Calcium (Ca) is essential for crustaceans, due to calcium carbonate (CaCO3) deposition in the new exoskeleton to harden it. The purpose of this work was to study short term Ca balance in terms of dietary Ca ingestion in two phylogenetically related crabs (Superfamily Grapsoidea) showing different degrees of terrestrial adaptations: Sesarma rectum Randall, 1840 and Neohelice granulata (Dana, 1851). Dietary Ca ingestion was studied using purified diets with different Ca concentrations (0, 2.2 and 6.66 % Ca), together with measurements of Ca excretion and Ca hemolymph levels. The results showed that both crabs had the same response to foods containing different levels of Ca, with both species eating more of the high Ca diet. However, S. rectum consumed more per mg body mass at all Ca concentrations (6 mg.g-1 for S. rectum against 3 mg.g-1 for N. granulata). Both species excreted/egested Ca differently: S. rectum excreted Ca proportionally to ingestion, whereas N. granulata maintained constant faecal Ca output at all dietary Ca levels. Moreover, Ca hemolymph levels for crabs fed the different diets were independent of dietary Ca. In conclusion, both S. rectum and N. granulata seem to regulate the consumption of diets containing more Ca, which suggests a fine balance for Ca intake.

O autor procurou estabelecer os dados relativos ao hemograma de coelho normal, do biotério do Instituto Oswaldo Cruz, encontrando as seguintes médias: hematias 5.186.000 por mm3, leucócitos 10.419 por mm3, e na contagem específica os seguintes valores para Heterófilos 40%, basófilos 2.5%, eosinófilos 0.2%, monócitos 6.5% e linfócitos 50%, e células de Tuerk 0.8%. Pode o autor confirmar o achado de AGOSIN, referente à presença de células morfológicamente semelhantes ás de TUERK, no sangue periférico dos coelhos normais. Realizou ainda, uma revisão bibliográfica do assunto.

O trabalho refere as observações feitas em 1947 em um foco de leishmaniose muco-cutânea na Baixada Fluminense (Estado do Rio de Janeiro, Brasil). A existência da moléstia como uma endemia na região foi comprovada pelo encontro de 21 cicatrizes típicas, reagindo positivamente à intradermo-reação com antígeno específico, algumas datando de 5 a 15 anos. Na época dos trabalhos, entretanto, foi constatado um "surto epidêmico", o qual coincidira com uma grande derrubada florestal para fabrico de carvão vegetal. De 306 pessoas examinadas (cêrca de 50% da população local), foram encontradas 39 com lesões leishmanióticas (12,7%). Dentro e fora dos domicílios foram capturados Phlebotomus intermedius. Em 12 cães examinados foi encontrado um com diagnóstico provável da moléstia. Quinze gatos examinados mostraram-se negativos e, do mesmo modo, 28 mamíferos silvestres de pequeno porte. Dos 39 paciente, 4 tinham lesões mucosas (10,3%); 16 apresentavam lesões múltiplas (41,0%); e 19 eram mulheres (48,7%). Havia absoluta predominância das lesões nas partes descobertas do corpo. Cêrca de 1/3 dos casos era em crianças até 10 anos, atestando uma intensa transmissão domiciliária. Existiam casas com 2 a 6 enfermos. Com base nos informes dos pacientes ou responsáveis quanto ao tempo de doença (e admitindo-se um período incubativo médio de 2 meses), conclui-se que, provàvelmente, a grande maioria das infecções se dera entre julho e novembro, coincidindo com a derrubada florestal acima citada. Em 36 casos foi feita a intradermo-reação de Montenegro, obtendo-se respostas duvidosas em 2 e positivas em 34, com intensidade variável. Foram feitas 18 biópsias. Na epiderme havia hiperacantose e, freqüentement, pseudoepiteliomatose com globos córneos e microabcessos; e na derme observaram-se 2 quadros característicos: ou um infiltrado de plasmócitos predominantes, ou uma reação granulomatosa, os quais, às vêzes, se associavam. Em geral, a granulomatose ocorria nos casos mais antigos, isolada ou associada à infiltração, que predominava nos casos mais recentes da enfermidade. A granulomatose traduziria um estado hiperérgico do organismo, uma vez que os indivífuos que a apresentaram tinham maior tempo de doença e reagiam fortemente à intradermo-reação. As leishmanias nunca se mostraram muito numerosas nos cortes estudados. Em 3 paciente foi observada cura espontânea. Foram tratados 26 doentes, sendo 18 com tártaro emético, 4 com "fuadina" e 4 com ambos os remédios. O tártaro mostrou-se tóxico, embora desse resultados tão bons quanto a "fuadina". Dois paciente com lesões da mucosa nasal não se curaram completamente, não obstante terem recebido ambos os remédios. Cinco anos depois dêste inquérito contatou-se pràticamente a extinção dêste foco de leishmaniose. Durante êsse tempo tinham sido feitas aspersões domiciliárias periódicas com o DDT para combate à malária (NERY GUIMARÃES & BUSTAMANTE, 1953).

Neste trabalho é feita a descrição anatômica e histológica do aparelho genital de Succiena meridionalis Orbigy, 1846, considerando-se também a concha, mandíbula, rádula e câmara paleal. As variações da concha, rádula e "talon", bem como a intensidade de pigmentação de diversas estruturas também são discutidas. Além dêstes aspectos, são relatados alguns dados biológicos da espécie e disposição anatômica do aparelho copulador por ocasião do ato sexual. Como caracteres importantes para diagnose desta espécie, dentro do gênero, consideramos: 1 - Conchas geralmente translúcidas e frágeis, como comprimento, nos adultos, variando de 6,0 a 13,1 mm e número de espiras compreendida entre 2 1/6 e 3 1/6. 2 - Ovotestis constituído por numerosos folículo reunidos num só grupo. 3 - Canal coletor do ovotestis unido à vesícula seminal subterminalmente. 4 - Vagina curta e mais dilatada na porção proximal, podendo formar um ângulo reto ou agudo com o oviduto. 5 - Bainha muscular do pênis envolvendo a porção distal do falus e sendo atravessada subrterminalmente pelo canal deferente. 6 - Porção proximal do falus bem dilatada, tendo dobrada contra si o epifalus. 7 - Presença de uma papila na comunicação do epifalus com a porção proximal do falus e que se projeta para a luz dêste último. 8 - Inserção terminal do músculo retrtor no epifalus.

Foi verificado que a esporogênese de B. licheniformis, amostra 23910, sòmente ocorre quando Mn[2+] ou Mg[2+] etão presentes, em meio sintético, nas concentrações de 0,1 mg%. Ambos os íons estimulam o crescimento, e no 14º dia de incubação a cultura apresenta endósporos e esporos livres, quando a concentração dos metais é de 0,1 mg%. Aumentando-se o teor dêstes íons para 1,0 mg%, esporos livres são obtidos no 10º dia de incubação. Outros íons testados como, Ca[2+], Fe[2+], Co[2+], Cu[2+], Mo[6+], Zn[2+] e B[3+] não mostraram influir no processo. Co[2+] e Cu[2+] foram tóxicos, inibindo o crescimento. O Ca[2+] pareceu não interferir na esporogênese, mas foi capaz de inibir o cresciemnto na concentração de 3mg%. O aparecimento de endósporos e esporos livres, no meio, quando Mn[2+] encontra-se a 1,0 mg%, coincide com um teor de glicose correspontente a cêrca de 12,5% da concentração inicial do açúcar. O autor sugere, também, a possibilidade do Mn[2+] ou Mg[2+] ativar a síntese do ácido dipicolínico.

Quatro frangos e três pombos (1ª e 2ª experiências) não se infectaram com o "T. (S.) cruzi" (amostra Y), injetados na pele com doses de 9,8 x10*5 a 3,8 x 10*6 parasitos (mistura de formas de cultivo tripomastigotos sangüíneos). Em outros 2 frangos (6ª experiência), a refratariedade não foi rompida com, respectivamente, 5 injeções peritoneais em dias alternados, de 2,8 x 10*6 a 6,1 x 10*6 parasitos, e 5 injeções peritoneais diárias do conteúdo de um cultivo suspenso em salina, adicionado de tripamastigotos sangüíneos, obtidos de 3 camundongos jovens infectados. E, ainda em outros 2 frangos (7ª experiência), a refratariedade não foi rompida associando-se a doses peritoneais de 8,1 x 10*6 e 3,5 x 10*7 parasitos, respectivamente, a ministração de dexametasona, seja com o início da droga no mesmo dia da inoculação (0,4 mg/8 doses/10 dias), seja com o ínicio da droga 3dias antes do dia da inoculação (0,4 mg/10 doses/12 dias). Nessas 11 aves a refratariedade foi comprovada pela inoculação do sangue em camundongos albinos recém-nascidos (total de 54 inoculaçoes negativas em 301 camundongos); e por 52 xenodiagnósticos negativos (total de 175 larvas e adultos de Triatoma infestans e Panstrongylys megistus). As inoculações e os xenodiagnósticos foram feitos entre 1 hora (h) e 96 h e aos 10, 20 e 30 dias depois da inoculação das aves. (Quadro 1). Portanto, além da refratariedade, ficou demonstrado que os parasitos não passam para o sangue circulante das aves. em 10 frangos (3ª, 4ª e 5ª experiências) inoculados na pele com, 1,9 x 10*6 a 3,7 x 10*6 parasitos, verificou-se a duração da viabilidade dos parasitos nas áreas injetadas. Estas foram puncionadas de 1/2 com 1/2 hora, e o material obtido microscopado e inoculado em camundongos recém-nascidos. De 17 inoculações feitas até às 8:30 h, doze foram positivas, inclusive a última; e de 13 inoculações feitas entre 9 e 90 h (as 4 primeiras ainda de 1/2 em 1/2 h), todas foram negativas. Por outro lado, de 10 xenodiagnósticos feitos nas áreas injetadas, entre 1/2h e 9 h após a inoculação das aves, apenas 2 foram positivos; e de 10 xenodiagnósticos feitos entre 9 e 90 h, todos foram negativos. (Quadro 2). Em 4 frangos (8ª experiência), 10 xenodiagnósticos foram feitos entre 1 e 10 h, sendo 2 positivos (7 h e 8 h). Apesar de dificuldades operacionais na execução dos xenodiagnósticos, nas condições experimentais deste trabalho, eles se mostraram inferiores ás inoculações em camundongos recém-nascidos, como prova diagnóstica laboratorial. Portanto, verificou-se que ainda são encontrados parasitos viáveis nas áreas injetadas, pelo menos até ás 8:30 h depois inoculação. todavia, a partir de 2 h após a inoculação os parasitos diminuem de número progressivamente nas áreas injetadas, ao mesmo tempo que mostram sensíveis alterações morfológicas, encontrando-se também muito deles mortos. Ao exame direto, não mais foram vistos depois das 5:30 h. As inoculações provocam na pele dos frangos...

PURPOSE: To evaluate a diagnostic strategy for pulmonary embolism that combined clinical assessment, plasma D-dimer measurement, lower limb venous ultrasonography, and helical computed tomography (CT). METHODS: A cohort of 965 consecutive patients presenting to the emergency departments of three general and teaching hospitals with clinically suspected pulmonary embolism underwent sequential noninvasive testing. Clinical probability was assessed by a prediction rule combined with implicit judgment. All patients were followed for 3 months. RESULTS: A normal D-dimer level (<500 microg/L by a rapid enzyme-linked immunosorbent assay) ruled out venous thromboembolism in 280 patients (29%), and finding a deep vein thrombosis by ultrasonography established the diagnosis in 92 patients (9.5%). Helical CT was required in only 593 patients (61%) and showed pulmonary embolism in 124 patients (12.8%). Pulmonary embolism was considered ruled out in the 450 patients (46.6%) with a negative ultrasound and CT scan and a low-to-intermediate clinical probability. The 8 patients with a negative ultrasound and CT scan despite a high clinical probability proceeded to pulmonary angiography (positive: 2; negative: 6). Helical CT was inconclusive in 11 patients (pulmonary embolism: 4; no pulmonary embolism: 7). The overall prevalence of pulmonary embolism was 23%. Patients classified as not having pulmonary embolism were not anticoagulated during follow-up and had a 3-month thromboembolic risk of 1.0% (95% confidence interval: 0.5% to 2.1%). CONCLUSION: A noninvasive diagnostic strategy combining clinical assessment, D-dimer measurement, ultrasonography, and helical CT yielded a diagnosis in 99% of outpatients suspected of pulmonary embolism, and appeared to be safe, provided that CT was combined with ultrasonography to rule out the disease.

Animal models of infective endocarditis (IE) induced by high-grade bacteremia revealed the pathogenic roles of Staphylococcus aureus surface adhesins and platelet aggregation in the infection process. In humans, however, S. aureus IE possibly occurs through repeated bouts of low-grade bacteremia from a colonized site or intravenous device. Here we used a rat model of IE induced by continuous low-grade bacteremia to explore further the contributions of S. aureus virulence factors to the initiation of IE. Rats with aortic vegetations were inoculated by continuous intravenous infusion (0.0017 ml/min over 10 h) with 10(6) CFU of Lactococcus lactis pIL253 or a recombinant L. lactis strain expressing an individual S. aureus surface protein (ClfA, FnbpA, BCD, or SdrE) conferring a particular adhesive or platelet aggregation property. Vegetation infection was assessed 24 h later. Plasma was collected at 0, 2, and 6 h postinoculation to quantify the expression of tumor necrosis factor (TNF), interleukin 1α (IL-1α), IL-1β, IL-6, and IL-10. The percentage of vegetation infection relative to that with strain pIL253 (11%) increased when binding to fibrinogen was conferred on L. lactis (ClfA strain) (52%; P = 0.007) and increased further with adhesion to fibronectin (FnbpA strain) (75%; P < 0.001). Expression of fibronectin binding alone was not sufficient to induce IE (BCD strain) (10% of infection). Platelet aggregation increased the risk of vegetation infection (SdrE strain) (30%). Conferring adhesion to fibrinogen and fibronectin favored IL-1β and IL-6 production. Our results, with a model of IE induced by low-grade bacteremia, resembling human disease, extend the essential role of fibrinogen binding in the initiation of S. aureus IE. Triggering of platelet aggregation or an inflammatory response may contribute to or promote the development of IE.

ABSTRACT: BACKGROUND: Recent data indicate a slight decrease in the prevalence of smoking in Switzerland, but little is known regarding the intention and difficulty to quit smoking among current smokers. Hence, we aimed to quantify the difficulty and intention to quit smoking among current smokers in Switzerland. METHODS: Cross-sectional study including 607 female and 658 male smokers. Difficulty, intention and motivation to quit smoking were assessed by questionnaire. RESULTS: 90% of women and 85% of men reported being "very difficult" or "difficult" to quit smoking. Almost three quarters of smokers (73% of women and 71% of men) intended to quit; however, less than 20% of them were in the preparation stage and 40% were in the precontemplation stage. On multivariate analysis, difficulty to quit was lower among men (Odds ratio and 95% [confidence interval]: 0.51 [0.35-0.74]) and increased with nicotine dependence and number of previous quitting attempts (OR=3.14 [1.75-5.63] for 6+ attempts compared to none). Intention to quit decreased with increasing age (OR=0.48 [0.30-0.75] for [greater than or equal to]65 years compared to <45 years) and increased with nicotine dependence, the number of previous quitting attempts (OR=4.35 [2.76-6.83] for 6+ attempts compared to none) and among non-cigarette smokers (OR=0.51 [0.28-0.92]). Motivation to quit was inversely associated with nicotine dependence and positively associated with the number of previous quitting attempts and personal history of lung disease. CONCLUSION: Over two thirds of Swiss smokers want to quit. However, only a small fraction wishes to do so in the short term. Nicotine dependence, previous attempts to quit or previous history of lung disease are independently associated with difficulty and intention to quit.

BACKGROUND:: Attenuated innate immune responses to the intestinal microbiota have been linked to the pathogenesis of Crohn's disease (CD). Recent genetic studies have revealed that hypofunctional mutations of NLRP3, a member of the NOD-like receptor (NLR) superfamily, are associated with an increased risk of developing CD. NLRP3 is a key component of the inflammasome, an intracellular danger sensor of the innate immune system. When activated, the inflammasome triggers caspase-1-dependent processing of inflammatory mediators, such as IL-1β and IL-18. METHODS:: In the current study we sought to assess the role of the NLRP3 inflammasome in the maintenance of intestinal homeostasis through its regulation of innate protective processes. To investigate this role, Nlrp3(-/-) and wildtype mice were assessed in the dextran sulfate sodium and 2,4,6-trinitrobenzenesulfonic acid models of experimental colitis. RESULTS:: Nlrp3(-/-) mice were found to be more susceptible to experimental colitis, an observation that was associated with reduced IL-1β, reduced antiinflammatory cytokine IL-10, and reduced protective growth factor TGF-β. Macrophages isolated from Nlrp3(-/-) mice failed to respond to bacterial muramyl dipeptide. Furthermore, Nlrp3-deficient neutrophils exhibited reduced chemotaxis and enhanced spontaneous apoptosis, but no change in oxidative burst. Lastly, Nlrp3(-/-) mice displayed altered colonic β-defensin expression, reduced colonic antimicrobial secretions, and a unique intestinal microbiota. CONCLUSIONS:: Our data confirm an essential role for the NLRP3 inflammasome in the regulation of intestinal homeostasis and provide biological insight into disease mechanisms associated with increased risk of CD in individuals with NLRP3 mutations. The migration of larval Schistosoma mansoni was tracked by means of autoradiographic analysis in naive rabbits percutaneously exposed to L-(**75 Se) selenomethionine-labeled cercariae on serial intervals of 1, 2, 4, 6, 8, 10, 15, 20, 25, 30, 40 and 50 days post-infection. Autoradiographic foci were detected from the 1st day in the skin, up to the 15th day in the liver. Adult and mature worms were recovered either paired or not 60 days after infection, by perfusion of hepatic and mesenteric veins. Morphometric analysis under optical microscopy, showed that worms were within regular dimention limits as compared to adult worms harboured by other host species. These observations extend previous informations on the S. mansoni-rabbit association and clearly demonstrate the post-liver phase of S.mansoni life-cycle in this host.

BACKGROUND: Low 24-hour urine volume (24 UV) may be a significant risk factor for decline in kidney function. We therefore aimed to study associated markers and possible determinants of 24 UV in a sample of the Swiss population. METHODS: The cross-sectional Swiss Salt Study included a population-based sample of 1535 (746 men and 789 women) individuals from three linguistic regions of Switzerland. Data from 1300 subjects were available for the present analysis. 24 UV was measured using 24-hour urine collection. Determinants of 24 UV were identified using multivariable linear regression models. RESULTS: In bivariate analysis, 24 UV was higher in women compared to men (2000 ml/24 h [interquartile range (IQR): 1354, 2562] versus 1780 ml/24 h [IQR: 1244, 2360], p = 0.002). In multivariable regression analyses, independent associated markers of 24 UV were female sex (β = 280, 95% confidence interval [CI]: 174, 386, p < 0.0001), fluid intake (β = 604, 95% CI: 539, 670, p < 0.0001), sodium excretion (β = 4.2, 95% CI: 3.4, 4.9, p < 0.0001) age (β = 6.6, CI: 3.4, 9.7, p < .0001), creatinine clearance (β = 2.4, CI: 0.2, 4.6, p = 0.04), living in the German-speaking part of Switzerland (β = 124, CI: 29, 219, p = 0.01), alcohol consumption (β = 41, CI: 9, 73, p = 0.01 for increasing categories of alcohol consumption), body mass index (β = -32, CI: -45, -18, p < 0.0001), current smoking (β = -146, CI: -265, -26, p = 0.02), and consumption of meat and cold cut (β = -56, CI: -108, -5, p = 0.03). CONCLUSION: In this large population-based, cross-sectional study, we found several strong and independent correlates for 24 UV. These findings may be important to improve our understanding in the development of chronic kidney disease.

BACKGROUND: Dissection during laparoscopic surgery produces smoke containing potentially toxic substances. The aim of the present study was to analyze smoke samples produced during laparoscopic colon surgery using a bipolar vessel sealing device (LigaSuretrade mark). METHODS: Four consecutive patients undergoing left-sided colectomy were enrolled in this pilot study. Smoke was produced by the use of LigaSuretrade mark. Samples (5,5l) were evacuated from the pneumoperitoneum in a closed system into a reservoir. Analysis was performed with CO2-laser-based photoacoustic spectroscopy and confirmed by a Fourier-transform infrared spectrum. The detected spectra were compared to the available spectra of known toxins. RESULTS: Samples from four laparoscopic sigmoid resections were analyzed. No relevant differences were noted regarding patient and operation characteristics. The gas samples were stable over time proven by congruent control measurements as late as 24 h after sampling. The absorption spectra differed considerably between the patients. One broad absorption line at 100 ppm indicating H2O and several unknown molecules were detected. With a sensitivity of alpha min ca 10-5 cm-1 no known toxic substances like phenol or indole were identified. CONCLUSION: The use of a vessel sealing device during laparoscopic surgery does not produce known toxic substances in relevant quantity. Further studies are needed to identify unknown molecules and to analyze gas emission under various conditions.
