Ankylosing spondylitis (AS) is a chronic inflammatory rheumatic disease of the axial skeleton. The major outcome of this disease is defined by new bone formation, commonly observed in the ligaments of the intervertebral joints, that can lead to the formation of bony spurs, known as syndesmophytes. Previous studies have shown that serum levels of TNF, IL-6 and IL-17 are increased in AS patients and may be implicated in the development of secondary osteoporosis, since these cytokines are able to induce osteoclast (OC) differentiation and, therefore, bone resorption. In this work we aimed to assess the effects of TNF-blocking therapy in the systemic inflammatory environment of AS patients with active disease as well as in OC differentiation and activity. To accomplish this objective, we cultured circulating monocytes from AS patients, before and after therapy, under osteoclastogenic conditions and we performed two functional assays (TRAP staining and resorption pit assay) and analyzed the expression of osteoclast specific genes. We have shown that AS patients with active disease have increased levels of pro-inflammatory cytokines when compared with healthy subjects. We also found that IL-17, TGF-β and osteoprotegerin are decreased after TNF-blocking therapy. Interestingly, we also observed that after TNF-blocking therapy the expression of some genes is favoring osteoclastogenesis and that differentiated OCs have increased resorption activity. These results suggest that in active AS there may be an uncoupling between inflammation and OC activity that is reset by TNF-blocking therapy.

The bacterium Geobacter sulfurreducens (Gs) is capable of oxidizing a large variety of compounds relaying electrons out of the cytoplasm and across the membrane in a process designated as extracellular electron transfer. The Gs genome was fully sequenced and a family composed by five periplasmic triheme cytochromes c7 (designated PpcA-E) was identified. These cytochromes play an important role in the reduction of extracellular acceptors. They contain approximately 70 amino acids, three heme groups with bis-histidinyl axial coordination, and share between 57 and 77% sequence identity. The triheme cytochrome PpcA is highly abundant in Gs and is most likely the reservoir of electrons destined for outer surface. In addition to its role in electron transfer pathways this protein can perform e-/H+ energy transduction, a process that is disrupted when the strictly conserved aromatic residue phenylalanine 15 is replaced by a leucine (PpcAF15L). This Thesis focuses on the expression, purification and characterization of these proteins using Nuclear Magnetic Resonance and ultraviolet-visible spectroscopy. The orientations of the heme axial histidine ring planes and the orientation of the heme magnetic axis were determined for each Gs triheme cytochrome. The comparison of the orientations obtained in solution with the crystal structures available showed significant differences. The results obtained provide the paramagnetic constraints to include in the future refinement of the solution structure in the oxidized state. In this work was also determined the solution structure and the pH-dependent conformational changes of the PpcAF15L allowing infer the structural origin for e-/H+ energy transduction mechanism as shown by PpcA. Finally, the backbone and side chain NH signals of PpcA were used to map interactions between this protein and the putative redox partner 9,10-anthraquinone-2,6-disulfonate (AQDS). In this work a molecular interaction was identified for the first time between PpcA and AQDS, constituting the first step toward the rationalization of the Gs respiratory chain.

Diffusion Kurtosis Imaging (DKI) is a fairly new magnetic resonance imag-ing (MRI) technique that tackles the non-gaussian motion of water in biological tissues by taking into account the restrictions imposed by tissue microstructure, which are not considered in Diffusion Tensor Imaging (DTI), where the water diffusion is considered purely gaussian. As a result DKI provides more accurate information on biological structures and is able to detect important abnormalities which are not visible in standard DTI analysis. This work regards the development of a tool for DKI computation to be implemented as an OsiriX plugin. Thus, as OsiriX runs under Mac OS X, the pro-gram is written in Objective-C and also makes use of Apple’s Cocoa framework. The whole program is developed in the Xcode integrated development environ-ment (IDE). The plugin implements a fast heuristic constrained linear least squares al-gorithm (CLLS-H) for estimating the diffusion and kurtosis tensors, and offers the user the possibility to choose which maps are to be generated for not only standard DTI quantities such as Mean Diffusion (MD), Radial Diffusion (RD), Axial Diffusion (AD) and Fractional Anisotropy (FA), but also DKI metrics, Mean Kurtosis (MK), Radial Kurtosis (RK) and Axial Kurtosis (AK).The plugin was subjected to both a qualitative and a semi-quantitative analysis which yielded convincing results. A more accurate validation pro-cess is still being developed, after which, and with some few minor adjust-ments the plugin shall become a valid option for DKI computation

Os elementos traço são elementos que existem no organismo humano e que se apresentam em concentrações variadas. A variação da concentração destes elementos pode levar a intoxicações ou mesmo à formação de tecidos cancerígenos no organismo humano. Neste trabalho efectuaram-se dois estudos de elementos traço no organismo, sendo que numa das situações foram utilizados ratos Wistar e na outra situação utilizaram-se amostras de tecidos humanos recolhidos em biópsias. Foi utilizada a técnica de fluorescência de raios-X na análise de tecidos, nomeadamente, a espectroscopia de micro-fluorecência de raios-X ( -XRF) e a espectroscopia de energia dispersiva de raios-X com um sistema tri-axial (EDXRF). No estudo dos ratosWistar, pretendeu-se analisar a intoxicação por chumbo no organismo através da adição de acetato de chumbo na água ingerida. Conclui-se deste estudo que existe uma correlação positiva entre a acumulação de chumbo no organismo e a sua excreção através das unhas e do pêlo, sendo que o pêlo poderá vir a servir como método de diagnóstico de intoxicação por metais pesados. No estudo de tecidos cancerígenos pretendeu-se observar as possíveis alterações de elementos no organismo quando o tecido contém uma neoplasia. Analisaram-se amostras de tecidos de rim, pulmão e cólon, saudáveis e com neoplasia e concluiu-se que efectivamente existem alterações nas concentrações de elementos traço de amostras saudáveis para amostras com neoplasia variando estas concentrações de tecido para tecido.

Nesta dissertação é realizado o estudo de um motor em disco polifásico com armadura convencional e rotor que pode ser convencional, em alumínio, ou composto por material supercondutor de alta temperatura multi‐semente. O motor com o rotor em alumínio apresenta um comportamento assíncrono bem definido, baseado na lei geral de indução de eletromotrizes no induzido. Contudo, ao ser mergulhado em azoto líquido, evidencia melhores características eléctricas, tanto do induzido como do indutor, mesmo que o núcleo ferromagnético convencional possa exibir degradação magnética. O motor com o rotor SAT exibe um comportamento distinto, evidenciando ambos os regimes síncrono e assíncrono estáveis. O regime síncrono fica caracterizado pelo fenómeno de aprisionamento de campo, que garante que o rotor gire síncrono com o campo girante do estator. Em regime assíncrono, para um determinado valor de binário resistente aplicado superior ao electromagnético máximo desenvolvido pelo motor, o fenómeno de escoamento de campo está presente, sendo o binário electromagnético motor reforçado por um binário extra de perdas de carácter resistivo desenvolvidas no rotor devido à interacção da densidade de corrente a campos eléctricos que se estabelecem no rotor. Neste caso, o fenómeno de escoamento de campo não é devido a forças de Lorentz que se definem na matriz de Abrikosov, mas sim devido a esta ser “puxada” pelo campo girante do estator, ficando a matriz a deslizar sobre o rotor. De modo a obter as características dos sistemas electromecânicos em estudo é realizada uma análise teórica, baseada na teoria convencional das máquinas eléctricas, com o objectivo de compreender alguns dos fenómenos do motor de fluxo axial, e uma análise baseada num programa comercial de elementos finitos onde o fenómeno da supercondutividade é simulado com base na relação entre o campo elétrico e a densidade de corrente pela lei da potenciação (E‐J power law). O ensaio dos motores referidos é realizado para comparar ambos os sistemas e de realçar o princípio de funcionamento e as características mais relevantes de cada um. Da análise realizada é desenvolvido um modelo que caracteriza o funcionamento da máquina com o rotor com SAT.

A crescente dependência da energia eléctrica tem proporcionado o desenvolvimento do actual sistema energético, garantindo a continuação da produção e distribuição da electricidade, mantendo sempre um adequado balanço na relação qualidade/preço. Este sistema encontra-se explorando novas fontes de energias, de caracter renováveis, elevando assim a complexidade da rede energética e a probabilidade de ocorrerem falhas devido a correntes de curto-circuito (CC). Como alternativas para resolver ou prevenir os problemas que advêm das correntes de CC, encontram-se disponíveis dispositivos frequentemente utilizados na limitação das correntes na rede, mas que têm custos importantes associados à substituição e/ou manutenção dos equipamentos. Novas tecnologias que utilizam supercondutividade como principal componente, têm apresentado desempenhos satisfatórios, nomeadamente os limitadores de corrente supercondutores, os quais têm como característica particular, serem invisíveis à rede em funcionamento normal e apresentar elevada impedância em situação de falha, assim como menores tempos de recuperação. De forma a contribuir para a optimização destes limitadores, é importante identificar e caracterizar os esforços mecânicos aos quais se encontram sujeitos em situações de falha, nomeadamente os seus enrolamentos. Neste trabalho de dissertação, estudam-se e simulam-se os principais esforços electrodinâmicos em transformadores e, principalmente, nos enrolamentos de alguns limitadores de corrente supercondutores.

The momentum anomaly has been widely documented in the literature. However, there are still many issues where there is no consensus and puzzles left unexplained. One is that strategies based on momentum present a level of risk that is inconsistent with the diversification that it offers. Moreover, recent studies indicate that this risk is variable over time and mostly strategy-specific. This work project hypothesises and proves that this evidence is explained by the portfolio constitution of the momentum strategy over time, namely the covariance and correlation between companies in the top and down deciles and across them.

Esta dissertação foi desenvolvida no Departamento de Engenharia Civil (DEC) da Faculdade de Ciências e Tecnologia (FCT) da Universidade Nova de Lisboa (UNL). Tem como objetivo estudar o comportamento da ligação pavimento/parede de edifícios antigos. Primeiramente, foi realizada uma pesquisa bibliográfica sobre alvenaria de pedra, pavimentos de madeira e ligações pavimento/parede em alvenarias antigas. De seguida, foram analisados alguns trabalhos de investigação relacionados com o tema da presente dissertação, sobre alvenarias de pedra, pavimentos de madeira e respetivas ligações. A parte experimental desta dissertação foi desenvolvida no laboratório de estruturas do DEC da FCT da UNL. Teve como objetivo estudar a simulação de duas ligações pavimento/parede: uma, entre um pavimento e o murete de alvenaria de pedra sem reforço; e, outra, entre um pavimento e um murete de alvenaria de pedra reforçado com uma camada de microbetão e rede metálica. Posteriormente aos ensaios de simulação da ligação, foram efetuados ensaios de compressão axial nos dois muretes. A seguir foi determinada a profundidade de carbonatação. A partir dos resultados obtidos foi estudado o mecanismo de ferrolho para alvenaria de pedra e feita a verificação das tensões nos dois muretes. Os resultados obtidos nesta dissertação foram comparados com os resultados de trabalhos anteriormente realizados por outros autores, em especial, com a dissertação anterior a esta (Martins, 2014), a qual deu origem à presente linha de investigação. Uma das principais conclusões do estudo mostrou que, as ligações pavimento/parede realizadas no murete de alvenaria reforçado com lâmina de microbetão resistiu aproximadamente o dobro da resistência que as ligações realizadas no murete de alvenaria não reforçado.

The first part of this research work regards the assessment of the mathematical modelling of reinforced concrete columns confined with carbon fibre (CFRP) sheets under axial loading. The purpose was to evaluate existing analytical models, contribute to possible improvements and choose the best model(s) to be part of a new model for the prediction of the behaviour of confined columns under bending and compression. For circular columns, a wide group of authors have proposed several models specific for FRP-confined concrete. The analysis of some of the existing models was carried out by comparing these with several tested columns. Although several models predict fairly the peak load only few can properly estimate the load-strain and dilation behaviour of the columns. Square columns confined with CFRP show a more complex interpretation of their behaviour. Accordingly, the analysis of two experimental programs was carried out to propose new modelling equations for the whole behaviour of columns. The modelling results show that the analytical curves are in general agreement with the presented experimental curves for a wide range of dimensions. An analysis similar to the one done for circular columns was this turn carried out for square columns. Few models can fairly estimate the whole behaviour of the columns and with less accuracy at all levels when compared with circular columns. The second part of this study includes seven experimental tests carried out on reinforced concrete rectangular columns with rounded corners, different damage condition and with confinement and longitudinal strengthening systems. It was concluded that the use of CFRP confinement is viable and of effective performance enhancement alone and combined with other techniques, maintaining a good ductile behaviour for established threshold displacements. As regards the use of external longitudinal strengthening combined with CFRP confinement, this system is effective for the performance enhancement and viable in terms of execution. The load capacity was increased significantly, preserving also in this case a good ductile behaviour for threshold displacements. As to the numerical nonlinear modelling of the tested columns, the results show a variation of the peak load of 1% to 10% compared with tests results. The good results are partly due to the inclusion of the concrete constitutive model by Mander et al. modified by Faustino, Chastre & Paula taking into account the confinement effect. Despite the reasonable approximation to tests results, the modelling results showed higher unloading, which leads to an overestimate dissipated energy and residualdisplacement.

Noonan syndrome is a multiple congenital anomaly syndrome, inherited in an autosomal dominant pattern. We studied 31 patients (18 males and 13 females) affected by this disorder regarding their clinical and genetic characteristics. The most frequent clinical findings were short stature (71%); craniofacial dysmorphisms, especially hypertelorism, ptosis, downslanting of the palpebral fissures; short or webbed neck (87%); cardiac anomalies (65%), and fetal pads in fingers and toes (70%). After studying the probands' first-degree relatives, we made the diagnosis of Noonan syndrome in more than one family member in three families. Therefore, the majority of our cases were sporadic.

PURPOSE: To report the experience of a radiology department in the use of computed tomography - guided cutting needle biopsy of pulmonary nodules, by evaluating diagnostic yield and incidence of complications. METHODS: This is a retrospective analysis of 52 consecutive patients who underwent lung lesion biopsy guided by computed tomography, performed between May 1997 and May 2000. Thirty-five patients were male and 17 were female, with ages ranging from 5 to 85 years (median, 62 years). The size of the lesions ranged from 1.8 to 15 cm (median, 5.4 cm). RESULTS: In a total of 52 biopsies of lung lesions, 51 biopsies (98.1%) supplied appropriate material for histopathological diagnosis, with 9 diagnosed (17.3%) as benign and 42 (80.8%) as malignant lesions. Specific diagnosis was obtained in 44 (84.6%) biopsies: 4 benign (9.1%) and 40 (90.9%) malignant lesions. The sensitivity, specificity, and accuracy of the cutting needle biopsies for determining presence of malignancy were 96.8%, 100%, and 97.2%, respectively. Complications occurred in 9 cases (17.3%), including 6 cases (11.5%) of small pneumothorax, 1 (1.9%) of hemoptysis, 1 (1.9%) of pulmonary hematoma, and 1 (1.9%) of thoracic wall hematoma. All had spontaneous resolution. There were no complications requiring subsequent intervention. CONCLUSION: The high sensitivity and specificity of the method and the low rate of complications have established cutting needle biopsy as an efficient and safe tool for the diagnosis of lung lesions. In our hospital, cutting needle biopsy is considered a reliable procedure for the evaluation of indeterminate pulmonary nodules.

OBJECTIVE: Noonan syndrome is a multiple congenital anomaly syndrome, and bleeding diathesis is considered part of the clinical findings. The purpose of this study was to determine the frequency of hemostatic abnormalities in a group of Noonan syndrome patients. METHOD: We studied 30 patients with clinical diagnosis of Noonan syndrome regarding their hemostatic status consisting of bleeding time, prothrombin time, activated partial thromboplastin time and thrombin time tests, a platelet count, and a quantitative determination of factor XI. RESULTS: An abnormal laboratory result was observed in 9 patients (30%). Although coagulation-factor deficiencies, especially factor XI deficiency, were the most common hematological findings, we also observed abnormalities of platelet count and function in our screening. CONCLUSIONS: Hemostatic abnormalities are found with some frequency in Noonan syndrome patients (30% in our sample). Therefore, we emphasize the importance of a more extensive hematological investigation in these patients, especially prior to an invasive procedure, which is required with some frequency in this disorder.
