The objective of this study was to investigate morphological variation in traits of systematic relevance and the phylogenetic position, ecology, and reproductive biology of the shrimp Lysmata rauli Laubenheimer and Rhyne, 2010 (Caridea: Hippolytidae), described based only on a single specimen collected in Salvador, Bahia, Brazil. We analyzed a total of 89 specimens from Camamu Bay, Bahia (n = 88) and from S3o Vicente estuary, São Paulo (n = 1). Considerable morphological variation was detected in the rostral spine series, number of segments on the carpus and merus of pereiopod 2, number of spiniform setae on the ventrolateral margin of merus and on the ventral margin of propodus of pereiopods 3-5. Importantly, L rauli can be distinguished neither using morphology, nor coloration from the Indo-Pacific L. vittata (Stimpson, 1860). Furthermore, molecular phylogenetic analyses (using the 16S mt DNA fragment) did not reveal any considerable genetic dissimilarities between L rauli and L vittata. Thus, our results clearly indicate that L rauli is not a new species but a junior synonym of L vittata. The high density observed within the structures of oyster farming indicates that the invasive L vittata lives in crowds in Brazil. The studied population was composed of males, hermaphrodites, and transitional individuals (having characteristics of males and hermaphrodites). The above information suggests that L rauli is a protandric simultaneous hermaphrodite, as it has been observed in all species of Lysmata that have been investigated. Dengue virus is a mosquito-borne flavivirus that has a large impact in global health. It is considered as one of the medically important arboviruses, and developing a preventive or therapeutic solution remains a top priority in the medical and scientific community. Drug discovery programs for potential dengue antivirals have increased dramatically over the last decade, largely in part to the introduction of high-throughput assays. In this study, we have developed an image-based dengue high-throughput/high-content assay (HT/HCA) using an innovative computer vision approach to screen a kinase-focused library for anti-dengue compounds. Using this dengue HT/HCA, we identified a group of compounds with a 4-(1-aminoethyl)-N-methylthiazol-2-amine as a common core structure that inhibits dengue viral infection in a human liver-derived cell line (Huh-7.5 cells). Compounds CND1201, CND1203 and CND1243 exhibited strong antiviral activities against all four dengue serotypes. Plaque reduction and time-of-addition assays suggests that these compounds interfere with the late stage of viral infection cycle. These findings demonstrate that our image-based dengue HT/HCA is a reliable tool that can be used to screen various chemical libraries for potential dengue antiviral candidates. © 2013 Cruz et al.

The influence of high hydrostatic pressure (HHP) on Pêra Rio orange juice was investigated using response surface methodology. A central composite design was used to evaluate the effects of three processing conditions (independent variables), namely pressure (100-600 MPa), temperature (30-60 °C) and time (30-360 s), on the native microflora and pectin methylesterase (PME) activity of orange juice. Analysis of variance showed that second-order polynomial models fitted well with the experimental data for PME residual activity (R2 = 0.9586, p < 0.001) and aerobic microorganism count (R2 = 0.9879, p < 0.001). The optimum HHP processing conditions to produce orange juice with PME residual activity of less than 20 % and low microorganism count (<2 log cycles CFU/mL) were 550 to 600 MPa, 55 to 60 °C and 330 to 360 s. © 2013 Springer Science+Business Media New York.

This article assesses the limitations and potentials of the National High School Exam (ENEM) as an indicator of school effectiveness in Brazil, and considers the effects of introducing contextual variables. A multilevel regression analysis was performed on three levels (individual, school and state) using microdata on 17,359 schools from 2009 and 2010. Contextual factors made it possible to explain 79% of the difference between schools. The raw average and value-added (random effect at the school level) produced contrasting evaluations in 34% of cases; and the average was more stable (r = 0.8) than value-added (r = 0.5) in both years. Various shortcomings in the ENEM as an indicator of school effectiveness were identified. The results show that this league table reveals more about socioeconomic conditions than the schools' own merit, in other words the value-added they are supposedly providing to the students.

Meio ambiente e Ética são temas recorrentes nas pautas de discussões sobre os desafios da educação brasileira. O mundo está imerso em uma crise ambiental e ética que nos convida ao movimento dialético da ação-reflexão-ação. O objetivo deste trabalho foi investigar as concepções e as relações entre os Temas Transversais Meio Ambiente e Ética dos professores de Ciências, técnicos educacionais e alunos de 5ª a 8ª série do Ensino Fundamental, de uma escola da rede particular de Belém. O estudo apoiou-se em Sauvé, Reigota, Sánchez Vásquez, Leff, Grün, Valls, Bardin, Zabala, Freire, Lüdke, André, entre outros. O caminho metodológico foi o da abordagem qualitativa. Para mergulhar nos diálogos intersubjetivos, que são as práticas pedagógicas, foram utilizadas as técnicas de pesquisa de campo e pesquisa documental. A primeira consistiu na realização de entrevistas semiestruturadas com 16 estudantes, 3 professores e 2 técnicos da Escola - “Alfa”; a segunda envolveu os Parâmetros Curriculares Nacionais, os livros didáticos de Ciências e os documentos entregues aos alunos e professores, em 2008, na Escola. A interpretação dos dados se baseou na análise de conteúdo, em sua modalidade temática. Foram trabalhados dois temas: Concepções de Meio Ambiente e Concepções de Ética. A unidade de contexto, o ―pano de fundo‖ do estudo, foi uma escola da rede particular de ensino, dotada de boa infraestrutura, cuja clientela é formada, em sua maioria, por alunos da classe média alta. Para definição das categorias, foi preparado um caderno com as entrevistas, o que facilitou o agrupamento, a classificação e a análise interpretativa dos dados. Isso resultou na síntese em sete categorias de análise: Concepção de Meio Ambiente como natureza; Concepção de Meio Ambiente como o local onde se vive; Concepção de Meio Ambiente como relação dos seres entre si e deles com o ambiente; Concepção de Meio Ambiente como sustentabilidade; Concepção de Ética como respeito; Concepção associada a princípios e valores e Concepção relacionada ao meio ambiente e ao respeito às pessoas. Os resultados revelam, contraditoriamente, a prevalência da lógica disciplinar nos PCN e na Escola ―Alfa‖; os livros adotados pela Escola não se limitam aos conteúdos factuais e conceituais; em relação às concepções de Meio Ambiente e Ética, os entendimentos do que vem a ser ambiente e ética são reducionistas. Os sujeitos, de uma maneira geral, não estabelecem conexão necessária entre Ambiente e Ética. Nas turmas de 5ª a 8ª série, a Escola não trabalha os Temas de forma efetiva. As reflexões finais pontificam um convite à Escola para refletir e buscar alternativas que revertam o quadro de suas ações educacionais relativas ao Meio Ambiente e à Ética. As escolas precisam se sentir desafiadas a inserir a Educação Ambiental em seu cotidiano, com o fito de colaborar na construção de uma educação cidadã.

Leprosy in children is correlated with community-level factors, including the recent presence of disease and active foci of transmission in the community. We performed clinical and serological examinations of 1,592 randomly selected school children (SC) in a cross-sectional study of eight hyperendemic municipalities in the Brazilian Amazon Region. Sixty-three (4%) SC, with a mean age of 13.3 years (standard deviation = 2.6), were diagnosed with leprosy and 777 (48.8%) were seropositive for anti-phenolic glycolipid-I (PGL-I). Additionally, we evaluated 256 house-hold contacts (HHCs) of the students diagnosed with leprosy; 24 (9.4%) HHC were also diagnosed with leprosy and 107 (41.8%) were seropositive. The seroprevalence of anti-PGL-I was significantly higher amongst girls, students from urban areas and students from public schools (p < 0.0001). Forty-five (71.4%) new cases detected amongst SC were classified as paucibacillary and 59 (93.6%) patients did not demonstrate any degree of physical disability at diagnosis. The results of this study suggest that there is a high rate of undiagnosed leprosy and subclinical infection amongst children in the Amazon Region. The advantages of school surveys in hyperendemic areas include identifying leprosy patients at an early stage when they show no physical disabilities, preventing the spread of the infection in the community and breaking the chain of transmission.

Less than half of adolescents reach the recommended 300 minutes per week of physical activity (PA). Physical education classes and sports participation provide opportunities for adolescents to accumulate more time for PA practice; however, little is known about the influence of these variables on the level of total physical activity of adolescents. The aim of this study was to investigate the association between the practice of physical education (PE) in schools and sports activities (SA) with the practice of total PA of adolescents. The study was cross-sectional and involved 467 adolescents of high school (15.8 ± 0.9 years-old) from the city of Rio Claro, in the State of São Paulo. Participants completed the Physical Activity Questionnaire to Older Children (PAQ-C) and questions related to the practice of PE and SA in schools. We performed a logistic regression with p<0.05 using SPSS. Girls had lower prevalence of PA than boys, 9.4% and 26.8%, respectively. Boys who did not participate of PE classes (OR=0.25, 95% CI=0.09-0.66) and SA in schools (OR=0.34, 95% CI=0.12-0.95) were less likely to be active in PA than boys who practiced these activities. The participation in PE classes or engagement in some SA were positively associated with the practice of total PA in boys.

Chikungunya virus (CHIKV) is a mosquito-borne arthrogenic alphavirus that causes acute febrile illness in humans accompanied by joint pains and in many cases, persistent arthralgia lasting weeks to years. The re-emergence of CHIKV has resulted in numerous outbreaks in the eastern hemisphere, and threatens to expand in the foreseeable future. Unfortunately, no effective treatment is currently available. The present study reports the use of resazurin in a cell-based high-throughput assay, and an image-based high-content assay to identify and characterize inhibitors of CHIKV-infection in vitro. CHIKV is a highly cytopathic virus that rapidly kills infected cells. Thus, cell viability of HuH-7 cells infected with CHIKV in the presence of compounds was determined by measuring metabolic reduction of resazurin to identify inhibitors of CHIKV-associated cell death. A kinase inhibitor library of 4,000 compounds was screened against CHIKV infection of HuH-7 cells using the resazurin reduction assay, and the cell toxicity was also measured in non-infected cells. Seventy-two compounds showing >= 50% inhibition property against CHIKV at 10 mu M were selected as primary hits. Four compounds having a benzofuran core scaffold (CND0335, CND0364, CND0366 and CND0415), one pyrrolopyridine (CND0545) and one thiazol-carboxamide (CND3514) inhibited CHIKV-associated cell death in a dose-dependent manner, with EC50 values between 2.2 mu M and 7.1 mu M. Based on image analysis, these 6 hit compounds did not inhibit CHIKV replication in the host cell. However, CHIKV-infected cells manifested less prominent apoptotic blebs typical of CHIKV cytopathic effect compared with the control infection. Moreover, treatment with these compounds reduced viral titers in the medium of CHIKV-infected cells by up to 100-fold. In conclusion, this cell-based high-throughput screening assay using resazurin, combined with the image-based high content assay approach identified compounds against CHIKV having a novel antiviral activity -inhibition of virus-induced CPE - likely by targeting kinases involved in apoptosis.

Objectives: To evaluate if the prevalence of cervical smear results varies between pregnant and nonpregnant women stratified by age group.Study design: Observational analytical study with a total sample of 1,336,180 pregnant and nonpregnant women, aged between 20 and 34 years, who underwent cervical cancer screening in the Primary Health Care of the national health system in the area of Campinas in Brazil during the period of 2005-2009. The source is the information system for cervical cancer screening. Data collected on abnormal cervical smears were analyzed using the Chi-square test and Fisher's exact test and the magnitude of the association between pregnancy and high-grade squamous epithelial lesions was analyzed by odds ratio (OR) and estimated values with confidence intervals (CI) of 95%.Results: 15,190 pregnant women and 395,961 non-pregnant women were analyzed and fulfilled the inclusion criteria. Regardless of age, no statistical differences were observed for high-grade squamous intraepithelial lesion prevalence (OR 0.90; CI 0.66-1.23). Taking into account the five-year age groups, however, low-grade squamous intraepithelial lesion was less prevalent in pregnant women aged 20-24 (OR 0.71; 0.54-0.95) and 25-29 years (OR 0.56; 0.35-0.89); also, atypical squamous cells of undetermined significance was more prevalent in non-pregnant women aged 25-29 years (OR 0.72; 0.54-0.97).Conclusion: The study showed that the cytological prevalence of high-grade squamous intraepithelial lesion was similar in pregnant and non-pregnant women, regardless of age. The results indicate that there are no reasons for specific approaches to cervical cancer screening for pregnant women. The examination should be carried out only on pregnant women who have not been tested according to current recommendations. (C) 2014 Elsevier Ireland Ltd. All rights reserved.

The aim of this study was to evaluate the effects of a Gallium Arsenide (GaAs) laser, using a high final energy of 4.8J, during muscle regeneration after cryoinjury. Thirty Wistar rats were divided into three groups: Control (C, n=10); Injured (I, n=10) and Injured and laser treated (Injured/LLLT, n=10). The cryoinjury was induced in the central region of the tibialis anterior muscle (TA). The applications of the laser (904nm, 50mW average power) were initiated 24h after injury, at energy density of 69Jcm(-1) for 48s, for 5days, to two points of the lesion. Twenty-four hours after the final application, the TA muscle was removed and frozen in liquid nitrogen to assess the general muscle morphology and the gene expression of TNF-, TGF-, MyoD, and Myogenin. The Injured/LLLT group presented a higher number of regenerating fibers and fewer degenerating fibers (P<0.05) without changes in the collagen remodeling. In addition, the Injured/LLLT group presented a significant decrease in the expression of TNF- and myogenin compared to the injured group (P<0.05). The results suggest that the GaAs laser, using a high final energy after cryoinjury, promotes muscle recovery without changing the collagen remodeling in the muscle extracellular matrix.
