A highly sensitive amperometric biosensor for determination of carbamate pesticides directly in water, fruit and vegetable samples has been evaluated, electrochemically characterized and optimized. The biosensor strip was fabricated in screen printed technique on a ceramic support using silver-based paste for reference electrode, and platinum-based paste for working and auxiliary electrodes. The working electrode was modified by a layer of carbon paste mixed with cobalt(II) phthalocyanine and acetylcellulose. Cholinesterase (ChE) enzymes with low enzymatic charge were immobilized on this layer. The operational simplicity of the biosensor consists in that a small drop (similar to 50 mu l) of substrate or sample is deposited on a horizontally positioned biosensor strip representing the microelectrochemical cell. The working potential of the biosensor was 370 mV versus Ag/AgI on a ship reference electrode preventing the interference of electroactive species which are oxidable at more positive potentials. The biosensor was applied to investigate the degradation of two reference ChE inhibitors in freeze dried water under different storage conditions and for direct determination of some N-methylcarbamates (NMCs) in fruit and vegetable samples at ppb concentration levels without any sample pretreatment. Despite their spiny nature, Aiphanes palms are popular as ornamentals. They can be grown in various soil conditions, but achieve fast, luxuriant growth in sheltered positions with plenty of water. Shaded conditions are the best when plants are young. The objective of this study was to evaluate the effect of fruit maturation stage and light on the germination of seeds of A. aculeata. Fruits were collected at three different maturation stages: (1) yellow, (2) red, (3) collected on the ground without the fleshy mesocarp (removed by birds and insects). The fleshy mesocarp was also removed in treatments (1) and (2). All fruits were then treated with thiram and 25 seeds per plot were seeded in trays full of sand. The treatments (2) and (3) were subdivided in two other treatments: (a) 12 h photoperiod, (b) continuous darkness. The temperature was maintained at 30 degrees C. Seed germination began 34 days after sowing. After 160 days, the final germination percentage was (1) 54,67%, (2a) 61,33%, (2b) 65,33%, (3a) 38,00% and (3b) 45,33%. The best treatment was (2) independently of the light conditions.

Peroxidase from peach fruit was purified 28.9-fold by DEAE-cellulose, Sephadex G-100 and hydroxylapatite chromatography. The purified enzyme showed only one peak of activity with an optimum pH of 5.0 and temperature of 40 degreesC. The calculated activation energy (Ea) for the reaction was 7.97 kcal/mol. The enzyme was heat-labile in the temperature range of 60 to 80 degreesC with a fast inactivation at 80 degreesC. PAGE of the inactivation course at 70 degreesC showed only one band of activity. Different sugars increased the heat stability of the activity in the following order: sucrose>lactose>glucose>fructose. Measurement of residual activity showed a stabilizing effect of sucrose at various temperature/sugar concentrations (10 to 40%, w/w) with the Ea for inactivation increasing with sucrose concentration from 0 to 20% (w/w). After inactivation at 70 degreesC and 75 degreesC the enzyme was able to be reactivated by up to 40% of the initial activity when stared at 30 degreesC.

A competitive enzyme-linked immunosorbent assay (ELISA) method for carbaryl quantitation in crop extracts was validated by liquid chromatography (LC) with diode array detection (DAD). For this purpose, six crops (banana, carrot, green bean, orange, peach and potato) were chosen for recovery and reproducibility studies. The general sample preparation included extraction with methanol followed by liquid-liquid partitioning and clean-up on Celite-charcoal adsorbent column of the vegetable extracts. ELISA samples consisted of a diluted LC extract in assay phosphate buffer (pH 7.5). The potential effect of methanol in these samples was evaluated. It was observed that a maximum content of 10% methanol present in the assay buffer could be tolerated without expressive losses in the ELISA performance. Under these conditions, a IC50 similar to 1.48 mu g l(-1) was obtained. A minimum matrix effect with a 1:50 dilution of the methanolic extracts in assay buffer was noticed, except for green bean samples that inhibited completely the assay. For the vegetable extracts, the ELISA sensitivities varied from 3.9 to 5.7 mu g l(-1), and good recoveries (82-96%) with R.S.D.s ranging from 5.7 to 12.1% were found. An excellent correlation between the LC-DAD and ELISA techniques was obtained. The confirmation of the carbaryl in less concentrated samples was achieved by LC-mass spectrometry interfaced with atmospheric pressure chemical ionisation. The [M + H](+)= 202 and [M + H-57](+)=145 ions, equivalent to the protonated molecular and l-naphthol ions, respectively, were used to carbaryl identification in these samples. (C) 1998 Elsevier B.V. Action of auxins on the rooting of stem cuttings of kiwi (Actinidia chinensis P. cv Monty). This work studies the effects of some synthetic auxins and B in the rooting of kiwi (Actinidia chinensis Planch cv Monty) stem cuttings. The treatments used were as follows: T1 (H2O); T2 (NAA 300 ppm); T3 (IBA 300 ppm); T4 (NAA 300 ppm + B); T5 (IBA 300 ppm + B); T6 (NAA 0,5%-talc) and T7 (IBA 0,5%-talc), applied to the bases of the cuttings. These were then placed in rooting dishes with pure vermiculite in a misty nebulization chamber until collection day (120 days). The evaluation of auxin and boric acid effects on kiwi stem cuttings were made based on the following observations: 1. The percentage of rooted stem cuttings; 2. reducing sugars and total sugar analyses (in g/100 g of dry matter); and 3. tryptophan analyses (in mu g/100 mg of dry matter). The results show that summer is the best season for rooting Actinidia chinensis Planch cv Monty stem cuttings. The use of IBA talc 0,5% on the bases of the cuttings shamed positive results too.

Esterases are known for their involvement in several physiological processes and high degree of polymorphism, in many organisms. Such polymorphism has been used to characterize species and species groups and to study genetic changes occurred in their evolutionary history. In the present study, the esterase patterns of 19 strains from 10 species representative of the five subgroups of the saltans species group were analyzed using polyacrylamide gel electrophoresis and alpha- and beta- naphthyl acetates as substrates. Fifty-one esterase bands were detected and classified as 31 alpha-esterases, 18 beta-esterases and two alpha/beta-esterases. on the basis of the inhibition patterns using Malathion and eserine sulfate, 34 bands were classified as carboxylesterases, 14 as acethylesterases and three as cholinesterases. Ten gene loci were tentatively established on the basis of data on band position in the gel, substrate preference and inhibition pattern. Twenty bands were species-specific, the remaining being shared by species from the same or different subgroups. Bands detected exclusively in males and bands with a different frequency or degree of expression between sexes were also detected. In the gels prepared for analysis of gene expression in the body parts (head, thorax and abdomen), the degree of expression of the beta-esterases was higher in the thorax, while the alpha-esterases were expressed predominantly in the abdomen and thorax. A global view of the data available at present on the esterases of the species from the saltans group and their degree of polymorphism are presented, as well as the possibility of using some beta-esterases, because of their characteristics in the gels, as markers for species identification.

The development of the ovule and of the fruit of Hypolytrum bullatum and H. schraderianum (Mapanioideae) and of Rhynchospora consanguinea and R. rugosa (Cyperoideae) are described. All species share anatropous, bitegmic and crassinucellate ovules, funicular obturator, megagametophyte of the Polygonum type, presence of starch grains in the mature megagametophyte, free-nuclear endosperm, Onagrad-type embryogeny, testal-tegmic seed, and a simple fruit of the achene type. Rhynchospora species have characters typical of the family: micropyle formed by the inner integument alone; 3-4-layered parietal tissue; and hard achene. Hypolytrum species differ in those characters by presenting a slightly zigzag micropyle formed by both integuments connected with the funicular obturator, 5-8-layered parietal tissue, and fibrous-spongy achene. The peculiar formation of the micropyle in Hypolytrum is a feature reported here for the first time in the family. The ontogeny provides evidence for a better understanding of the dispersal unit in Hypolytrum supporting the classification as a true achene, like that of Rhynchospora, which is characteristic of the family.

Se estudió la ocurrencia de Lonchaeidae en variedades de melocotón, conducidos sobre los patrones ‘Okinawa’ y Umê: Tropical, Ouromel 3, Jóia 4, Régis, Talismã, Aurora 2, Aurora 1, Dourado 2 y Doçura 2. Se colectaron muestras de 30 frutos por planta en el Banco de Germoplasma en Presidente Prudente, Brasil. Se obtuvieron 633 especímenes de Lonchaeidae; 394 de ellos en frutos de variedades sobre ‘Okinawa’ y 239 sobre Umê. Se capturaron especies como Neosilba zadolicha, N. inesperata, N. pendula, N. certa y Neosilba spp. (hembras). N. zadolicha y N. inesperata, se observaron en 77,78% de las muestras de las variedades sobre ‘Okinawa’. Sobre el patrón Umê la mayor incidencia fue de N. zadolicha , N. inesperata y N. pendula (55,6%, 33,3% y 33,3% respectivamente). Las variedades Ouromel 3, Talismã, Doçura 2 y Aurora 2 presentaron mayores infestaciones por Lonchaeidae. N. certa tuvo una menor incidencia y sólo se observó en la variedad Doçura 2 sobre ‘Okinawa’. Plantas con mayor número de frutos presentaron mayor incidencia de moscas por fruto; sin embargo, no hubo correlación entre peso del fruto y número de moscas. No se observó diferencia para peso del fruto y número de moscas por fruto entre los dos patrones, ‘Okinawa’ y Umê. Melocotones crecidos en los patrones ‘Okinawa’ y Umê en Presidente Prudente están infestados por especies de Lonchaeidae que presentan preferencia independientemente del peso de los frutos. El aporte de este trabajo al conocimiento de la comunidad de especies de Lonchaeidae ofrece una base para su control.

Farmers of the Submedio Sao Francisco Region (Brazil) have been spraying amino acids on mango trees with the objective of increasing panicle length and improving fruit retention and quality. This study, done in two experiments, tested the effect of amino acids sprayings at concentrations of 0.0%; 0.02%; 0.04% and 0.06%, on mango plants, 'Tommy Atkins', on the budding phase (panicles with 5 cm), fruit set and fruit growth (5 cm diameter). They were carried out from June to October in 2003, that is the natural period for harvest in the region, and from January to May, in 2004. There were no statistical differences in the first experiment among treatments regarding panicle length and fruit production, probably due to an appropriate management of nutrition, water and plant growth regulators, besides climatic conditions, mainly temperature and solar radiation. In the second experiment, significant increments in the panicle length of 13.37%, 11.70% and 21.4% were observed with amino acids concentrations, compared to the control. Increasing amino acids doses also enhanced the number of fruits per plant, thirty days before the harvest, in 16.17%, 45.32% and 37.38%, respectively, compared to the control, but there were no significant statistical differences. Characteristics of fruit quality during storage, as weight loss, total soluble solids, total titratable acidity and pulp firmness were not significantly affected by amino acids spraying. Changes on those variables were registered as a consequence of fruit ripening. Amino acids sprays lightly delayed the evolution of skin luminosity and Hue of pulp, but the differences could not be visually identified. The concentrations of amino acids were not efficient for improving the natural concentrations of these substances in the leaves, which could be the reason for the non significant effects on the variables analyzed.

From field observations on Drimys brasiliensis, principally in the Botucatu region of São Paulo State, Brazil, new data on the reproductive biology, the rhythm of growth, and the development of lateral cymose inflorescences, flowers and fruits are presented. Pollination accelerates the rate of flower-development for about 4-6 days. Pollination experiments show that D. brasiliensis is not self-sterile; because of mechanical devices the sticky pollen grains do not normally come into contact with the stigmata unless an animal pollen vector is involved. The pollinators are diurnal Coleoptera, Diptera and Thysanoptera which eat from the pollen, lick from the stigmatic exudates and (in case of the flies) probably also from the staminal glands. Fruit- and seedeaters are birds which seem to be the main dispersal agents. Establishment of new individuals normally is through seedlings, but also by vegetative propagation through plagiotropous branches which may root and separate from the mother plant. The morphological, developmental and reproductive aspects in D. brasiliensis are discussed in a wider context, compared with data from other Magnoliidae, and related to aspects of early Angiosperm evolution. © 1980 Springer-Verlag.

Soluble and bound peroxidases were isolated from the pulp of ripening papaya fruit. During papaya ripening, soluble and bound peroxidase activities increased 2.5- and 4.2-fold, respectively. Soluble peroxidase was purified 59-fold by ammonium sulphate precipitation and chromatography on Sephadex G-25, DEAE-cellulose and Sephadex G-100. Bound peroxidase was purified 140-fold by ammonium sulphate precipitation and chromatography on Sephadex G-100 and DEAE-cellulose. Polyacrylamide gel electrophoresis of the purified preparations revealed that both enzymes were highly purified by the procedures adopted. The soluble and bound forms had a Mr of 41 000 and 54 000, respectively. Soluble and bound peroxidases showed optimum activity at pH 6.0 and 5.5, respectively, and were inhibited by p-chloromercuribenzoate, iodoacetamide, N-ethylmaleimide, potassium cyanide and Fe2+. Soluble peroxidase was activated by ammonium sulphate and this activation was prevented by cyanide. © 1990.

The persistence and metabolism of fenthion in orange fruit were studied in field conditions. The fenthion was transformed to fenthion sulfoxide and fenthion sulfone. Sunlight photodegradation experiments showed that this transformation is due to the action of sunlight. Residues were found only in the fruit peel. Fenthion showed a rapid degradation rate with a half-life of ca. 6 days. Fenthion sulfoxide was degraded more slowly with a half-life of ca. 14 days and represented the major residue. Fenthion sulfone was present in low quantities.

One hundred and eleven samples of processed fruit juices (apple, grape, pineapple, papaya, guava, banana and mango) and 38 samples of sound fruits (apple, papaya, mango, pear and peach) produced and marketed in Brazil, were analysed for patulin by HPLC. Only one out of 30 samples of apple juice was found positive at 17 μg/l. Patulin was not detected in the other foodstuffs. It was found in 14 samples of spoiled fruit samples of apple (150-267 μg/kg), pear (134-245 μg/kg) and peach (92-174 μg/kg). Confirmation of the identity of patulin was based on the UV spectrum obtained by the HPLC diode array detector, compared with that of standard patulin, TLC developed by several solvent systems and sprayed with 3-methyl-2-benzothiazolinone hydrazone, and by acetylation with acetic anhydride.
