymes. As most thermostable proteins, RmLamR contains a relatively large number of salt bridges, which are not randomly distributed on the structure. On the contrary, they form clusters interconnecting beta-sheets of the catalytic domain. Not all salt bridges, however, are beneficial for the protein thermostability: the existence of charge-charge interactions permeating the hydrophobic core of the enzymes actually contributes to destabilize the structure by facilitating water penetration into hydrophobic cavities, as can be seen in the case of mesophilic enzymes. Furthermore, we demonstrate that the mobility of the side-chains is perturbed differently in each class of enzymes. The side-chains of loop residues surrounding the catalytic cleft in the mesophilic laminarinase gain mobility and obstruct the active site at high temperature. By contrast, thermophilic laminarinases preserve their active site flexibility, and the active-site cleft remains accessible for recognition of polysaccharide substrates even at high temperatures. The present results provide structural insights into the role played by salt-bridges and active site flexibility on protein thermal stability and may be relevant for other classes of proteins, particularly glycosyl hydrolases.

Cdc25 phosphatases involved in cell cycle checkpoints are now active targets for the development of anti-cancer therapies. Rational drug design would certainly benefit from detailed structural information for Cdc25s. However, only apo- or sulfate-bound crystal structures of the Cdc25 catalytic domain have been described so far. Together with previously available crystalographic data, results from molecular dynamics simulations, bioinformatic analysis, and computer-generated conformational ensembles shown here indicate that the last 30-40 residues in the C-terminus of Cdc25B are partially unfolded or disordered in solution. The effect of C-terminal flexibility upon binding of two potent small molecule inhibitors to Cdc25B is then analyzed by using three structural models with variable levels of flexibility, including an equilibrium distributed ensemble of Cdc25B backbone conformations. The three Cdc25B structural models are used in combination with flexible docking, clustering, and calculation of binding free energies by the linear interaction energy approximation to construct and validate Cdc25B-inhibitor complexes. Two binding sites are identified on top and beside the Cdc25B active site. The diversity of interaction modes found increases with receptor flexibility. Backbone flexibility allows the formation of transient cavities or compact hydrophobic units on the surface of the stable, folded protein core that are unexposed or unavailable for ligand binding in rigid and densely packed crystal structures. The present results may help to speculate on the mechanisms of small molecule complexation to partially unfolded or locally disordered proteins.

Coal mining and incineration of solid residues of health services (SRHS) generate several contaminants that are delivered into the environment, such as heavy metals and dioxins. These xenobiotics can lead to oxidative stress overgeneration in organisms and cause different kinds of pathologies, including cancer. In the present study the concentrations of heavy metals such as lead, copper, iron, manganese and zinc in the urine, as well as several enzymatic and non-enzymatic biomarkers of oxidative stress in the blood (contents of lipoperoxidation = TBARS, protein carbonyls = PC, protein thiols = PT, alpha-tocopherol = AT, reduced glutathione = GSH, and the activities of glutathione S-transferase = GST, glutathione reductase = GR, glutathione peroxidase = GPx, catalase = CAT and superoxide dismutase = SOD), in the blood of six different groups (n = 20 each) of subjects exposed to airborne contamination related to coal mining as well as incineration of solid residues of health services (SRHS) after vitamin E (800 mg/day) and vitamin C (500 mg/day) supplementation during 6 months, which were compared to the situation before the antioxidant intervention (Avila et al., Ecotoxicology 18:1150-1157, 2009; Possamai et al., Ecotoxicology 18:1158-1164, 2009). Except for the decreased manganese contents, heavy metal concentrations were elevated in all groups exposed to both sources of airborne contamination when compared to controls. TBARS and PC concentrations, which were elevated before the antioxidant intervention decreased after the antioxidant supplementation. Similarly, the contents of PC, AT and GSH, which were decreased before the antioxidant intervention, reached values near those found in controls, GPx activity was reestablished in underground miners, and SOD, CAT and GST activities were reestablished in all groups. The results showed that the oxidative stress condition detected previously to the antioxidant supplementation in both directly and indirectly subjects exposed to the airborne contamination from coal dusts and SRHS incineration, was attenuated after the antioxidant intervention.

Reactive oxygen species and nitrogen species have been implicated in the pathogenesis of coal dust-induced toxicity. The present study investigated several oxidative stress biomarkers (Contents of lipoperoxidation = TBARS, reduced = GSH, oxidized = GSSG and total glutathione = TG, alpha-tocopherol, and the activities of glutathione S-transferase = GST, glutathione reductase = GR, glutathione peroxidase = GPx, catalase = CAT and superoxide dismutase = SOD), in the blood of three different groups (n = 20 each) exposed to airborne contamination associated with coal mining activities: underground workers directly exposed, surface workers indirectly exposed, residents indirectly exposed (subjects living near the mines), and controls (non-exposed subjects). Plasma TBARS were increased and whole blood TG and GSH levels were decreased in all groups compared to controls. Plasma alpha-tocopherol contents showed approximately half the values in underground workers compared to controls. GST activity was induced in workers and also in residents at the vicinity of the mining plant, whilst CAT activity was induced only in mine workers. SOD activity was decreased in all groups examined, while GPx activity showed decreased values only in underground miners, and GR did not show any differences among the groups. The results showed that subjects directly and indirectly exposed to coal dusts face an oxidative stress condition. They also indicate that people living in the vicinity of the mine plant are in health risk regarding coal mining-related diseases.

In this work, the electronic and structural characterization of polyaniline (PANI) formed in cavities of zeolites Y (ZY) and Mordenite (MOR) and montmorillonite (MMT) clay having Cu(II) as oxidant agent are presented. The formation of PANI and its structure is analyzed by Resonance Raman, UV-Vis-NIR, FT-IR and N K XANES techniques. In all cases the structure of PANT formed is different from the ""free"" polymer. The presence of azo bonds linked to phenazine-like rings are observed only for PANI-MMT composites, independent of the kind of oxidant agent employed in the synthesis. The presence of Cu(II) ions leads to the formation of Phenosafranine-like rings. The presence of these phenazine-like rings in the structure of confined PANT chains can also contribute to the enhancement of the thermal stability observed for all composites. (C) 2008 Elsevier Ltd. Objective: To evaluate the flexural strength, microleakage, and degree of conversion of a microhybrid resin polymerized with argon laser and halogen lamp. Method and Materials: For both flexural test and degree of conversion analysis, 5 bar samples of composite resin were prepared and polymerized according to ISO 4049. The halogen light-curing unit was used with 500 MW/cm(2) for 20 seconds and the argon laser with 250 mW for 10 and 20 seconds. Samples were stored in distilled water in a dark environment at 37 degrees C for 24 hours. The flexural property was quantified by a 3-point loading test. For the microleakage evaluation, 60 bovine incisors were used to prepare standardized Class 5 cavities, which were restored and polished. Specimens were stored in distilled water for 24 hours at 37 degrees C and thermocycled 500 times (6 degrees C to 60 degrees C). Specimens were then immersed in art aqueous solution of basic fuchsin for 24 hours. Longitudinal sections of each restoration were obtained and examined with a stereomicroscope for qualitative evaluation of microleakage. Fourier transform (FT)-Raman RFS 100/S spectrometer (Bruker) was used to analyze the degree of conversion. Results: ANOVA showed no statistically significant differences of flexural strength between the photoactivation types evaluated in the flexural study. Microleakage data were statistically analyzed by Mann-Whitney and Kruskal-Wallis tests. Enamel margins resulted in a statistically lower degree of leakage than dentin margins. No statistically significant difference was found among the 3 types of photocuring studied. ANOVA also showed no statistically significant difference in the degree of conversion among the studied groups. Conclusion: According to the methodology used in this research, the argon laser is a possible alternative for photocuring, providing the same quality of polymerization as the halogen lamp. None of the photocured units tested in this study completely eliminated microleakage.

The genus Candida includes different species that have the potential to invade and colonize the human body and C. albicans is the most common cause of skin, nail and mucous infections. The increasing resistance against antifungal drugs has renewed the search for new treatment procedures and antimicrobial photodynamic inactivation (PDI) is a propitious candidate. Hypericin (HY) has several wanted properties to be used as a photosensitizer in this technique including a high quantum yield of singlet oxygen generation, a high extinction coefficient near 600 nm, and a relatively low dark toxicity. Although the phototoxicity of HY on several tumor cells has been reported, the data concerning its photoactivity on microorganisms are scarce. The aim of this study was to obtain the experimental parameters to achieve an acceptable selective hypericinphotoinactivation of two species of Candida comparing with fibroblasts and epithelial cells which are the constituents of some potential host tissues, such mucosas, skin and cavities. Microorganisms and cells were incubated with the same HY concentrations and short incubation time followed by irradiation with equal dose of light. The best conditions to kill just Candida were very low HY concentration (0.1-0.4 mu g ml(-1)) incubated by 10 min and irradiated with LED 590 nm with 6 J cm(-2).

Denna uppsats handlar om förekomsten av Antisemitism i den svenska s.k. Vit makt-musiken. Uppsatsens syfte är att kartlägga förekomsten av antisemitism i tre vit makt- band: Pluton Svea, Storm och Dirlewanger. Analysen sker genom att texterna till två album av varje band studeras. Metoden är både kvalitativ som kvantitativ och bland annat så undersöks olika typer av antisemitiska uttalanden som därefter nivåsätts enligt en, av författaren, modifierad skala konstruerad av forskaren John C.G. Röhl. Resultatet av denna undersökning visar att antisemitism är ständigt närvarande i denna musikstil. Dock tar den sig olika uttryck beroende på vilket band det gäller. I de album som är producerade under 2000-talet tycks även en viss nedtoning av den våldsammaste formen av antisemitism kunna ses.

Denna uppsats problematiserar hur kollektiva förvaltningsorganisationer fördelar pengar insamlade från offentligt framförd klubbmusik samt undersöker hur etablerade dj’s och producenter ser på dagens system och tror sig påverkas av detta. Med djupintervjuer av fyra personer undersöker uppsatsen även hur dessa ser på rapportering av den spelade musiken, vilka problem som kan uppstå och om det finns tekniska möjligheter att underlätta för korrekt fördelning. Resultaten visar att intervjupersonerna skiljer sig åt i vilken typ av karriär de har och därmed hur man förhåller sig till dagens system. Ingen ställde sig dock positiv till att rapportera in spellistor med vad de spelat som dj’s, man lyfte även fram problemet med att ersätta musik som spelas men ännu inte släppts officiellt. Det framkom också att vi idag har tekniska möjligheter att övervaka vad som spelas genom användandet av akustiska fingeravtryck. Jag diskuterade att det är först när ett sådant system är på plats som vi kan se hur mycket fel dagens pengar fördelas.

The history of a gay and lesbian student community at Colby seems to point to the difficulty of visibility. For students who were able to find others like themselves, their group of lesbian and gay friends had to remain underground. For students who were grappling with their newly found, socially stigmatized sexuality, the experience was isolating if they did not know where to find others like themselves. This paper seeks to address the social forces that kept sexually variant students from expressing their sexual identities openly on campus. Part of this difficulty is attributable to the compulsory heterosexuality assumed by general American society at the time, manifested in the silence or outright hostility directed against homosexuals. Naturally, Colby students replicated this assumption. Some of the students we interviewed seemed to internalize compulsory heterosexuality, while it was forced upon others. Religion and psychology were two methods of enforcing heterosexuality that were relevant to the people we interviewed. Another significant obstacle to visibility was Colby's location and the nature of Colby's student body. Waterville, unlike more urban cities, did not have a history of gay life, and thus an established gay community or gay identity into which one could be socialized. Colby, as a small, homogeneous and isolated space, posed difficulties in establishing a gay community as the population to draw from was small and regulated.

As a result of urbanization, stormwater runoff flow rates and volumes are significantly increased due to increasing impervious land cover and the decreased availability of depression storage. Storage tanks are the basic devices to efficiently control the flow rate in drainage systems during wet weather. Presented in the paper conception of vacuum-driven detention tanks allows to increase the storage capacity by usage of space above the free surface water elevation at the inlet channel. Partial vacuum storage makes possible to gain cost savings by reduction of both the horizontal area of the detention tank and necessary depth of foundations. Simulation model of vacuum-driven storage tank has been developed to estimate potential profits of its application in urban drainage system. Although SWMM5 has no direct options for vacuum tanks an existing functions (i.e. control rules) have been used to reflect its operation phases. Rainfall data used in simulations were recorded at raingage in Czestochowa during years 2010÷2012 with time interval of 10minutes. Simulation results gives overview to practical operation and maintenance cost (energy demand) of vacuum driven storage tanks depending of the ratio: vacuum-driven volume to total storage capacity. The following conclusion can be drawn from this investigations: vacuum-driven storage tanks are characterized by uncomplicated construction and control systems, thus can be applied in newly developed as well as in the existing urban drainage systems. the application of vacuum in underground detention facilities makes possible to increase of the storage capacity of existing reservoirs by usage the space above the maximum depth. Possible increase of storage capacity can achieve even a few dozen percent at relatively low investment costs. vacuum driven storage tanks can be included in existing simulation software (i.e. SWMM) using options intended for pumping stations (including control and action rules ).

With the service life of water supply network (WSN) growth, the growing phenomenon of aging pipe network has become exceedingly serious. As urban water supply network is hidden underground asset, it is difficult for monitoring staff to make a direct classification towards the faults of pipe network by means of the modern detecting technology. In this paper, based on the basic property data (e.g. diameter, material, pressure, distance to pump, distance to tank, load, etc.) of water supply network, decision tree algorithm (C4.5) has been carried out to classify the specific situation of water supply pipeline. Part of the historical data was used to establish a decision tree classification model, and the remaining historical data was used to validate this established model. Adopting statistical methods were used to access the decision tree model including basic statistical method, Receiver Operating Characteristic (ROC) and Recall-Precision Curves (RPC). These methods has been successfully used to assess the accuracy of this established classification model of water pipe network. The purpose of classification model was to classify the specific condition of water pipe network. It is important to maintain the pipeline according to the classification results including asset unserviceable (AU), near perfect condition (NPC) and serious deterioration (SD). Finally, this research focused on pipe classification which plays a significant role in maintaining water supply networks in the future.

Tendo como inspiração o comix underground norte-americano, os quadrinhos alternativos brasileiros revelaram vários artistas de talento, muitos deles reconhecidos inclusive no exterior, e abordaram criticamente temas relevantes da realidade brasileira. O mercado editorial de Histórias em Quadrinhos no Brasil sempre esteve sujeito às intempéries políticas e econômicas, mas, ao longo das décadas de 1980 e 1990, com o processo de redemocratização e apesar da crise inflacionária, a produção alternativa de histórias em quadrinhos conquistou o público-leitor. Uma das editoras mais expressivas nesse contexto foi a Circo Editorial.

Este trabalho apresenta uma técnica de programação e controle da construção repetitiva denominada Linha de Balanço. E uma técnica, surgida na indústria fabril, onde a produção em série e em grande escala exige a organização dos métodos de produção e a racionalização do trabalho. O estudo, inicialmente, modela o trabalho de construção de unidades repetitivas, salientando os problemas de organização e administração destes empreendimentos. Num segundo momento, são apresentados os princípios teóricos da técnica com aplicações práticas da programação. Por fim, é analisada a metodologia usual de construção de conjuntos habitacionais através de um estudo de caso e, observadas as condições de aplicação da técnica da Linha de Balanço, não exigindo mudificações muito profundas na atual ação gerencial neste tipo de obra. As conclusões esboçadas confirmam as vantagens da técnica da Linha de Balanço para estes empreendimentos e propõem a sua utilização na organização do trabalho nos canteiros repetitivos.

A alface é a hortaliça folhosa mais consumida no mundo. É uma rica fonte de vitaminas e sais minerais que está presente no cardápio diário de grande parte da população brasileira. O nitrogênio é o nutriente mais importante para a produção de alface. O excesso ou falta desse nutriente acarretam queda na produção e problemas de qualidade nutritiva, por excesso de nitrato. A dose recomendada de nitrogênio varia muito entre autores e órgãos oficiais. Uma parte do nitrogênio aplicado ao solo é perdida por lixiviação, contaminando os mananciais de água. Devido a isso é recomendada a sua aplicação em diversas etapas, cujo número também é variável nas recomendações. Este trabalho teve por objetivo determinar qual a dose de nitrogênio e seu modo de aplicação que proporcionam a maior produção, sem comprometer a qualidade da alface do tipo “lisa”, nas condições de clima e solo do município de Eldorado do Sul, região da Grande Porto Alegre, RS. Foram realizados dois estudos, um testando doses crescentes de nitrogênio sob a forma de uréia e o outro testando o parcelamento em diversas épocas do cultivo. Foram avaliados a produção de matéria fresca e seca, os teores de nitrogênio, amônio e nitrato no tecido seco, a extração e a eficiência do uso do fertilizante. Os resultados revelam que a dose de 200 kgha-1 foi a que proporcionou maior rendimento da cultura e que a dose de 400 kgha-1 diminui a produção. A extração de nitrogênio foi da ordem de 35,7 kgha-1 e os teores de nitrogênio, amônio e nitrato foram respectivamente de 2,45%; 651,7 mg.kg-1 e 153,6 mg.kg-1. O teor de nitrato na alface foi muito menor do que o considerado prejudicial a saúde humana. A eficiência do uso do fertilizante foi de 12,06% quando aplicado 200 kgha-1 de N em 3 épocas. A eficiência do uso do adubo foi aumentada em 26,35% com o parcelamento em 4 doses iguais de 50 kg ha-1, aplicadas no plantio e aos 15, 30 e 45 dias após, sem alterar a qualidade visual e nutricional da alface.
