Glycogen synthase, an enzyme involved in glycogen biosynthesis, is regulated by phosphorylation and by the allosteric ligand glucose-6-phosphate (G6P). In addition, enzyme levels can be regulated by changes in gene expression. We recently cloned a cDNA for glycogen synthase (gsn) from Neurospora crassa, and showed that gsn transcription decreased when cells were exposed to heat shock (shifted from 30degreesC to 45degreesC). In order to understand the mechanisms that control gsn expression, we isolated the gene, including its 5' and 3' flanking regions, from the genome of N. crassa. An ORF of approximately 2.4 kb was identified, which is interrupted by four small introns (II-V). Intron I (482 bp) is located in the 5'UTR region. Three putative Transcription Initiation Sites (TISs) were mapped, one of which lies downstream of a canonical TATA-box sequence (5'-TGTATAAA-3'). Analysis of the 5'-flanking region revealed the presence of putative transcription factor-binding sites, including Heat Shock Elements (HSEs) and STress Responsive Elements (STREs). The possible involvement of these motifs in the negative regulation of gsn transcription was investigated using Electrophoretic Mobility Shift Assays (EMSA) with nuclear extracts of N. crassa mycelium obtained before and after heat shock, and DNA fragments encompassing HSE and STRE elements from the 5'-flanking region. While elements within the promoter region are involved in transcription under heat shock, elements in the 5'UTR intron may participate in transcription during vegetative growth. The results thus suggest that N. crassa possesses trans-acting elements that interact with the 5'-flanking region to regulate gsn transcription during heat shock and vegetative growth.

Over the last decades, increasing water demands have fostered research to obtain high well yields in crystalline terrains where, besides the intrinsic properties of rocks, the groundwater flow depends on several factors. The depth of the wells, the lithotypes, the presence and thickness of sedimentary coverings and weathered layers, the landforms, the geological structures, and the effects of tectonic stresses are among the most investigated factors considered as determinant of well productivity. The influence of these factors on productivity of wells that exploit the Crystalline Aquifer System in the Jundiai-River Catchment, southeastern Brazil, is investigated in this work. The largest region of the studied area is located on the Precambrian Basement, partially covered by sedimentary deposits. The results show that the sedimentary deposits and the weathered layer are important for high well yield, but it also depends on the existence of a net of open fractures, in order to maintain high productivity. The sites that have more possibility of occurrence of such structures are the regional shear and fault zones and other minor structures with NW-SE and E-W directions, which characterize areas subjected to transtensional stress related to the neotectonics.

O presente trabalho teve por objetivo avaliar o ganho de peso vivo de bovinos submetidos a três tipos de pastagens de capim-colonião (Panicum maximum Jacq.): colonião testemunha (sem adubação nitrogenada); colonião adubado com 100 kg/ha de nitrogênio; e colonião consociado com soja perene (Neonotonia wightii Verde.) e centrosema (Centrosema pubescens Benth.). Foram usados novilhos da raça Guzerá, com idade variável de 12 a 15 meses e peso vivo médio de 210 kg. O sistema de pastejo foi rotativo, sendo o ajuste pasto-animal realizado pelo método de carga variável. As pesagens dos animais e amostragens do material vegetativo foram realizadas a cada 35 dias. Os resultados revelaram que, durante a época seca, os ganhos diários e por hectare referentes ao tratamento consociado (0,264 kg/animal/dia e 85,73 kg/ha, respectivamente) foram superiores aos das pastagens adubadas com nitrogênio (0,114 kg/animal/dia e 19,93 kg/ha) e testemunha (0,140 kg/animal/dia e 36,60 kg/ha). Ao final de quinze meses, as pastagens adubadas com nitrogênio e consociadas não apresentaram diferenças nos ganhos de peso vivo por hectare, sendo estes, entretanto, 16% superiores em relação ao da pastagem testemunha.

A frequency upconversion process in Pr(3+) doped TeO(2)-ZnO glasses containing silver nanoparticles is studied under excitation with a nanosecond laser operating at 590 nm, in resonance with the (3)H(4)-->(1)D(2) transition. The excited Pr(3+) ions exchange energy in the presence of the nanoparticles, originating efficient conversion from orange to blue. The enhancement in the intensity of the luminescence at similar to 482 nm, corresponding to the (3)P(0)-->(3)H(4) transition, is due to the influence of the large local field on the Pr(3+) ions, which are located near the metallic nanoparticles. The synthesis and crystal structure of two complexes resulting from interaction between NiBr2 and triphenylarsine oxide (Ph3AsO) is described. Green and orange complexes can be obtained from the blue, probably tetrahedral complex [NiBr2(Ph3AsO)2], depending on the solvents used for recrystallization. NiBr2·4[(C6H5)3AsO]·8H2O (green): M = 1650.2, P21/c, a = 13.731(2), b = 16.267(3), c = 17.647(2) Å, β = 112.04(1)°, V = 3651.4 Å3, Z = 2, Dx = 1.501 g cm-3, CuKα, λ = 1.54184 Å, μ = 38.67 cm-1, R = 0.039, 3741 unique reflections, 3203 with I > 3σ(I). NiBr2·4[(C6H5)3AsO]·3|2(C6H5CH3)·H2O (orange): M = 1663.7, P1, a = 12.647(8), b = 13.953(5), c = 22.853(6) Å, α = 90.91(3), β = 96.70(4), γ = 111.16(4)°, V = 3727.4 Å3, Z = 2, Dx = 1.482 g cm-3, MoKα, λ = 0.71073 Å, μ = 30.48 cm-1, R = 0.087, 8600 unique reflections, 4293 with I > 3σ(I). In the green complex the Ni(II) ion is sited on a center of symmetry and is octahedrally coordinated to six water molecules, hydrogen bonded to the Ph3AsO molecules and to the bromide anions forming a second coordination sphere in a nearly octahedral arrangement. In the orange complex the cation is pentacoordinated with the four oxygen atoms of the Ph3AsO ligands forming the basis of a tetragonal pyramid and with one Br- anion in the apical position. The absorption spectrum of the orange complex is compared with the spectra of other Ni(II) square pyramidal complexes described in the literature. © 1984.

This model connects directly the radar reflectivity data and hydrological variable runoff. The catchment is discretized in pixels (4 Km × 4 Km) with the same resolution of the CAPPI. Careful discretization is made so that every grid catchment pixel corresponds precisely to CAPPI grid cell. The basin is assumed a linear system and also time invariant. The forecast technique takes advantage of spatial and temporal resolutions obtained by the radar. The method uses only the measurements of the factor reflectivity distribution observed over the catchment area without using the reflectivity - rainfall rate transformation by the conventional Z-R relationships. The reflectivity values in each catchment pixel are translated to a gauging station by using a transfer function. This transfer function represents the travel time of the superficial water flowing through pixels in the drainage direction ending at the gauging station. We illustrate the sensitivities of LEP experiments to leptonic signals associated to models where supersymmetry (SUSY) is realized with spontaneous breaking of R-parity. We focus on missing transverse momentum plus acoplanar muon events (p Τ + μ + μ -) arising from lightest neutralino single production xv as well as pair production xx, followed by x decays, where x denotes the lightest neutralino. We show that the integrated luminosity achieved at LEP already starts probing the basic parameters of the theory. We discuss the significance of these constraints for the simplest spontaneous R-parity breaking models and their relevance for future searches of SUSY particles.

The objectives of this study were to evaluate the effect of type of cuttings (apical, intermediate and basal) and different concentrations of IBA (indole-3-butyric acid) on rooting of Red success rose (Rosa sp.) leafy cuttings, during two season of cuttings collection (summer and winter). The investigation was carried out in the farm Irmaos Van Schaik in Holambra-SP, Brazil, from February to April and August to October/1993. The experimental design was a randomized block in a factorial arrangement. It consisted of 12 treatments (3 types of cuttings combined with IBA powder in 4 different concentrations - 0, 1000, 2000 and 4000 ppm) with 3 replicates during 2 seasons. The investigation permitted the following conclusions: the apical and intermediate cuttings showed better results in general when compared to the basal ones in both seasons; the average rooting at the transplanting time were 76%, 70% and 47% (summer) and 80%, 69% and 33% (winter) for apical, intermediate and basal cuttings, respectively; the utilization of IBA did not stimulate rooting. The average rooting for the control cuttings reached 85% (summer) and 78% (winter), regardless the type of cuttings.

The objectives of this study were to evaluate the effects of rooting media (vermiculite, carbonized rice bark, fenolic spume and sand), presence of IBA (indole-3-butyric acid) and presence of fertilization during the rooting of rose (Rosa sp.) leafy cuttings, Dalas cultivar. This investigation was carried out in Jaboticabal (Sao Paulo State - Brazil) during the period of March to April 1997. The experimental design was a randomized block in a factorial arrangement. It consisted of 16 treatments (the 4 rooting media combined with IBA concentrated solutions - 0 and 1.000 ppm and fertilizer - 0 and commercial product with macro and micronutrients, applied weekly) with 3 replicates. The evaluations were 30 days after cuttings showed that the best rooting percentage occurs in sand (98%), followed by vermiculite (90%) and fenolic spume (87%) and cuttings treated with IBA (95%), while the fertilization showed no promoting effects. The number of cuttings roots wasn't affected with rooting media or DBA treatment, while the fertilization showed effective.

We introduce and study new integrable models (IMs) of An (1)-nonabelian Toda type which admit U(1) ⊗ U(1) charged topological solitons. They correspond to the symmetry breaking SU(n + 1) → SU(2) ⊗ SU(2) ⊗ U(1)n-2 and are conjectured to describe charged dyonic domain walls of N = 1 SU(n + 1) SUSY gauge theory in large n limit. It is shown that this family of relativistic IMs corresponds to the first negative grade q = -1 member of a dyonic hierarchy of generalized cKP type. The explicit relation between the 1-soliton solutions (and the conserved charges as well) of the IMs of grades q = -1 and q = 2 is found. The properties of the IMs corresponding to more general symmetry breaking SU(n + 1) → SU(2)⊗p ⊗ U(1)n-p as well as IM with global SU(2) symmetries are discussed. © 2002 Elsevier Science B.V. Being our objective to characterize demographic and morbidity patterns of the 60 years old or more attended at the UNESP Clinic Hospital in 1997 a sample consisting of 482 handbooks has been analyzed in order to evaluate ambulatory attendance, other consisting of 101 handbooks to evaluate first aid clinic demand and statistics from Medical Informatic Center (Centro de Informática da Medicina - CIMED) refering to 2652 internments of such populations this year. Ten and a half per cent from the total of first aid clinic attended patients refers to this old people and the majority (57,4%) lives in Botucatu County. More frequently diagnoses during consultation are Chronic Obstructive Lung Disease (10.1%), Congestive Cardiac Disease (4.7%), Asthma and Cataract (3.1 % for both). Such population is also referred to be 19.8% from the number of all patients attended at the UNESP ambulatories, 65.8% coming from other city in São Paulo State, being in average 69.6 years old and attending 4.5 consultations in average by year. More frequently found diagnoses referred to Essential Primary Hypertension (4.2%), Prostatic Nodular Hyperplasia (4.0%) and Nonspecified Cataract (3.3%). Regarding to internments, they referred to be 18.4% from the number of all interned patients. More frequent diagnoses at internmemt referred to Nonspecified Cataract (12%), Glaucoma (3.0%), Prostatic Nodular Hyperplasia (2.4%) and Limbs Arteriosclerosis (2.3%). Results indicates the existence of an tertiary hospital divided into specialties, also attending morbidity that should be in control into levels of primary and secondary attention.

Aim: To evaluate the influence of coronal filling and apical perforation on the induction of periapical inflammation. Methodology: Fifty-eight root canals in the teeth of dogs were divided into four groups. Groups I and II: root canals were exposed for 180 days; groups III and IV: root canals were exposed for 7 days and then the access cavity filled for 53 days. The root apices of groups I and III were perforated after the coronal opening, whilst those of groups II and IV remained intact. Standard radiographs were taken before and after the experimental periods. Digital images of the radiographs were created and then analysed by three examiners. After induction of periapical inflammation, the root canal contents were collected using paper points. Microbiologic evaluation of the type of microorganism was carried out by culture in different growth media. The radiographic and microbiologic data were statistically analysed using ANOVA at a 5% significance level. Results: There were a greater total number of microorganisms in groups I and II (P < 0.05). The number of anaerobes was greater than the number of aerobes (P < 0.05). The size of the periapical radiolucencies were not significantly different between the experimental groups. Conclusions: The different methods analysed induced similar areas of periapical radiolucency in dogs with predominantly anaerobic bacteria. However, the time required for induction was less when the method with coronal filling was used. © 2005 International Endodontic Journal.
