Juniperus virginiana (eastern redcedar) is encroaching into mesic prairies of the southern Great Plains, USA, and is altering the hydrologic cycle. We used the thermal dissipation technique to quantify daily water use of J. virginiana into a mesic prairie by measuring 19 trees of different sizes from different density stands located in north-central Oklahoma during 2011. We took the additional step to calibrate our measurements by comparing thermal dissipation technique estimates to volumetric water use for a subset of trees. Except for days with maximum air temperature below -3 degrees C, J. virginiana trees used water year round, reached a peak in late May, and exhibited reduced water use in summer when soil water availability was low. Overall daily average water use was 24 l (+/- 21.81 s.d.) per tree. Trees in low density stands used more water than trees with similar diameters from denser stands. However, there was no difference in water use between trees in different density stands when expressed on a canopy area basis. Approximately 50% of variation in water use that remained after accounting for the factors site, tree, and day was explained using a physiologically-based model that included daily potential evapotranspiration, maximum vapour pressure deficit, maximum temperature, solar radiation, and soil water storage between 0 and 10 cm. Our model suggested that a J. virginiana woodland with a closed canopy is capable of transpiring almost all precipitation reaching the soil in years with normal precipitation, indicating the potential for encroachment to reduce water yield for streamflow and groundwater recharge. Hot spots, hot moments, and spatio-temporal controls on soil CO2 efflux in a water-limited ecosystem

Soil CO2 efflux is the primary source of CO2 emissions from terrestrial ecosystems to the atmosphere. The rates of this flux vary in time and space producing hot moments (sudden temporal high fluxes) and hot spots (spatially defined high fluxes), but these high reaction rates are rarely studied in conjunction with each other. We studied temporal and spatial variation of soil CO2 efflux in a water-limited Mediterranean ecosystem in Baja California, Mexico. Soil CO2 efflux increased 522% during a hot moment after rewetting of soils following dry summer months. Monthly precipitation was the primary driver of the seasonal trend of soil CO2 efflux (including the hot moment) and through changes in soil volumetric water content (VWC) it influenced the relationship between CO2 efflux and soil temperature. Geostatistical analyses showed that the spatial dependence of soil CO2 efflux changed between two contrasting seasons (dry and wet). During the dry season high soil VWC was associated with high soil CO2 efflux, and during the wet season the emergence of a hot spot of soil CO2 efflux was associated with higher root biomass and leaf area index. These results suggest that sampling designs should accommodate for changes in spatial dependence of measured variables. The spatio-temporal relationships identified in this study are arguably different from temperate ecosystems where the majority of soil CO2 efflux research has been done. This study provides evidence of the complexity of the mechanisms controlling the spatio-temporal variability of soil CO2 efflux in water-limited ecosystems. (C) 2014 Elsevier Ltd. Wood basic density (BD) is related to other wood characteristics and its determination is important in forest inventory, though BD must be differentiated from the apparent density (AD), which relates to the moisture content (MC) of wood. The aim of this study is to demonstrate a reliable conversion from BD to AD for any MC of Eucalyptus grandis wood based on two exponential and linear models that relate volumetric shrinkage to MC. To this end, wood specimens were submitted to drying and the volumetric shrinkage was determined as a function of MC. The two models proved to be efficient in the conversion of BD to AD and vice versa.

In the second half of the last century the automobile industries suffered from the petroleum crisis caused mainly by the wars in the Middle East. These crises led the automakers rethink their vehicles. One of the most important events after that was the adoption of new steels by the industry. One example is the TRIP steels (Transformationinduced plasticity). It is known that the macroscopic behavior of a material is strongly dependent on its microstructure and therefore the quantitative metallography is important to understand and relate the material properties to its microstructure. In this work, different specimens of TRIP steels were etched using LePera reagent. The obtained images were analyzed using digital processing. Using the ImageJ software the methods threshold and watershed were studied as well as a comparison with the ASTM E562 standard. The methods were compared and finally the morphological characteristics and volumetric fraction of the retained austenite and martensite phases were analyzed. The results showed that the threshold led to a higher number of identified grains with lower mean area and total area fraction than the watershed method and ASTM standard

Objective The aim of this study was to evaluate repair after endodontic surgery using two- and tridimensional imaging methods. Materials and methods Periapical radiographs and cone beam computed tomography (CBCT) were performed before the surgeries and after 48 h (baseline), 4 months, and 8 months. The area (square millimeters) of periapical lesions in CBCT and in radiographs was compared regarding the percentage of repair. In the CBCT, multiple areas were converted to volume. Repeated-measures analyses and paired t tests (α=0.05) were used to compare the methods. Correlation coefficients were calculated between the periods of evaluation within the CBCT volumetric analysis. Bland-Altman plots were used to compare the methods, based on the 95 % limits of agreement for the difference of the means. Results Baseline showed a larger lesion volume (192.54 mm3 ) than 4-month (79.79 mm3 ) and 8-month (47.51 mm3 ) periods. No differences were found in the percentage of repair in the first 4 months and after 8 months. The volumetric analysis showed a higher percentage of repair when the first and last 4 months were compared. No differences were found in the percentage of repair by area in the CBCTs. Repair of 73 % was obtained after 8 months. Similar results were observed by the Bland-Altman agreement analyses. Conclusions The percentage of repair varied after 8 months, when lower values were obtained by volumetric evaluation. Clinical relevance Considering the outcome at follow-up periods over 4 months, tridimensional evaluation by CBCT is more capable of determining the absence of periapical bone repair than conventional two-dimensional radiographs. Therefore, the use of CBCT would be suggested only for more complex cases with slower evolution of repair or for the association of factors that make prognosis difficult after surgery.

This study used histomorphometric analysis to investigate the effect of sodium alendronate, used for the treatment of osteoporosis, on the repair of surgically-induced bone defects in the tibia of castrated rats. Methods: The castrated animals were given subcutaneous injections of sodium alendronate (0.7mg/Kg) diluted in saline once a week; the control animals were given the same dose of saline. At 16, 30 and 44 days after the first injection of sodium alendronate, the animals were sacrificed and the right tibias were removed and processed for histomorphometric analysis. The volumetric bone mineral density was estimated by a reticular grid (25 points) attached to a light microscope. The number of points on the bone tissue was counted in the histological sections, totaling 100 points/animal. Results: The results revealed that sodium alendronate stimulated bone formation in castrated rats in all occasions, mainly at 16 and 30 days. Conclusion: Sodium alendronate affects mineral homeostasis, promoting bone repair.

Objective To assess the expression of rabies virus G-glycoprotein (RVGP) expression using Semliki Forest virus as a vector in combination with BHK-21 cells cultured in suspension. Results A multilevel factorial design was used to quantify effects of temperature (33–37 C), fresh medium addition after the viral adsorption step (100–200 % with respect to the initial cell suspension volume before infection) and harvest time (8–40 h) on RVGP production. Experimental runs were performed in 24-well cell culture plates at a multiplicity of infection (MOI) of 16. An additional experiment in spinner-flask was performed at MOI of 9, using the optimal conditions determined in cell culture plates. Values for temperature, fresh medium addition and harvest time of 33 C, 100 % and 16 h, respectively, ensured the optimal RVGP production in culture plates. The volumetric yield (239 ng ml-1 ) in these conditions was higher than that reported previously for adherent cell culture. In spinner-flasks, the volumetric yield was improved (559 ng ml-1 ). Conclusion These results establish the basis for designing bioprocess to produce RVGP.
