In this thesis, we explore constraints which can be put on the primordial power spectrum of curvature perturbations beyond the scales probed by anisotropies of the cosmic microwave background and galaxy surveys. We exploit present and future measurements of CMB spectral distortions, and their synergy with CMB anisotropies, as well existing and future upper limits on the stochastic background of gravitational waves. We derive for the first time phenomenological templates that fit small-scale bumps in the primordial power spectrum generated in multi-field models of inflation. By using such templates, we study for the first time imprints of primordial peaks on anisotropies and spectral distortions of the cosmic microwave background and we investigate their contribution to the stochastic background of gravitational waves. Through a Monte Carlo Markov Chain analysis we infer for the first time the constraints on the amplitude, the width and the location of such bumps using Planck and FIRAS data. We also forecast how a future spectrometer like PIXIE could improve FIRAS boundaries. The results derived in this thesis have implications for the possibility of primordial black holes from inflation.

Extracts from malagueta pepper (Capsicum frutescens L.) were obtained using supercritical fluid extraction (SFE) assisted by ultrasound, with carbon dioxide as solvent at 15MPa and 40°C. The SFE global yield increased up to 77% when ultrasound waves were applied, and the best condition of ultrasound-assisted extraction was ultrasound power of 360W applied during 60min. Four capsaicinoids were identified in the extracts and quantified by high performance liquid chromatography. The use of ultrasonic waves did not influence significantly the capsaicinoid profiles and the phenolic content of the extracts. However, ultrasound has enhanced the SFE rate. A model based on the broken and intact cell concept was adequate to represent the extraction kinetics and estimate the mass transfer coefficients, which were increased with ultrasound. Images obtained by field emission scanning electron microscopy showed that the action of ultrasonic waves did not cause cracks on the cell wall surface. On the other hand, ultrasound promoted disturbances in the vegetable matrix, leading to the release of extractable material on the solid surface. The effects of ultrasound were more significant on SFE from larger solid particles.

The electrocardiogram (ECG) is the simplest and most effective non-invasive method to assess the electrical activity of the heart and to obtain information on the heart rate (HR) and rhythm. Because information on the HR of very small reptiles (body mass <10 g) is still scarce in the literature, in the present work we describe a procedure for recording the ECG in non-anesthetized geckos (Hemidactylus mabouia, Moreau de Jonnès, 1818) under different conditions, namely manual restraint (MR), spontaneous tonic immobility (TI), and in the non-restrained condition (NR). In the gecko ECG, the P, QRS and T waves were clearly distinguishable. The HR was 2.83 ± 0.02 Hz under MR, which was significantly greater (p < 0.001) than the HR under the TI (1.65 ± 0.09 Hz) and NR (1.60 ± 0.10 Hz) conditions. Spontaneously beating isolated gecko hearts contracted at 0.84 ± 0.03 Hz. The in vitro beating rate was affected in a concentration-dependent fashion by adrenoceptor stimulation with noradrenaline, as well as by the muscarinic cholinergic agonist carbachol, which produced significant positive and negative chronotropic effects, respectively (p < 0.001). To our knowledge, this is the first report on the ECG morphology and HR values in geckos, particularly under TI. The methodology and instrumentation developed here are useful for non-invasive in vivo physiological and pharmacological studies in small reptiles without the need of physical restraint or anesthesia.

The cerebral cysticercosis can produce intracranial hypertension by inflammatory obstruction of the basal cysterns or by expansive lesion in the cerebral parenchima or ventricular cavities. In the latter and in tumor cases the clinical picture is very similar and only after surgery can the etiology be determined. We present 11 operated cases of intracranial cysticercosis which presented the clinical picture of an expansive lesion. There were 7 females and 4 males with ages between 4 and 65 years. Nine patients were admitted because of headache, vomiting and visual disturbances suggestive of intracranial hypertension. One patient was admited with lymphocytic meningitis and another with focal seizures following hemiparesis. Five patients presented focal signs and six edema of the papilla. Epileptic manifestations were present in 45.5% of the cases. A plain X-ray films of the skull failed to reveal calcificatons, however signs of chronic hypertension were present in three cases. The electroencephalogram showed slow focal waves in 8 patients The spinal fluid examination revealed lymphocytosis in 4 cases, increased protein content in another 4 and complement fixation for cysticercosis was positive in 2 cases. The expansive lesions were localized by angiograph and ventriculography. In these the location was temporal in 4, frontal in 3, parietal in 2, in the third ventricle in one and in the fourth ventricle in another. At surgery we removed a large cyst from the cerebral parenchyma in six cases. Around the cyst a thick glial reaction was present. In the other cases the cyst was small but fixed to the ventricular trigone and produced dilatation of the inferior horn of the lateral ventricle. In two cases we removed a solitary intraventricular cyst from the third and fourth ventricles. In the two children operated upon there were several small hard cysts involving the cerebral parenchyma which displayed intense gliosis. There were no postoperative complications.

Neste trabalho estudou-se a influência dos padrões de onda extratropicais, que favorecem o desenvolvimento de eventos extremos frios no sudeste Sul-Americano, e em particular na região conhecida como Pampa Úmida. O aquecimento anômalo observado na região do oceano Pacífico tropical ocidental a nordeste da Austrália, durante os invernos de máxima freqüência de ocorrência de Geadas Generalizadas (GG) no centro-leste da Argentina, (região conhecida como Pampa Úmida - PU), atua como disparador de ondas de Rossby, as quais se propagam até o continente, favorecendo assim a ocorrência daqueles eventos. O padrão de propagação obtido nas simulações numéricas com um modelo baroclínico global, mostra o predomínio de um número de onda 3. Adicionalmente, foram analisadas as correlações do vento meridional em altos e baixos níveis observados para os eventos de GG, selecionados dentro dos invernos de máxima freqüência de ocorrência desses eventos. O vento meridional global em 250hPa apresenta regiões com correlação estatisticamente significativa com o vento meridional médio na PU. A configuração obtida no caso do vento meridional global em 250hPa, correlacionado com o vento meridional na PU, pode estar associada ao padrão de propagação das ondas simuladas numericamente a partir da forçante tropical. Igualmente importantes e significativos são os valores de correlação do vento sul nos baixos níveis, em particular para toda região da PU. O padrão de ondas simulado está bem representado pelas significativas correlações entre o vento meridional hemisférico em altos níveis e a temperatura no dia de evento de GG.

Este trabalho faz uma análise das estimativas de teores de umidade obtidas com o método Ground Penetrating Radar (GPR) comparativamente às determinadas com os métodos Time Domain Reflectometry (TDR) e gravimétrico. Os dados foram obtidos em dois experimentos diferentes: um experimento controlado em laboratório buscando reproduzir um meio homogêneo onde foram obtidas as medidas de umidade com GPR (antenas de 1 GHz) e TDR, e outro experimento de campo onde foram obtidos dados com GPR (antenas de 200 MHz) e de amostras de solos do local. Para a obtenção das estimativas a partir do método GPR foram analisados os eventos relativos à onda de transmissão direta entre as antenas, onda refratada criticamente e onda refletida em interfaces com diferentes propriedades elétricas.O GPR mostrou-se sensível às variações de umidades presentes nos dois experimentos e apresentou boa correlação com os dados obtidos com TDR (REQM de0,007 m³m-3) e das amostras (REQM de 0,039 m³m-3).

Mixing layers are present in very different types of physical situations such as atmospheric flows, aerodynamics and combustion. It is, therefore, a well researched subject, but there are aspects that require further studies. Here the instability of two-and three-dimensional perturbations in the compressible mixing layer was investigated by numerical simulations. In the numerical code, the derivatives were discretized using high-order compact finite-difference schemes. A stretching in the normal direction was implemented with both the objective of reducing the sound waves generated by the shear region and improving the resolution near the center. The compact schemes were modified to work with non-uniform grids. Numerical tests started with an analysis of the growth rate in the linear regime to verify the code implementation. Tests were also performed in the non-linear regime and it was possible to reproduce the vortex roll-up and pairing, both in two-and three-dimensional situations. Amplification rate analysis was also performed for the secondary instability of this flow. It was found that, for essentially incompressible flow, maximum growth rates occurred for a spanwise wavelength of approximately 2/3 of the streamwise spacing of the vortices. The result demonstrated the applicability of the theory developed by Pierrehumbet and Widnall. Compressibility effects were then considered and the maximum growth rates obtained for relatively high Mach numbers (typically under 0.8) were also presented.

The absorption spectra of DPH at fixed concentration do not change with water content in organic solvents. It exhibits monomer bands, such as those obtained in ethanol. The absorption did not change for solutions up to 54 and 46% of water in ethanol and DMSO, respectively, for [DPH] = 5.0 × 10-6 mol L-1 at 30 °C. However, at the same experimental conditions, a gradual sharp decay of the DPH fluorescence is observed. It is proposed that water molecules below these water concentration limits act as quenchers of the excited states of DPH. Stern-Volmer quenching constants by intensities measurements are 7.4 × 10-2 (water/ethanol) and 2.6 × 10-2 L mol-1 (water/DMSO). DPH lifetime measurements in the absence and presence of water resulted in 7.1 × 10-2 L mol-1 in water/ethanol, which pointed out that the process is a dynamic quenching by water molecules. For experiments using DPH as probe, this process can affect data, leading to misunderstanding interpretation.

The [Ru3O(Ac)6(py)2(CH3OH)]+ cluster provides an effective electrocatalytic species for the oxidation of methanol under mild conditions. This complex exhibits characteristic electrochemical waves at -1.02, 0.15 and 1.18 V, associated with the Ru3III,II,II/Ru3III,III,II/Ru 3III,III,III /Ru3IV,III,III successive redox couples, respectively. Above 1.7 V, formation of two RuIV centers enhances the 2-electron oxidation of the methanol ligand yielding formaldehyde, in agreement with the theoretical evolution of the HOMO levels as a function of the oxidation states. This work illustrates an important strategy to improve the efficiency of the oxidation catalysis, by using a multicentered redox catalyst and accessing its multiple higher oxidation states.

Desembocaduras são ambientes bastante dinâmicos e sujeitos à complexa interação entre fatores estabilizadores e desestabilizadores. Dependendo dessa interação, desembocaduras podem apresentar a tendência de migração ao longo de barreiras arenosas. Um dos mecanismos mais eficientes de transporte de sedimento paralelo à costa, e consequentemente migração de canais, são as correntes longitudinais geradas pelas ondas se aproximando obliquamente à costa. A motivação do presente trabalho é entender o comportamento morfodinâmico do sistema de desembocadura do rio Itapocú, localizado no centro-norte de Santa Catarina (SC), frente aos processos forçantes que atuam na sua migração ao longo da linha de costa. A morfologia dos pontais arenosos foi obtida a partir de levantamentos morfológicos com o uso de DGPS. Para analisar a refração de ondas foi utilizado o modelo numérico MIKE 21 SW, sendo considerados como condições de contorno os dados de ondas referentes ao ano de 2002 e os dados de ondas previstos referentes ao período de coleta. Os dados de saída do modelo foram utilizados para estimar a deriva litorânea potencial na região. Os resultados morfológicos obtidos demonstraram uma migração da desembocadura para o norte durante o período analisado, sendo mais intenso durante o inverno e o verão. Ondas incidentes do quadrante sul sofreram mais o fenômeno da refração e as ondas de leste apresentaram menor variação angular ao se aproximarem à costa. A deriva litorânea potencial anual para os dados de ondas de 2002 apresentou sentido norte-sul, com inversão de sentido durante o outono. Utilizando os dados de ondas previstas para o período dos levantamentos, a deriva litorânea potencial estimada apresentou sentido sul-norte, concordando com a migração observada. Na região próxima a desembocadura, nos pontais arenosos, a deriva potencial apresentou direção para o norte durante todas as estações. Os dados de descarga fluvial não apresentaram influência na migração do canal, porém apresentaram uma relação com a largura do mesmo sazonalmente.Os dados de morfologia juntamente com os dados de deriva litorânea referentes às ondas de 2004/2005 mostraram claramente a migração do canal para o norte sendo a deriva a principal contribuinte para a migração da desembocadura.

This paper provides a description of the wave climate off the Brazilian coast based on an eleven-year time series (Jan/1997-Dec/2007) obtained from the NWW3 operational model hindcast reanalysis. Information about wave climate in Brazilian waters is very scarce and mainly based on occasional short-term observations, the present analysis being the first covering such temporal and spatial scales. To define the wave climate, six sectors were defined and analyzed along the Brazilian shelf-break: South (W1), Southeast (W2), Central (W3), East (W4), Northeast (W5) and North (W6). W1, W2 and W3 wave regimes are determined by the South Atlantic High (SAH) and the passage of synoptic cold fronts; W4, W5 and W6 are controlled by the Intertropical Convergence Zone (ITCZ) and its meridional oscillation. The most energetic waves are from the S, generated by the strong winds associated to the passage of cold fronts, which mainly affect the southern region. Wave power presents a decrease in energy levels from south to north, with its annual variation showing that the winter months are the most energetic in W1 to W4, while in W5 and W6 the most energetic conditions occur during the austral summer. The information presented here provides boundary conditions for studies related to coastal processes, fundamental for a better understanding of the Brazilian coastal zone.

Em função de suas condições de interface entre águas doces e salinas, desembocaduras estuarinas e lagunares constituem sistemas geomorfológicos altamente complexos e dinâmicos. Como conseqüência da variabilidade espacial e temporal dos fluxos de maré, o leito responde com uma grande variabilidade nas características morfológicas e sedimentares. Neste sentido, é possível relacionar diretamente a circulação de fundo e o transporte sedimentar com as feições submersas geradas. Perfis de ecossondagem, sonar de varredura lateral e sísmica de alta resolução, executados na desembocadura lagunar de Cananéia, revelaram a existência de uma dinâmica de fundo extremamente complexa, caracterizada por marcas onduladas e ondas de areia de alturas métricas. As maiores ondas de areia, localizadas em uma depressão na desembocadura lagunar, apresentam inversão de polaridade em sua assimetria, com a presença de ondas simétricas de grande tamanho no ponto de inversão. Este padrão morfológico não apresenta variação temporal em escala anual, sugerindo a persistência de um padrão de fluxos sobre o leito. Esta dinâmica revela, também, a constância de fluxos convergentes que aparentemente independem das condições de maré enchente ou vazante. Os resultados permitiram o estabelecimento de um primeiro modelo qualitativo de circulação de fundo na área, com aplicações potenciais na navegação e estudos de proteção da costa.
