The paper offers a critical discussion of Simon Keller’s individuals view concerning the grounds of special obligations. It is argued that the view offers a misleading account of agent-relative reasons, which leads to problems with the arguments Keller offers for his view – the Kantian, consequentialist, particularistic argument. Furthermore, it argues that Keller’s argument from phenomenology suffers from not distinguishing between reasons for partiality and reasons from partiality. At best, it can be applied to reasons from partiality – reasons that agents have in virtue of standing in a relationship of partiality with some other person. Thus, the argument does not support the individuals view, as it is compatible with the other approaches Keller discusses. The paper suggests a modified relationship view to ground reasons of partiality.

BACKGROUND Listeria (L.) monocytogenes causes fatal infections in many species including ruminants and humans. In ruminants, rhombencephalitis is the most prevalent form of listeriosis. Using multilocus variable number tandem repeat analysis (MLVA) we recently showed that L. monocytogenes isolates from ruminant rhombencephalitis cases are distributed over three genetic complexes (designated A, B and C). However, the majority of rhombencephalitis strains and virtually all those isolated from cattle cluster in MLVA complex A, indicating that strains of this complex may have increased neurotropism and neurovirulence. The aim of this study was to investigate whether ruminant rhombencephalitis strains have an increased ability to propagate in the bovine hippocampal brain-slice model and can be discriminated from strains of other sources. For this study, forty-seven strains were selected and assayed on brain-slice cultures, a bovine macrophage cell line (BoMac) and a human colorectal adenocarcinoma cell line (Caco-2). They were isolated from ruminant rhombencephalitis cases (n = 21) and other sources including the environment, food, human neurolisteriosis cases and ruminant/human non-encephalitic infection cases (n = 26). RESULTS All but one L. monocytogenes strain replicated in brain slices, irrespectively of the source of the isolate or MLVA complex. The replication of strains from MLVA complex A was increased in hippocampal brain-slice cultures compared to complex C. Immunofluorescence revealed that microglia are the main target cells for L. monocytogenes and that strains from MLVA complex A caused larger infection foci than strains from MLVA complex C. Additionally, they caused larger plaques in BoMac cells, but not CaCo-2 cells. CONCLUSIONS Our brain slice model data shows that all L. monocytogenes strains should be considered potentially neurovirulent. Secondly, encephalitis strains cannot be conclusively discriminated from non-encephalitis strains with the bovine organotypic brain slice model. The data indicates that MLVA complex A strains are particularly adept at establishing encephalitis possibly by virtue of their higher resistance to antibacterial defense mechanisms in microglia cells, the main target of L. monocytogenes.

This article analyzes the Jakobsonian classification of aphasias. It aims to show on the one hand the non-linguistic character of this classification and on the other hand its asymmetry, in spite of the fact that its author had conceived his structural construction as symmetrical. The non-linguistic character of Jakobson’s formulation is due to the absence of any definition of language, this absence being the main characteristic of Jakobsonian linguistics: concerning the aphasia problem, the Jakobsonian formulation is linguistic solely by virtue of its object, aphasia, which is already considered as a linguistic concern because it belongs to the field of « language », but which is not defined as such (as linguistic). As for asymmetry, it demonstrates first the circularity of the Jakobsonian representation of language (the duality between structure and functioning), and secondly the non-linguistic character – in the Saussurean sense of the term – of the aphasia problem. Thus it appears that breaking (in the sense of Gaston Bachelard) with idiom is the prerequisite of a scientific apprehension of language, and therefore of any interdisciplinarity, this being one of Jakobson’s favorite topics but one that this linguist failed to render fruitful because he did not offer a real definition of language.

The Ca2+-binding protein calmodulin (CaM) is a key transducer of Ca2+ oscillations by virtue of its ability to bind Ca 2+ selectively and then interact specifically with a large number of downstream enzymes and proteins. It remains unclear whether Ca2+ -dependent signaling alone can activate the full range of Ca 2+/CaM regulated processes or whether other regulatory schemes in the cell exist that allow specific targeting of CaM to subsets of Ca 2+/CaM binding sites or regions of the cell. Here we investigate the possibility that alterations of the availability of CaM may serve as a potential cellular mechanism for regulating the activation of CaM-dependent targets. By utilizing sensitive optical techniques with high spatial and temporal resolution, we examine the intracellular dynamics of CaM signaling at a resolution previously unattainable. After optimizing and characterizing both the optical methods and fluorescently labeled probes for intracellular measurements, the diffusion of CaM in the cytoplasm of HEK293 cells was analyzed. It was discovered that the diffusion characteristics of CaM are similar to that of a comparably sized inert molecule. Independent manipulation of experimental parameters, including increases in total concentrations of CaM and intracellular Ca2+ levels, did not change the diffusion of CaM in the cytoplasm. However, changes in diffusion were seen when the concentration of Ca2+/CaM-binding targets was increased in conjunction with elevated Ca2+. This indicates that CaM is not normally limiting for the activation of Ca 2+/CaM-dependent enzymes in HEK293 cells but reveals that the ratio of CaM to CaM-dependent targets is a potential mechanism for changing CaM availability. Next we considered whether cellular compartmentalization may act to regulate concentrations of available Ca2+/CaM in hippocampal neurons. We discovered changes in diffusion parameters of CaM under elevated Ca2+ conditions in the soma, neurite and nucleus which suggest that either the composition of cytoplasm is different in these compartments and/or they are composed of unique families of CaM-binding proteins. Finally, we return to the HEK293 cell and for the first time directly show the intracellular binding of CaM and CaMKII, an important target for CaM critical for neuronal function and plasticity. Furthermore, we analyzed the complex binding stoichiometry of this molecular interaction in the basal, activated and autophosphorylated states of CaMKII and determined the impact of this binding on CaM availability in the cell. Overall these results demonstrate that regulation of CaM availability is a viable cellular mechanism for regulating the output of CaM-dependent processes and that this process is tuned to the specific functional needs of a particular cell type and subcellular compartment. ^

In 2008, 132 law enforcement officers were killed in the line of duty in The United States. Additionally, some have explored both the public health implications of interactions with law enforcement as well as the potential benefits of the use of law enforcement officers as public health and emergency healthcare providers. By virtue of these novel analyses and techniques, professional medical direction of the emerging specialty of law enforcement medicine is needed. This paper, an analysis of law enforcement medical direction through a look at the Dallas Police Medical Direction Program, seeks to examine origins of law enforcement medicine through a comprehensive literature review, as well as begin to define to core competencies of law enforcement medical direction. ^ The unique intersection of public health, medicine and law enforcement, and the subsequent specialty that is developing to manage this interface, is in its relative infancy. An analysis of this nature is in order to begin to lay down the foundations necessary for future study and improvements in the field. ^

Among Mexican Americans, the second largest minority group in the United States, the prevalence of gallbladder disease is markedly elevated. Previous data from both genetic admixture and family studies indicate that there is a genetic component to the occurrence of gallbladder disease in Mexican Americans. However, prior to this thesis no formal genetic analysis of gallbladder disease had been carried out nor had any contributing genes been identified.^ The results of complex segregation analysis in a sample of 232 Mexican American pedigrees documented the existence of a major gene having two alleles with age- and gender-specific effects influencing the occurrence of gallbladder disease. The estimated frequency of the allele increasing susceptibility was 0.39. The lifetime probabilities that an individual will be affected by gallbladder disease were 1.0, 0.54, and 0.00 for females of genotypes "AA", "Aa", and "aa", respectively, and 0.68, 0.30, and 0.00 for males, respectively. This analysis provided the first conclusive evidence for the existence of a common single gene having a large effect on the occurrence of gallbladder disease.^ Human cholesterol 7$\alpha$-hydroxylase is the rate-limiting enzyme in bile acid synthesis. The results of an association study in both a random sample and a matched case/control sample showed that there is a significant association between cholesterol 7$\alpha$-hydroxylase gene variation and the occurrence of gallbladder disease in Mexican Americans males but not in females. These data have implicated a specific gene, 7$\alpha$-hydroxylase, in the etiology of gallbladder disease in this population.^ Finally, I asked whether the inferred major gene from complex segregation analysis is genetically linked to the cholesterol 7$\alpha$-hydroxylase gene. Three pedigrees predicted to be informative for linkage analysis by virtue of supporting the major gene hypothesis and having parents with informative genotypes and multiple offspring were selected for this linkage analysis. In each of these pedigrees, the recombination fractions maximized at 0 with a positive, albeit low, LOD score. The results of this linkage analysis provide preliminary and suggestive evidence that the cholesterol 7$\alpha$-hydroxylase gene and the inferred gallbladder disease susceptibility gene are genetically linked. ^

Background. Cancer cachexia is a common syndrome complex in cancer, occurring in nearly 80% of patients with advanced cancer and responsible for at least 20% of all cancer deaths. Cachexia is due to increased resting energy expenditure, increased production of inflammatory mediators, and changes in lipid and protein metabolism. Non-steroidal anti-inflammatory drugs (NSAIDs), by virtue of their anti-inflammatory properties, are possibly protective against cancer-related cachexia. Since cachexia is also associated with increased hospitalizations, this outcome may also show improvement with NSAID exposure. ^ Design. In this retrospective study, computerized records from 700 non-small cell lung cancer patients (NSCLC) were reviewed, and 487 (69.57%) were included in the final analyses. Exclusion criteria were severe chronic obstructive pulmonary disease, significant peripheral edema, class III or IV congestive heart failure, liver failure, other reasons for weight loss, or use of research or anabolic medications. Information on medication history, body weight and hospitalizations was collected from one year pre-diagnosis until three years post-diagnosis. Exposure to NSAIDs was defined if a patient had a history of being treated with NSAIDs for at least 50% of any given year in the observation period. We used t-test and chi-square tests for statistical analyses. ^ Results. Neither the proportion of patients with cachexia (p=0.27) nor the number of hospitalizations (p=0.74) differed among those with a history of NSAID use (n=92) and those without (n=395). ^ Conclusions. In this study, NSAID exposure was not significantly associated with weight loss or hospital admissions in patients with NSCLC. Further studies may be needed to confirm these observations.^

To identify more mutations that can affect the early development of Myxococcus xanthus, the synthetic transposon TnT41 was designed and constructed. By virtue of its special features, it can greatly facilitate the processes of mutation screening/selection, mapping, cloning and DNA sequencing. In addition, it allows for the systematic discovery of genes in regulatory hierarchies using their target promoters. In this study, the minimal regulatory region of the early developmentally regulated gene 4521 was used as a reporter in the TnT41 mutagenesis. Both positive (P) mutations and negative (N) mutations were isolated based on their effects on 4521 expression.^ Four of these mutations, i.e. N1, N2, P52 and P54 were analyzed in detail. Mutations N1 and N2 are insertion mutations in a gene designated sasB. The sasB gene is also identified in this study by genetic and molecular analysis of five UV-generated 4521 suppressor mutations. The sasB gene encodes a protein without meaningful homology in the databases. The sasB gene negatively regulates 4521 expression possibly through the SasS-SasR two component system. A wild-type sasB gene is required for normal M. xanthus fruiting body formation and sporulation.^ Cloning and sequencing analysis of the P52 mutation led to the identification of an operon that encodes the M. xanthus high-affinity branched-chain amino acid transporter system. This liv operon consists of five genes designated livK, livH, livM, livC, and livF, respectively. The Liv proteins are highly similar to their counterparts from other bacteria in both amino acid sequences, functional motifs and predicted secondary structures. This system is required for development since liv null mutations cause abnormality in fruiting body formation and a 100-fold decrease in sporulation efficiency.^ Mutation P54 is a TnT41 insertion in the sscM gene of the ssc chemotaxis system, which has been independently identified by Dr. Shi's lab. The sscM gene encodes a MCP (methyl-accepting chemotaxis protein) homologue. The SscM protein is predicted to contain two transmembrane domains, a signaling domain and at least one putative methylation site. Null mutations of this gene abolish the aggregation of starving cells at a very early stage, though the sporulation levels of the mutant can reach 10% that of wild-type cells. ^

En este nuevo número de los Cuadernos del CILHA que el lector tiene en sus manos se ha reunido un conjunto de trabajos que, por la diversa procedencia institucional de cada uno de sus autores, nos permite asomarnos panorámicamente a algunas líneas de trabajo que en la actualidad se desarrollan en torno a la literatura hispanoamericana. Así por ejemplo resultan interesantes los trabajos de Mariana Catalin “La proliferación del yo…", Mariana Libertad Suárez “Perdón o condena: discurso y subjetividad…" y Betina Keizman “Entre el testimonio y la autobiografía…", que se ocupan de la subjetividad hispanoamericana, a través de una abordaje que va de la problemática genérica a la lectura crítica de los textos narrativos o autobiográficos. Se vincula con esta línea mencionada, el artículo de Ramiro Zó que aborda en “Funciones de la novela sentimental…", la cuestión de la novela sentimental durante el siglo XIX. Cabe indicar al respecto que el trabajo de Zó puede situarse en la categoría de los pioneros, ya que no abundan las investigaciones sobre esta temática. Pablo Martínez Gramuglia se ocupa, por su lado, de la obra de Leopoldo Marechal destacando las relaciones entre el mito y la política. Los trabajos restantes de la Sección Misceláneas se refieren a Arturo Uslar Pietri, de quien en el 2006 se cumplió el primer centenario de su nacimiento. María Antonia Zandanel (“ Pizarrón. Una escritura…") se encarga de analizar la escritura ensayística del notable escritor venezolano y Laura Febres (“Arturo Uslar Pietri el artífice de la…") de la narrativa. Finalmente, se publica el texto de Mario Cámara “Algunos elogios posibles para Glauco Mattoso". La decisión de ir sumando investigaciones sobre la literatura brasilera resulta para nosotros muy valioso, de manera que esperamos incrementar el número de artículos sobre la literatura y la cultura del Brasil. Reglón aparte merece el dossier “El ensayo latinoamericano". Marcos Olalla ha sido el responsable de reunir estos textos. No vamos a sobreabundar en lo ya escrito por él en la excelente introducción al dossier. Nos importa subrayar únicamente el interés que puede tener este esfuerzo en virtud de que los artículos concretan notables aportes a la problemática del género como también actualizan la lectura de algunos textos a esta altura canónicos del ensayo.

Algunos autores de la filosofía contemporánea ha criticado a la antropología medieval aduciendo que ella ha sustentando en una visión sustancialista que no da razón de la riqueza de la realidad personal (como es el caso de Zubiri que, con sus palabras, propone que la persona es sustantividad y supra-estante). Sin embargo, en la respuesta de Tomás de Aquino al problema planteado por Pedro Lombardo en las Sententiae de qué es y cómo está la caridad en el ser humano se plantea la posibilidad de que en el hombre haya una virtud o hábito, insertado en la estructura psicológica del hombre, que tenga por origen un don creado de carácter divino, que perfecciona el alma humana y haciéndole capaz de un fin sobrenatural, pero que responde a la dinámica de facultades, hábitos y actos que estructura la realidad del acto verdaderamente humano. La respuesta tomista que justifica la existencia de una virtud como la caridad revela un planteamiento antropológico con una noción de persona de gran riqueza.

Resumen: Augusto llegó al poder por ser hijo adoptivo de César. Hasta recibir el título de Augusto en el año 27 a. C., fue conocido como G. Iulius Caesar Diui filius. Ser heredero de César tenía que ser algo más que recibir su nombre y sus poderes: también sus cualidades personales debían recordar al Dictador. Por eso, durante la mitad del triunvirato, Augusto intentó emular a su padre adoptivo también en sus cualidades como amante. Su tumultuosa vida afectiva y conyugal, en efecto, se concentra entre los años 44 y 35 a. C., con varios matrimonios o esponsales, y el nacimiento de su única hija. De esta época proceden la mayoría de las historias que –ciento cincuenta años después- contaría Suetonio a partir de una fuente cercana a Marco Antonio. A partir de la sacrosantidad promulgada para Livia en el año 35 a. C., sin embargo, comienza una nueva etapa en la imagen moral de Augusto, centrada en la virtud y la monogamia: una etapa que le llevaría años después a desterrar a su propia hija y, después, a su nieta, por adulterio. De esta doble imagen de Augusto, la primera debida a la emulación de su padre adoptivo, y la segunda debida a una necesidad de justificación ideológica de su régimen frente al de su enemigo Marco Antonio, proceden las noticias aparentemente contradictorias sobre los “vicios privados y públicas virtudes” de Gayo Julio César Augusto.

En la década de 1990 se produjo en Argentina la ruptura de la matriz estadocéntrica que regía la relación Estado-sociedad. Ese proceso, denominado Reforma del Estado, consistió en la aplicación de políticas neoliberales elaboradas por el Consenso de Washington. Para que esa decisión política lograra legitimación social fue muy importante el rol desempeñado por los medios de comunicación pertenecientes al bloque hegemónico, los que, en virtud de su intencionalidad editorial, contribuyeron a instalar la idea del "Estado demoníaco", y la imprescindible necesidad de reducirlo y transformarlo para dotarlo de eficiencia y modernizar a nuestro país.

En diversos pasajes de su Comentario a las Sentencias Buenaventura aborda el tema de las faltas veniales incorporando, en un marco teológico, una acepción del concepto de tolerancia heredada de los victorinos, el de ‘cohibitio’, para indicar un aspecto de la permisión que, por oposición al concepto de ‘prohibitio’, señalaría la posibilidad de tolerar una falta venial con el fin de evitar un mal mayor. Este trabajo presenta un seguimiento del concepto de cohibitio en los usos previos a Buenaventura, y señala en este último la vinculación de la tolerancia con el resto de las virtudes mediante la forma común o ‘forma directiva’: la caridad, que otorga a las virtudes un complemento en su ordenación natural al Bien como fin último. En este contexto, se muestra asimismo cómo la estructura conceptual de la teoría de las virtudes en Buenaventura sigue un esquema filosófico que encuentra su fundamentación metafísica en la unidad de los trascendentales.

La preocupación por definir una pauta de comportamiento éticamente correcto una vez que se cuestiona la trascendencia es central en todo el pensamiento ilustrado francés. El objetivo del presente trabajo es reconstruir algunas de las ideas sobre la moral expuestas en sus discursos por el líder jacobino, Robespierre, y demostrar de qué modo son desarmadas y puestas en ridículo en la literatura del Marqués de Sade mediante una particular concepción de la naturaleza. En ella, como veremos, se condensa una doble crítica: por un lado, al ideal rousseauniano que supone que el hombre es originariamente bueno y depravado por las instituciones y, por el otro, a la posición de Robespierre que sostiene que una República puede erigirse sobre la virtud. La elaboración sadiana del concepto de “naturaleza", expresada fundamentalmente en La philosophie dans le boudoir a través de la voz de Dolmancé, se convierte en la herramienta fundamental para socavar los presupuestos iluministas sobre los que se asienta la moralidad revolucionaria.
