Neural connections in the adult central nervous system are highly precise. In the visual system, retinal ganglion cells send their axons to target neurons in the lateral geniculate nucleus (LGN) in such a way that axons originating from the two eyes terminate in adjacent but nonoverlapping eye-specific layers. During development, however, inputs from the two eyes are intermixed, and the adult pattern emerges gradually as axons from the two eyes sort out to form the layers. Experiments indicate that the sorting-out process, even though it occurs in utero in higher mammals and always before vision, requires retinal ganglion cell signaling; blocking retinal ganglion cell action potentials with tetrodotoxin prevents the formation of the layers. These action potentials are endogenously generated by the ganglion cells, which fire spontaneously and synchronously with each other, generating "waves" of activity that travel across the retina. Calcium imaging of the retina shows that the ganglion cells undergo correlated calcium bursting to generate the waves and that amacrine cells also participate in the correlated activity patterns. Physiological recordings from LGN neurons in vitro indicate that the quasiperiodic activity generated by the retinal ganglion cells is transmitted across the synapse between ganglion cells to drive target LGN neurons. These observations suggest that (i) a neural circuit within the immature retina is responsible for generating specific spatiotemporal patterns of neural activity; (ii) spontaneous activity generated in the retina is propagated across central synapses; and (iii) even before the photoreceptors are present, nerve cell function is essential for correct wiring of the visual system during early development. Since spontaneously generated activity is known to be present elsewhere in the developing CNS, this process of activity-dependent wiring could be used throughout the nervous system to help refine early sets of neural connections into their highly precise adult patterns.

Cytokines are now recognized to play important roles in the physiology of the central nervous system (CNS) during health and disease. Tumor necrosis factor alpha (TNF-alpha) has been implicated in the pathogenesis of several human CNS disorders including multiple sclerosis, AIDS dementia, and cerebral malaria. We have generated transgenic mice that constitutively express a murine TNF-alpha transgene, under the control of its own promoter, specifically in their CNS and that spontaneously develop a chronic inflammatory demyelinating disease with 100% penetrance from around 3-8 weeks of age. High-level expression of the transgene was seen in neurons distributed throughout the brain. Disease is manifested by ataxia, seizures, and paresis and leads to early death. Histopathological analysis revealed infiltration of the meninges and CNS parenchyma by CD4+ and CD8+ T lymphocytes, widespread reactive astrocytosis and microgliosis, and focal demyelination. The direct action of TNF-alpha in the pathogenesis of this disease was confirmed by peripheral administration of a neutralizing anti-murine TNF-alpha antibody. This treatment completely prevented the development of neurological symptoms, T-cell infiltration into the CNS parenchyma, astrocytosis, and demyelination, and greatly reduced the severity of reactive microgliosis. These results demonstrate that overexpression of TNF-alpha in the CNS can cause abnormalities in nervous system structure and function. The disease induced in TNF-alpha transgenic mice shows clinical and histopathological features characteristic of inflammatory demyelinating CNS disorders in humans, and these mice represent a relevant in vivo model for their further study.

Many features of Down syndrome might result from the overdosage of only a few genes located in a critical region of chromosome 21. To search for these genes, cosmids mapping in this region were isolated and used for trapping exons. One of the trapped exons obtained has a sequence very similar to part of the Drosophila single-minded (sim) gene, a master regulator of the early development of the fly central nervous system midline. Mapping data indicated that this exonic sequence is only present in the Down syndrome-critical region in the human genome. Hybridization of this exonic sequence with human fetal kidney poly(A)+ RNA revealed two transcripts of 6 and 4.3 kb. In situ hybridization of a probe derived from this exon with human and rat fetuses showed that the corresponding gene is expressed during early fetal life in the central nervous system and in other tissues, including the facial, skull, palate, and vertebra primordia. The expression pattern of this gene suggests that it might be involved in the pathogenesis of some of the morphological features and brain anomalies observed in Down syndrome.

Trans-synaptic activation of gene expression is linked to long-term plastic adaptations in the nervous system. To examine the molecular program induced by synaptic activity, we have employed molecular cloning techniques to identify an immediate early gene that is rapidly induced in the brain. We here report the entire nucleotide sequence of the cDNA, which encodes an open reading frame of 396 amino acids. Within the hippocampus, constitutive expression was low. Basal levels of expression in the cortex were high but can be markedly reduced by blockade of N-methyl-D-aspartate receptors. By contrast, synaptic activity induced by convulsive seizures increased mRNA levels in neurons of the cortex and hippocampus. High-frequency stimulation of the perforant path resulted in long-term potentiation and a spatially confined dramatic increase in the level of mRNA in the granule cells of the ipsilateral dentate gyrus. Transcripts were localized to the soma and to the dendrites of the granule cells. The dendritic localization of the transcripts offers the potential for local synthesis of the protein at activated postsynaptic sites and may underlie synapse-specific modifications during long-term plastic events.

Background e scopi dello studio. Il carcinoma renale rappresenta circa il 3% delle neoplasie e la sua incidenza è in aumento nel mondo. Il principale approccio terapeutico alla malattia in stadio precoce è rappresentato dalla chirurgia (nefrectomia parziale o radicale), sebbene circa il 30-40% dei pazienti vada incontro a recidiva di malattia dopo tale trattamento. La probabilità di recidivare può essere stimata per mezzo di alcuni noti modelli prognostici sviluppati integrando sia parametri clinici che anatomo-patologici. Il limite principale all’impiego nella pratica clinica di questi modelli è legata alla loro complessità di calcolo che li rende di difficile fruizione. Inoltre la stratificazione prognostica dei pazienti in questo ambito ha un ruolo rilevante nella pianificazione ed interpretazione dei risultati degli studi di terapia adiuvante dopo il trattamento chirurgico del carcinoma renale in stadio iniziale. Da un' analisi non pre-pianificata condotta nell’ambito di uno studio prospettico e randomizzato multicentrico italiano di recente pubblicazione, è stato sviluppato un nuovo modello predittivo e prognostico (“score”) che utilizza quattro semplici parametri: l’età del paziente, il grading istologico, lo stadio patologico del tumore (pT) e della componente linfonodale (pN). Lo scopo del presente studio era quello di validare esternamente tale score. Pazienti e Metodi. La validazione è stata condotta su due coorti retrospettive italiane (141 e 246 pazienti) e su una prospettica americana (1943 pazienti). Lo score testato prevedeva il confronto tra due gruppi di pazienti, uno a prognosi favorevole (pazienti con almeno due parametri positivi tra i seguenti: età < 60 anni, pT1-T3a, pN0, grading 1-2) e uno a prognosi sfavorevole (pazienti con meno di due fattori positivi). La statistica descrittiva è stata utilizzata per mostrare la distribuzione dei diversi parametri. Le analisi di sopravvivenza [recurrence free survival (RFS) e overall survival (OS)] sono state eseguite il metodo di Kaplan-Meier e le comparazioni tra i vari gruppi di pazienti sono state condotte utilizzando il Mantel-Haenszel log-rank test e il modello di regressione di Cox. Il metodo di Greenwood è stato utilizzato per stimare la varianza e la costruzione degli intervalli di confidenza al 95% (95% CI), la “C-statistic” è stata utilizzata per descrivere l’ accuratezza dello score. Risultati. I risultati della validazione dello score condotta sulle due casistiche retrospettive italiane, seppur non mostrando una differenza statisticamente significativa tra i due gruppi di pazienti (gruppo favorevole versus sfavorevole), sono stati ritenuti incoraggianti e meritevoli di ulteriore validazione sulla casistica prospettica americana. Lo score ha dimostrato di performare bene sia nel determinare la prognosi in termini di RFS [hazard ratio (HR) 1.85, 95% CI 1.57-2.17, p < 0.001] che di OS [HR 2.58, 95% CI 1.98-3.35, p < 0.001]. Inoltre in questa casistica lo score ha realizzato risultati sovrapponibili a quelli dello University of California Los Angeles Integrated Staging System. Conclusioni. Questo nuovo e semplice score ha dimostrato la sua validità in altre casistiche, sia retrospettive che prospettiche, in termini di impatto prognostico su RFS e OS. Ulteriori validazioni su casistiche internazionali sono in corso per confermare i risultati qui presentati e per testare l’eventuale ruolo predittivo di questo nuovo score.

In the chemical textile domain experts have to analyse chemical components and substances that might be harmful for their usage in clothing and textiles. Part of this analysis is performed searching opinions and reports people have expressed concerning these products in the Social Web. However, this type of information on the Internet is not as frequent for this domain as for others, so its detection and classification is difficult and time-consuming. Consequently, problems associated to the use of chemical substances in textiles may not be detected early enough, and could lead to health problems, such as allergies or burns. In this paper, we propose a framework able to detect, retrieve, and classify subjective sentences related to the chemical textile domain, that could be integrated into a wider health surveillance system. We also describe the creation of several datasets with opinions from this domain, the experiments performed using machine learning techniques and different lexical resources such as WordNet, and the evaluation focusing on the sentiment classification, and complaint detection (i.e., negativity). Despite the challenges involved in this domain, our approach obtains promising results with an F-score of 65% for polarity classification and 82% for complaint detection.

We provide a complete characterization of the astrophysical properties of the σ Ori Aa, Ab, B hierarchical triple system and an improved set of orbital parameters for the highly eccentric σ Ori Aa, Ab spectroscopic binary. We compiled a spectroscopic data set comprising 90 high-resolution spectra covering a total time span of 1963 days. We applied the Lehman-Filhés method for a detailed orbital analysis of the radial velocity curves and performed a combined quantitative spectroscopic analysis of the σ Ori Aa, Ab, B system by means of the stellar atmosphere code FASTWIND. We used our own plus other available information on photometry and distance to the system for measuring the radii, luminosities, and spectroscopic masses of the three components. We also inferred evolutionary masses and stellar ages using the Bayesian code BONNSAI. The orbital analysis of the new radial velocity curves led to a very accurate orbital solution of the σ Ori Aa, Ab pair. We provided indirect arguments indicating that σ Ori B is a fast-rotating early B dwarf. The FASTWIND+BONNSAI analysis showed that the Aa, Ab pair contains the hottest and most massive components of the triple system while σ Ori B is a bit cooler and less massive. The derived stellar ages of the inner pair are intriguingly younger than the one widely accepted for the σ Orionis cluster, at 3 ± 1 Ma. The outcome of this study will be of key importance for a precise determination of the distance to the σ Orionis cluster, the interpretation of the strong X-ray emission detected for σ Ori Aa, Ab, B, and the investigation of the formation and evolution of multiple massive stellar systems and substellar objects.

State and international entities can have profound effects on the development of a country’s nursing profession. Through a global health governance lens, this paper explores the development of nursing in Brazil during the early twentieth century, and its intersections with national and international interests. Accordingly, we will show how state policies established an environment that fostered the institutionalization of nursing as a profession in Brazil and supported it as a means to increase the presence of females in nation building processes. The State focused on recruiting elite women for nursing, in part due to the Rockefeller Foundation’s involvement in the country. Nurses who worked for Rockefeller came from well-educated classes within US society with specific ideas about who should be a nurse and the roles of nurses in a healthcare system. These women served as the primary vehicles for interacting with Brazilian health authorities responsible for health system development. Their early efforts did not, however, ensure a system capable of producing nursing human resources at a rate that, in present day Brazil, could meet the health needs of the country. Findings from this paper offer new avenues for historians to explore the early roots of professional nursing through a global health governance lens, improve the understanding of the intersection between international politics and professionalization, and highlight how these factors may impact nursing human resources production in the long term.

The institutionalisation of early retirement has become a universal feature of postwar industrial economies, though there are significant cross-national variations. This paper studies the impact of different types of welfare regimes, production systems and labour relations on early exit from work. After an analysis of the main trends, the paper discusses the costs and benefits of early retirement for the various actors — labour, capital and the state — at different levels. The paper outlines both the "pull” and "push” factors of early exit. It first compares the distinct welfare state regimes and private occupational pensions in their impact on early retirement. Then it looks at the labour-shedding strategies inherent to particular employment regimes, production systems and financial governance structures. Finally, the impact of particular industrial relations systems, and especially the role of unions is discussed. The paper finds intricate "institutional complementarities” between particular welfare states, production regimes and industrial relations systems, and these structure the incentives under which actors make decisions on work and retirement. The paper argues that the "collusion” between capital, labour and the state in pursuing early retirement is not merely following a labour-shedding strategy to ease mass unemployment, but also caused by the need for economic restructuration, the downsizing pressures from financial markets, the maintenance of peaceful labour relations, and the consequences of a seniority employment system.

The eruption of Tambora (Indonesia) in April 1815 had substantial effects on global climate and led to the ‘Year Without a Summer’ of 1816 in Europe and North America. Although a tragic event — tens of thousands of people lost their lives — the eruption also was an ‘experiment of nature’ from which science has learned until today. The aim of this study is to summarize our current understanding of the Tambora eruption and its effects on climate as expressed in early instrumental observations, climate proxies and geological evidence, climate reconstructions, and model simulations. Progress has been made with respect to our understanding of the eruption process and estimated amount of SO2 injected into the atmosphere, although large uncertainties still exist with respect to altitude and hemispheric distribution of Tambora aerosols. With respect to climate effects, the global and Northern Hemispheric cooling are well constrained by proxies whereas there is no strong signal in Southern Hemisphere proxies. Newly recovered early instrumental information for Western Europe and parts of North America, regions with particularly strong climate effects, allow Tambora’s effect on the weather systems to be addressed. Climate models respond to prescribed Tambora-like forcing with a strengthening of the wintertime stratospheric polar vortex, global cooling and a slowdown of the water cycle, weakening of the summer monsoon circulations, a strengthening of the Atlantic Meridional Overturning Circulation, and a decrease of atmospheric CO₂. Combining observations, climate proxies, and model simulations for the case of Tambora, a better understanding of climate processes has emerged.

The chronostratigraphy, the calcareous nannofossil biochronology, and the biostratigraphy of the Miocene and Pliocene sediments retrieved during Leg 115 in the equatorial western Indian Ocean are presented and discussed. Most of the zonal boundaries of the standard 1971 zonation of Martini and the 1973 zonation of Bukry are easily recognized in these low-latitude sediments. We also comment on the secondary events that are proposed in the literature to improve the biostratigraphic resolution provided by the standard zonations. The study of calcareous nannofossil biostratigraphy and taphonomy of sequences from the Northern Mascarene Plateau area, which was drilled to investigate the Neogene history of carbonate flux and dissolution, indicate that the accumulation of carbonates in this area results from a complex interplay among carbonate bioproductivity, carbonate removal by chemical dissolution and mechanical erosion, and carbonate addition by mass and current transport. In spite of these drawbacks, major changes and trends in carbonate accumulation can be recognized, most of which, if not all, correlate with major steps in the evolution of the Neogene climatic system.

The early Eocene represents a time of major changes in the global carbon cycle and fluctuations in global temperatures on both short- and long-time scales. These perturbations of the ocean-atmosphere system have been linked to orbital forcing and changes in net organic carbon burial, but accurate age models are required to disentangle the various forcing mechanisms and assess causal relationships. Discrepancies between the employed astrochronological and radioisotopic dating techniques prevent the construction of a robust time frame between ~49 and ~54 Ma. Here we present an astronomically tuned age model for this critical time period based on a new high-resolution benthic d13C record of ODP Site 1263, SE Atlantic. First, we assess three possible tuning options to the stable long-eccentricity cycle (405-kyr), starting from Eocene Thermal Maximum 2 (ETM2, ~54 Ma). Next we compare our record to the existing bulk carbonate d13C record from the equatorial Atlantic (Demerara Rise, ODP Site 1258) to evaluate our three initial age models and compare them with alternative age models previously established for this site. Finally, we refine our preferred age model by expanding our tuning to the 100-kyr eccentricity cycle of the La2010d solution. This solution appears to accurately reflect the long- and short-term eccentricity-related patterns in our benthic d13C record of ODP Site 1263 back to at least 52 Ma and possibly to 54 Ma. Our time scale not only aims to provide a new detailed age model for this period, but it may also serve to enhance our understanding of the response of the climate system to orbital forcing during this super greenhouse period as well as trends in its background state.

Knowledge of the evolution of North Atlantic Deep Water (NADW) is key to understanding the past evolution of the climatic system. We developed a new rock-magnetic method to determine the constituent magnetic minerals of sediments and report on the evolution of NADW during 2.2-2.9 Ma. We measured isothermal remanence acquisition curves of North Atlantic deep-sea sediments drilled at the Gardar Drift and decomposed the first derivatives of these curves into high-coercivity and low-coercivity components. Residuals of the decomposition were sufficiently small throughout the study interval, confirming that the Gardar Drift sediments represent a mixing of the two end-members. Fractional changes of the high-coercivity component represent variation of the Iceland-Scotland Overflow Water, a branch of NADW formed at the Nordic Seas. The high-coercivity component increased significantly during an interglacial period just after ~2.68 Ma, which suggests that NADW formation in the Nordic Seas abruptly intensified at this time.
