Os desreguladores endócrinos são compostos micropoluentes de ação deletéria que causam efeitos aos animais e aos seres humanos. Estes vêm trazendo a várias décadas, problemas relacionados à má formação ou ainda infertilidade de várias espécies. A presença destes compostos na ordem de ng L-1 já é capaz de gerar efeitos nocivos ao sistema endócrino. Este trabalho visou à determinação da presença e quantidade de alguns destes compostos como 17α-etinilestradiol, bisfenol A e estrona entre outros, nas águas do rio Arroio Fundo, localizado em Jacarepaguá no RJ que possui um índice de qualidade de água ruim. A primeira etapa deste trabalho foi a implementação e validação de metodologia para determinação do 17α-etinilestradiol por cromatografia líquida acoplada a detector de fluorescência onde foram estabelecido limite de detecção, de quantificação e linearidade conforme orientação do Inmetro (2010). Além da avaliação das amostras ambientais através da metodologia estabelecida. Na etapa subsequente foram realizados os experimentos relativos à determinação dos desreguladores endócrinos através da espectrometria de massas por detector tipo tandem quadrupolo, onde também foram realizados os procedimentos de validação e determinação dos compostos bisfenol A e estrona que não foram suscetíveis a efeito de matriz durante a ionização, que acabou impedindo a determinação de outros desreguladores endócrinos. Os resultados indicam que não há redução da presença dos desreguladores endócrinos detectados, e de outros compostos orgânicos também detectados, mas não identificados, entre os pontos a montante e a jusante da Unidade de Tratamento do Rio Arroio Fundo. Todavia em função do grande efeito de matriz oriundo do fato da amostra ser de extrema complexidade, não foi possível realizar uma identificação e quantificação de todos os compostos alvos.

Estudos em animais experimentais evidenciaram associações significativas entre esquistossomose mansoni e hipercolesterolemia. Estudos in vitro e in vivo já demonstraram que o colesterol é essencial para Schistosoma mansoni, embora este não tenha capacidade de sintetizá-lo. A captação é realizada a partir do ambiente (cultivo ou hospedeiro) através do tegumento. O colesterol está envolvido nos mecanismos de evasão do helminto contra a resposta imunológica, além de poder participar na modulação da sinalização celular e reprodução, estimulando os órgãos reprodutores dos helmintos adultos como observado na fase aguda da infecção experimental. Este trabalho tem como objetivo avaliar se o mesmo fenômeno ocorre na fase crônica. Os helmintos foram recuperados de dez camundongos submetidos à dieta hiperlipídica ou padrão (controle) foram corados pelo carmin cloridrico e montados, individualmente, em lâmina histológica com bálsamo do Canadá. A preparação foi analisada por microscopia de campo claro nos seguintes caracteres: tegumento e o sistema reprodutor nos vermes machos (lobos testiculares, vesícula seminal, lobos testiculares supranumerários e canal ginecóforo) e, nas fêmeas (ovário, oótipo, útero, ovo, glândulas vitelínicas e espermateca). Posteriormente, algumas lâminas foram separadas para visualização pela microscopia confocal dos órgãos do sistema reprodutores acima descritos. Apesar de ter sido observado uma maior quantidade de espermatozoides, uma maior quantidade de oócitos sendo liberados no grupo da dieta, não houve diferença estatística significativa (p>0,05) entre os grupos analisados. Houve um aumento na oogênese como observado na fase aguda. Dessa forma, o colesterol pode estar relacionado com a estimulação na atividade dos órgãos reprodutores dos helmintos adultos na fase crônica da infecção.

A nicotina é considerada o principal componente psicoativo do tabaco e esta induz seus efeitos farmacológicos centrais atuando em receptores nicotínicos colinérgicos (nAChRs). A maturação dos sistemas colinérgicos é consolidada durante o período da periadolescência, o que sugere que o cérebro do adolescente é vulnerável aos efeitos de estimulantes colinérgicos. Dados os efeitos deletérios do consumo de tabaco, incluindo a dependência, têm sido desenvolvidas estratégias terapêuticas para facilitar a interrupção do uso. A mais recente é o uso da vareniclina, um agonista parcial dos nAChRs α4β2. Pouco se sabe sobre os efeitos da exposição à nicotina na adolescência e menos ainda dos efeitos de curto e longo prazos dos tratamentos disponíveis para a reversão da dependência ainda durante este período. Neste sentido, os objetivos desse trabalho foram o de estudar os comportamentos associados à ansiedade e busca por novos estímulos e a função adrenal em animais expostos à fumaça do cigarro durante a adolescência e subsequentemente tratados com procedimento para reversão de dependência à nicotina. Esse estudo utilizou 150 camundongos Suíços adolescentes (de ambos os sexos). Os animais foram expostos à solução aquosa de nicotina (50g/ml - NIC) ou de sacarina (2% - SAC) v.o. do 30o dia de vida pós-natal (PN) à PN45 e submetidos aos seguintes tratamentos por gavagem de PN45 à PN56: 1) vareniclina (0,1 ou 1,0 mg/kg/dia, VAR1 e VAR2 respectivamente); 2) veículo (VEH); 3) nicotina na dose utilizada entre PN30 e PN45 (NIC). Sete grupos experimentais foram utilizados: SACVEH, SACVAR1, SACVAR2, NICVEH, NICVAR1, NICVAR2 e NICNIC. Em PN55, os animais foram submetidos ao teste do labirinto em cruz elevado (LCE) por 5 min, e duas depois ao teste do campo vazado (CV) também por 5 min. Os tecidos coletados em PN56 (após o sacrifício dos animais) foram: adrenal esquerda para a verificação do conteúdo de catecolaminas, adrenal direita para avaliação da expressão da tirosina hidroxilase e soro para a dosagem de corticosterona. Os nossos resultados demonstraram que no final da vigência do tratamento não foram encontradas diferenças entre os grupos no comportamentos associados à ansiedade do LCE e nos associados à busca por novos estímulos no CV. Por outro lado, a análise do número de orifícios explorados no centro do CV, também utilizado como medida de ansiedade, sugerem que a vareniclina por si só e a exposição continuada à nicotina são ansiogênicas mas que que o tratamento com vareniclina após a exposição à nicotina mantém os resultados dentro da normalidade. Do ponto de vista endócrino, o conteúdo adrenal de catecolaminas foi corrigido pelo tratamento com vareniclina, os níveis séricos de corticosterona foram aumentados pela exposição à nicotina enquanto o tratamento com vareniclina aumentou a expressão de tirosina hidroxilase. Nosso estudo indica que, no final da vigência do tratamento, tanto aspectos comportamentais como endócrinos estão significativamente afetados em nosso modelo de exposição à nicotina durante a adolescência e tratamento subsequente com vareniclina. Estudos futuros deverão avaliar estes parâmetros em períodos posteriores, com o objetivo de verificar se as alterações observadas persistem na vida adulta.

A erva-mate (Ilex paraguariensis) contribui para a perda de peso e reversão da resistência à insulina, mesmo em combinação com dieta hiperlipídica, despertando interesse do seu uso como uma possível estratégia no controle da obesidade. Previamente demonstramos que ratos desmamados precocemente desenvolvem sobrepeso, obesidade central, hiperleptinemia, resistência à insulina e à leptina na idade adulta. Propomos então avaliar os benefícios do extrato aquoso do Ilex paraguariensis sobre a composição corporal, perfis lipídico e hormonal, sinalização da leptina, neuropeptídeos envolvidos com o controle do consumo alimentar, marcadores inflamatórios e do estresse oxidativo. Para indução do desmame precoce, as tetas das ratas lactantes foram envolvidas com uma atadura para bloquear o acesso da prole ao leite materno nos últimos 3 dias da lactação (grupo DP). Os filhotes do grupo controle tiveram livre acesso ao leite durante todos os 21 dias do período de lactação normal (grupo C). Aos 150 dias de idade, as proles DP foram subdivididas em: DP e DP+Mate, de acordo com o tratamento (água ou extrato aquoso de erva-mate 1g/kg PC, respectivamente), por gavagem, diariamente, por 30 dias. As proles C receberam água por gavagem por 30 dias. Aos 180 dias de idade, os 3 grupos foram sacrificados por decapitação. Dados foram considerados significativos quando p <0,05. Aos 180 dias (1 mês após o tratamento), o grupo DP+Mate corrigiu várias alterações observadas no grupo DP, tais como maior massa corporal (+9%), massa de tecido adiposo retroperitoneal e epididimal (+34% e +39%, respectivamente), gordura corporal total (+76%), gordura subcutânea (+61%), área de adipócitos viscerais (+34%), triglicerídeos séricos (+35%), TNF-α no núcleo ARC (+57%), expressão de IL-1β no adipócito (+3,6 vezes), percentual de esteatose hepática (+4,8 vezes) e TBARS em fígado e plasma (+56%, +87%). A atividade SOD diminuída no grupo DP é normalizada pelo tratamento com mate, enquanto, paradoxalmente, a catalase que estava aumentada no grupo DP é normalizada com o tratamento com mate. As alterações de glicemia e HDL-c sérico no grupo DP não se modificaram ao final do tratamento com a erva-mate. A hiperfagia não foi observada na prole DP+Mate, provavelmente devido a normalização parcial da sensibilidade central à leptina (confirmada pelo teste do efeito anorexigênico da leptina) e pela redução de NPY e aumento de POMC. Assim, até o momento evidenciamos que o tratamento por 30 dias com extrato de erva-mate foi capaz de impedir a gênese de obesidade abdominal, a resistência à leptina, a hipertrigliceridemia e algumas alterações do perfil inflamatório e do estresse oxidativo nos ratos com obesidade programada pelo DP, tornando o uso da erva-mate como uma estratégia promissora no controle de peso e das desordens metabólicas presentes na obesidade.

Pituitary extract is in use for fish breeding since a long. Due to the difficulties in procuring pituitary glands and its preservation, scientists were looking for its substitute. Results on the use of human chorionic gonadotrophin in combination with pituitary for breeding of Cirrhina mrigala and Labeo rohita were encouraging. It has been possible to breed C. mrigala and L. rohita by use of HCG also. An increase in hatching percentage was observed with HCG & PG mixture in modern carp hatchery CIFE D-81.

The paper presents a resume on experiments carried on mass breeding of the silver carp Hypophthalmichthys molitrix in a cement cistern through administration of crude human chorionic gonadotropin. The experiments proves that human chorionic gonadotropin (H.C.G.) can be used for mass breeding of the fish in simple enclosures, with facilities of some input and output of water and a dose of 5 mg/kg and 18 mg/kg of body weight, for males and females respectively are required for the purpose. Observations were made by using pituitary glands in similar way. For successful breeding of about 80 kg female together, the expenditure towards cost of pituitary gland and H.C.G. are respectively 275 and 117 Rs.

The study deals with a series of experiments to investigate feeding and excretion in three species of bivalves: Perna viridis (Linné), Marcia cor (Sowerby) and Cassostrea gryphoides (Gould) from Manora Channel, Karachi. Bivalves were fed with suspensions of Ankistrodesmes labelled with P³². These animals showed a considerable variation in the average filtration rates depending upon species and the body lenght. Exceptionally high content of the P³² introduced with Akistrodesmes, got excreted as pseudofaeces and faeces within first three days following its absorption as a meal. The assimilated P³² is partly released as faecal material and its major proportion is directly transferred to the solution. As expexted the gonad and kidney are the main organs found responsible for excretion as comared to other body parts. Although, the assimilated P³² is mostly concentrated in the digestive glands, the results also show a significant presence of P³² in the gonads. Accumulation of P³² was the least in the foot.

Studies on reproductive biology and artificial propagation including larval rearing of freshwater mud eel, Monopterus cuchia and spiny eel, Mastacembelus armatus were attempted. The gonadosomatic index (GSI) of mud eel ranged from 0.41 (August) to 5.52 (June) in males and 0.53 (August) to 7.61 (June) in females. In both cases the GSI showed a peak in June. Fecundity ranged from 228 (TL - 396 mm; W - 78g) to 5510 (TL - 865 mm; W - 630 g). In case of spiny eel, the GSI varied from 0.65 (August) to 8.30 (July) in males and 0.70 (August) to 10.46 (July) in females. GSI showed single peak in July. Fecundity ranged from 570 (TL - 240 mm; W - 30 g) to 10870 (TL - 601; W - 350g). Histology of the testes and ovaries of the eels were carried out to investigate the gonadal development stages during the reproductive months (August to November 2003). In case of male M. cuchia, the secondary primordial germ cells, primary spermatogonium, some spermatogonia A and clone of spermatogonium B in testis were observed in September. In October-males different sized lobules having spermatogonia, spermatocytes and spermatids were observed. In the ovary of M. cuchia, polygonal shaped oocytes were seen during September. The oogonia were reduced with dense and irregular shaped during October. Numerous pycnotic cells were visible during November. In male M. armatus numerous broken lobule walls were found in testes during September. In October, abundant primary germ cells, pycnotic nests of degenerating cells, spermatogonia and spermatids were observed. In females, ovaries had distinct yolk vesicles stage and yolk granules stages in August. In September, the follicular cells of the oogonia were ruptured, shrunk forming irregular shaped in October. Oogonia were also shrunk with thin, irregular shaped structure but broken parts of the ruptured follicular cells were scattered in case of M. armatus. Experimental attempts on artificial propagation indicated that both freshwater eels were difficult to breed using inducing agents like pituitary glands (PG) of 10, 20, 50, 100 and 150 mg per kg of body weight. Same doses were used for both sexes with equal sex-ratio. In both cases, brood fish died at higher doses of injection given at 100 and 150 mg PG/kg bodyweight. However, M. cuchia breed naturally in cisterns when provided with water hyacinths and tunnel in muddy bottom. M. cuchia fed with chopped cooked fish attained a mean weight of 18.75 ± 2.3 g and cent percent survival. While in case of M. armatus best growth by weight (12.0 ± 2.48 g) and cent percent survival were achieved using chopped raw fish. Car tyre was observed as best shelter for attaining the mean weight gain 22.53 ± 2.24 g and cent percent survival of M. cuchia. While PVC pipe was found to be the best shelter for M. armatus, where it attained the mean weight of 12.73 ± 1.88 g and cent percent survival.

A novel plasminogen activator from Trimeresurus stejnegeri venom (TSV-PA) has been identified and purified to homogeneity. It is a single chain glycoprotein with an apparent molecular weight of 33,000 and an isoelectric point of pH 5.2. It specifically activates plasminogen through an enzymatic reaction. The activation of human native GIu-plasminogen by TSV-PA is due to a single cleavage of the molecule at the peptide bond Arg(561)-Val-(562). Purified TSV-PA, which catalyzes the hydrolysis of several tripeptide p-nitroanilide substrates, does not activate nor degrade prothrombin, factor X, or protein C and does not clot fibrinogen nor show fibrino(geno)lytic activity in the absence of plasminogen. The activity of TSV-PA was readily inhibited by phenylmethanesulfonyl fluoride and by p-nitrophenyl-p-guanidinobenzoate. Oligonucleotide primers designed on the basis of the N-terminal and the internal peptide sequences of TSV-PA were used for the amplification of cDNA fragments by polymerase chain reaction. This allowed the cloning of a full-length cDNA encoding TSV-PA from a cDNA library prepared from the venom glands. The deduced complete amino acid sequence of TSV-PA indicates that the mature TSV-PA protein is composed of 234 amino acids and contains a single potential N-gIycosylation site at Asn(1G1). The sequence of TSV-PA exhibits a high degree of sequence identity with other snake venom proteases: 66% with the protein C activator from Aghistrodon contortrix contortrix venom, 63% with batroxobin, and 60% with the factor V activator from Russell's viper venom. On the other hand, TSV-PA shows only 21-23% sequence similarity with the catalytic domains of u-PA and t-PA. Furthermore, TSV-PA lacks the sequence site that has been demonstrated to be responsible for the interaction of t-PA (KHRR) and u-PA (RRHR) with plasminogen activator inhibitor type 1.

The Ag5 proteins are the most abundant and immunogenic proteins in the venom secretory ducts of stinging insects. An antigen 5-like protein (named tabRTS) composed of 221 amino acid residues was purified and characterized from the salivary glands of the horsefly, Tabanus yao (Diptera, Tabanidae). Its cDNA was cloned from the cDNA library of the horsefly's salivary gland. TabRTS containing the SCP domain (Sc7 family of extracellular protein domain) was found in insect antigen 5 proteins. More interestingly, there is an Arg-Thr-Ser (RTS) disintegrin motif at the C-terminus of tabRTS. The RTS motif is positioned in a loop bracketed by cysteine residues as those found in RTS-disintegrins of Crotalidae and Viperidae snake venoms, which act as angiogenesis inhibitors. Endothelial Cell Tube formation assay in vitro and chicken chorioallantoic membrane (CAM) angiogenesis assay in vivo were performed as to investigate the effect of tabRTS on angiogenesis. It was found that tabRTS could significantly inhibit angiogenesis in vitro and in vivo. Anti-alpha(1)beta(1) monoclonal antibody could dose-dependently inhibit the anti-angiogenic activity of tabRTS. This result indicated that tabRTS possibly targets the alpha(1)beta(1) integrin to exert the anti-angiogenic activity as snake venom RTS-/KTS-disintegrins do. The current work revealed the first angiogenesis inhibitor protein containing RTS motif from invertebrates, a possible novel type of RTS-disintegrin. (C) 2009 Elsevier Ltd. All rights reserved.

By Sephadex G-50 gel filtration, cation-exchange CM-Sephadex C-25 chromatography and reversed phase high-performance liquid chromatography (HPLC), a novel serine protease inhibitor named bungaruskunin was purified and characterized from venom of Bungarus fasciatus. Its cDNA was also cloned from the cDNA library of B. fasciatus venomous glands. The predicted precursor is composed of 83 amino acid (aa) residues including a 24-aa signal peptide and a 59-aa mature bungaruskunin. Bungaruskunin showed maximal similarity (64%) with the predicted serine protease inhibitor blackelin deduced from the cDNA sequence of the red-bellied black snake Pseudechis porphyriacus. Bungaruskunin is a Kunitz protease inhibitor with a conserved Kunitz domain and could exert inhibitory activity against trypsin, chymotrypsin, and elastase. By screening the cDNA library, two new B chains of beta-bungarotoxin are also identified. The overall structures of bungaruskunin and beta -bungarotoxin B chains are similar; especially they have highly conserved signal peptide sequences. These findings strongly suggest that snake Kunitz/BPTI protease inhibitors and neurotoxic homologs may have originated from a common ancestor. (c) 2007 Elsevier Inc. All rights reserved.

About 3600 specimens were collected by bottom trawl at 15 sampling stations. 24 biometric characters were measured for each specimens at the laboratory.. Microscopic cross – sections of statolith were used for age determination. Sex determination and fecundity were determined. Population dynamics parameters as well as stock as stock assessment including cohort analysis were estimated using FISAT software. The findings showed that Dorsal Mantle Length (DML) and Body weight (BW) of the Indian squid were 133.9 ± 0.78 mm and 99.61 ± 0.95 g respectively. Strong correlation was found between these 2 variables (R2 = 0.90). The maximum age was 5 years. Relationship between DML and age was highly significantly of p ≤ 0.05. Overall sex ratio (M: F = 0.52) was significantly different from the expected 1:1 ratio (p ≤ 0.05). The ovary weight and nidamental glands weight were 7.72 ± 0.0006 g and 3.07 ± 0.0003g respectively. Absolute and relative fecundity of the Indian squid were found to be 122733 ± 30.87 and 2348 ± 0.4 respectively. GSI were 14.35 in April and 8.63 in July. This squid is therefore a spring spawner. The infinite dorsal mantle length were 258.62 mm for females, 194.72 mm for males and 252.02 for both sexes respectively. For population growth and mortality parameters; K (0.65 per year for both sexes, 0.85 per year for males, 0.65 per year for females); t0 (0.24year for both sexes, 0.22 year in females, 0.26 year in male); φ` (2.30 in both sexes, 2.47 for males, 2.37 for females); Z (1.17 per year for both sexes, 1.10 per year in females, 1.39 per year, in males); M (0.70 per year for both sexes, 0.90 for males, 0.67 for females); F(0.27 per year for both sexes, 0.27 per year in males, 0.195 per year in females). Exploitation coefficient were 0.51 per year for both sexes, 0.57 per year males and 0.51 per year females respectively. The results indicates that since the Indian squid is a short live aquatic organism, therefore, the exploitation coefficient could be raised to 0.7 per year. The analysis showed that total biomass and MSY were 10103.5 ton and 2576.4 ton respectively. These findings are the first study of its sort about the Indian squid in the coastal waters of Oman Sea as well as North-West of Indian Ocean.

Distribution of polychaetes worms Nereidae in intertidal zone of Bandar Abbass indicated that distribution rate of worms from the west to east for the reason status of seabed type, habitat and feeding substrate used to worms increased. Also investigation description that last-mentioned worms are belong to Nereididae family, Perinereis genus and species of Perinereis nuntia. By viewpoint seabed soil types experiments implement indicated Nereididae worms have higher survival with sandy clay loam soil texture. Statistical analysis showed positive relation coefficient correlation of Pearson between substrate type to frequency of worms and too substrate type with soil organic matter value (P<0.05). By viewpoint feeding also experiments with different feed treatment indicated that Nereid worms is Omnivore but in natural condition more utilized algae substrate special Entermorpha and survival rate had the worms last-mentioned in dietary treatments on this algae 93/3% different significant with other treatments (P<0.05). In this project pond-reared white Indian shrimp (Fenneropenaeus indicus) were used in four maturation and spawning trials lasting 60 days. one group of shrimps was fed a formulated pelleted special of broodstock diet only, a second group was fed squid meat diet; a third group of shrimps with mollusca (Solen vagina) meat diet and fourth group was fed polychaete worms Nereidae family, collected in intertidal zone of Bandar Abbass. Four types of dietary treatments (M1, M4) were given to separate batches that were run in three duplicate. Results of experiments demonstrate that more grew rate and maturation and spawning rate and we found that shrimps fed with polychaete worms have the best condition and then shrimps fed with solen. This is demonstrate furthermore environmental circumstance, endocrine hormone, types of feed important of maturation and spawning of shrimps and in polychaete worms used HUFA help to maturation ovarian in shrimp.

A total of 592 individuals of Loligo brasiliensis from the Mar del Plata coastal fishing area (Buenos Aires prov., Argentina) have been studied during the 1961-1964 period. From a morphological point of view the population appears to be uniform and homogeneus. A brief description of this species is given in this paper since references in the literature are scarce from the time at which Blainville (1923) first described it. The only further references are found in D'Orbigny (1835), and Ferrusac (1839), and in Hoyle (1886), and Tyron Pilsbry (1879). In this paper the species was mentioned only as a bibliographical reference on morphological or biological conditions has been found in the literature. The distribution of this species ranges from Cuba, Brazil, Uruguay to the Argentine coast, probably down to the Gulf of San Jorge. The samples had been studied with respect to various body measurements by classifing the individuals in total length classes, since body length was considered the most significant measurement. The condition factor K has been calculated for different sexes and ages, for the various length classes. The results lead to the conclusion that the smaller the length the higher is the value obtained for K and viceversa. This is due to the fact that the length of the tentacle increases considerably with increasing size. Since the tentacle are quite light the factor K diminishes accordingly. The condition factor increases considerably from December to April with an average of 0.42, decrease and becomes stable from March to October, with an average of 0.30. This is a consequence of the ripening of the sex glands. The sex-ratios are as following: year 1961, 42 % female, 42 % male; year 1962, 51 % female, 45 % male; year 1963, 46 % female, 53 % male; year 1964, 26 % female, 42 % male, 32 % indif. The great percentage of 72 undifferentiated young individuals in the 1964 (March) sampling increases the ratio of undifferentiation. A short morphological description of both ovules and spermatozoos is given. An examination of the sex glands leads to the following conclusions: a) male and female sex gland in a preparatory stage during the whole year; b) the highest percentage of ripe glands is found through, November-March; e) the spawning appears to precede rather slowly, but this certain since the spawning environment does not coincide with the natural habitat of the species. Few spawning individuals were found; d) sexual differentiation begins at body lenght from 30 to 40 mm; i.e. a total length of approximately 145 mm. At a body length of 70 mm. the hectocotilication (sexual character) begins to appear. In June 1962, a sample gathered at Rawson (Chubut) was analyzed. The conclusion was reached that the sex glands in this population are in an earlier stage of development in comparison with those from the Mar del Plata area.
