Nas últimas décadas a quiralidade tornou-se essencial na conceção, descoberta, desenvolvimento e comercialização de novos medicamentos. A importância da quiralidade na eficácia e segurança dos fármacos tem sido globalmente reconhecida tanto pelas indústrias farmacêuticas como pelas agências reguladoras de todo o mundo. De forma a produzir eficazmente medicamentos seguros e dar resposta à demanda da indústria de compostos enantiomericamente puros, a pesquisa de novos métodos de síntese assimétrica, assim como o desenvolvimento estratégico dos métodos já disponíveis tem sido um dos principais objetos de estudo de diversos grupos de investigação tanto na academia como na indústria farmacêutica No primeiro capítulo desta dissertação são introduzidos alguns dos conceitos fundamentais associados à síntese de moléculas quirais e descritas algumas das estratégias que podem ser utilizadas na sua síntese. Apresenta-se ainda uma breve revisão bibliográfica acerca dos antecedentes do grupo de investigação e sobre a ocorrência natural, atividade biológica e métodos de síntese e transformações de compostos do tipo (E,E)-cinamilidenoacetofenona. O segundo capítulo centra-se na adição de Michael enantiosseletiva de diversos nucleófilos a derivados de (E,E)-cinamilidenoacetofenona. Inicialmente descreve-se a síntese de derivados de (E,E)-cinamilidenoacetofenona através de uma condensação aldólica de acetofenonas e cinamaldeídos apropriadamente substituídos. Estes derivados são posteriormente utilizados como substratos na adição de Michael enantiosseletiva de três diferentes nucleófilos: nitrometano, malononitrilo e 2-[(difenilmetileno)amino]acetato de metilo. Nestas reações são utilizados diferentes organocatalisadores de forma a induzir enantiosseletividade nos aductos de Michael para serem utilizados na síntese de compostos com potencial interesse terapêutico. É descrita ainda uma nova metodologia de síntese de Δ1-pirrolinas através de um procedimento one-pot de redução/ciclização/desidratação mediada por ferro na presença de ácido acético de (R,E)-1,5-diaril-3-(nitrometil)pent-4-en-1-onas com bons rendimentos e excelentes excessos enantioméricos. O terceiro capítulo centra-se no estabelecimento de novas rotas de síntese e transformação de derivados do ciclo-hexano. Após uma breve revisão bibliográfica, são descritas três metodologias enantiosseletivas distintas, sendo que a primeira envolve a utilização de organocatalisadores e catalisadores de transferência de fase derivados de alcaloides cinchona. Os derivados do ciclo-hexano foram obtidos a partir da reação entre as (E,E)-cinamilidenoacetofenonas e o malononitrilo com bons rendimentos, mas baixas enantiosseletividades independentemente do catalisador utilizado. De forma a contornar este problema e uma vez que a formação do derivado do ciclo-hexano envolve inicialmente a formação in-situ do aducto de Michael, a segunda e terceira metodologias de síntese envolvem a utilização dos aductos de Michael enantiomericamente puros preparados no segundo capítulo. Assim, a reação do (S,E)-2-(1,5-diaril-1-oxopent-4-en-3-il)malononitrilo com os derivados de (E,E)-cinamilidenoacetofenona organocatalisada pela hidroquinina permitiu obter os compostos pretendidos com excelentes excessos enantioméricos. A utilização de um catalisador de transferência de fase não foi tão eficiente em termos de enantiosseletividades obtidas na reação entre as (R,E)-1,5-diaril-3-(nitrometil)pent-4-en-1-onas e os derivados de (E,E)-cinamilidenoacetofenona, apesar de estes terem sido obtidos em bons rendimentos. A preparação destes derivados levou ainda à idealização de uma nova metodologia de síntese de análogos do ácido γ-aminobutírico (GABA) devido à presença de um grupo nitro em posição gama relativamente a um grupo carboxílico. No entanto, apesar de terem sido testadas várias metodologias, não foi possível obter os compostos pretendidos. No quarto capítulo apresenta-se uma breve revisão bibliográfica acerca da ocorrência natural, atividade biológica e métodos de síntese de derivados de di-hidro- e tetra-hidropiridinas, assim como um enquadramento teórico acerca das reações pericíclicas utilizadas na síntese dos compostos pretendidos. Inicialmente é descrita a preparação de N-sulfonilazatrienos substituídos através da condensação direta de derivados de (E,E)-cinamilidenoacetofenona e sulfonamidas. Estes compostos são posteriormente utilizados na síntese de derivados de 1,2-di-hidropiridinas através de uma aza-eletrociclização-6π por duas metodologias distintas: utilização de organocatalisadores quirais e utilização de complexos metálicos de bisoxazolinas. Na síntese das tetra-hidropiridinas os N-sulfonilazatrienos são utilizados como dienos e o étoxi-eteno como dienófilo numa reação hetero-Diels-Alder inversa utilizando também os complexos metálicos de bisoxazolinas como catalisadores. Todos os novos compostos sintetizados foram caracterizados estruturalmente recorrendo a estudos de espetroscopia de ressonância magnética nuclear (RMN), incluindo espetros de 1H e 13C e estudos bidimensionais de correlação espetroscópica homonuclear e heteronuclear e de efeito nuclear de Overhauser (NOESY). Foram também efetuados, sempre que possível, espetros de massa (EM) e análises elementares ou espetros de massa de alta resolução (EMAR) para todos os novos compostos sintetizados.

All the evidence indicates that distillation and liqueurs preparation began in Monchique mountain because this place was pointed as a possible capital of the oldest population of Algarve and an important Arabic village (Barreto, 1972: 19). It was possible to find lots of vestiges like the alembic produced by Arabic population near the X century (Telo, 1988: 77). Traditionally the Algarvian people produce the Arbutus unedo L., fig, carob, grape distillates. At the same time they produce liqueur-using maceration of parts of plants or fruits in some kinds of distillates. Most of the work about Algarvian distillates started by studying the basic compounds of Arbutus unedo spirits by gas chromatography (GC) and mass spectrometry (MS) as well as other physical-chemical properties. In a second phase aged distillates were studied by their phenolic compounds evolution using high resolution liquid chromatography (HPLC). Volatile compounds of traditional liqueurs were identified by head space micro extraction solid phase (HE-SPME) and also analysed by gas chromatography mass spectrometry (GC-MS) and when possible confirmed with standards. Total phenols were determined by Folin-Ciocalteur method. Flavenoids were studied by high performance liquid chromatography (HPLC). Sensorial analysis was also done in every drink studies. The results showed that the arbutus distillate doesn’t present a high level of methanol according to the current legislation. The excesses of acidity or ethyl acetate present normal values when the fermentation is well done (Galego, et al. 1995: 341; Galego, et al. 1995: 685). During the aging process, the colour of spirits tend to become darker, the colour changes occurred more rapidly in the arbutus spirits located in cellars with higher temperatures (Galego, et al. 2001: 432). In the sensory evaluation of samples aged during 12 months into 50 L medium toasting level oak wood barrels, panellists considered that samples of arbutus spirit had too much wood flavour and they were not able to detect the characteristic aroma of arbutus fruit (Galego, et al., 2001: 183). Differences in liqueurs were observed using HS-SPME-GC, HS-SPME-GC-MS or HPLC analysis and this observation was confirmed by a sensorial panel (Galego, et al. 2003: 60).

Aims: Thymus species are wild species mostly found in the arid lands of Portugal. Possible antimicrobial properties of Thymus essential oils have been investigated. The chemical composition of the essential oils and the antimicrobial activity of Thymus mastichina (L) L. subsp. mastichina , T. camphoratus and T. lotocephalus from different regions of Portugal were analysed. Methods and Results: Hydrodistillation was used to isolate the essential oils and the chemical analyses were performed by gas chromatography (GC) and GC coupled to mass spectrometry. The antimicrobial activity was tested by the disc agar diffusion technique against Candida albicans , Escherichia coli , Listeria monocytogenes , Proteus mirabilis , Salmonella spp. and Staphylococcus aureus . Pure linalool, 1,8-cineole and a mixture (1:1) of these compounds were included. Linalool, 1,8-cineole or linalool/1,8-cineole and linalool/1,8-cineole/linalyl acetate were the major components of the essential oils, depending on the species or sampling place. The essential oils isolated from the Thymus species studied demonstrated antimicrobial activity but the micro-organisms tested had significantly different sensitivities. Conclusions: The antimicrobial activity of essential oils may be related to more than one component. Significance and Impact of the Study: Portuguese endemic species of Thymus can be used for essential oil production for food spoilage control, cosmetics and pharmaceutical use. Further studies will be required to elucidate the cell targets of the essential oil components.

Objective: To investigate the effect of nutrient stimulation of gut hormones by oligofructose supplementation on appetite, energy intake (EI), body weight (BW) and adiposity in overweight and obese volunteers. Methods: In a parallel, single-blind and placebo-controlled study, 22 healthy overweight and obese volunteers were randomly allocated to receive 30 g day−1 oligofructose or cellulose for 6 weeks following a 2-week run-in. Subjective appetite and side effect scores, breath hydrogen, serum short chain fatty acids (SCFAs), plasma gut hormones, glucose and insulin concentrations, EI, BW and adiposity were quantified at baseline and post-supplementation. Results: Oligofructose increased breath hydrogen (P < 0.0001), late acetate concentrations (P = 0.024), tended to increase total area under the curve (tAUC)420mins peptide YY (PYY) (P = 0.056) and reduced tAUC450mins hunger (P = 0.034) and motivation to eat (P = 0.013) when compared with cellulose. However, there was no significant difference between the groups in other parameters although within group analyses showed an increase in glucagon-like peptide 1 (GLP-1) (P = 0.006) in the cellulose group and a decrease in EI during ad libitum meal in both groups. Conclusions: Oligofructose increased plasma PYY concentrations and suppressed appetite, while cellulose increased GLP-1 concentrations. EI decreased in both groups. However, these positive effects did not translate into changes in BW or adiposity.

GOAL: The manufacturing and distribution of strips of instant thin - layer chromatography with silica gel (ITLC - SG) (reference method) is currently discontinued so there is a need for an alternative method f or the determination of radiochemical purity (RCP) of 99m Tc - tetrofosmin. This study aims to compare five alternative methods proposed by the producer to determine the RCP of 99m Tc - tetrofosmin. METHODS: Nineteen vials of tetrofosmin were radiolabelled with 99m Tc and the percentages of the RCP were determined. Five different methods were compared with the standard RCP testing method (ITLC - SG, 2x20 cm): Whatman 3MM (1x10 cm) with acetone and dichloro - methane (method 1); Whatman 3MM (1x1 0 cm) with ethyl acetate (method 2); aluminum oxide - coated plastic thin - layer chromatography (TLC) plate (1x10 cm) and ethanol (method 3); Whatman 3MM (2x20 cm) with acetone and dichloro - methane (method 4); solid - phase extraction method C18 cartridge (meth od 5). RESULTS: The average values of RCP were 95,30% ± 1,28% (method 1), 93,95 ± 0,61% (method 2), 96,85% ± 0,93% (method 3), 92,94% ± 0,99% (method 4) and 96,25% ± 2,57% (method 5) (n=12 each), and 93,15% ± 1,13% for the standard method (n=19). There we re statistical significant differences in the values obtained for methods 1 (P=0,001), 3 (P=0,000) and 5 (P=0,004), and there were no statistical significant differences in the values obtained for methods 2 (P=0,113) and 4 (P=0,327). CONCLUSION: From the results obtained, methods 2 and 4 showed a higher correlation with the standard method. Unlike method 4, method 2 is less time - consuming than the reference method and can overcome the problems associated with the solvent toxicity. The remaining methods (1, 3 and 5) tended to overestimate RCP value compared to the standard method.

A flow injection analysis (FIA) system comprising a tartrate- (TAT) selective electrode has been developed for determination of tartaric acid in wines. Several electrodes constructed for this purpose had a PVC membrane with a complex of quaternary ammonium and TAT as anion exchanger, a phenol derivative as additive, and a more or less polar mediator solvent. Characterization of the electrodes showed behavior was best for membranes with o-nitrophenyl octyl ether as solvent. On injection of 500 μL into a phosphate buffer carrier (pH = 3.1; ionic strength 10–2 mol/L) flowing at 3 mL/min, the slope was 58.06 ± 0.6 with a lower limit of linear range of 5.0 × 10–4 mol/L TAT and R2 = 0.9989. The interference of several species, e.g. chloride, bromide, iodide, nitrate, gallic acid, tannin, sucrose, glucose, fructose, acetate, and citrate, was evaluated in terms of potentiometric selectivity coefficients. The Hofmeister series was followed for inorganic species and the most interfering organic ion was citrate. When red and white wines were analyzed and the results compared with those from an independent method they were found to be accurate, with relative standard deviations below 5.0%.

O crescimento populacional esperado para os próximos anos conduzirá à necessidade de aumentar a produção agrícola de modo a satisfazer o aumento da procura. Nos últimos anos tem-se assistido a uma evolução tecnológica nos sistemas de produção que tem permitido aumentar a produtividade agrícola, por vezes à custa de elevados consumos de energia e com práticas nem sempre ambientalmente corretas. Os desafios que se colocam atualmente são no sentido de melhorar a conservação de recursos escassos, como o solo e a água, de aumentar a eficiência de uso de fatores de produção, de encontrar novas culturas, do desenvolvimento da biotecnologia, da diminuição dos consumos energéticos e de melhorar ainda mais as tecnologias associadas à produção. De maneira a responder aos desafios emergentes da procura por alimentos, da escassez de terrenos agrícolas aráveis bem como da existência de pragas de insetos e de ervas daninhas, os pesticidas tem vindo a ser usados com maior frequência, tendo-se assistido a uma contaminação dos solos e águas subterrâneas, causando deste modo um risco para a saúde dos seres vivos. Neste sentido, vários fabricantes de pesticidas estão a desenvolver novas formulações contendo pesticidas encapsulados em nanopartículas como modo de aumentar a sua solubilidade em água, biodisponibilidade, volatilidade, estabilidade e eficácia. tendo por objetivo um desenvolvimento sustentável. Neste trabalho, procedeu-se ao estudo do encapsulamento do herbicida Oxadiargil (5-terc-butil-3-[2,4-dicloro-5-(2-propiniloxi)fenil]-1,3,4-oxadiazol-2(3H)-ona) com a 2-hidroxipropil-β-ciclodextrina (HP-β-CD). O estudo da formação do complexo de inclusão Oxadiargil - HP-β-CD foi realizado em diferentes meios, água desionizada, tampão acetato pH = 3,46 e pH = 5,34 e tampão fosfato pH = 7,45, com o objetivo de determinar e comparar a sua constante de estabilidade. Verificou-se, em qualquer dos casos, a ocorrência de uma relação linear entre o aumento da solubilidade do Oxadiargil e o aumento da concentração de HP-β-CD, com um declive inferior a um, o que indicia a formação de um complexo na proporção estequiométrica de 1:1. Os resultados obtidos permitiram concluir que o processo de complexação Oxadiargil - HP-β-CD não é muito influenciado pela constituição e pelo pH do meio. De facto, as constantes de estabilidade obtidas para a água desionizada e soluções-tampão pH = 3,46, pH = 5,34 e pH = 7,45 foram de 919 ± 25, 685 ± 13, 623 ± 17 e 753 ± 9, respetivamente. A solubilidade do complexo obtido nos estudos realizados, em diferentes meios, é cerca de 23 a 32 vezes superior à observada para o Oxadiargil livre. De forma a caracterizar o complexo Oxadiargil - HP-β-CD procedeu-se à sua síntese utilizando o método de “kneading”. O composto obtido foi caracterizado por Ressonância Magnética Nuclear (RMN) tendo-se confirmado a formação de um complexo de inclusão na proporção estequiométrica de 1:1. O complexo obtido é mais solúvel e porventura mais estável quimicamente. O encapsulamento permite uma redução da aplicação dos pesticidas diminuindo assim os custos e o impacto negativo no ambiente. Com a nanotecnologia é possível a libertação controlada dos pesticidas, aumentando a sua eficácia e fornecendo os meios necessários para um desenvolvimento sustentável.

Sulfadiazine is an antibiotic of the sulfonamide group and is used as a veterinary drug in fish farming. Monitoring it in the tanks is fundamental to control the applied doses and avoid environmental dissemination. Pursuing this goal, we included a novel potentiometric design in a flow-injection assembly. The electrode body was a stainless steel needle veterinary syringe of 0.8-mm inner diameter. A selective membrane of PVC acted as a sensory surface. Its composition, the length of the electrode, and other flow variables were optimized. The best performance was obtained for sensors of 1.5-cm length and a membrane composition of 33% PVC, 66% onitrophenyloctyl ether, 1% ion exchanger, and a small amount of a cationic additive. It exhibited Nernstian slopes of 61.0 mV decade-1 down to 1.0×10-5 mol L-1, with a limit of detection of 3.1×10-6 mol L-1 in flowing media. All necessary pH/ionic strength adjustments were performed online by merging the sample plug with a buffer carrier of 4-(2-hydroxyethyl)-1-piperazineethanesulfonic acid, pH 4.9. The sensor exhibited the advantages of a fast response time (less than 15 s), long operational lifetime (60 days), and good selectivity for chloride, nitrite, acetate, tartrate, citrate, and ascorbate. The flow setup was successfully applied to the analysis of aquaculture waters. The analytical results were validated against those obtained with liquid chromatography–tandem mass spectrometry procedures. The sampling rate was about 84 samples per hour and recoveries ranged from 95.9 to 106.9%.

Enrofloxacin (ENR) is an antimicrobial used both in humans and in food producing species. Its control is required in farmed species and their surroundings in order to reduce the prevalence of antibiotic resistant bacteria. Thus, a new biomimetic sensor enrofloxacin is presented. An artificial host was imprinted in specific polymers. These were dispersed in 2-nitrophenyloctyl ether and entrapped in a poly(vinyl chloride) matrix. The potentiometric sensors exhibited a near-Nernstian response. Slopes expressing mV/Δlog([ENR]/M) varied within 48–63. The detection limits ranged from 0.28 to 1.01 µg mL−1. Sensors were independent from the pH of test solutions within 4–7. Good selectivity was observed toward potassium, calcium, barium, magnesium, glycine, ascorbic acid, creatinine, norfloxacin, ciprofloxacin, and tetracycline. In flowing media, the biomimetic sensors presented good reproducibility (RSD of ± 0.7%), fast response, good sensitivity (47 mV/Δlog([ENR]/M), wide linear range (1.0 × 10−5–1.0 × 10−3 M), low detection limit (0.9 µg mL−1), and a stable baseline for a 5 × 10−2 M acetate buffer (pH 4.7) carrier. The sensors were used to analyze fish samples. The method offered the advantages of simplicity, accuracy, and automation feasibility. The sensing membrane may contribute to the development of small devices allowing in vivo measurements of enrofloxacin or parent-drugs.

Enhanced biological phosphorus removal (EBPR) is the most economic and sustainable option used in wastewater treatment plants (WWTPs) for phosphorus removal. In this process it is important to control the competition between polyphosphate accumulating organisms (PAOs) and glycogen accumulating organisms (GAOs), since EBPR deterioration or failure can be related with the proliferation of GAOs over PAOs. This thesis is focused on the effect of operational conditions (volatile fatty acid (VFA) composition, dissolved oxygen (DO) concentration and organic carbon loading) on PAO and GAO metabolism. The knowledge about the effect of these operational conditions on EBPR metabolism is very important, since they represent key factors that impact WWTPs performance and sustainability. Substrate competition between the anaerobic uptake of acetate and propionate (the main VFAs present in WWTPs) was shown in this work to be a relevant factor affecting PAO metabolism, and a metabolic model was developed that successfully describes this effect. Interestingly, the aerobic metabolism of PAOs was not affected by different VFA compositions, since the aerobic kinetic parameters for phosphorus uptake, polyhydroxyalkanoates (PHAs) degradation and glycogen production were relatively independent of acetate or propionate concentration. This is very relevant for WWTPs, since it will simplify the calibration procedure for metabolic models, facilitating their use for full-scale systems. The DO concentration and aerobic hydraulic retention time (HRT) affected the PAO-GAO competition, where low DO levels or lower aerobic HRT was more favourable for PAOs than GAOs. Indeed, the oxygen affinity coefficient was significantly higher for GAOs than PAOs, showing that PAOs were far superior at scavenging for the often limited oxygen levels in WWTPs. The operation of WWTPs with low aeration is of high importance for full-scale systems, since it decreases the energetic costs and can potentially improve WWTP sustainability. Extended periods of low organic carbon load, which are the most common conditions that exist in full-scale WWTPs, also had an impact on PAO and GAO activity. GAOs exhibited a substantially higher biomass decay rate as compared to PAOs under these conditions, which revealed a higher survival capacity for PAOs, representing an advantage for PAOs in EBPR processes. This superior survival capacity of PAOs under conditions more closely resembling a full-scale environment was linked with their ability to maintain a residual level of PHA reserves for longer than GAOs, providing them with an effective energy source for aerobic maintenance processes. Overall, this work shows that each of these key operational conditions play an important role in the PAO-GAO competition and should be considered in WWTP models in order to improve EBPR processes.
