Crotamine is one of four major components of the venom of the South American rattlesnake Crotalus durissus terrificus. Similar to its counterparts in the family of the myotoxins, it induces myonecrosis of skeletal muscle cells. This paper describes a new NMR structure determination of crotamine in aqueous solution at pH 5.8 and 20 degrees C, using standard homonuclear (1)H NMR spectroscopy at 900 MHz and the automated structure calculation software ATNOS/CANDID/DYANA. The automatic NOESY spectral analysis included the identification of a most likely combination of the six cysteines into three disulfide bonds, i.e. Cys4-Cys36, Cys11-Cys30 and Cys18-Cys37; thereby a generally applicable new computational protocol is introduced to determine unknown disulfide bond connectivities in globular proteins. A previous NMR structure determination was thus confirmed and the structure refined. Crotamine contains an alpha-helix with residues 1-7 and a two-stranded anti-parallel beta-sheet with residues 9-13 and 34-38 as the only regular secondary structures. These are connected with each other and the remainder of the polypeptide chain by the three disulfide bonds, which also form part of a central hydrophobic core. A single conformation was observed, with Pro13 and Pro21 in the trans and Pro20 in the cis-form. The global fold and the cysteine-pairing pattern of crotamine are similar to the beta-defensin fold, although the two proteins have low sequence homology, and display different biological activities. Two common South American species of lynx spiders, Peucetia rubrolineata and P. flava (Oxyopidae), were surveyed on three localities in southeastern Brazil to determine plant choice. Both species were found to be associated with plants bearing glandular trichomes. A literature review and complementary data show that ten Peucetia species are associated with up to 55 plant species bearing glandular trichomes in at least 20 distinct vegetation types (phytophysiognomies) in more than 36 localities in the Neotropical, Neartic, Afrotropical, and Paleartic regions. The main plant families used by the spiders were Solanaceae, Asteraceae, and Melastomataceae. The specialization of the Peucetia species for plants bearing glandular trichomes may have evolved because insects adhered to these sticky structures may be used as prey by the spiders.

Microfacies analysis of marine carbonates cored by Petrobras well 1-SPS-6 in the offshore Santos Basin (southeastern Brazil) has revealed a remarkable fossil assemblage of calpionellids (colomiellids), favusellids, hedbergellids, globigerinelloidids, buliminids, radiolarians, inoceramid prisms, roveacrinids, and saccocomids(?) preserved in lower Albian calcimudstones-wackestones of the lower part of the Guaruja Formation. This assemblage represents an allochtonous accumulation in a deep neritic to shallow bathyal hypoxic environment. Besides 'saccocomid-like' sections, the only determinable sections of roveacrinids are thecal plates of Poecilocrinus dispandus elongatus Peck, 1943. This species was previously only known from the Weno Formation of Texas. The Brazilian material extends its records farther south from at least the lower Albian, which then represents the earliest occurrence of this peculiar family in the South Atlantic region. Taking into account their Albian global distribution and the location of their oldest representative (Hauterivian near Alicante, Spain), the Roveacrinidae dispersed westward throughout all of Cretaceous Tethys. The Tethyan origin of Roveacrinidae is further evidence that, during late Aptian-Albian times, the northern South Atlantic (north of the Walvis-SBo Paulo Ridge) was supplied by a Tethyan water mass. To address understanding the organization of the 5S rRNA multigene family in the fish genome, the nucleotide sequence and organization array of 5S rDNA were investigated in the genus Leporinus, a representative freshwater fish group of South American fauna. PCR, subgenomic library screening, genomic blotting, fluorescence in situ hybridization, and DNA sequencing were employed in this study. Two arrays of 5S rDNA were identified for all species investigated, one consisting of monomeric repeat units of around 200 bp and another one with monomers of 900 bp. These 5S rDNA arrays were characterized by distinct NTS sequences (designated NTS-I and NTS-II for the 200- and 900-bp monomers, respectively); however, their coding sequences were nearly identical. The 5S rRNA genes were clustered in two chromosome loci, a major one corresponding to the NTS-I sites and a minor one corresponding to the NTS-II sites. The NTS-I sequence was variable among Leporinus spp., whereas the NTS-II was conserved among them and even in the related genus Schizodon. The distinct 5S rDNA arrays might characterize two 5S rRNA gene subfamilies that have been evolving independently in the genome.

The habitat of the mycelial saprobic form of Paracoccidio ides brasiliensis, which produces the infectious propagula, has not been determined and has proven difficult for mycologists to describe. The fungus has been rarely isolated from the environment, the disease has a prolonged latency period and no outbreaks have been reported. These facts have precluded the adoption of preventive measures to avoid infection. The confirmation of natural infections in nine-banded armadillos (Dasypus novemcinctus) with P. brasiliensis, in high frequency and wide geographic distribution, has opened new avenues for the study and understanding of its ecology. Armadillos belong to the order Xenarthra, which has existed in South America ever since the Paleocene Era (65 million years ago), when the South American subcontinent was still a detached land, before the consolidation of what is now known as the American continent. on the other hand, strong molecular evidence suggests that P. brasiliensis and other dimorphic pathogenic fungi - such as Blastomyces dermatitidis, Coccidioides immitis and Histoplasma capsulatum - belong to the family Onygenaceae sensu Into (order Onygenales, Ascomycota), which appeared around 150 million years ago.P. brasiliensis ecology and relation to its human host are probably linked to the fungal evolutionary past, especially its long coexistence with and adaptation to animal hosts other than Homo sapiens, of earlier origin. Instead of being a blind alley, the meaning of parasitism for dimorphic pathogenic fungi should be considered as an open two-way avenue, in which the fungus may return to the environment, therefore contributing to preserve its teleomorphic (sexual) and anamorphic (asexual) forms in a defined and protected natural habitat. Haemoglobin, the 'honorary enzyme' [Brunori (1999), Trends Biochem. Sci. 24, 158-161], constitutes a prime prototype for allosteric models. Here, the crystallization and preliminary X-ray analysis of haemoglobin I from the South American fish Brycon cephalus are reported. X-ray diffraction data have been collected to 2.5 Angstrom resolution using synchrotron radiation (LNLS). Crystals were determined to belong to the space group P6(1)22 and preliminary structural analysis revealed the presence of one dimer (alpha beta) in the asymmetric unit. The structure was determined using standard molecular-replacement techniques.

The Vazante Group, located in the northwestern part of Minas Gerais, hosts the most important zinc mine in Brazil, the Vazante Mine, which represents a major known example of a hypogene nonsulfide zinc deposit. The main zinc ore is represented by willemite and differs substantially from other deposits of the Vazante-Paracatu region, which are sulfide-dominated zinc-lead ore. The age of the Vazante Group and the hosted mineralization is disputable. Metamorphosed mafic dikes (metabasites) that cut the metasedimentary sequence and are affected by hydrothermal processes recently were found and may shed light on the geochronology of this important geological unit. Zircon crystals recovered from the metabasites are xenocrystic grains that yield U-Pb conventional ages ranging from 2.1 to 2.4 Ga, so the basement of the Vazante Group is Paleoproterozoic or has metasedinientary rocks whose source area was Paleoproterozoic. Pb isotopes determined for titanite separated from the metabasites have common, nonradiogenic Ph compositions, which prevents determination of their crystallization age. However, the Pb signatures observed for the titanite crystals are in agreement with those determined for galena from the carbonate-hosted Zn-Pb deposits hosted by the Vazante Group, including galena from minor sulfide ore bodies of the Vazante deposit. These similarities suggest that the metalliferous fluids that affected the metabasites may have been those responsible for galena formation, which could imply a similar lead source for both nonsulfide and sulfide zinc deposits in the Vazante-Paracatu district. This common source could be related to deep-seated, basin-derived, metalliferous fluids associated with a long-lived hydrothermal system related to diagenesis and deformation of the Vazante Group during the Neoproterozoic. Background: the paca is a South American rodent with potential as a commercial food animal. We examined paca placenta as part of a wider effort to understand the reproductive biology of this species.Methods: Thirteen specimens between midgestation and term of pregnancy were studied by light and transmission electron microscopy.Results: the placenta is divided into several lobes separated by interlobular trophoblast. Maternal arterial channels and fetal veins are found at the centre of each lobe. In the labyrinth, maternal blood flows through trophoblast-lined lacunae in close proximity to the fetal capillaries. The interhaemal barrier is of the haemomonochorial type with a single layer of syncytiotrophoblast. Caveolae occur in the apical membrane of the syncytiotrophoblast and recesses in the basal membrane, but there is no evidence of transtrophoblastic channels. The interlobular areas consist of cords of syncytiotrophoblast defining maternal blood channels that drain the labyrinth. Yolk sac endoderm covers much of the fetal surface of the placenta. The subplacenta comprises cytotrophoblast and syncytiotrophoblast. There are dilated intercellular spaces between the cytotrophoblasts and lacunae lined by syncytiotrophoblast. In the junctional zone between subplacenta and decidua, there are nests of multinucleated giant cells with vacuolated cytoplasm. The entire placenta rests on a pedicle of maternal tissue. An inverted yolk sac placenta is also present. The presence of small vesicles and tubules in the apical membrane of the yolk sac endoderm and larger vesicles in the supranuclear region suggest that the yolk sac placenta participates in maternal-fetal transfer of protein.Conclusion: the paca placenta closely resembles that of other hystricomorph rodents. The lobulated structure allows for a larger exchange area and the development of precocial young.

Crotoxin (CTX). a neurotoxin isolated from the venom of the South American rattlesnake Crotalus durissus terrificus. induces analgesia. In this study, we evaluated the antinociceptive effect of CTX in a model of neuropathic pain induced by rat sciatic nerve transection. Hyperalgesia was detected 2 h after nerve transection and persisted for 64 days. Immersion of proximal and distal nerve stumps in CTX solution (0.01 mM for 10 s), immediately after nerve transection, blocked hyperalgesia. The antinociceptive effect of CTX was long-lasting, since it was detected 2 h after treatment and persisted for 64 days. CTX also delayed, but did not block, neurectomy-induced neuroma formation. The effect of CTX was blocked by zileuton (100 mg/kg, p.o.) and atropine (10 mg/kg. i.p.), and reduced by yohimbine (2 mg/kg, i.p.) and methysergide (5 mg/kg, i.p.). on the other hand. indomethacin (4 mg/kg, i.v.). naloxone (1 mg/kg, i.p.). and N-methyl atropine (30 mg/kg, i.p.) did not interfere with the effect of CTX. These results indicate that CTX induces a long-lasting antinociceptive effect in neuropathic pain, which is mediated by activation of central muscarinic receptors and partially, by activation of alpha-adrenoceptors and 5-HT receptors. The present investigation examined the extracutaneous pigmentation pattern of three species of anuran amphibians, Dendropsophus nanus, Physalaemus cuvieri, and Rhinella schneideri, during the course of their breeding seasons. Pigmentation intensity in the different organs was graded on a 4-category scale, in which category 0 refers to organs without pigment and category 3 refers to intensely pigmented organs, with 2 intermediate stages of progressively stronger pigmentation. Rhinella schneideri showed testicular pigmentation, with intra-specific variation (categories 0 and 1). In P. cuvieri the pigmentation in the lungs varied in time: all the animals showed pigmentation (category 1) at the beginning of the breeding season, and as the season progressed the absence of pigments became the most common pattern. In the liver of the first animals collected, the pigment intensity was high (category 2) with many iridophores present, but in the last specimens collected no iridophores were found. The variation in D. nanus occurred in the kidneys, where animals collected at the beginning of the season did not show pigmentation. Renal veins displayed few melanocytes (category 1); animals collected at the end of the season showed more pigmentation in the kidneys (category 2), whereas in the renal veins the intensity remained the same. The changes observed in the extracutaneous pigment system in some organs, during the reproductive period, may be due to physiological alterations or may represent a species-specific characteristic.

The Dendropsophus marmoratus group is composed of eight species known for their explosive breeding habits and morphologically characterized by a bark-like dorsum, warty skin around the lower lips and an extremely large vocal sac. Within this group, D. nahdereri is the only species with distribution restricted to the southern region of Brazil. Apart from the original description and its tadpole, nothing else is known about this species. Using a mechanistic definition of note, we describe the advertisement call of D. nahdereri, which is similar to the advertisement calls of other species of the group and frequently has "final pulseclusters" as defined in the text.

The Rondonian-San Ignacio Province (1.56-1.30 Ga) is a composite orogen created through successive accretion of arcs, ocean basin closure and final oblique microcontinent-continent collision. The effects of the collision are well preserved mostly in the Paragua Terrane (Bolivia and Mato Grosso regions) and in the Alto Guapore Belt and the Rio Negro-Juruena Province (Rondonia region), considering that the province was affected by later collision-related deformation and metamorphism during the Sunsas Orogeny (1.25-1.00 Ga). The Rondonian-San Ignacio Province comprises: (1) the Jauru Terrane (1.78-1.42 Ga) that hosts Paleoproterozoic basement (1.78-1.72 Ga), and the Cachoeirinha (1.56-1.52 Ga) and the Santa Helena (1.48-1.42 Ga) accretionary orogens, both developed in an Andean-type magmatic arc; (2) the Paragua Terrane (1.74-1.32 Ga) that hosts pre-San Ignacio units (>1640 Ma: Chiquitania Gneiss Complex, San Ignacio Schist Group and Lomas Manechis Granulitic Complex) and the Pensamiento Granitoid Complex (1.37-1.34 Ga) developed in an Andean-type magmatic arc; (3) the Rio Alegre Terrane (1.51-1.38 Ga) that includes units generated in a mid-ocean ridge and an intra-oceanic magmatic arc environments; and (4) the Alto Guapore Belt (<1.42-1.34 Ga) that hosts units developed in passive marginal basin and intra-oceanic arc settings. The collisional stage (1.34-1.32 Ga) is characterized by deformation, high-grade metamorphism, and partial melting during the metamorphic peak, which affected primarily the Chiquitania Gneiss Complex and Lomas Manechis Granulitic Complex in the Paragua Terrane, and the Colorado Complex and the Nova Mamore Metamorphic Suite in the Alto Guapore Belt. The Paragua Block is here considered as a crustal fragment probably displaced from its Rio Negro-Juruena crustal counterpart between 1.50 and 1.40 Ga. This period is characterized by extensive A-type and intra-plate granite magmatism represented by the Rio Crespo Intrusive Suite (ca. 1.50 Ga), Santo Antonio Intrusive Suite (1.40-1.36 Ga), and the Teotonio Intrusive Suite (1.38 Ga). Magmatism of these types also occur at the end of the Rondonian-San Ignacio Orogeny, and are represented by the Alto Candeias Intrusive Suite (1.34-1.36 Ga), and the Sao Lourenco-Caripunas Intrusive Suite (1.31-1.30 Ga). Advertisement, territorial, and distress calls of Hypsiboas exastis, a species of the Hypsiboas faker group, are described for the first time. Acoustic records were obtained of one specimen found at the Municipality of Urucuca, state of Bahia, Brazilian eastern cost. We compared the advertisement calls of H. exastis with those of H. lundii and H. pardalis (the closest related species) and suggested a closer similarity with H. lundii.
