Optimal conditions for the extraction of casearins from Casearia sylvestris were determined using response surface methodology. The maceration and sonication extraction techniques were performed using a 3 x 3 x 3 full factorial design including three acidity conditions, three solvents of different polarities and three extraction times. The yields and selectivities of the extraction of casearins were significantly influenced by acidity conditions. Taking into account all variables tested, the optimal conditions for maceration extraction were estimated to involve treatment with dichloromethane saturated with ammonium hydroxide for 26 h. Similar yields and selectivities for casearins were determined for sonication extraction using the same solvent but for the much shorter time of I h. The best results for stabilisation of the fresh plant material were obtained using leaves that had been oven dried at 40 degrees C for 48 h. Copyright (c) 2006 John Wiley & Sons, Ltd.

Yttrium-aluminum oxides are interesting compounds and they have been extensively used as host for lasers and phosphors, due to their stable physical and chemical properties. The fabrication of yttrium-aluminum garnet (YAG) has been investigated thoroughly. Single-crystal YAG is expensive and to produce it a new way has been investigated. This process consists of modifying the methodology of reagents mixture and the process of heating them. The microwave irradiation is used to heat-treat the oxide mixture. The traditional synthesis of YAG powders occurs through the reaction of aluminum and yttrium powders at high temperatures. With this work we investigated the preparation of YAG by non-hydrolytic sol-gel route as an alternative methodology to obtain yttrium-aluminum matrix from inorganic precursors (yttrium and aluminum chloride). The preparation of the gel was carried out in an oven-dried glassware. The AlCl3, YCl3 and ethanol were reacted in reflux under argon atmosphere. Europium III chloride was added as a structural probe. The powder was dried and heat-treated in modified microwaves. The samples were pre-treated at 50 and 800 C during I h and then heated in microwaves for 30 s, 2 and 4 min. The formation process and structure of the powders were studied by means of X-ray diffraction (XRD), photoluminescence (PL) and transmission electronic microscopy (TEM). XRD presents only picks corresponding to the YAG phase and confirmed by TEM. PL date showed that the YAG phase was formed in 2 min with the samples pre-treated at 50 C. For the samples pretreated at 800 degrees C, the YAG phase appears in 30s. The excitation spectra present a maximum of 394 nm corresponding to the L-5(6) level and emission spectra of Eu III ion present bands characteristic transitions arising from the D-5(0) -> F-7(J) (J= 1, 2, 3, 4) monifolds excited at their maximum. The magnetic dipole D-5(0) -> F-7(1) transition presents more intensity than the electric dipole D-5(0) -> F-7(2) transition. This methodology showed efficiency in obtaining YAG phase. This work develops a methodology (using the degree-days concept and linear regression), to forecast the duration of phenological phases in crops. An experiment was conducted in the greenhouse with three cultivars of cowpea (Vigna unguiculata (C.) Walp.), cv. California-781, Tvx 5058-09C and IT 81D-1032. The methodology was based on the relative thermal efficiency rate, determined for each species or cv. The results show that the proposed methodology may be a good alternative in works involving crops, especially because it does not require the repetition of the experiments.

This work describes a methodology for power factor control and correction of the unbalanced currents in four-wire electric circuits. The methodology is based on the insertion of two compensation networks, one wye-grounded neutral and other in delta, in parallel to the load. The mathematical development has been proposed in previous work [3]. In this paper, however, the determination of the compensation susceptances is based on the instantaneous values of load currents. The results are obtained using the MatLab-Simulink enviroment

This paper describes a methodology for solving efficiently the sparse network equations on multiprocessor computers. The methodology is based on the matrix inverse factors (W-matrix) approach to the direct solution phase of A(x) = b systems. A partitioning scheme of W-matrix , based on the leaf-nodes of the factorization path tree, is proposed. The methodology allows the performance of all the updating operations on vector b in parallel, within each partition, using a row-oriented processing. The approach takes advantage of the processing power of the individual processors. Performance results are presented and discussed.

The objective of this paper is to present a methodology to analyze a transmission line model used in electromagnetic transitory simulators, called equivalent impedance test. Initially the definition of equivalent impedance reference test is shown. Soon after this methodology is applied to a transmission line model, the Quasi-Modes model. The studies were accomplished in a hypothetical non-transposed three-phase transmission fine of 440 kV. The line length is 500 km, and it was modeled through cascades of pi-circuits (with 50 pi's circuits, each with 10 km length).

In this work the copper(II) complexation parameters of aquatic organic matter, aquatic and soil humic substances from Brazilian were determined using a new versatile approach based on a single-stage tangential-flow ultrafiltration (TF-UF) technique (cut-off 1 kDa) and sensitive atomic spectrometry methods. The results regarding the copper(II) complexation capacity and conditional stability constants obtained for humic materials were compared with those obtained using direct potentiometry with a copper-ion-selective electrode. The analytical procedure based on ultrafiltration is a good alternative to determine the complexation parameters in natural organic material from aquatic and soil systems. This approach presents additional advantages such as better sensibility, applicability for multi-element capability, and its possible to be used under natural conditions when compared with the traditional ion-selective electrode.

Response surface methodology was employed to optimize the production of a snack food from chickpea. The independent variables, process temperature (123-137-degrees-C) and feed moisture (13-27% d.s.b.) were selected at five levels (rotatable five level composite design: - square-root 2, -1, 0, 1, + square-root 2) in the extrusion of defatted chickpea flour. Response variables were expansion ratio, shear strength of the extrudate and sensory preference assessed by an untrained panel. Expansion ratio increased steadily with decrease in feed moisture similar to cereal extrusion. Regions of maxima were observed for sensory preference and shear strength, and these two product attributes were linearly related. The most acceptable chickpea snack was rated higher than a commercial corn snack.

BACKGROUND: The use of Trichogramma species is a potential key strategy in integrated pest management. However, its effectiveness depends on the use of chemicals that do not interfere with parasitism and parasite population growth. Here, a study was made of the effects of synthetic insecticides on Trichogramma pretiosum and Trichogramma exiguum in different hosts (Ephestia kuehniella, Plutella xylostella and Spodoptera frugiperda) and the influence of International Organisation for Biological Control (IOBC/WPRS) methodology in selectivity studies using different Trichogramma species. The insecticides used were commercial formulations (triflumuron at a concentration of 0.2 mL L-1 water, etofenprox at a concentration of 0.47 mL L-1 water and endosulfan at a concentration of 7.5 mL L-1 water); the control treatment consisted of distilled water. Eggs attached to cardboard cards were offered to parasitoids inside glass cages. Parasitised eggs, parasitism and adult emergence rates and parasitism reduction were evaluated.RESULTS: Endosulfan and etofenprox, classified as class-4 toxic products, were extremely toxic to the parasitoids. Triflumuron, classified as a non-toxic product, was selective to the parasitoids in eggs of all hosts.CONCLUSIONS: The methodology recommended by IOBC/WPRS influenced results regarding the use of different species of parasitoids, and the use of a single parasitoid species in their experiment is questionable. The present study reports on an alternative methodology for the extraction of Humic Substances (HS) from the soil. The extractions were carried out with 0.5 M KOH for 3 h, at a ratio of 1:20 (m/v) under a nitrogen atmosphere. The HS were separated by centrifugation based on their solubility in alcaline solution. This methodology was compared with the usual procedure in three different soil samples and in one sample from vermicompost. The yield, E4/E6 ratio, ash content, and the elemental composition (C, H, O, N) of the extracted HS have been determined. The functional groups were identified by Fourier-transform infrared spectroscopy. This novel procedure adds a new perspective to the extraction of humic substances due to the short time and high performance of the extraction in relation to the usual procedure.

In the laboratory of cytogenetics of the DBAA-UNESP we are studying the karyotipe of some Brazilian Palms. To determine the best protocol, methods of seed germination, inhibition of mitosis, time to pick up the roots and staining were analyzed. The results shown that the seed germination in sphagnum is effective to achieve good roots. The best time to collect the root tips is between 11 to 12 AM., when there are more cell metaphases. The inhibition of mitosis cycle at metaphases may be effective both with 8-hydroxiquinoleine (0,03% -5 hours) or with cold water (0°C - 18-20 hours). The staining with Giemsa 2% showed the best chromosome figures in the metaphases. Now, to get good metaphases slides the method in use in the lab is: 1) seed germination in sphagnum at room temperature and high humidity; 2) The major roots are cut maintaining at least 5 cm, because this technic allows new emergence of roots, increasing the number of roots collected per germinated seed, that is very important in some species with poor germination rates; 3) To get the mitosis inhibition we are using cold water (0°C) treatment for 18-20 hours, following the standard protocols for conservation and hydrolysis; or enzyme digestion with pectinasecellulase 4) the staining procedures are made using Giemsa 2%. The Brazilian palms species studied and their respective chromosome number were: Aiphanes acanthophylla (2n=30), A. caryotaefolia (2n=30), Syagrus quinquifaria (2n=32), S. coronata (2n=32), S. romanzoffiana (2n=32), Euterpe edulis (2n=36), E. oleracea (2n=36), Copernicia prunifera (2n=36), Scheelea lauromuelleriana (2n=32) and Bactris gasipaes (2n=30).

Differences of venom peptide composition as function of two collection methodologies, electrical stimulation (ES) and reservoir disruption (RD), were analyzed by reverse-phase HPLC in Apis mellifera races - A. m. adansonii, A. m. ligustica and Africanized honeybee. The analyses were performed through determination of the relative number and percentage of each molecular form associated to the peaks eluted by chromatography. Comparison of these profiles revealed qualitative and quantitative differences related to the venom collection methodology as well to the three races analyzed. In contrast to data usually found for venom proteins, the three races presented a major number of peaks or molecular forms when venom was collected by ES. Besides, in general, the relative concentration of each peak was higher for ES in relation to RD. That indicates the presence of molecular precursors in the venom obtained by RD. The presence/absence pattern of the peaks, such as their relative concentrations showed a closer similarity between A. m. adansonii and the Africanized honeybees than that observed between these and A. m. ligustica. The obtained data allowed a discussion about the differences in the relative concentration of each venom component according to the collection methodology, and finally the biological action of the venom in different races. So, these results, apart from being useful to establish a peptide profile for each bee race as a function of the venom collection methodology, pointed out once more that the chromatographic techniques are a great tool for the identification of A. mellifera subspecies.

Among the positioning systems that compose GNSS (Global Navigation Satellite System), GPS has the capability of providing low, medium and high precision positioning data. However, GPS observables may be subject to many different types of errors. These systematic errors can degrade the accuracy of the positioning provided by GPS. These errors are mainly related to GPS satellite orbits, multipath, and atmospheric effects. In order to mitigate these errors, a semiparametric model and the penalized least squares technique were employed in this study. This is similar to changing the stochastical model, in which error functions are incorporated and the results are similar to those in which the functional model is changed instead. Using this method, it was shown that ambiguities and the estimation of station coordinates were more reliable and accurate than when employing a conventional least squares methodology.
