A medida dos diâmetros longitudinal e transverso de uma lesão ulcerada na leishmaniose tegumentar não reflete a profundidade, a qual também tem importância na avaliação da cura. A proposta deste estudo foi avaliar o volume da úlcera leishmaniõtica, através da moldagem seriada, antes e após o tratamento. A moldagem da cavidade, apôs assepsia local com água e sabão, foi feita com material plástico gelatinoso ã base de sílica, alginato de potássio, sulfato de cálcio e óxido de magnésio, comercializado sob o nome de jeltrate® (Dentsply Laboratory). Dissolveu-se 9,5g de jeltrate® em 20ml de água e aplicou-se o gel sobre a úlcera, o qual se solidificou em cinco minutos. Este molde foi então preenchido com acrílico autopolimerizante e o seu volume mensurado através do peso, usando-se balança analítica (Técnica 1) ou através do deslocamento da água, aplicando o princípio de Arquimedes (Técnica 2). Mostramos os dados de um estudo i-ealizado numa área de transmissão de Leishmania (Viannia) braziliensis onde 20 pacientes foram tratados com três diferentes esquemas: sete pacientes receberam isotianato depentamidina, sete receberam sulfato de aminosidine e seis antimoniato de meglumina. Os resultados mostram em todos os pacientes redução uniforme do volume das úlceras melhor avaliada por qualquer uvta das duas técnicas utilizadas. Em relação aos esquemas terapêuticos houve diferença estatística significante entre os três esquemas usando o teste T de student, o qual mostrou que o sulfato de aminosidine produziu maior redução de volume que os outros compostos. Moldagens seriadas são registros permanentes e refletem melhor a cura clínica. Concluímos que a medida da lesão ulcerada é útil na avaliação terapêutica como procedimento prático, barato e que pode ser utilizado em estudos de campo.

Due to the importance and wide applications of the DNA analysis, there is a need to make genetic analysis more available and more affordable. As such, the aim of this PhD thesis is to optimize a colorimetric DNA biosensor based on gold nanoprobes developed in CEMOP by reducing its price and the needed volume of solution without compromising the device sensitivity and reliability, towards the point of care use. Firstly, the price of the biosensor was decreased by replacing the silicon photodetector by a low cost, solution processed TiO2 photodetector. To further reduce the photodetector price, a novel fabrication method was developed: a cost-effective inkjet printing technology that enabled to increase TiO2 surface area. Secondly, the DNA biosensor was optimized by means of microfluidics that offer advantages of miniaturization, much lower sample/reagents consumption, enhanced system performance and functionality by integrating different components. In the developed microfluidic platform, the optical path length was extended by detecting along the channel and the light was transmitted by optical fibres enabling to guide the light very close to the analysed solution. Microfluidic chip of high aspect ratio (~13), smooth and nearly vertical sidewalls was fabricated in PDMS using a SU-8 mould for patterning. The platform coupled to the gold nanoprobe assay enabled detection of Mycobacterium tuberculosis using 3 8l on DNA solution, i.e. 20 times less than in the previous state-of-the-art. Subsequently, the bio-microfluidic platform was optimized in terms of cost, electrical signal processing and sensitivity to colour variation, yielding 160% improvement of colorimetric AuNPs analysis. Planar microlenses were incorporated to converge light into the sample and then to the output fibre core increasing 6 times the signal-to-losses ratio. The optimized platform enabled detection of single nucleotide polymorphism related with obesity risk (FTO) using target DNA concentration below the limit of detection of the conventionally used microplate reader (i.e. 15 ng/μl) with 10 times lower solution volume (3 μl). The combination of the unique optical properties of gold nanoprobes with microfluidic platform resulted in sensitive and accurate sensor for single nucleotide polymorphism detection operating using small volumes of solutions and without the need for substrate functionalization or sophisticated instrumentation. Simultaneously, to enable on chip reagents mixing, a PDMS micromixer was developed and optimized for the highest efficiency, low pressure drop and short mixing length. The optimized device shows 80% of mixing efficiency at Re = 0.1 in 2.5 mm long mixer with the pressure drop of 6 Pa, satisfying requirements for the application in the microfluidic platform for DNA analysis.

Gene therapy presents an ideal strategy for the treatment of genetic as well as acquired diseases, such as cancer and typically involves the insertion of a functioning gene into cells to correct a cellular dysfunction or to provide a new cellular function. Gene delivery vectors are based in two models: viral and non-viral. Viral vectors have high transfection efficiency but their major barrier is immunogenicity. Since the non-viral vectors have no immunogenicity, these have been widely studied. Gold nanoparticles have been proposed as optimal delivery systems of genetic material, due their small size, high surface-to-volume ratio and the ability to be functionalized with multiple molecules. In the present work, an AuNP-based formulation was developed to deliver a plasmid in a colorectal cancer cell line, containing as reporter gene the gene encoding to EGFP. The delivery system resulted from the functionalization of 14 nm AuNP with a PEG layer (4300114 PEG chains/AuNP), which increases stability and biocompatibility of AuNPs; quaternary ammonium groups which provide positive charges that allow electrostatic binding of plasmid, which is considered the therapeutic agent to be transported into cells. The system developed was characterized by UV-vis spectroscopy, DLS, TEM and by electrophoretic mobility, yielding a formulation with 113.5 nm.Transfection efficiency of the formulation developed was evaluated through PCR and through EGFP expression by fluorescence microscopy and fluorescence spectroscopy. The internalization was observed 3h post transfection; however a low level of EGFP expression was achieved. After 24h of incubation, EGFP expression increases just 3 times compared to non-transfected cells. The commercial system (Lipofectamine) expressed EGFP 5 times more than the system developed AuNP@PEG@R4N+@pEGFP. This difference could be related to lower translocation to the nucleus.

Spin-lattice Relaxation, self-Diffusion coefficients and Residual Dipolar Couplings (RDC’s) are the basis of well established Nuclear Magnetic Resonance techniques for the physicochemical study of small molecules (typically organic compounds and natural products with MW < 1000 Da), as they proved to be a powerful and complementary source of information about structural dynamic processes in solution. The work developed in this thesis consists in the application of the earlier-mentioned NMR techniques to explore, analyze and systematize patterns of the molecular dynamic behavior of selected small molecules in particular experimental conditions. Two systems were chosen to investigate molecular dynamic behavior by these techniques: the dynamics of ion-pair formation and ion interaction in ionic liquids (IL) and the dynamics of molecular reorientation when molecules are placed in oriented phases (alignment media). The application of NMR spin-lattice relaxation and self-diffusion measurements was applied to study the rotational and translational molecular dynamics of the IL: 1-butyl-3-methylimidazolium tetrafluoroborate [BMIM][BF4]. The study of the cation-anion dynamics in neat and IL-water mixtures was systematically investigated by a combination of multinuclear NMR relaxation techniques with diffusion data (using by H1, C13 and F19 NMR spectroscopy). Spin-lattice relaxation time (T1), self-diffusion coefficients and nuclear Overhauser effect experiments were combined to determine the conditions that favor the formation of long lived [BMIM][BF4] ion-pairs in water. For this purpose and using the self-diffusion coefficients of cation and anion as a probe, different IL-water compositions were screened (from neat IL to infinite dilution) to find the conditions where both cation and anion present equal diffusion coefficients (8% water fraction at 25 ºC). This condition as well as the neat IL and the infinite dilution were then further studied by 13C NMR relaxation in order to determine correlation times (c) for the molecular reorientational motion using a mathematical iterative procedure and experimental data obtained in a temperature range between 273 and 353 K. The behavior of self-diffusion and relaxation data obtained in our experiments point at the combining parameters of molar fraction 8 % and temperature 298 K as the most favorable condition for the formation of long lived ion-pairs. When molecules are subjected to soft anisotropic motion by being placed in some special media, Residual Dipolar Couplings (RDCs), can be measured, because of the partial alignment induced by this media. RDCs are emerging as a powerful routine tool employed in conformational analysis, as it complements and even outperforms the approaches based on the classical NMR NOE or J3 couplings. In this work, three different alignment media have been characterized and evaluated in terms of integrity using 2H and 1H 1D-NMR spectroscopy, namely the stretched and compressed gel PMMA, and the lyotropic liquid crystals CpCl/n-hexanol/brine and cromolyn/water. The influence that different media and degrees of alignment have on the dynamic properties of several molecules was explored. Different sized sugars were used and their self-diffusion was determined as well as conformation features using RDCs. The results obtained indicate that no influence is felt by the small molecules diffusion and conformational features studied within the alignment degree range studied, which was the 3, 5 and 6 % CpCl/n-hexanol/brine for diffusion, and 5 and 7.5 % CpCl/n-hexanol/brine for conformation. It was also possible to determine that the small molecules diffusion verified in the alignment media presented close values to the ones observed in water, reinforcing the idea of no conditioning of molecular properties in such media.

The Corporate world is becoming more and more competitive. This leads organisations to adapt to this reality, by adopting more efficient processes, which result in a decrease in cost as well as an increase of product quality. One of these processes consists in making proposals to clients, which necessarily include a cost estimation of the project. This estimation is the main focus of this project. In particular, one of the goals is to evaluate which estimation models fit the Altran Portugal software factory the most, the organization where the fieldwork of this thesis will be carried out. There is no broad agreement about which is the type of estimation model more suitable to be used in software projects. Concerning contexts where there is plenty of objective information available to be used as input to an estimation model, model-based methods usually yield better results than the expert judgment. However, what happens more frequently is not having this volume and quality of information, which has a negative impact in the model-based methods performance, favouring the usage of expert judgement. In practice, most organisations use expert judgment, making themselves dependent on the expert. A common problem found is that the performance of the expert’s estimation depends on his previous experience with identical projects. This means that when new types of projects arrive, the estimation will have an unpredictable accuracy. Moreover, different experts will make different estimates, based on their individual experience. As a result, the company will not directly attain a continuous growing knowledge about how the estimate should be carried. Estimation models depend on the input information collected from previous projects, the size of the project database and the resources available. Altran currently does not store the input information from previous projects in a systematic way. It has a small project database and a team of experts. Our work is targeted to companies that operate in similar contexts. We start by gathering information from the organisation in order to identify which estimation approaches can be applied considering the organization’s context. A gap analysis is used to understand what type of information the company would have to collect so that other approaches would become available. Based on our assessment, in our opinion, expert judgment is the most adequate approach for Altran Portugal, in the current context. We analysed past development and evolution projects from Altran Portugal and assessed their estimates. This resulted in the identification of common estimation deviations, errors, and patterns, which lead to the proposal of metrics to help estimators produce estimates leveraging past projects quantitative and qualitative information in a convenient way. This dissertation aims to contribute to more realistic estimates, by identifying shortcomings in the current estimation process and supporting the self-improvement of the process, by gathering as much relevant information as possible from each finished project.

O adequado tratamento da água residual revela-se de extrema importância na preservação dos recursos naturais e da qualidade de vida. Esse tratamento é caracterizado pela produção de subprodutos que incluem resíduos removidos da água residual e lama produzida no tratamento da mesma. Ao longo do tempo, as directrizes europeias têm vindo a ser cada vez mais restritivas quanto aos valores limite que determinam a qualidade de emissão da água residual ou de deposição da lama, por forma a minimizar os impactes negativos no local de recepção dos mesmos e redução dos custos de transporte, no caso da lama, pelo que a ETAR deve ser planeada de forma a garantir o cumprimento da legislação em vigor, de acordo com a qualidade da água residual afluente à estação. O tratamento na estação divide-se em linha de fase líquida e sólida. No tratamento convencional da fase sólida, a lama é tratada segundo a sequência de etapas espessamento, estabilização e desidratação. Nesta dissertação será dada especial relevância à linha de fase sólida, com o intuito de apresentar, caracterizar e comparar as tecnologias (além do tratamento convencional) que se encontram em fase experimental ou já implantadas em ETAR e têm como objectivo a redução do volume da lama produzida. Será desenvolvida uma aplicação informática de apoio à decisão quanto à tecnologia indicada, de acordo com as variáveis seleccionadas pelo utilizador. As tecnologias dividem-se em: processos biológicos, hidrólise, oxidação avançada, química, térmica e mecânica. Nos biológicos a redução do volume da lama pode ser garantida pela predação dos microrganismos, pela manutenção/inibição do metabolismo e pela alteração/introdução de um reactor biológico na estação. A hidrólise é optimizada após doseamento de enzimas ou por aumento da temperatura. A oxidação avançada é caracterizada pelas tecnologias de ozonização, processo de Fenton, oxidação por ar húmido e oxidação electroquímica. A tecnologia química depende da adição de reagente ácido, alcalino ou outro. A tecnologia mecânica abrange a sonificação, o moinho de rotação, o reactor de homogeneização, a aplicação de campo eléctrico, a aplicação de radiação gama, recurso a técnicas centrífugas e aplicação de pressão na lama. A elevação da temperatura de reacção por meio convencional, por aplicação de radiação de micro-ondas ou garantindo o ciclo de congelamento/aquecimento pertencem à tecnologia térmica. Estas tecnologias podem ser aplicadas numa fase anterior ou posterior da estabilização da lama por forma a melhorar a biodegradabilidade dos compostos orgânicos e reduzir o número de microrganismos viáveis. As tecnologias actuam por meio de libertação de energia, radiação ou pressão no sentido de garantir os objectivos descritos e podem ser aplicadas individualmente ou combinadas (sistema híbrido). Assim o consumo energético, o incremento de matéria orgânica e a capacidade de redução da quantidade de lama são factores que devem ser considerados na escolha da melhor tecnologia que se adequa a cada ETAR.

Programmes supporting micro and small enterprises in developing countries have been showing that capital is not enough to allow business success: survival and growth. Literature does not provide comprehensive and practical tool to support business development in this context, but allowed the collection of forty-nine success variables that were studied in a sample of successful and unsuccessful businesses in the Island of Mozambique to discover what were the key factors affecting those businesses’ performance. Empirical data gave the insights for the development of a model to screen and improve business potential of micro and small enterprises in this context.
