Bacillus thuringiensis (Bt) transgenic cotton has shown changes of vegetative and reproductive growth characteristics. The objective of this study was to investigate the physiological change of nitrogen metabolism that related closely to the growth in Bt cotton cultivars. The study Was undertaken on two 131 transgenic cotton cultivars and their parents, one conventional (Xingyang822) and recurrent parent (Sumian No. 9), the other a hybrid (Kumian No. 1) and female parent (Yumian No. 1), during the 2001 and 2002 growing seasons at the Yangzhou University Farm, Yangzhou, China. In the 2001 study, The results indicated that the Bt cotton cultivars were higher than their parents in leaf total nitrogen, free amino acid and soluble protein content, greater in NR and GPT activity, and lower in protease activity, during peak square and boll developing period. The biggest increase of total nitrogen was at peak boll period, which increased by 36.01 and 18.96% for Kumian No. I and Xingyang822, respectively. There were similar results for free amino acid and soluble protein content. The results showed further in 2002 study that NR activity increased dramatically at peak square and early boll open period, the biggest increase at early boll open period, with Kumian No. I and Xingyan,822 being 87.5 and 61.4% higher than their parent, respectively, the biggest increase of GPT activity was at peak boll period, with Kumian No. I and Xingyang822 being 39.1 and 29.1% higher than their parent, respectively. However, protease activity of Bt cultivars reduced significantly before flowering and early boll open period, the biggest decrease was before flowering period, with Kumian No. I being more than 30%, Xingyang822 being 26.5% at peak square period. Moreover, the boll total nitrogen content reduced sharply. The results suggest that the Bt cotton cultivars have higher intensity of leaf nitrogen metabolism than their parent, especially during square and boll development period. It is disadvantage for square development and earlier boll maturity under high nitrogen condition. The cultural practice should aim at reducing leaf nitrogen metabolic strength and keep the balance of vegetative and reproductive growth. (C) 2003 Elsevier B.V. All rights reserved.

Bt transgenic cotton has not shown the same level of resistance to bollworm in China, as in other major Bt cotton growing areas of the world. The objective of this study was to investigate the effects of high temperature on the CryIA insecticidal protein content and nitrogen metabolism, in the leaf of Bt transgenic cotton. The study was undertaken on two transgenic cotton cultivars, one conventional (Xinyang 822) and the other a hybrid (Kumian No. 1), during the 2001 and 2002 growing seasons at the Yangzhou University Farm, Yangzhou, China. In the 2001 study, potted cotton plants were exposed to 37 C for 24 h under glasshouse conditions at three growth stages peak square, peak flowering and peak boll developing periods. Based on the 2001 results, in 2002 the same two cultivars were exposed to the same temperature for 48 h at two growth stages-peak flowering and boll developing periods. The results of the study indicated that the insecticidal protein content of the leaf was not significantly affected by the stress during the square and flowering periods. However, exposure to high temperature for 24h during the boll period reduced the CryIA protein content by approximately 51% in the cultivar Kumian No 1, and 30% in Xinyang 822 in the 2001 study, and by approximately 73 and 63% for 48 h with the same cultivars, respectively, in the 2002 study. Glutamic-pyruvic transaminase (GPT) activity, total free amino acid and soluble protein content, and the activity of protease in the leaf, showed relatively little change in response to high temperature in the flowering period. However, exposure to high temperature in the boll period resulted in the following changes - a reduction of GPT activity, a sharp increase in free amino acid content, a significant decrease in soluble protein content, and significant increases in the activity of protease. The results suggest that high temperature may result in the degradation of soluble protein in the leaf, with a resulting decline in the level of the toxin CryIA. It is believed that this may be the cause of the reduced efficacy of Bt cotton in growing conditions in China, where temperatures during the boll period often reach 36-40° C. © 2004 Elsevier B.V All rights reserved.

OBJECTIVE: To compare the accuracy, costs and utility of using the National Death Index (NDI) and state-based cancer registries in determining the mortality status of a cohort of women diagnosed with ovarian cancer in the early 1990s. METHODS: As part of a large prognostic study, identifying information on 822 women diagnosed with ovarian cancer between 1990 and 1993, was simultaneously submitted to the NDI and three state-based cancer registries to identify deceased women as of June 30, 1999. This was compared to the gold standard of "definite deaths". A comparative evaluation was also made of the time and costs associated with the two methods. RESULTS: Of the 450 definite deaths in our cohort the NDI correctly identified 417 and all of the 372 women known to be alive (sensitivity 93%, specificity 100%). Inconsistencies in identifiers recorded in our cohort files, particularly names, were responsible for the majority of known deaths not matching with the NDI, and if eliminated would increase the sensitivity to 98%. The cancer registries correctly identified 431 of the 450 definite deaths (sensitivity 96%). The costs associated with the NDI search were the same as the cancer registry searches, but the cancer registries took two months longer to conduct the searches. CONCLUSIONS AND IMPLICATIONS: This study indicates that the cancer registries are valuable, cost effective agencies for follow-up of mortality outcome in cancer cohorts, particularly where cohort members were residents of those states. For following large national cohorts the NDI provides additional information and flexibility when searching for deaths in Australia. This study also shows that women can be followed up for mortality with a high degree of accuracy using either service. Because each service makes a valuable contribution to the identification of deceased cancer subjects, both should be considered for optimal mortality follow-up in studies of cancer patients.

Esta pesquisa evidencia o uso de instrumentos de comunicação interna e dirigida e de ações de relações públicas e marketing de relacionamento dentro das igrejas evangélicas históricas. Também enfatiza a importância da comunicação interna e das estratégias de comunicação mercadológicas utilizadas dentro das igrejas inclusive com o uso do marketing boca a boca. Para a realização desta pesquisa foi feito um recorte das quatro maiores igrejas evangélicas históricas, em número de membros, situadas na cidade de Curitiba: Batista, Luterana, Presbiteriana e Metodista. O estudo foi realizado por meio de pesquisas de observação participante, análise de materiais diversos, entrevista com os líderes e uma pesquisa de campo, quantitativa, envolvendo os membros e freqüentadores das igrejas. Com o auxilio de um questionário estruturado foram analisados: 1) o comportamento dos membros e freqüentadores diante dos instrumentos de comunicação interna e dirigida que estão sendo utilizados e percebidos; e 2) os relacionamentos dentro da igreja, tanto entre membros quanto entre estes e seus líderes. A pesquisa concluiu que estes instrumentos de comunicação interna possuem relação com a retenção, perda e crescimento numérico de membros nas igrejas pesquisadas, frente a uma nova configuração religiosa onde a fidelização está cada vez mais difícil de ser alcançada.(AU)

Background and Purpose Although it is established that the receptor activity modifying proteins (RAMPs) can interact with a number of GPCRs, little is known about the consequences of these interactions. Here the interaction of RAMPs with the glucagon-like peptide 1 receptor (GLP-1 receptor), the human vasoactive intestinal polypeptide/pituitary AC-Activating peptide 2 receptor (VPAC) and the type 1 corticotrophin releasing factor receptor (CRF) has been examined. Experimental Approach GPCRs were co-transfected with RAMPs in HEK 293S and CHO-K1 cells. Cell surface expression of RAMPs and GPCRs was examined by elisa. Where there was evidence for interactions, agonist-stimulated cAMP production, Ca mobilization and GTPγS binding to G, G, G and G were examined. The ability of CRF to stimulate adrenal corticotrophic hormone release in Ramp2 mice was assessed. Key Results The GLP-1 receptor failed to enhance the cell surface expression of any RAMP. VPAC enhanced the cell surface expression of all three RAMPs. CRF enhanced the cell surface expression of RAMP2; the cell surface expression of CRF was also increased. There was no effect on agonist-stimulated cAMP production. However, there was enhanced G-protein coupling in a receptor and agonist-dependent manner. The CRF: RAMP2 complex resulted in enhanced elevation of intracellular calcium to CRF and urocortin 1 but not sauvagine. In Ramp2 mice, there was a loss of responsiveness to CRF. Conclusions and Implications The VPAC and CRF receptors interact with RAMPs. This modulates G-protein coupling in an agonist-specific manner. Peptides are of great therapeutic potential as vaccines and drugs. Knowledge of physicochemical descriptors, including the partition coefficient logP, is useful for the development of predictive Quantitative Structure-Activity Relationships (QSARs). We have investigated the accuracy of available programs for the prediction of logP values for peptides with known experimental values obtained from the literature. Eight prediction programs were tested, of which seven programs were fragment-based methods: XLogP, LogKow, PLogP, ACDLogP, AlogP, Interactive Analysis's LogP and MlogP; and one program used a whole molecule approach: QikProp. The predictive accuracy of the programs was assessed using r(2) values, with ALogP being the most effective (r( 2) = 0.822) and MLogP the least (r(2) = 0.090). We also examined three distinct types of peptide structure: blocked, unblocked, and cyclic. For each study (all peptides, blocked, unblocked and cyclic peptides) the performance of programs rated from best to worse is as follows: all peptides - ALogP, QikProp, PLogP, XLogP, IALogP, LogKow, ACDLogP, and MlogP; blocked peptides - PLogP, XLogP, ACDLogP, IALogP, LogKow, QikProp, ALogP, and MLogP; unblocked peptides - QikProp, IALogP, ALogP, ACDLogP, MLogP, XLogP, LogKow and PLogP; cyclic peptides - LogKow, ALogP, XLogP, MLogP, QikProp, ACDLogP, IALogP. In summary, all programs gave better predictions for blocked peptides, while, in general, logP values for cyclic peptides were under-predicted and those of unblocked peptides were over-predicted.
