Since 2007, 96 wild Queensland groupers, Epinephelus lanceolatus, (Bloch), have been found dead in NE Australia. In some cases, Streptococcus agalactiae (Group B Streptococcus, GBS) was isolated. At present, a GBS isolate from a wild grouper case was employed in experimental challenge trials in hatchery-reared Queensland grouper by different routes of exposure. Injection resulted in rapid development of clinical signs including bilateral exophthalmia, hyperaemic skin or fins and abnormal swimming. Death occurred in, and GBS was re-isolated from, 98% fish injected and was detected by PCR in brain, head kidney and spleen from all fish, regardless of challenge dose. Challenge by immersion resulted in lower morbidity with a clear dose response. Whilst infection was established via oral challenge by admixture with feed, no mortality occurred. Histology showed pathology consistent with GBS infection in organs examined from all injected fish, from fish challenged with medium and high doses by immersion, and from high-dose oral challenge. These experimental challenges demonstrated that GBS isolated from wild Queensland grouper reproduced disease in experimentally challenged fish and resulted in pathology that was consistent with that seen in wild Queensland grouper infected with S. agalactiae.

The occurrence of high-pressure mafic-ultramafic bodies within major shear zones is one of the indicators of paleo-subduction. In mafic granulites of the Andriamena complex (north-eastern Madagascar) we document unusual textures including garnet-clinopyroxene-quartz coronas that formed after the breakdown of orthopyroxene-plagioclase-ilmenite. Textural evidence and isochemical phase diagram calculations in the Na2O-CaO-K2O-FeO-MgO-Al2O3-SiO2-H2O-TiO2 system indicate a pressure-temperature (P-T) evolution from an isothermal (780 degrees C) pressure up to c. 24 kbar to decompression and cooling. Such a P-T trajectory is typically attained in a subduction zone setting where a gabbroic/ultramafic complex is subducted and later exhumed to the present crustal level during oceanic closure and final continental collision. The present results suggest that the presence of such deeply subducted rocks of the Andriamena complex is related to formation of the Betsimisaraka suture. LA-ICPMS U-Pb zircon dating of pelitic gneisses from the Betsimisaraka suture yields low Th/U ratios and protolith ages ranging from 2535 to 2625 Ma. A granitic gneiss from the Alaotra complex yields a zircon crystallization age of ca. 818 Ma and Th/U ratios vary from 1.08 to 2.09. K-Ar dating of muscovite and biotite from biotite-kyanite-sillimanite gneiss and garnet-biotite gneiss yields age of 486 +/- 9 Ma and 459 +/- 9 Ma respectively. We have estimated regional crustal thicknesses in NE Madagascar using a flexural inversion technique, which indicates the presence of an anomalously thick crust (c. 43 km) beneath the Antananarivo block. This result is consistent with the present concept that subduction beneath the Antananarivo block resulted in a more competent and thicker crust. The textural data, thermodynamic model, and geophysical evidence together provide a new insight to the subduction history, crustal thickening and evolution of the high-pressure Andriamena complex and its link to the terminal formation of the Betsimisaraka suture in north-eastern Madagascar. The (He3, n) reactions on B11, N15, O16, and O18 targets have been studied using a pulsed-beam time-of-flight spectrometer. Special emphasis was placed upon the determination of the excitation energies and properties of states with T = 1 (in Ne18), T = 3/2 (in N13 and F17) and T = 2 (in Ne20). The identification of the T = 3/2 and T = 2 levels is based on the structure of these states as revealed by intensities and shapes of angular distributions. The reactions are interpreted in terms of double stripping theory. Angular distributions have been compared with plane and distorted wave stripping theories. Results for the four reactions are summarized below:
1) O16 (He3, n). The reaction has been studied at incident energies up to 13.5 MeV and two previously unreported levels in Ne18 were observed at Ex = 4.55 ± .015 MeV (Γ = 70 ± 30 keV) and Ex = 5.14 ± .018 MeV (Γ = 100 ± 40 keV).
2) B11 (He3, n). The reaction has been studied at incident energies up to 13.5 MeV. Three T = 3/2 levels in N13 have been identified at Ex = 15.068 ± .008 MeV (Γ ˂ 15 keV), Ex = 18.44 ± .04, and Ex 18.98 ± .02 MeV (Γ = 40 ± 20 keV).
3) N15 (He3, n). The reaction has been studied at incident energies up to 11.88 MeV. T = 3/2 levels in F17 have been identified at Ex = 11.195 ± .007 MeV (Γ ˂ 20 keV), Ex = 12.540 ± .010 MeV (Γ ˂ 25 keV), and Ex = 13.095 ± .009 MeV (Γ ˂ 25 keV).
4) O18 (He3, n). The reaction has been studied at incident energies up to 9.0 MeV. The excitation energy of the lowest T = 2 level in Ne20 has been found to be 16.730 ± .006 MeV (Γ ˂ 20 keV).
Angular distributions of the transitions leading to the above higher isospin states are well described by double stripping theory. Analog correspondences are established by comparing the present results with recent studies (t, p) and (He3, p) reactions on the same targets.

An attempt was made to conduct spatial assessment of the pattern and extent of damage to coastal aquaculture ponds along the east coast of Aceh province in Sumatra, Indonesia, resulting from the tsunami event of 26 December 2004. High-resolution satellite imagery, i.e., SPOT-5 multispectral scenes covering the 700 km stretch of the coast, acquired before and after the tsunami, were digitally enhanced and visually interpreted to delineate pockets of aquaculture ponds that were discerned to be damaged and relatively intact. Field checks were conducted at 87 sites in the four eastern coastal districts. The results indicate that SPOT-5 multispectral imagery was minimally sufficient to detect areas of damaged and relatively intact aquaculture ponds, but the 10-m spatial resolution poses limitations to evaluating the extent of pond damage. Nevertheless, the 60 km swath of the imagery makes it reasonably affordable for large-area assessment to identify pockets of severe damage for targeting more detailed assessments. The image maps produced from a mosaic of the SPOT-5 scenes can also serve as base maps for spatial planning in the challenging task of reconstruction and rehabilitation of the disrupted livelihoods of the coastal communities.

Stomach contents of 110 franciscanas (Pontoporia blainvillei), from northern Argentina were analysed in order to improve our knowledge about the feeding habits of this species and to better characterise the lactation period. The samples included calves, juveniles and adults of both sexes. Evidence of predation by franciscanas is seen at a very young age (2.5-3 months), with a transition diet composed by both milk and solid food, mainly represented by crustaceans. Weaning seems to begin by April, when franciscanas are about 6-7 months old. Franciscanas inhabiting two different habitats were analysed in this study: a brackish water estuary and an adjacent marine coastal system. The diet of Pontoporia blainvillei in northern Argentina was composed by a total of 26 prey species: 20 teleosts, 4 crustaceans and 2 cephalopods. Based on the Index of Relative Importance (IRI) the main prey species were Cynoscion guatucupa, Micropogonias furnieri, Loligo sanpaulensis and Urophycis brasiliensis. Estuarine franciscanas preyed mainly on Micropogonias furnieri (dominant species), Cynoscion guatucupa, Odonthestes argentinensis and Macrodon ancylodon, while dolphins from marine areas preyed mainly on Cynoscion guatucupa (dominant species), Loligo sanpaulensis and Urophycis brasiliensis. Our results confirm that franciscanas prey mainly on juvenile fish (< 8cm) and small loliginid squids, in close agreement with previous results obtained in southern Brazil and Uruguay. Qualitative and quantitative differences observed in the diet of dolphins from each habitat emphasise the need to discriminate between samples from different habitats and environmental parameters. SPANISH: Se analizaron 110 contenidos estomacales de franciscanas (Pontoporia blainvillei) provenientes de la costa norte de Argentina, para extender en conocimiento sobre su dieta y caracterizar la lactancia. Las muestras incluyeron cachorros, juveniles y adultos de ambos sexos. Las primeras etapas de predación se inician a muy temprana edad (2,5-3 meses), presentando una dieta de transición compuesta tanto por leche como por presas sólidas, principalmente crustáceos; el destete se iniciaría a partir de abril, a una edad estimada entre 6 y 7 meses. Las franciscanas estudiadas provienen de dos habitats diferentes: un área estuarial de baja salinidad y la region marina adyacente. La dieta de Pontoporia blainvillei de Argentina estuvo compuesta por un total de 26 especies: 20 teleósteos, 4 crustáceos y 2 cefalópodos. Basados en el Indice de Importancia Relativa (IIR), las presas más importantes fueron Cynoscion guatucupa, Micropogonias furnieri, Loligo sanpaulensis y Urophycis brasiliensis. Las franciscanas provenientes del área estuarial predaron principalmente sobre Micropogonias furnieri (especie dominante), Cynoscion guatucupa, Odonthestes argentinensis y Macrodon ancylodon, mientras que los delfines marinos predaron sobre Cynoscion guatucupa (especie dominante), Loligo sanpaulensis y Urophycis brasiliensis. Nuestros resultados confirman que la franciscana preda sobre peces juveniles (< 8cm) y pequeños calamares Loliginidae, coincidiendo con resultados previos obtenidos en el sur del Brasil y Uruguay. Las diferencias cualitativas y cuantitativas observadas en la dieta de cada uno de las áreas analizadas, nos sugieren que los futuros estudios sobre ecología trófica de la franciscana deberían discriminarse de acuerdo al origen de los ejemplares y a la tipificación del ambiente.

In Cambodia, fish provide a major source of animal protein for rural households. Capture fisheries have declined and aquaculture has been identified as playing an important role in food and nutritional security and rural income generation. In 2011, WorldFish, in partnership with the Stung Treng Fishery Administration Cantonment and the Culture and Environment Preservation Association, aimed at improving the uptake of small-scale aquaculture by communities with limited experience in fish culture in Stung Treng Province in northeast Cambodia. The system was given the name “WISH ponds,” derived from the combination of the words "water" and "fish" to reflect the integration of fish cultivation with water for storage and vegetable growing. It was targeted towards households with limited space to construct large aquaculture ponds, such as peri-urban households. The study indicated that WISH ponds can create an important learning platform for communities to address challenges associated with small-scale aquaculture development by using scientific data generated and owned by the participants. Results from this 2011 study provided important insights into the challenges and constraints for introducing small-scale aquaculture into rural households in Cambodia. In mid-2013, WorldFish won a Feed the Future Partnering for Innovation grant, funded by the United States Agency for International Development, to build upon its successful engagement with communities in northeast Cambodia where WISH ponds had already been introduced and investigate scaling this technology to establish more WISH ponds in these communities.

Sodium rutin sulfate (SRS) is a sulfated rutin modified from the natural flavonol glycoside rutin. Here, we investigated its in vitro anti-HIV and -HSV activities and its cytotoxic profile. Fifty percent inhibitory concentration (IC50) values of SRS against HIV-1 X4 virus IIIB, HIV-1 R5 isolates Ada-M and Ba-L were 2.3 +/- 0.2, 4.5 +/- 2.0 and 8.5 +/- 3.8 mu M with a selectivity index (SI) of 563, 575 and 329, respectively. Its IC50 against primary R5 HIV-1 isolate from Yunnan province in China was 13.1 +/- 5.5 mu M, with a Sl of 197. In contrast, unsulfated rutin had no activity against any of the HIV-1 isolates tested. Further study indicated that SRS blocked viral entry and virus-cell fusion likely through interacting with the HIV- I envelope glycoprotein. SRS also demonstrated some activity against human herpes simplex virus (HSV) with an IC50 of 88.3 +/- 0.1 mu M and a Sl of 30. The 50% cytotoxicity concentration (CC50) of SRS was >3.0 mM, as determined in human genital ME 180, HeLa and primary human foreskin fibroblast cells. Minimum inhibitory concentration of SRS for vaginal lactobacilli was >3.0 mM. These results collectively indicate that SRS represents a novel candidate for anti-HIV-1/HSV microbicide development. The seasonal population dynamics and maturation cycle of the nematode Camallanus cotti in the posterior intestine of Chinese hooksnout carp Opsariichthys bidens have been studied in the Danjiangkou Reservoir of the Hubei Province in central China from September 2004 to November, 2005. The overall prevalence, mean abundance and intensity of C cotti among fish sampled (n = 700 fish) were 47%, 2.29 +/- 12.38 ( +/- S.D.) and 1-307 (average 4.89 +/- 17.74), respectively. The overall sexual ratio of female to male nematodes (excluding L3 and L4 juveniles) was 1.17:1. Statistical results showed weakly positive correlations betweerl fish length and the number of nematodes per host. The dynamics of infection of the nematode exhibited significant seasonal pattern in changes in mean abundance. A similar pattern was found for changes in nematode prevalence, although this was not statistically significant. Higher levels of infection were observed among fish sampled in summer months and the lower in the winter. Neither the prevalence nor the abundance of the parasite was significantly different between male and female hosts. The pattern of frequency distribution of the parasite in the host was found to be over-dispersed throughout the sampling period. In addition, studies on the development and maturation of the parasite in O. bidens revealed that development (maturation), recruitment of the next generation, and reproduction may be continuous year-round, although reproduction may peak during the winter. Prenatal exposures to persistent organic pollutants were assessed using the levels of PCBs and organochlorine pesticides (OCPs) measured in cord blood and meconium samples from Luqiao and two other localities of the Zhejiang province in China. Luqiao is a town with the largest site for disassembly of PCB-containing obsolete transformers and electrical waste in China. The other two localities Pingqiao (100 km NW of Luqiao) and Lin'an (500 km NW of Luqiao) are towns without known electronic or electrical waste sites. A total of 23 PCB congeners (including 12 dioxin-like) and 6 OCPs were measured using the traditional GC-mu ECD technique. Micro-EROD bioassay was additionally used to measure TCDD-based TEQ levels of the 12 dioxin-like PCBs. Significant correlations were found between the TEQs measured by the two methods, supporting the application of micro-EROD as a practical toot for complementing the chemical analysis. The data showed that beta-HCH, p,p'-DDE, and 6 PCB congeners (101, 138 153, 180, 183, and 187) were the predominant pollutants, with PCB 138 being the best indicator (predictor) for total PCB levels. Cord blood and meconium from Luqiao have higher levels of PCBs than those from the other two localities, suggesting that a disassembly site for electronic and electric waste would provide an environment for greater exposure to these chemicals. The cord blood or meconium levels of beta-HCH, though likewise considerably high, were comparable in the three localities. Similar findings were observed for p,p'-DDE. Pollution by these OCPs might have come from past use of agricultural pesticides in the three localities. (c) 2007 Published by Elsevier B.V.

Lake Dianchi is in Yunnan Province in southwestern China. In recent years, significant cyanobacterial blooms have occurred in this lake nearly every year because of eutrophication. Monitoring data for the past 5 years acquired by our research group showed that phytoplankton composition alternated between species of Microcystis sp. during warm seasons and those of Aphanizomenon sp. during cool seasons. In March 2003, when phytoplankton composition was highly dominated by Aphanizomenon sp., samples were taken from the lake for toxin detection and immediate strain isolation. A mouse bioassay with extracts from the lyophilized field material showed obvious intoxication from paralytic shellfish poisons (PSPs), and all mice died within 30 min. Further analysis of both field and isolated algal strain Aphanizomenon DC-1 by the postcolumn HPLC-FLD method confirmed its PSP-producing ability The analogues found in the extracts from the field material were neoSTX, dcSTX, and dcGTX3, with contents of 2.279, 1.135, and 0.547 ng/mg DW, respectively. Under laboratory culture condition, toxin content in the Aphanizomenon strain DC-1 varied greatly during different growth phases, with two peaks: in the early-exponential and late-stationary growth phases. When the culture grew at a relatively high rate during the mid- to late-exponential growth phase, toxin content declined gradually. Moreover, the types of toxin in the DC-1 strain varied greatly during a single culture cycle. The HPLC results showed that dcSTX was the only toxin isomer detected throughout the culture period, and its level remained stable. On the other hand, dcGTX2 and GTX4 were the major toxins during the early-exponential and stationary phases, respectively. This article presents the first data on the identification and detection of paralytic shellfish toxins from cyanobacteria in Lake Dianchi. As far as we know, this is also the first report of this type of toxin in inland water bodies in China. Our study indicates the threat associated with PSP toxins in Lake Dianchi and suggests that necessary measures and programs for control are urgently needed to prevent the spread of toxic cyanobacterial blooms.
