Epigenetic inheritance is more widespread in plants than in mammals, in part because mammals erase epigenetic information by germline reprogramming. We sequenced the methylome of three haploid cell types from developing pollen: the sperm cell, the vegetative cell, and their precursor, the postmeiotic microspore, and found that unlike in mammals the plant germline retains CG and CHG DNA methylation. However, CHH methylation is lost from retrotransposons in microspores and sperm cells and restored by de novo DNA methyltransferase guided by 24 nt small interfering RNA, both in the vegetative nucleus and in the embryo after fertilization. In the vegetative nucleus, CG methylation is lost from targets of DEMETER (DME), REPRESSOR OF SILENCING 1 (ROS1), and their homologs, which include imprinted loci and recurrent epialleles that accumulate corresponding small RNA and are premethylated in sperm. Thus genome reprogramming in pollen contributes to epigenetic inheritance, transposon silencing, and imprinting, guided by small RNA.

Alachlor has been a commonly applied herbicide and is a substance of ecotoxicological concern. The present study aims to identify molecular biomarkers in the eukaryotic model Saccharomyces cerevisiae that can be used to predict potential cytotoxic effects of alachlor, while providing new mechanistic clues with possible relevance for experimentally less accessible eukaryotes. It focuses on genome-wide expression profiling in a yeast population in response to two exposure scenarios exerting effects from slight to moderate magnitude at phenotypic level. In particular, 100 and 264 genes, respectively, were found as differentially expressed on a 2-h exposure of yeast cells to the lowest observed effect concentration (110 mg/L) and the 20% inhibitory concentration (200 mg/L) of alachlor, in comparison with cells not exposed to the herbicide. The datasets of alachlor-responsive genes showed functional enrichment in diverse metabolic, transmembrane transport, cell defense, and detoxification categories. In general, the modifications in transcript levels of selected candidate biomarkers, assessed by quantitative reverse transcriptase polymerase chain reaction, confirmed the microarray data and varied consistently with the growth inhibitory effects of alachlor. Approximately 16% of the proteins encoded by alachlor-differentially expressed genes were found to share significant homology with proteins from ecologically relevant eukaryotic species. The biological relevance of these results is discussed in relation to new insights into the potential adverse effects of alachlor in health of organisms from ecosystems, particularly in worst-case situations such as accidental spills or careless storage, usage, and disposal.

A polymorphic inversion that lies on chromosome 17q21 comprises two major haplotype families (H1 and H2) that not only differ in orientation but also in copy-number. Although the processes driving the spread of the inversion-associated lineage (H2) in humans remain unclear, a selective advantage has been proposed for one of its subtypes. Here, we genotyped a large panel of individuals from previously overlooked populations using a custom array with a unique panel of H2-specific single nucleotide polymorphisms and found a patchy distribution of H2 haplotypes in Africa, with North Africans displaying a higher frequency of inverted subtypes, when compared with Sub-Saharan groups. Interestingly, North African H2s were found to be closer to "non-African" chromosomes further supporting that these populations may have diverged more recently from groups outside Africa. Our results uncovered higher diversity within the H2 family than previously described, weakening the hypothesis of a strong selective sweep on all inverted chromosomes and suggesting a rather complex evolutionary history at this locus.

Ediacaran fronds are key components of terminal-Proterozoic ecosystems. They represent one of the most widespread and common body forms ranging across all major faunal localities and time slices postdating the Gaskiers glaciation, but uncertainty over their phylogenetic affinities has led to uncertainty over issues of homology and functional morphology between, and within, organisms displaying this ecomorphology. Here we present the first large scale, multi-group cladistic analysis of Ediacaran organisms sampling 20 ingroup taxa with previously asserted affinities to the Arboreomorpha, Erniettomorpha and Rangeomorpha. Using a newly derived morphological character matrix that incorporates multiple axes of potential phylogenetically informative data, including architectural, developmental, and structural qualities, we seek to illuminate the evolutionary history of these organisms. We find strong support for existing classification schema and devise apomorphy-based definitions for each of the three frondose clades examined here. Through a rigorous cladistics framework it is possible to discern the pattern of evolution within, and between, these clades, including the identification of homoplasies and functional constraints. This work both validates earlier studies of Ediacaran groups and accentuates instances where previous assumptions of their natural history are uninformative.

In this seminar, I will share my experience in the early process of becoming an entrepreneur from a research background. Since 2008, I have been working with Prof. Mike Wald on an innovative video annotation tool called Synote. After about eight years of research around Synote, I have applied for the Royal Acadamy of Engineering Enterprise Fellowship in order to focus on developing Synote for real clients and making Synote sustainable and profitable. Now, it is already eight months into the fellowship, which has totally changed my life. It is very exciting, but at the same time I'm struggling all the time. The seminar will briefly go through my experience so far on the way of commercializing Synote from a research background. I will also discuss the valuable resources you can get from RAEng Enterprise Hub and Future Worlds, which is a Southampton based organization to help startups. If you are a Ph.D. student or research fellow in the University, and you want to start your own business, this is the seminar you want to attend.
