Proteolytic enzymes have evolved several mechanisms to cleave peptide bonds. These distinct types have been systematically categorized in the MEROPS database. While a BLAST search on these proteases identifies homologous proteins, sequence alignment methods often fail to identify relationships arising from convergent evolution, exon shuffling, and modular reuse of catalytic units. We have previously established a computational method to detect functions in proteins based on the spatial and electrostatic properties of the catalytic residues (CLASP). CLASP identified a promiscuous serine protease scaffold in alkaline phosphatases (AP) and a scaffold recognizing a beta-lactam (imipenem) in a cold-active Vibrio AP. Subsequently, we defined a methodology to quantify promiscuous activities in a wide range of proteins. Here, we assemble a module which encapsulates the multifarious motifs used by protease families listed in the MEROPS database. Since APs and proteases are an integral component of outer membrane vesicles (OMV), we sought to query other OMV proteins, like phospholipase C (PLC), using this search module. Our analysis indicated that phosphoinositide-specific PLC from Bacillus cereus is a serine protease. This was validated by protease assays, mass spectrometry and by inhibition of the native phospholipase activity of PI-PLC by the well-known serine protease inhibitor AEBSF (IC50 = 0.018 mM). Edman degradation analysis linked the specificity of the protease activity to a proline in the amino terminal, suggesting that the PI-PLC is a prolyl peptidase. Thus, we propose a computational method of extending protein families based on the spatial and electrostatic congruence of active site residues.

The hydrological response of a catchment to rainfall on different timescales is result of a complex system involving a range of physical processes which may operate simultaneously and have different spatial and temporal influences. This paper presents the analysis of streamflow response of a small humid-temperate catchment (Aixola, 4.8 km(2)) in the Basque Country on different timescales and discusses the role of the controlling factors. Firstly, daily time series analysis was used to establish a hypothesis on the general functioning of the catchment through the relationship between precipitation and discharge on an annual and multiannual scale (2003-2008). Second, rainfall-runoff relationships and relationships among several hydrological variables, including catchment antecedent conditions, were explored at the event scale (222 events) to check and improve the hypothesis. Finally, the evolution of electrical conductivity (EC) during some of the monitored storm events (28 events) was examined to identify the time origin of waters. Quick response of the catchment to almost all the rainfall events as well as a considerable regulation capacity was deduced from the correlation and spectral analyses. These results agree with runoff event scale data analysis; however, the event analysis revealed the non-linearity of the system, as antecedent conditions play a significant role in this catchment. Further, analysis at the event scale made possible to clarify factors controlling (precipitation, precipitation intensity and initial discharge) the different aspects of the runoff response (runoff coefficient and discharge increase) for this catchment. Finally, the evolution of EC of the waters enabled the time origin (event or pre-event waters) of the quickflow to be established; specifically, the conductivity showed that pre-event waters usually represent a high percentage of the total discharge during runoff peaks. The importance of soil waters in the catchment is being studied more deeply.

Shear banding characterization of Zr64.13Cu15.75Ni10.12Al10 and Zr65Cu15Ni10Al10 bulk metallic glasses (BMGs) with significant difference in inherent plasticity and quite similar chemical composition was studied by depth sensitive macroindentaion tests with conical indenter. Well-developed shear band pattern can be found for both BMGs after indentation. Distinct difference in the shear band spacing, scale of plastic deformation region and the shear band branching in the two BMGs account for the different plasticity.

A research was conducted in thirty approximately 100 sq.m earthern ponds of the Brackishwater Aquaculture Centre (BAC), College of Fisheries, University of the Philippines, Leganes Iloilo from November 7, 1982 to March 7, 1983 to evaluate the effects of nine supplemental feeds containing different protein: energy ratios on the growth and survival of Tilapia nilotica in brackishwater ponds. Nine supplemental feeds formulated were with protein levels of 20%, 25%, and 30% each at three energy levels of 3,000 kcals; 3,500 kcals; and 4,000 kcals. There was a control treatment with no feeding so that mean weight gain growth rate, feed conversion rate, and survival were determined. Fish fingerlings were acclimated from 0-29 ppt. salinity before the experiment and 20% of fish in each treatment were sampled after every 30 days. Growth rates were significantly different and increased with increasing energy level at the 30% protein feeds but decreased at high energy levels in the 20% and 25% protein feeds. Feed conversion was significantly different due to interaction between protein and energy levels in the feeds, and was better at the 30:3,500 kcals feeds having a feed conversion of 1.55 g. Survival was not significantly different

In near wall measurements with microPIV/PTV, whether seeding particles can be effectively used to detect local fluid velocity is a crucial problem. This talk presents our recent measurements in microchannels [1][2]. Based on measured velocity profiles with 200nm and 50nm in pure water, we found that the measured velocity profiles are agreed with the theoretical values in the middle of channel, but large deviations between measured data and theoretical prediction appear close to wall (0.25mm

Seven groups of fingerling rainbow trout (S. gairdneri ) were fed for 10 weeks on 0%, 10%, 20% and 30% of cassava or rice in isonitrogenous diets. Optimum growth and food utilization was at 20% dietary cassava. High fiber content of the control diet did not suppress protein digestibility in this group. Rather, at all levels, protein digestibility was good and remained between 84.4% and 90.1%. However, in the control group, carbohydrate digestibility was very poor. The cassava diets which had the highest digestible energy as carbohydrate produced the best growth performance, food utilization and protein sparing. At the levels studied, the dietary carbohydrates produced no hyperglycamic effect on the fish

An in-situ visualization of two-phase flow inside anode flow bed of a small liquid fed direct methanol fuel cells in normal and reduced gravity has been conducted in a drop tower. The anode flow bed consists of 11 parallel straight channels. The length, width and depth of single channel, which had rectangular cross section, are 48.0, 2.5 and 2.0 mm, respectively. The rib width was 2.0 mm. The experimental results indicated that when the fuel cell orientation is vertical, two-phase flow pattern in anode channels can evolve from bubbly flow in normal gravity into slug flow in microgravity. The size of bubbles in the reduced gravity is also bigger. In microgravity, the bubbles rising speed in vertical channels is obviously slower than that in normal gravity. When the fuel cell orientation is horizontal, the slug flow in the reduced gravity has almost the same characteristic with that in normal gravity. It implies that the effect of gravity on two-phase flow is small and the bubbles removal is governed by viscous drag. When the gas slugs or gas columns occupy channels, the performance of liquid fed direct methanol fuel cells is failing rapidly. It infers that in long-term microgravity, flow bed and operating condition should be optimized to avoid concentration polarization of fuel cells.

Three fertilizer types (NPK, Super-phosphate and cow dung) were applied at two levels (Low, 0.3 kg/25m super(2)/2weeks and High, 0.7kg/25 m super(2)/2weeks) to 12 ponds with two ponds serving as control. Each pond had an area of 25 m super(2). Application of fertilizers and monitoring of plankton productivity and water quality parameters continued fortnightly for 52 days. Results obtained were subjected to Statistical Variance Analysis. The abundance of phytoplankton was in the order: Chlorophyceae > Bacillariophyceae > Cyanophyceae > Desmideaceae. While that of zooplankton followed the order: Crustacean > Rotifer > Protozoan. Primary productivity showed a variation between treatments with lowest value of 5592 mg/O sub(2)/m super(3)/day obtained in the control and cow dung low application rates (1.5 kg/25 m super(2)/2weeks). The highest value for primary productivity was obtained at M sub(2) (0.7 kg/25 m super(2)/2weeks, N.P.K) with primary productivity value of 7200 mg/O sub(2)/m super(3)/day, closely followed by M sub(4) (0.7 kg/25 m super(2)/2weeks, super phosphate) with 6792 mg/O sub(2)/m super(3)/day.

Fish farming practices in the Lake Kainji Area of Nigeria are categorized under seven main cultural facilities, namely, earthen ponds/reservoirs, indoor/outdoor concrete tanks, plastic tanks, floating cages/hapas, aquaria, sewage and feral conditions. The presence of Bacteria isolates associated with diseased fish conditions varied significantly (P<0.05) with different cultural facilities. The highest bacteria isolates and bacterial disease incidence, 33% and 46% respectively, was associated with diseased fish in the indoor/outdoor concrete tanks. The least incidence of bacteria isolates (3.5%) and blue bacterial disease (3%) was associated with diseased fish in the aquaria and feral conditions. Nine Gram-negative and two Gram-positive bacteria genera were isolated during this investigation. Pseudomonas spp. (23.6%) and Staphylococcus spp. (14.3%), were the predominant Gram-negative and Gram-positive bacteria genera in the different cultural facilities, respectively. This paper highlights the relevance of occurrence and distribution of bacteria isolates associated with diseased fish to bacterial fish diseases under different cultural facilities

Fry of Heterobranchus bidorsalis (mean weight, 0.03g) were fed at 10, 20, 30, 40 or 50% of their body weight (bw)/day in outdoor concrete tanks in New Bussa, Nigeria. The fry attained the highest average weight of 6.66g in 10 weeks when fed at 40% bw/day. Feed conversion ratio ranged from 2.13 to 9.65, the protein efficiency ratio from 1.33 to 5.93. Results indicated that H. bidorsalis are efficient feed converters. Feed intake was significantly different (p<0.05) between the treatments, specific growth rate (SGR), daily growth rate (DGR), production and average weight gain indicated that 40% of fish biomass was the optimum feeding level

The biomass yields of duck week (Lemna minor(L) was monitored in hydroponic media prepared by variously extracting 0.50, 1.00 and 2.00g of dried chicken manure per liter of city water (tap water) supply. The culture media consisting of aqueous extract of the various manure treatments were made up to 12 liters in all cases with tap water as control. Plastic baths of 25 liters capacity with 0.71 super(m2) surface area were used as culture facility. Each bath was stocked at a density of 30g super(m-2) with fresh weed samples (i.e 21.30g/bath). Maximum yields were obtained at all treatment levels and control on day 3 and based on the highest yield of 0.37gm super(-2)d super(-1) (dry matter) obtained at 1.00gL manure treatment which was however not significantly higher (P>0.05) than the 0.36gm super(-2)d super(-1) (dry matter) at 0.05gl super(-1) media manure content, an average manure level of 0.75l super(-1) was selected and used to determine the operational plant density. Thus fresh weights of 30 to 300gm super(-2) was grown in triplicate at 30g intervals for a period of 3 days. A regression equation of Y=2.6720+0.0021x with a corresponding maximum density or operational plant density of 266gm super(-2) and yield of 0.98gm super(-2), d super(-1) (dry matter) were obtained. Further growth trials were carried out at the operational density and manure levels of 0.50, 0.75, 1.00, 1.25, 1.50, 1.75 and 2.00gl super(-1) media manure concentration giving a significantly higher yield (P<0.05) of 17gm super(-2), d super(-1) (dry matter). This yield was however doubled to between 2.21 and 2.24gm super(-2) d super(-1) (equivalent to 7.96 to 8.06mt.ha-1, Yr-1 dry matter on extrapolation) if 25% and 75% respectively of the total weed cover were harvested daily within the experimental period. The study was carried out to asses the nutritional qualities of smoked O. niloticus and to discover the best methods of storage to minimize spoilage and infestation of smoked fish. Result showed that the protein contents in A and D decreased while the protein contents of b and C increased. The lipid content increased only in A while it decreased in B-C and D. The moisture content generally increased over the period of storage and there was an increase in ash content only in C while it decreased in A, B and D. The samples packed in polythene bag suffered about 35% mould infection and a few were attached by rodents with some fouling. Samples packed in jute bag were in good condition but were slightly attached by insect. All samples packed in carton and basket were still in good state but there were insect attack in those packed in carton
