The application of disk shaped gold ultramicroelectrode for nitrite determination with and without addition of supporting electrolyte was studied using the differential pulse voltammetric method. The well-defined peak for nitrite oxidation near 0.8V (vs. Ag/AgCl reference electrode) was used to obtain analytical plots in the concentration range from 0.1 to 0.6 mmol L-1 and from 10.0 to 50.0 mu mol.L-1. The calculated detection limit was 0.65 mu mol.L-1 in purified water, in the absence of supporting electrolyte, with relative standard deviation of 1.36% (n=6) for analyzing 10.0 mu mol L-1 nitrite solutions, and accuracy of 100.9 %, based on recovery studies. The application of this analytical method to mineral and river water samples of natural pH also showed improved sensitivity when compared with the linear sweep voltammetric method previously reported.

Electron microscopy and immunolabelling with antiserum specific to cucumber mosaic virus coat protein were used to examine tobacco leaf cells infected by cucumber mosaic virus isolated from Catharanthus roseus (CMV-Cr). Crystalline and amorphous inclusions in the vacuoles were the most obvious cytological modifications seen. Immunogold labelling indicated that the crystalline inclusion was made up of virus particles and amorphous inclusions contained coat protein. Rows of CMV-Cr particles were found between membranes of dictyosomes, but membranous bodies and tonoplast-associated vesicles were not evident. Virus particles and/or free coat protein were easily detected in the cytoplasm by immunolabelling. No gold labelling was found within nuclei, chloroplasts and mitochondria.

Hypochlorous acid (HOCl) released by activated leukocytes has been implicated in the tissue damage that characterizes chronic inflammatory diseases. In this investigation, 14 indole derivatives, including metabolites such as melatonin, tryptophan and indole-3-acetic acid, were screened for their ability to inhibit the generation of this endogenous oxidant by stimulated leukocytes. The release of HOCl was measured by the production of taurine-chloramine when the leukocytes (2 x 10(6) cells/mL) were incubated at 37ºC in 10 mM phosphate-buffered saline, pH 7.4, for 30 min with 5 mM taurine and stimulated with 100 nM phorbol-12-myristate acetate. Irrespective of the group substituted in the indole ring, all the compounds tested including indole, 2-methylindole, 3-methylindole, 2,3-dimethylindole, 2,5-dimethylindole, 2-phenylindole, 5-methoxyindole, 6-methoxyindole, 5-methoxy-2-methylindole, melatonin, tryptophan, indole-3-acetic acid, 5-methoxy-2-methyl-3-indole-acetic acid, and indomethacin (10 µM) inhibited the chlorinating activity of myeloperoxidase (MPO) in the 23-72% range. The compounds 3-methylindole and indole-3-acetic acid were chosen as representative of indole derivatives in a dose-response study using purified MPO. The IC50 obtained were 0.10 ± 0.03 and 5.0 ± 1.0 µM (N = 13), respectively. These compounds did not affect the peroxidation activity of MPO or the production of superoxide anion by stimulated leukocytes. By following the spectral change of MPO during the enzyme turnover, the inhibition of HOCl production can be explained on the basis of the accumulation of the redox form compound-II (MPO-II), which is an inactive chlorinating species. These results show that indole derivatives are effective and selective inhibitors of MPO-chlorinating activity.

Platinum-Pd nuggets from the Born Sucesso stream alluvium Minas Gerais, Brazil. have been investigated by electron-probe microanalysis. The nuggets attain 1 mm in maximum dimension and have a botryoidal habit, with pronounced compositional core-to-margin zoning of internal structures, Although there is a wide variation in internal morphology and individual zones vary markedly in thickness (<1-100 mum), a typical composite arborescent nugget comprises a broad irregular core region of massive auriferous Pd-Hg alloy (potarite delta-PdHg) or cavity space + relict potarite enclosed by a narrow zone of platiniferous, palladium or alloy of near Pt50Pd50 composition, and is progressively oscillatory zoned by palladian platinum, with growth eventually enveloping the whole botryoidal colony, to a narrow rim of palladian platinum or pure platinum. Other nuggets comprise an arborescent to dendritic core of auriferous potarite, a broad internal zone of either pure platinum or palladian platinum, and a narrow rim of platinum, the mineral palladium contains up to about 65 at.% Pd; this is the first detailed modern confirmation of palladium in its type locality. Auriferous potarite ranges in composition from similar toPd(3)Hg(2) to near Pd(Hg,Au). The origin of these nuggets remains unclear, but their mineralogy is broadly equivalent to that of palladian gold. potarite and platinum in alluvial sediments and overburden from Devon, England, which are considered to be detrital, and their platinum-group-clement geochemistry is consistent with precipitation from hydrothermal fluids. We suggest that the Bom Sucesso nuggets resulted from high-level episodic hydrothermal alteration of mafic and ultramafic rocks within the drainage basin, with the remobilized Pt and Pd precipitated in open spaces in the enclosing metaquartzites.

The electrochemical oxidation of (benzylideneacetone)dicarbonyl(phosphine)iron(0) and benzylideneacetone)dicarbonyl(phosphite)iron(0) complexes was studied by cyclic voltammetry and controlled potential electrolysis in 0.5 M NaClO4 (dimethyl formamide). The results suggest that the electrode process involves a complicated mechanism, the species formed in the first oxidation step being highly unstable and its decomposition producing free benzylideneacetone, free phosphine or phosphite, solvated iron(II) species and carbon monoxide which adsorbs on the platinum electrode. A linear relationship between E(p/2)ox and the ligand parameter P(L) was obtained with E(s) = 0.41 V and beta = 0.964, where E(s) and beta-denote electron-richness and polarizability of the metal centre, respectively.

Kinetic studies of hydrogen evolution reaction (HER) at the surface of Pt in alkaline conditions, reported in this paper, show that electrocatalytic activity is enhanced after adsorption of S-2 ions. EIS and steady-state polarization curve data pointed to an undoubted improvement in performance with the Pt-S cathode that was attributed to higher adsorbed hydrogen coverage. Experimental findings suggested an increase in the electronic density of the modified surface sites that may strengthen the interaction between H2O and the adsorption site and, consequently, accelerates the Volmer step. We performed immunogold labeling with an ST-1 monoclonal antibody (IgM), specific for intact heparin, to define the subcellular localization of heparin in mast cells. Rat peritoneal mast cells were fixed by a modified Karnovsky method and embedded in Araldite. Ultrathin sections were first treated with sodium periodate and then sequentially incubated with MAb ST-1, rabbit anti-mouse IgM, and protein A-gold. By transmission electron microscopy, gold particles were localized inside cytoplasmic granules of peritoneal mast cells. In contrast, with the same procedure, no labeling was observed in mast cells from rat intestinal mucosa. Control sections of rat peritoneal or intestinal mucosa mast Mast cells cells treated with an irrelevant MAb (IgM) did not show any labeling. Treatment with nitrous Heparin acid abolished the reactivity of MAb ST-1 with peritoneal mast cells. These results Granules show that different mast cells can be identified regarding their heparin content by immunochemical procedures using MAb ST-1.

Brazil does not have working platinum mines, nor even large reserves of the platinum metals, but there is platinum in Brazil. In this paper, four massifs (mafic/ultramafic complexes) in eastern Brazil, in the states of Minas Gerais and Ceara, where platinum is found will be described. Three of these massifs contain concentrations of platinum group minerals or platinum group elements, and gold, associated with the chromitite rock found there. In the fourth massif, in Minas Gerais State, the platinum group elements are found in alluvial deposits at the Bom Sucesso occurrence. This placer is currently being studied.

A new platinum(II) complex with deoxyalliin was synthesized and characterized by chemical and spectroscopic techniques. Elemental and mass spectrometry analyses of the solid complex fit to the composition [Pt(C6H11NO2S)Cl-2] center dot H2O. C-13 NMR, N-15 NMR and infrared spectra of the complex are consistent with coordination of deoxyalliin to Pt( II) through the nitrogen and sulfur atoms forming a square-planar geometry. The complex is soluble in dimethylsulfoxide. Biological analysis for evaluation of a potential cytotoxic effect of the complex was performed using HeLa cells, a human cervix adenocarcinoma-derived cell line. The results were compared with those of a palladium( II) complex previously described.

Synthesis and X-ray structure of a dinuclear platinum(II) complex with the ligand saccharin(sac) are described. The structure shows two approximately square-planar platinum centers. Each platinum atom is coordinated to one water molecule and three N-bonded saccharinate ligands. The two centers are linked through two potassium atoms. Each potassium atom interacts with six oxygen atoms from hydration and coordinated water molecules and from carbonyl and sulfonate groups of the ligands. It is suggested that, in aqueous solution, the dimeric structure of the complex is dissociated and the monomeric species K[Pt(sac)(3)(H2O)] is formed. The complex was dissolved in water and submitted to in vitro cytotoxic analyses using HeLa cells (human cervix cancer). It was shown that the monomeric complex elicited a potent cytotoxic activity when compared to the vehicle-treated cells. The IC50 value for the monomeric complex is 6.8 mu M, a little bit higher than that obtained for cisplatin. (c) 2007 Elsevier B.V. All rights reserved.

The biooxidation of ferrous ion into ferric ion by Acidithiobacillus ferrooxidans can be potentially used for the removal of H2S from industrial gases. In this work, Fe3+ ions were obtained through the oxidation of Fe2+ using the LR strain of At. ferrooxidans immobilized in PVC stands in a pilot-scale bioreactor, while H2S was removed in an absorption tower equipped with Rasching rings. At. ferrooxidans LR strain cells were immobilized by inoculating the bacterium in a Fe2+-mineral medium and percolating it through the support. After complete Fe2+ oxidation, which took around 90 h, the reactor was washed several times with sulfuric acid (pH 1.7) before a new cycle was started. Four additional cycles using fresh Fe2+ mineral medium were then run. During these colonization cycles, the time required for complete iron oxidation decreased, dropping to about 60 h in the last cycle. The batch experiments in the H2S gas removal trials resulted in a gas removal rate of about 98-99% under the operational conditions employed. In the continuous experiments with the bioreactor coupled to the gas absorption column, a gas removal efficiency of almost 100% was reached after 500 min. The construction of a flow-through cell incorporating an array of gold microelectrodes is described and its application to flow injection analysis with amperometric detection is presented, Simple modification of almost any conventional integrated circuit chip, used as an inexpensive source of pre-assembled gold micro-wires, leads to the rapid and successful preparation of arrays of 8-48 elements, the polymeric encapsulation material from the top face of the chip is removed by abrasion until the gold micro-mires (used to interconnect the silicon circuit to the external contact pins of the chip) are disrupted and their transversal (elliptical) sections become exposed. Once polished, the flat and smooth top surface of the gold microelectrode-array chip (MEAC) is provided with a spacer and fitted under pressure against an acrylic block with the reference and auxiliary electrodes, to form the electrochemical (thin-layer) flow cell, while the contact pins are plugged into a standard IC socket, This design ensures autonomous electric contact with each electrode and allows fast dismantling for polishing or substitution, the performance of flow cells with MEACs was investigated utilizing the technique of reverse pulse amperometry without oxygen removal, A method was established for the determination of the copper concentration in sugar cane spirit, regulated by law for beverages, Samples from industrial producers and small-scale (alembic) brewers were compared, With a 24 MEAC, a detection limit of 30 mu g I-l of copper (4.7 x 10(-7) mol l(-1) of Cu-II for 100 mu l injections) was calculated, Routine operation was established at a frequency of 60-90 determinations per hour, Intercomparison with atomic absorption spectrometric determinations resulted in excellent agreement.

A procedure for the standardization of ensembles of gold nanodisk electrodes (NEE) of 30 nm diameter is presented, which is based on the analytical comparison between experimental cyclic voltammograms (CV) obtained at the NEEs in diluted solutions of redox probes and CV patterns obtained by digital simulation. Possible origins of defects sometimes found in NEEs are discussed. Selected NEEs are then employed for the study of the electrochemical oxidation of iodide in acidic solutions. CV patterns display typical quasi-reversible behavior which involves associated chemical reactions between adsorbed and solution species. The main CV characteristics at the NEE compare with those observed at millimeter sized gold disk electrodes (Au-macro), apart a slight shift in E1/2 values and slightly higher peak to peak separation at the NEE. The detection limit (DC) at NEEs is 0.3 mu M, which is more than one order of magnitude lower than DL at the Au-macro (4 RM). The mechanism of the electrochemical oxidation of iodide at NEEs is discussed. Finally, NEEs are applied to the direct determination of iodide at micromolar concentration levels in real samples, namely in some ophthalmic drugs and iodized table salt. (c) 2006 Elsevier B.V.
