Living bacteria or yeast cells are frequently used as bioreporters for the detection of specific chemical analytes or conditions of sample toxicity. In particular, bacteria or yeast equipped with synthetic gene circuitry that allows the production of a reliable non-cognate signal (e.g., fluorescent protein or bioluminescence) in response to a defined target make robust and flexible analytical platforms. We report here how bacterial cells expressing a fluorescence reporter ("bactosensors"), which are mostly used for batch sample analysis, can be deployed for automated semi-continuous target analysis in a single concise biochip. Escherichia coli-based bactosensor cells were continuously grown in a 13 or 50 nanoliter-volume reactor on a two-layered polydimethylsiloxane-on-glass microfluidic chip. Physiologically active cells were directed from the nl-reactor to a dedicated sample exposure area, where they were concentrated and reacted in 40 minutes with the target chemical by localized emission of the fluorescent reporter signal. We demonstrate the functioning of the bactosensor-chip by the automated detection of 50 μgarsenite-As l(-1) in water on consecutive days and after a one-week constant operation. Best induction of the bactosensors of 6-9-fold to 50 μg l(-1) was found at an apparent dilution rate of 0.12 h(-1) in the 50 nl microreactor. The bactosensor chip principle could be widely applicable to construct automated monitoring devices for a variety of targets in different environments.

The Laboratory Waste Management Program of the Chemistry Department of UFPR started on 1997 and was developed to meet the requirements of co-processing in cement kiln and those of the respective regulation. The in-lab procedures for waste collection and treatment were devised taking into account their cost, simplicity and wide range of application to the various types of residues generated. The program works with a five step annual journey : 1) Waste collection and treatment, 2) Bulk Storage, 3) Licensing (for transportation and co-processing), 4) Transportation and 5) Co-processing.

A didactic experiment is proposed in order to identify inorganic phosphates in home laundry detergents by ascending paper chromatography. Chemical principles related with hard water are also investigated. The role of polyphosphates in detergent formulations is discussed and the softening of hard water by sodium tripolyphosphate is illustrated. Several chemical concepts, related to the daily experience of the students, can be explored in post-lab discussions.

La reconstrucción de la historia de la madre del conocido Estanislao Severo Zeballos es el objetivo de este artículo que se centra en la búsqueda del rol ejercido por Felisa Jordán durante la organización del Estado-Nación de la República Argentina. Zeballos divulgó algunos datos de su vida privada, como su parentesco con los Urquiza y los Ruiz Moreno, o su condición de propietario de 351.562 hectáreas en el Chaco Boreal gracias a la mediación que practicó en el transcurso de la firma del tratado boliviano-paraguayo que fijó la línea de statu-quo de 1907. Pero el relato más creativo del rosarino Estanislao fue la gestión de su madre Felisa Jordán de Zeballos en el marco del ámbito asociativo de la ciudad de Rosario. Para describir esta llamativa vinculación entre Zeballos y su madre Felisa Jordán me he centrado en dos fuentes documentales singulares que nos permiten abordar las prácticas asociativas, los espacios públicos y la proyección femenina en Argentina entre 1869 y 1872. En primer lugar en los documentos privados de las mujeres de élite de la Ciudad de Rosario, en particular las actas y la correspondencia de la asociación benéfica Damas de Caridad. Esta documentación signó durante años mi investigación sobre el Asilo de Huérfanos y Expósitos cuyo nombre fue modificado por Hogar del Huérfano por el peronismo, una institución que hoy día se encuentra en un futuro incierto. La secretaria de aquella asociación femenina fue la llamativa Felisa Jordán, la viuda del militar Estanislao Zeballos, que colaboraba activamente con la presidenta de la asociación Damas de Caridad, María de los Ángeles Rodríguez de Rosas, y con la tesorera Blanca Musch de Villegas en cuyo domicilio se reunieron por primera vez para formar la entidad en el año 1869, es decir, en plena Guerra de la Triple Alianza. La documentación de las hoy llamadas Damas de Protección al Huérfano se conserva en el archivo del Hogar del Huérfano, otrora denominado Hogar de Huérfanos y Expósitos, y hace referencia a aspectos de la vida cotidiana de la entidad, de la asociación femenina y de sus socias.

El artículo que presentamos a continuación recoge el proyecto Trans formare. Lecturas de la Transformación del Museo Frederic Marès, relatando la experiencia de ocho artistas vinculados al Equipo Confluencias [The Crossing Lab] de la Facultat de Belles Arts de la Universidad de Barcelona (convenios ref. UB 007095 y UB 008992), que a lo largo del período de renovación del Museu Frederic Marès de Barcelona (2009-2011) han revisitado las salas de este espacio museográfico en tránsito. Durante estas circunstancias especiales, las obras, protagonistas del Museo, carentes de su aura habitual y de la rigidez del contexto expositivo, han permitido una proximidad y una cercanía poco corrientes; siendo esta particularidad el punto de partida de Trans formare: una investigación en torno al objeto artístico y su imagen, a través de las experiencias de transformación y memoria. Asimismo, el proyecto ha buscado releer el trabajo de Aby Warburg, sus propuestas de catalogación iconográfica y, muy especialmente, sus Mnemosyne, permitiendo con ello aportar nuevas lecturas y perspectivas interpretativas al recorrido museográfico. En este artículo se pretende, pues, analizar y describir el proyecto, sus bases ideológicas y sus resultados, que han dado lugar a una exposición temporal con la que el Museo Marès ha reabierto sus puertas, y que puede visitarse hasta enero de 2012.

A simple and low cost device (ca. US$ 150) that comprises two photodiodes fixed in lab-made Perspex flow cell is proposed for chemiluminescence measurements. The characteristics of the device (large observation window and reduced thickness) allow maximizing the amount of the emitted radiation detected. A sensitivity improvement of ca. 50 % was observed by employing two photodiodes for signal measurements. The performance of the device was assessed by the oxidation of luminol by hydrogen peroxide, yielding a linear response within the range of 2.50 to 500 µmol L-1 H2O2. The detection limit was estimated as 0.8 µmol L-1 hydrogen peroxide which is comparable with those obtained by using equipments based on photomultipliers.

An experiment is proposed to introduce some fundamentals of flow analysis, chemiluminescence and kinetic monitoring of enzymatic reactions in undergraduate courses. Chemiluminescence detection is performed with a simple spectrophotometer equipped with a lab-made spiral flow cell constructed from a polyethylene tube. The hydrogen peroxide produced by the glucose oxidation in the presence of glucose oxidase is continuously monitored by the reaction with luminol in alkaline media in a flow injection system. The exercise allows also the discussion of important analytical features and the comparison with different optical methods of analysis.

The discussion based on surfactant cleaning action is commonly used in chemistry classrooms for the comprehension of theoretical concepts related to surface tension and micelle formation. The experimental quantification of surface tension of aqueous solutions of surfactants provides the practical instruments for this discussion. The present paper describes a simplification on the common apparatus employed for the drop-weight method, making the determination of surface tension accessible to any chemistry lab. The surface tension of various liquids and the critical micelle concentration, CMC, of three commercial surfactants were measured with this modified method, and proved to be consistent with literature values.

Theoretical and practical aspects of the use of microwave-assisted strategies in chemistry are introduced for students using simple and safe experiments employing a domestic oven. Three procedures are proposed for evaluating the distribution of microwave radiation inside the microwave oven cavity: (1) variation of the volume of marshmallows; (2) drying of filter paper wetted with Co(II) solution, and (3) variation of water temperature, after microwave-assisted heating. These experiments establish the position with the highest incidence of microwave radiation in the oven cavity, which was chosen for the synthesis of salicylic acid acetate. This synthesis was performed in 5 min of heating and the yield was around 85%. All experiments can be carried out in a 4 h lab-session using low-cost instrumentation.

The environmental impact of detergents and other consumer products is behind the continued interest in the chemistry of the surfactants used. Of these, linear alkylbenzene sulfonates (LASs) are most widely employed in detergent formulations. The precursors to LASs are linear alkylbenzenes (LABs). There is also interest in the chemistry of these hydrocarbons, because they are usually present in commercial LASs (due to incomplete sulfonation), or form as one of their degradation products. Additionally, they may be employed as molecular tracers of domestic waste in the aquatic environment. The following aspects are covered in the present review: The chemistry of surfactants, in particular LAS; environmental impact of the production of LAS; environmental and toxicological effects of LAS; mechanisms of removal of LAS in the environment, and methods for monitoring LAS and LAB, the latter in domestic wastes. Classical and novel analytical methods employed for the determination of LAS and LAB are discussed in detail, and a brief comment on detergents in Brazil is given.

By the year 2005 the world biochemical market will reach an estimated $ 100 billion and separation processes are a vital link between lab discoveries and the fulfillment of this commercialization potential. The practical application of aqueous two-phase systems (ATPS) to extraction processes has been exploited for several years for the recovery of biological products. Unfortunately, this has not resulted in an extensive presence of the technique in commercial processes. In this paper a critical overview of the fundamental thermodynamic properties related to formation of aqueous two-phase systems and their application to extraction and purification of bioparticules is presented.

Actualment a Catalunya existeixen zones amb importants limitacions per l’aplicació de purins al sòl, pel que és imprescindible trobar alternatives de gestió i tractament que permetin l’aprofitament adequat dels recursos continguts a les dejeccions ramaderes sense afectar el medi. La digestió anaeròbia és una de les tècniques utilitzades en el tractament de les dejeccions ramaderes. L’efluent líquid que s’obté d’aquest tractament no modifica el contingut de nitrogen i fòsfor i per tant ha de ser gestionat correctament. L’objectiu general d’aquest projecte és avaluar la precipitació d’estruvita (sal de magnesi, amoni i fosfat) com una alternativa de gestió de l’efluent líquid d’una planta de digestió anaeròbia i compostatge que tracta dejeccions ramaderes conjuntament amb altres residus orgànics. S’han avaluat els efectes dels diferents paràmetres operacionals en la formació d’estruvita (pH, temperatura, velocitat d’agitació, alcalinitat), mitjançant assaigs en discontinu amb solució sintètica. A continuació s’ha procedit a obtenir estruvita a partir de la fracció líquida digerida de purí (FLD), en assaigs en discontinu per estudiar l’efecte del contingut de matèria orgànica i sòlids Totals (ST), així com el contingut en fosfats i el pH de reacció. Finalment, s’han optimitzat els paràmetres de procés en continu, mitjançant la posada en marxa d’un reactor a escala de laboratori i estudi de l’efecte de la velocitat d’agitació i de la introducció del stripping de CO2, tant amb solució sintètica com amb la fracció líquida digerida del purí. Dels resultats obtinguts es pot concloure que els factors que tenen una major influència en el procés d’obtenció d’estruvita són el pH (el pH òptim es situa al voltant de 9), i la presència de matèria orgànica i sòlids ens suspensió, que interfereix de forma quantitativa i qualitativa en la formació de l’estruvita. En el procés en continu s’ha aconseguit reduccions d’un 84% i 98% d’amoni i fòsfor respectivament, obtenintse estruvita que pot ser utilitzada com a fertilitzant d’alliberació lenta. Es pot concloure que la precipitació d’estruvita és una bona alternativa per millorar la gestió de les dejeccions ramaderes alhora que permet recuperar nutrients i tancar cicles. La combinació amb un tractament previ que elimini la matèria orgànica, com podria ser la digestió anaeròbia, i una separació de fases, per eliminar els sòlids en suspensió, es presenta com una configuració amb molts avantatges.

The performances of eight kinds of insulators from electrical distribution lines in Salvador-BA, Brazil, were evaluated considering the chemical and physical local environmental pollution. The parameters that were chosen as characteristic for the insulators' properties were leakage current and partial electrical discharge. A data storage processing system and a communication link to the lab were built for data acquisition. The results show that the main contribution to the poor performance of the insulators is settleable magnetite particulate matter on the insulator in addition to the long term wetness time, t4.

The present paper reports a bacteria autonomous controlled concentrator prototype with a user-friendly interface for bench-top applications. It is based on a micro-fluidic lab-on-a-chip and its associated custom instrumentation, which consists in a dielectrophoretic actuator, to pre-concentrate the sample, and an impedance analyser, to measure concentrated bacteria levels. The system is composed by a single micro-fluidic chamber with interdigitated electrodes and a instrumentation with custom electronics. The prototype is supported by a real-time platform connected to a remote computer, which automatically controls the system and displays impedance data used to monitor the status of bacteria accumulation on-chip. The system automates the whole concentrating operation. Performance has been studied for controlled volumes of Escherichia coli (E. coli) samples injected into the micro-fluidic chip at constant flow rate of 10 μL/min. A media conductivity correcting protocol has been developed, as the preliminary results showed distortion of the impedance analyser measurement produced by bacterial media conductivity variations through time. With the correcting protocol, the measured impedance values were related to the quantity of bacteria concentrated with a correlation of 0.988 and a coefficient of variation of 3.1%. Feasibility of E. coli on-chip automated concentration, using the miniaturized system, has been demonstrated. Furthermore, the impedance monitoring protocol had been adjusted and optimized, to handle changes in the electrical properties of the bacteria media over time.
