Biochemical composition of the spent ovaries of 23 different species of freshwater fishes showed a much lower value for protein and fat with higher moisture content than those of the ripe eggs, suggesting an inverse relationship between fat and moisture. Ovaries during the recovering phase showed fairly high values of phosphorus, but lower than those for the ripe eggs. Higher values of calcium and iron in spent ovaries than those in ripe eggs probably suggest that there is a greater requirement of calcium and iron during the spent phase of the gonads.

A laboratory trial was conducted to determine the digestible protein and energy value of fish meal, dextrin, fish oil and soybean oil for Thai sharpunti (Puntius gonionotus Bleeker). A reference diet containing 35% protein was formulated in which fish meal was the sole source of protein. Five test diets were formulated using reference diet and individual test ingredients (fish meal, dextrin, fish oil and soybean oil). Each treatment had three replicates with 15 fish per replicate. Fish were fed twice daily at the rate of 5% of their body weight. The result of the study indicated that the dietary protein in both reference and test diets were well digested and the apparent protein digestibility (APD) values of test diets ranged between 82.81 and 85.99%. The APD value of fish meal protein was 88.05%. The apparent digestible energy (ADE) value for the test ingredients ranged between 70.79 and 85.80% with soybean oil having the highest and fish meal the lowest value. The ADE values calculated in terms of Kcal/g of ingredients were 3.68, 3.22, 4.38 and 4.44 Kcal/g for fish meal, dextrin, fish oil and soybean oil respectively.

During the low temperature setting of fish paste, myosin heavy chain (MHC) is polymerized to cross-linked myosin heavy chain (CMHC), which is considered to occur by the action of endogenous transglutaminase (TGase). In this study the contribution of TGase on the setting of Alaska pollack surimi at different temperatures was studied. Alaska pollack surimi was ground with 3% NaCl, 30% h2o and with or without ethylene glycol bis (β-aminoethylether) N, N, N¹,N¹- tetra acetic acid (EGTA), an inhibitor of TGase. Among the pastes without EGTA, highest TGase activity was observed at 25°C but breaking force of the gel set at 25°C was lower than that set at 30°, 35°, and 40°C. Addition of EGTA (5m mol/kg) to the paste suppressed TGase activity at all setting temperatures from 20° to 40°C. Gelation of the pastes and cross-linking of MHC on addition of EGTA were suppressed completely at 20° and 25°C, partially at 30° and 35°C, and not at all at 40°C. The findings suggested that during the setting of Alaska pollack surimi TGase mediated cross-linking of MHC was strong at around 25°C but the thermal aggregation of MHC by non-covalent bonds was strong at above 35°C. Setting of surimi at 40°C and cross-linking of its MHC did not involve TGase.

A laboratory trial was conducted to determine the optimum dietary protein to energy (P/E) ratio of African catfish, Clarias gariepinus. The experiment was carried out in a flow-through system for 6 weeks. There were 12 treatments each with two replicates having 10 fish each with a mean initial weight of 1.80 ± 0.02g. Twelve semi-purified diets were formulated with four digestible crude protein levels (23, 26.5, 30 and 33.5%) and three digestible energy levels (2.25, 2.75 and 3.25 Kcal/g). The fish were fed three times daily at satiation level. The results of the study showed that, diet containing 33.5% digestible protein and 2.75 kcal/g digestible energy with a protein to energy ratio of 121 .8 (mg protein/kcal) appeared to be best utilized for growth.

Effect of gibbing process on the protein hydrolysis in terms of free alpha amino nitrogen (FAN) content during the ripening of barrel salted herring at low temperature (4°C) was investigated. For this purpose North Sea herring (Clupea harengus) from north-east British coast was salted in polypropylene barrels and allowed to ripen at 4°C. This process of barrel salting was carried out for whole fish in one batch and gibbed fish in another batch. The investigation was performed by using new salt and used salt in separate barrels for each batch of experimental fish. Results of the present study show that protein hydrolysis was significantly higher in the ripened salt-herring produced from whole fish which was found to have more characteristic sensory properties than those produced from gibbed fish. Similar result (proteolysis) was obtained when the investigation was repeated for the spent herring although the spent herring fails to produce a ripened product with the desired characteristic sensory attributes, compared to those of pre-spawning herring.

A laboratory trial was conducted in a sea water recirculatory system to study the nutrient digestibility coefficients of diets with varying energy to protein ratios in Japanese flounder Paralicthys olivaceus. Six different experimental diets with two protein levels (45 and 55%) having six different energy to protein ratio of 87, 90, 94, 107, 110 and 114 were formulated using white fish meal and casein as protein sources. The results of the study showed that the apparent protein digestibility (APD) value ranged between 90.59 to 91.61% and there were no significant differences (P>0.05) between the APD values of diets 1, 2, 3, 4 and 6. The apparent lipid digestibility (ALD) values of diets ranged between 88.24 to 90.18%. The apparent energy digestibility (AED) values ranged between 80.55 to 87.52% with diet 3 producing significantly the highest AED value. In general, except in diet 1 the ALD and AED values increased with the increase of dietary lipid at both protein levels. The results of the present investigation indicated that Japanese flounder can efficiently digest the dietary nutrients at varying energy to protein ratios.

A 30-day experiment was conducted to determine the effects of varying feeding rates on the growth of fry of silver dollar, Metynnis schreitmulleri (Ahl). Silver dollar fry with an average initial body weight of 1.100 ± 0.029 g were collected from a local fish breeder and fed a diet (35% protein and 6% fat) at the rate of 3, 6 and 9% of body weight per day in two equal meals. Per cent weight gain increased from 54.54 to 118.18 with increased feeding rates, which were significantly different (p<0.05) from each other. The highest specific growth rate was obtained in the fry fed at 9% body weight per day. In another study for 30 days, the effects of feeding frequency on growth, feed conversion and protein efficiency of silver dollar fry were evaluated. Groups of silver dollar fry with an initial individual weight of 0.700 ± 0.019 g were offered feed continuously for 24 hours during the day or night at different time intervals with varying meal sizes. The night time feeding with two meals gave the lowest weight and length gains (0.985 g and 0.30 cm). The growth rates of fish fed during day with three equal-sized meals are significantly (p<0.05) higher (4.66%) than the other treatments.

,The molecular dynamics research of the core domain of p53 protein crystal structure shows that besides the stability in biochemistry this domain also shows a high stability in molecular mechanics. Based on that work, the residue R249 was substituted with amino acids Gly and Ser respectively, and molecular dynamics researches were performed separately. The results show that these substitutions cause a relax tendency between loop2 and 3 domains, leading to an alteration of the whole conformation of p53 core domain and ruining its stability. The results visually explains the mechanism of p53 changes in immunological and biochemical reactions, which are caused by 249 residue substitutions from 3-D structure variations.

Stejnulxin, a novel snake C-type lectin-like protein with potent platelet activating activity, was purified and characterized from Trimeresurus stejnegeri venom. Under non-reducing conditions, it migrated on a SDS-polyacrylamide gel with an apparent molecular mass of 120 kDa. On reduction, it separated into three polypeptide subunits with apparent molecular masses of 16 kDa (alpha), 20 kDa (beta(1)) and 22 kDa (beta(2)), respectively. The complete amino acid sequences of its subunits were deduced from cloned cDNAs. The N-terminal sequencing and cDNA cloning indicated that beta(1) and beta(2) subunits of stejnulxin have identical amino acid sequences and each contains two N-glycosylation sites. Accordingly, the molecular mass difference between 1 and 2 is caused by glycosylation heterogenity. The subunit amino acid sequences of stejnulxin are similar to those of convulxin, with sequence identities of 52.6% and 66.4% for the U. and beta, respectively. Stejnulxin induced human platelet aggregation in a dose-dependent manner. Antibodies against UNA inhibited the aggregation response to stejnulxin, indicating that activation of alpha(IIb)beta(3) and binding of fibrinogen are involved in stejnulxin-induced platelet aggregation. Antibodies against GPIbalpha or alpha(2)beta(1) as well as echicetin or rhodocetin had no significant effect on stejnulxin-induced platelet aggregation. However, platelet activation induced by stejnulxin was blocked by anti-GPVI antibodies. In addition, stejnulxin induced a tyrosine phosphorylation profile in platelets that resembled that produced by convulxin. Biotinylated stejnulxin bound specifically to platelet membrane GPVI.

A platelet glycoprotein Ib-binding protein, termed TSV-GPIb-BP, was isolated from the venom of Trimeresurus stejnegeri. On SDS-polyacrylamide gel electrophoresis, TSV-GPIb-BP showed a single band with an apparent molecular weight of 28,000 and two distinct bands with apparent molecular weights of 16,000 and 15,000 under non-reducing and reducing conditions, respectively. cDNA clones containing the coding sequences for both TSV-GPIb-BP subunits were isolated and sequenced. The deduced amino acid sequences of TSV-GPIb-BP subunits were confirmed by N-terminal protein sequencing and trypsin-digested peptide mass fingerprinting. Interestingly, the a subunit of TSV-GPIb-BP is identical to that of alboaggregin-B, and the sequence identity of their beta subunits is 94.3%. TSV-GPIb-BP inhibited ristocetin-induced human platelet agglutination in platelet-rich plasma under lower dosages (<5 mug/ml). On the other hand, it directly aggregated washed human platelets in the absence of additional Ca2+ or any other cofactors under higher dosages (>5 mug/ml). This platelet aggregation activity was dose-dependently inhibited by specific GPIbalpha antibodies, but not by those antibodies against platelet GPIa, GPIIa, GPIIb and GPIIIa. To find out a suitable balance diet for Cirrhinus. cirrhosus fry substituting fish meal protein by full-fat soybean meal protein as dietary protein source for C. cirrhosus fry (1.29g) was studied in 12 aquaria for 60 days. The fishes were fed with four iso-nitrogenous (30% protein) and iso-caloric experimental diets viz. diet 1 (100% fish meal protein), diet 2 (75% fishmeal protein and 25% soybean meal protein), diet 3 (50% fishmeal protein and 50% soybean meal protein) and diet 4 (25% fish meal protein and 75% soybean meal protein). Among the diets the best growth, FCR and PER were obtained with diet 3 fed fishes and no significant difference (p<0.05) was observed between diet 1 and 2 and diet 4 offered the worst performance. Therefore, considering fish growth, FCR and PER, it can be suggested that fish meal protein might be replaced 50% by soybean meal protein in the diet of C. cirrhosus fry for better growth.

In mammals, trefoil factor family (TFF) proteins are involved in mucosal maintenance and repair, and they are also implicated in tumor suppression and cancer progression. A novel two domain TFF protein from frog Bombina maxima skin secretions (Bm-TFF2) has been purified and cloned. It activated human platelets in a dose-dependent manner and activation of integrin a(11b)beta(3) was involved. Aspirin and apyrase did not largely reduce platelet response to Bm-TFF2 (a 30% inhibition), indicating that the aggregation is not substantially dependent on ADP and thromboxane A2 autocrine feedback. Elimination of external Ca2+ with EGTA did not influence the platelet aggregation induced by Bm-TFF2, meanwhile a strong calcium signal (cytoplasmic Ca2+ release) was detected, suggesting that activation of phospholipase C (PLC) is involved. Subsequent immunoblotting revealed that, unlike in platelets activated by stejnulxin (a glycoprotein VI agonist), PLC gamma 2 was not phosphorylated in platelets activated by Bm-TFF2. FITC-labeled Bm-TFF2 bound to platelet membranes. Bm-TFF2 is the first TFF protein reported to possess human platelet activation activity. (c) 2005 Elsevier Inc. All rights reserved.

Mucetin (Trimeresurus mucrosquamatus venom activator, TMVA) is a potent platelet activator purified from Chinese habu (Trimeresurus mucrosquamatus) venom. It belongs to the snake venom heterodimeric C-type lectin family and exists in several multimeric forms. We now show that binding to platelet glycoprotein (GP) lb is involved in mucetin-induced platelet aggregation. Antibodies against GPIb as well as the GPIb-blocking C-type lectin echicetin inhibited mucetin-induced platelet aggregation. Binding of GPIb was confirmed by affinity chromatography and Western blotting. Antibodies against GPVI inhibited convulxin- but not mucetin-induced aggregation. Signalling by mucetin involved rapid tyrosine phosphorylation of a number of proteins including Syk, Src, LAT and PLCgamma2. Mucetininduced phosphorylation of the Fcgamma chain of platelet was greatly promoted by inhibition of alpha(llb)beta(3) by the peptidomimetic EMD 132338, suggesting that phosphatases downstream Of alpha(llb)beta(3) activation are involved in dephosphorylation of Fcgamma. Unlike other multimeric snake C-type lectins that act via GPIb and only agglutinate platelets, mucetin activates alpha(llb)beta(3). Inhibition Of alpha(llb)beta(3) strongly reduced the aggregation response to mucetin, indicating that activation Of alpha(llb)beta(3) and binding of fibrinogen are involved in mucetin-induced platelet aggregation. Apyrase and aspirin also inhibit platelet aggregation induced by mucetin, suggesting that ADP and thromboxaneA(2) are involved in autocrine feedback. Sequence and structural comparison with closely related members of this protein family point to features that may be responsible for the functional differences.

The nuclear RNA binding protein, FCA, promotes Arabidopsis reproductive development. FCA contains a WW protein interaction domain that is essential for FCA function. We have identified FY as a protein partner for this domain. FY belongs to a highly conserved group of eukaryotic proteins represented in Saccharomyces cerevisiae by the RNA 3' end-processing factor, Pfs2p. FY regulates RNA 3' end processing in Arabidopsis as evidenced through its role in FCA regulation. FCA expression is autoregulated through the use of different polyadenylation sites within the FCA pre-mRNA, and the FCA/FY interaction is required for efficient selection of the promoter-proximal polyadenylation site. The FCA/FY interaction is also required for the downregulation of the floral repressor FLC. We propose that FCA controls 3' end formation of specific transcripts and that in higher eukaryotes, proteins homologous to FY may have evolved as sites of association for regulators of RNA 3' end processing.

The author reviews the advances in the oil and meal industries related to the oil sardine fishery (Sardinella longiceps) since the 1920s. Data on the production of by-produced produced in Kerala over the period 1964- 69 are tabulated. Details of the properties of the commercial oil are given, and the values compared to those for other similar oils. The use of oil sardine for industrial purposes - the oil has been used to cure leather, temper metals and as fungicides or insecticides - and the production of fish meal and fish protein concentrate is considered.
