Objective-To develop and apply the liquid-phase blocking sandwich ELISA (BLOCKING-ELISA) for the quantification of antibodies against foot-and-mouth disease virus (FMDV) strains O-1 Campos, A(24) Cruzeiro, and C-3 Indaial.Design-Antibody quantification.Sample Population-158 water buffalo from various premises of São Paulo Stale-Brazil. The sera were collected either from systemically vaccinated or nonvaccinated animals.Procedure-The basic reagents of BLOCKING-ELISA (capture and detector antibodies, virus antigens, and conjugate) were prepared and the reaction was optimized and standardized to quantify water buffalo antibodies against FMDV. An alternative procedure based on mathematical interpolation was adopted to estimate more precisely the antibody 50% competition liters in the BLOCKING-ELISA. These titers were compared with the virus-neutralization test (VNT) titers to determine the correlation between these techniques. The percentages of agreement, cutoff points, and reproducibility also were determined.Results-The antibody liters obtained in the BLOCKING-ELISA had high positive correlation coefficients with VNT, reaching values of 0.90 for O-1 Campos and C-3 Indaial, and 0.82 for the A(24) Cruzeiro (P < 0.0005). The cutoff points obtained by use of the copositivity and conegativity curves allowed determination of high levels of agreement between BLOCKLNG-ELISA and VNT antibody titers against the 3 FMDV strains analyzed.Conclusions-The results characterized by high cor relation coefficients, levels of agreement, and reproducibility indicate that the BLOCKING-ELISA may replace the conventional VNT for detection and quantification of antibodies from water buffalo sera to FMDV.

A trial was conducted during 1994-95 to study the effect of potassium fertilization on a guava (Psidium guajava L.) culture for 3 years. The control plots (without K) showed fruit production and potassium exportation that did not agree with the levels obtained by chemical analysis of the soil. Physical, chemical, mineralogic and morphologic analyses were performed on the red yellow latosol to identify minerals able to supply potassium, with emphasis on the fact that guava trees have a considerably widespread root system. The results obtained confirmed the presence of minerals in this soil that can supply potassium to the trees through weathering. Feldspars were identified in the silt fraction and micas in the clay fraction by X-ray diffractometry. The determination of total potassium revealed that the silt fraction of the soil had the largest absolute amounts of potassium, followed by clay. However, in view of its greater content, clay was the fraction that contributed most to the total amounts of potassium detected.

The objective of this paper was to evaluate the relevance of environmental and genetics effects on milk production of buffalo cows in Brazil. The data were based on the Buffalo Genetic Improvement Program - PROMEBUL, using information of 1,911 cows (107 Jafarabadi, 101 Mediterranean, 1,056 Mu/Tab and 647 crossbred females) with parturition between 1982 and 2003. The mathematic model for evaluating milk production included the fixed effects of herd, parturition year (1982 to 2003) and month (January to December), calf's sex (male or female), genetic group (Jafarabadi, Mediterranean, Murrah, and crossbreed), number of milking (one or two), lactation order (1 to 12) and duration of lactation (as a linear effect). The mean milk production in herds was 1,590.36 +/- 609.25 kg. All sources of variation were significant (P<0.05) for the studied characteristics, except calf's sex. The mean milk production per genetic group was 1,651.4; 1,592.2; 1,578.3 and 1,135.5 kg, for Murrah, Mediterranean, Crossbred and Jafarabadi, respectively. The duration of lactation was the most important source of variation over milk production, followed by the year of parturition, herd, parturition order, genetic group and month of parturition.

Through the point-centered quarter method, a phytosociological study was carried out in an area of semideciduous mesophytic forest with 190 ha, in the municipality of Jahu,SP. Due to the fact of the forest be crossed by three small rivers, for the sampling process, the Total Area was divided in two sampling areas: the Area I (under the influence of the rivers) and the Area II (without this influence). Some differences between the two regions were observed.

This paper describes the anatomy and morphology of the complex nectary systems of the tropical tree Guarea macrophylla Vahl (Meliaceae) and presents the first record of extrafloral nectaries occurring on reproductive organs (fruits) of a member of the order Sapindales. The extrafloral nectaries of G. macrophylla occur on petioles, petiolules, the abaxial surface of all leaflets, leaf buds, and over the surface of fruits. All extrafloral nectaries are distinctly raised above the surface. Foraging ants collect extrafloral nectar on Guarea trees both day and night. We suggest that the presence of extrafloral nectaries might be a useful taxonomic character for the identification of Guarea species.

Data concerning genealogy, milk yield, lactation length, caving interval and lac at first calving, obtained from Murrah buffaloes on a farm In São Paulo state, Brazil, were studied in order to determine the presence of inbreeding in the herd and to evaluate its effects as well as the effects of some environmental factors on productive and reproductive traits. The mean inbreeding coefficient estimated for 2414 animals, born from 1965 to 1995 was 2.94%. of these animals, 47% mere considered inbred with a mean inbreeding coefficient equal 6.25%. Milk production, lactation length, calving interval and age at first calving averaged 1493.3 +/- 60.7 kg, 271.0 +/- 37.3 days, 385.0 +/- 53.4 days and 33.05 +/- 6.05 months, respectively. Among all four traits, only lactation length was affected by inbreeding.

The floristic composition was studied in an area of semideciduous mesophytic forest, with 120 ha, in the municipality of Botucatu, SP, Brazil. This forest lies on the ascent of the Cuesta of Botucatu. Due to the heterogeneity of the relief, for the sampling process, the forest was divided in three regions: the upper one (comprising the strip of forest on the plateau), middle on (the forestal area lied on the ascent properly said) and the lower one (the strip of forest on the plain). The tree sampling regions showed some differences among them and this must be connected to the differences in the soil.

Statecharts are an extension to finite state machines with capability for expressing hierarchical decomposition and parallelism. They also have a mechanism called history, to remember the last visit to a superstate. An algorithm to create a reachability tree for statecharts is presented. Also shown is how to use this tree to analyse dynamic properties of statecharts; reachability from any state configuration, usage of transitions, reinitiability, deadlocks, and valid sequence of events. Owing to its powerful notation, building a reachability tree for statecharts presents some difficulties, and we show how these problems were solved in the tree we propose.

(CO2)-C-14 production from [1-C-14] glucose, the rate of glycolysis measured by the value of lactate production and the activities of various enzymes were determined in buffalo erythrocytes. Buffalo red cell glycolytic metabolites were estimated and used for the calculation of the mass action ratios of reactions catalyzed by the glycolytic enzymes of Bubalus bubalis. A comparison of the values of the mass action ratios with the equilibrium constants of the various glycolytic reactions indicate that hexokinase, phosphofructokinase, phosphoglycerate kinase and pyruvate kinase reactions are displaced from equilibrium, suggesting a regulatory role for each of these enzymes in buffalo erythrocyte glycolysis. The cytogenetic study of 182 river buffalo (Bubalus bubalis L., 2n=50) of Murrah, Mediterranean and Jaffarabadi breeds, from the State of São Paulo, was carried out to characterize their chromosomes and to detect possible chromosomal abnormalities. The karyotypes were indistinguishable with conventional staining as well as with C and replication R banding techniques. In about 44% of the sample (8 males and 72 females), an X marker chromosome due to a fragile site was shown. The frequency of metaphases expressing the fragility site on the X was highly variable, from 2.86 to 41.03%. In females, the fragile site, rarely appeared on both X chromosomes. Most of the metaphases showed only 1 marker chromosome. In R-banded metaphases using 5-bromodeoxyuridine (BrdU) treatment, it corresponded in general to the late replicating X chromosome. No correlation between the X fragile site and altered phenotype was found. Structural and numerical chromosome rearrangements were ruled out in the present sample of buffalo. Yellows diseases associated with phytoplasmas cause high mortality in China-tree (Melia azedarach) in Argentina, but there has been no previous large-scale survey to determine their diversity and geographical distribution. To assess the presence and identity of phytoplasmas affecting this species throughout the country, 425 samples of symptomatic trees collected at different geographic locations were analysed by a polymerase chain reaction (using universal and group-specific primers) and restriction fragment length polymorphism. Phytoplasmas belonging to 16SrIII-B group were detected at almost every location sampled, whereas 16SrXIII-C group phytoplasmas, reported for the first time in Argentina, were only found in two regions sharing similar agro-ecological characteristics (Northeast provinces and Tucuman). Double infections with 16SrIII-B and 16SrXIII-C group phytoplasmas were also recorded. Nucleotide sequencing of the 16S rDNA of three Argentinian 16SrXIII-C group phytoplasma isolates revealed high identity (99.6-99.3%) with the CbY1 isolate reported from Bolivia.
