mitochondrial genomes are generally thought to be under selection for compactness, due to their small size, consistent gene content, and a lack of introns or intergenic spacers. As more animal mitochondrial genomes are fully sequenced, rearrangements and partial duplications are being identified with increasing frequency, particularly in birds (Class Ayes). In this study, we investigate the evolutionary history of mitochondrial control region states within the avian order Psittaciformes (parrots and cockatoos). To this aim, we reconstructed a comprehensive multi-locus phylogeny of parrots, used PCR of three diagnostic fragments to classify the mitochondrial control region state as single or duplicated, and mapped these states onto the phylogeny. We further sequenced 44 selected species to validate these inferences of control region state. Ancestral state reconstruction using a range of weighting schemes identified six independent origins of mitochondrial control region duplications within Psittaciformes. Analysis of sequence data showed that varying levels of mitochondrial gene and tRNA homology and degradation were present within a given clade exhibiting duplications. Levels of divergence between control regions within an individual varied from 0-10.9% with the differences occurring mainly between 51 and 225 nucleotides 3' of the goose hairpin in domain I. Further investigations into the fates of duplicated mitochondrial genes, the potential costs and benefits of having a second control region, and the complex relationship between evolutionary rates, selection, and time since duplication are needed to fully explain these patterns in the mitochondrial genome. Centronuclear myopathy (CNM) is a genetically heterogeneous disorder associated with general skeletal muscle weakness, type I fiber predominance and atrophy, and abnormally centralized nuclei. Autosomal dominant CNM is due to mutations in the large GTPase dynamin 2 (DNM2), a mechanochemical enzyme regulating cytoskeleton and membrane trafficking in cells. To date, 40 families with CNM-related DNM2 mutations have been described, and here we report 60 additional families encompassing a broad genotypic and phenotypic spectrum. In total, 18 different mutations are reported in 100 families and our cohort harbors nine known and four new mutations, including the first splice-site mutation. Genotype-phenotype correlation hypotheses are drawn from the published and new data, and allow an efficient screening strategy for molecular diagnosis. In addition to CNM, dissimilar DNM2 mutations are associated with Charcot-Marie-Tooth (CMT) peripheral neuropathy (CMTD1B and CMT2M), suggesting a tissue-specific impact of the mutations. In this study, we discuss the possible clinical overlap of CNM and CMT, and the biological significance of the respective mutations based on the known functions of dynamin 2 and its protein structure. Abstract Background The molecular phylogenetic relationships and population structure of the species of the Anopheles triannulatus complex: Anopheles triannulatus s.s., Anopheles halophylus and the putative species Anopheles triannulatus C were investigated. Methods The mitochondrial COI gene, the nuclear white gene and rDNA ITS2 of samples that include the known geographic distribution of these taxa were analyzed. Phylogenetic analyses were performed using Bayesian inference, Maximum parsimony and Maximum likelihood approaches. Results Each data set analyzed septely yielded a different topology but none provided evidence for the seption of An. halophylus and An. triannulatus C, consistent with the hypothesis that the two are undergoing incipient speciation. The phylogenetic analyses of the white gene found three main clades, whereas the statistical parsimony network detected only a single metapopulation of Anopheles triannulatus s.l. Seven COI lineages were detected by phylogenetic and network analysis. In contrast, the network, but not the phylogenetic analyses, strongly supported three ITS2 groups. Combined data analyses provided the best resolution of the trees, with two major clades, Amazonian (clade I) and trans-Andean + Amazon Delta (clade II). Clade I consists of multiple subclades: An. halophylus + An. triannulatus C; trans-Andean Venezuela; central Amazonia + central Bolivia; Atlantic coastal lowland; and Amazon delta. Clade II includes three subclades: Panama; cis-Andean Colombia; and cis-Venezuela. The Amazon delta specimens are in both clades, likely indicating local sympatry. Spatial and molecular variance analyses detected nine groups, corroborating some of subclades obtained in the combined data analysis. Conclusion Combination of the three molecular markers provided the best resolution for differentiation within An. triannulatus s.s. and An. halophylus and C. The latest two species seem to be very closely related and the analyses performed were not conclusive regarding species differentiation. Further studies including new molecular markers would be desirable to solve this species status question. Besides, results of the study indicate a trans-Andean origin for An. triannulatus s.l. The potential implications for malaria epidemiology remain to be investigated.

Durante o processo de projeto, o arquiteto transpõe suas ideias para o campo real, do concreto. Os diversos modos de expressão e representação têm como função mediar essa interação, diminuindo a distância entre esses dois campos. Vive-se hoje, um momento de intensa transformação das estratégias projetuais, propiciada pelos novos meios digitais. Assim, esta pesquisa, centra-se na comparação entre diversos momentos do uso de modelos nos processos projetivos contemporâneos, através de uma investigação em escritórios de arquitetura paulistanos que utilizam o modelo físico como parte de seus processos de projeto. Busca-se entender qual o papel dessa ferramenta de representação e suas potencialidades nos dias atuais. Como estudo de caso, faz-se uma análise comparativa entre o uso das maquetes digital e física, destacando dois estudos: a maquete do Conjunto Ponte dos Remédios, do arquiteto Marcos Acayaba e as maquetes de estudos elaboradas pelo escritório Andrade Morettin Arquitetos, para o concurso para o Instituto Moreira Salles/SP. Entre os objetivos desse trabalho também se encontra uma análise da contribuição dos modelos físicos no Ensino de Arquitetura.

Os diversos modos de expressão e representação têm como função mediar essa interação, diminuindo a distância entre esses dois campos. Vive-se hoje, um momento de intensa transformação das estratégias projetuais, propiciadas pelos novos meios digitais. Esta pesquisa tem como objetivo o estudo do uso das representações tridimensionais, especificamente dos modelos físicos e digitais. Pretende-se flagrar os momentos contributivos dos modelos no processo projetivo e as características intrínsecas a eles. A discussão busca não apenas destacar a importância dessa ferramenta, como também, traçar uma breve comparação entre a tecnologia digital e a feitura manual. Para este trabalho foram selecionados alguns arquitetos significativos do cenário da arquitetura paulista em cujos projetos comparecem o uso de modelos. Como estudos de caso tem-se a Residência do arquiteto Marcos Acayaba e o projeto vencedor do Concurso para o Instituto Moreira Salles/ SP, do escritório Andrade Morettin Arquitetos. Soma-se a estes objetivos, a apresentação do uso de modelos físicos e digitais em uma experiência didática projetiva.

Pineal melatonin synthesis can be modulated by many peptides, including insulin. Because melatonin appears to alter leptin synthesis, in this work we aimed to investigate whether leptin would have a role on norepinephrine- (NE-)mediated melatonin synthesis in cultured rat pineal glands. According to our data, cultured rat pineal glands express leptin receptor isoform b (Ob-Rb). Pineal expression of Ob-Rb mRNA was also observed in vivo. Administration of leptin (1 nM) associated with NE ( 1 µM) reduced melatonin content as well as arylalkylamine-N-acetyl transferase (AANAT) activity and expression in cultured pineal glands. Leptin treatment per se induced the expression of STAT3 in cultured pineal glands, but STAT3 does not participate in the leptin modulation of NE-mediated pineal melatonin synthesis. In addition, the expression of inducible cAMP early repressor (ICER) was further induced by leptin challenge when associated with NE. In conclusion, leptin inhibition of pineal melatonin synthesis appears to be mediated by a reduction in AANAT activity and expression as well as by increased expression of Icer mRNA. Peptidergic signaling within the pineal gland appears to be one of the most important signals which modulates melatonin synthesis; leptin, as a member of this system, is not an exception

Massmedia har en stor makt över tonåringar idag, vilka spenderar en tredjedel av sin vakna tid med att använda media. Dokusåpor är ett populärt mediefenomen där det kontinuerligt lanseras idealbilder. Det underliggande budskapet i idealbilderna handlar om att den graciösa kroppen och smilgroparna i ett attraktivt ansikte är nyckel till lycka. I takt med medias expansion har det skett en oroväckande ökning av den psykiska ohälsan bland tonåringar, detta innefattar oro, sömnproblem och andra stressrelaterade problem. Syftet med denna uppsats har varit att undersöka om det finns någon samvariation mellan de icke-hälsobefrämjande budskapen i dokusåporna Big Brother samt Top Model och tonårstjejers självkänsla, bruk av alkohol samt upplevda stress. Denna kvantitativa enkätundersökning har baserats på ett kvoturval och avgränsats till 50 tonårstjejer i åldern 16-19 år på en gymnasieskola i Jämtlands län. Empiri har sedan kopplats till tidigare forskning som sökts i olika databaser, KASAM-teorin, den sociala jämförelse teorin samt andra teoretiska begrepp av relevans. I slutsatsen kan vi se positiva resultat i form av ingen eller liten samvariation mellan de undersökta variablerna. Dock finns det även negativa resultat att finna angående hälsa. Detta då majoriteten av tjejerna bland annat är stressade, har sömnproblem, dricker alkohol regelbundet, har varit berusade samt att de känner press på sig att vara snygg. Jag kan inte argumentera för att dessa resultat är generaliserbara, men det är nog så intresseväckande och betänkligt att det ser ut så här i denna grupp av tonårstjejer.

SAMMANFATTNING: Medias makt att påverka individers sätt att tolka verkligheten är stor. Betydelsen för hur man väljer att skriva någonting blir viktigt eftersom det påverkar den sociala praktiken i fråga om makt och inflytande för individer och grupper. Vi har i denna uppsats lagt fokus på hur kvinnor med utländsk bakgrund uppfattar den mediala bilden av invandrarkvinnor och hur det påverkar deras identitetsskapande. Vårt syfte med uppsatsen är att empiriskt undersöka hur kvinnor med utländsk bakgrund uppfattar den bild som förmedlas i media av invandrarkvinnan. Vi har använt oss av en kvalitativ metod och samlat in ett empiriskt material bestående av åtta intervjuer. Resultaten visar att media i allt för stor utsträckning visar en gestaltning som bibehåller svenskheten och den svenska kvinnan som norm och invandrare och invandrarkvinnan som det avvikande. Våra intervjupersoner anser att bilden som media framställer av invandrarkvinnan är en negativ bild och en väldigt ensidig sådan. De uttrycker också att de inte identifierar sig med den getsaltningen av invandrarkvinnan samt att de ibland känner en oro att bli betraktad av andra, som den invandrarkvinna media skapat.
