Fifty-four different sugarcane resistance gene analogue (RGA) sequences were isolated, characterized, and used to identify molecular markers linked to major disease-resistance loci in sugarcane. Ten RGAs were identified from a sugarcane stem expressed sequence tag (EST) library; the remaining 44 were isolated from sugarcane stem, leaf, and root tissue using primers designed to conserved RGA motifs. The map location of 31 of the RGAs was determined in sugarcane and compared with the location of quantitative trait loci (QTL) for brown rust resistance. After 2 years of phenotyping, 3 RGAs were shown to generate markers that were significantly associated with resistance to this disease. To assist in the understanding of the complex genetic structure of sugarcane, 17 of the 31 RGAs were also mapped in sorghum. Comparative mapping between sugarcane and sorghum revealed syntenic localization of several RGA clusters. The 3 brown rust associated RGAs were shown to map to the same linkage group (LG) in sorghum with 2 mapping to one region and the third to a region previously shown to contain a major rust-resistance QTL in sorghum. These results illustrate the value of using RGAs for the identification of markers linked to disease resistance loci and the value of simultaneous mapping in sugarcane and sorghum.

Prevalence of low temperature at sowing results in poor rice seed germination, seedling establishment and vigour in several temperate rice growing countries around the world. Rice seed of four cultivars (Sasanishiki, H433, HSC-55 and Doongara) was soaked in various combinations of gibberellic acid(3) (GA(3)) and glycinebetaine (GB) in petri dishes placed in a low temperature glasshouse (18/13 degrees C; day/night) for 2 days. After the 2 days soak, 10 treated seed were transferred into plastic pots filled with soil and seedlings were grown in the same glasshouse, where seed was treated. Seedling emergence was least affected by low temperature in cold tolerant cultivar, HSC-55, while other three cultivars showed reduced seedling emergence. However, seedling emergence increased significantly in some cultivars in response to seed treatment with GA(3) and/or GB. Seedlings emerged faster even in the cold tolerant cultivar, HSC-55, as measured by reduced mean emergence time (MET), in response to GB. Seedling height and seedling dry matter also increased in response to both GA(3) and GB. Combined treatment of both GA(3) and GB was more beneficial in increasing seedling emergence and vigour than the treatment with only GA3 or GB. We demonstrated significant genotypic differences for seedling emergence and vigour and not all cultivars responded to the treatment with GA(3) and GB, under low temperature.

Anthracnose, caused by Colletotrichum trifolii, is one of the most serious diseases influencing lucerne persistence and productivity in eastern Australia. The disease is largely controlled by plant resistance; however, new pathotypes of C. trifolii have developed in Australia, seriously limiting the productive life of susceptible cultivars. This paper describes an incompletely recessive and quantitatively inherited resistance to C. trifolii identified in a clone (W116) from cv. Sequel. S-1, F-1, F-2 and backcross populations of W116 and D (highly susceptible clone) were studied for their reaction to C. trifolii race 1. Resistance was found to be quantitatively inherited, and quantitative trait loci associated with resistance and susceptibility were identified in a backcross population (D x W116) x D using random amplified polymorphic DNA and amplified fragment length polymorphic markers. A multi-locus region on linkage group 4 was found to contribute significantly to the resistance phenotype. The application of DNA markers to allow exploitation of this quantitatively inherited resistance in lucerne breeding is discussed.

Microsatellite markers were developed for Cannabis sativa L. (marijuana) to estimate the level of polymorphism, usefulness for DNA typing (genotype identification), and to measure the genetic relationships between the different plants. Twelve different oligonucleotide probes were used to screen an enriched microsatellite library of Cannabis sativa in which 49% of the clones contained microsatellite sequences. Characterization of microsatellite loci in Cannabis revealed that GA/CT was the most abundant class of isolated microsatellites representing 50% overall. Eleven polymorphic SSR markers were developed, derived from dinucleotide motifs and eight from trinucleotide motifs. A total of 52 alleles were detected averaging 4.7 alleles/locus. The expected heterozygosity of the eleven loci ranged between 0.368 and 0.710 and the common probability of identical genotypes was 1.8 x 107. The loci identified 27 unique profiles of the 41 Cannabis samples. The eleven microsatellite markers developed in this study were found to be useful for DNA fingerprinting and for assessing genetic relationships in Cannabis.

Young trees of two carobs (Ceratonia siliqua L.) cultivars, Mulata and Galhosa, ,propagated in vitro and grown for 3-4 years in a greenhouse were moved into a growth chamber under environmental controlled conditions where they were exposed to 25/15º C (control), 10/5º C (chilling) and again to 25/15º C (recovery).

Avaliaram-se, neste trabalho, em condições de baixo e alto teor de fósforo (P) no solo, 51 linhagens de arroz provenientes do Banco Ativo de Germoplasma da Embrapa Arroz e Feijão, com o objetivo de identificar indicadores de eficiência no uso de P e linhagens com eficiência diferenciada no uso desse nutriente. O Ãndice de eficiência do uso de fósforo mostrou ser mais adequado que a produtividade em baixo teor de fósforo no solo, na avaliação da eficiência diferenciada das linhagens no uso do P. Três linhagens, CNA 4634, CNA 4120 e CNA 3178, apresentam bom comportamento produtivo, tanto em alto como em baixo teor de P no solo e 22 mostraram bom comportamento produtivo em alto teor de P no solo; entretanto, não foram eficientes no aproveitamento de baixo P disponÃvel no solo enquanto as demais foram desconsideradas em razão de indicarem baixo comportamento produtivo sob boa disponibilidade de P.

The covering of the soil is an agricultural practice that intends to control the harmful herbs, to reduce the losses of water by evaporation of the soil, and to facilitate the harvest and the commercialization, once the product is cleaner and healthier. However, when the soil is covered important microclimatic parameters are also altered, and consequently the germination of seeds, the growth of roots, the absorption of water and nutrients, the metabolic activity of the plants and the carbohydrates storage. The current trial intended to evaluate the effect of soil covering with blue colored film on consumptive water-use in a lettuce crop (Lactuca sativa, L.). The experiment was carried out in a plastic greenhouse in Araras - São Paulo State, Brazil from March 3rd, 2001 to May 5th, 2001. The consumptive water-use was measured through two weighing lysimeter installed inside the greenhouse. Crop spacing was 0.25 m x 0.25 m and the color of the film above soil was blue. Leaf area index (IAF), was measured six times (7; 14; 21; 28; 35; 40 days after transplant) and the water-use efficiency (EU) was measured at the end. The experimental design was subdivided portions with two treatments, bare soil and covered soil. The average consumptive water-use was 4.17 mm day-1 to the bare soil treatment and 3.11 mm day-1 to the covered soil treatment. The final leaf area index was 25.23 to the bare soil treatment and 24.39 to the covered soil treatment, and there was no statistical difference between then.

A trial was carried out to evaluate the chemical composition in the aerial part of lettuce, cv. 'Elisa', irrigated with wastewater treated with constructed wetland and source deposit water, grown on a Rhodic Hapludox Soil, using the irrigation systems sprinkle, subsurface drip and surface drip irrigation. The experiment was carried out from August 17th to October 3rd of 2001 and the chemical analyses of the lettuce were accomplished to 47 days after transplanting of the seedling. The aerial part of the lettuce was analyzed as for the levels of total nitrogen, nitrate, phosphorus, potassium, calcium, magnesium, sulfur, iron, manganese, copper, zinc, sodium, boron, cobalt and molybdenum. The sodium and the sulfur presented higher levels than the maximum suitable in the aerial part of the lettuce and the smallest level of magnesium, while other chemical elements analyzed were normal and appropriate considering the standard for well-nourished plants, not being influenced by the water type. The sodium was the chemical element that presented the highest levels in the aerial part of the lettuce in the treatments irrigated with wastewater, presenting significant difference in relationship to the treatments irrigated with source deposit water in the three irrigation systems. The use of the different irrigation systems by the application of wastewater treated with constructed wetland did not interfere in the levels of nutrients in the aerial part of the lettuce.

Avaliou-se o efeito do processamento do feno de alfafa (Medicago sativa L.) e da adição de óleo de soja em dietas sobre a digestibilidade total de matéria seca, matéria orgânica, proteÃna bruta, extrato etéreo, fibra em detergente neutro e fibra em detergente ácido em eqüinos. Utilizaram-se quatro potros machos, sem raça definida, com aproximadamente 14 meses de idade e 197,25 kg, em delineamento quadrado latino (4 × 4) e em arranjo fatorial (2 × 2), composto de duas formas de fenação da alfafa (em cubos ou em ramas) e da adição ou não de óleo de soja em dietas contendo concentrado comercial na forma de péletes. A digestibilidade dos nutrientes foi determinada pelo método de coleta total de fezes durante três dias. A adição de óleo de soja refinado aumentou a digestibilidade total de matéria seca, matéria orgânica, extrato de etéreo e fibra em detergente neutro. A fenação da alfafa em cubos aumentou a digestibilidade total da proteÃna bruta, fibra em detergente neutro e da fibra em detergente ácido. A adição de óleo de soja nas dietas avaliadas foi um modo prático e seguro de aumentar a densidade calórica sem reduzir a digestibilidade dos macronutrientes orgânicos. O processamento da alfafa na forma de cubos melhorou a digestibilidade total da proteÃna bruta, fibra em detergente e fibra em detergente ácido da dieta em eqüinos.

Objetivou-se quantificar as frações de carboidratos pelas equações do Cornell Net Carbohydrate and Protein System (CNCPS) de três cultivares de girassol (Helianthus annuus L.) cultivados na presença ou não de irrigação. A utilização de uma preparação fibrosa, denominada parede celular (PC), nas equações da CNCPS, em substituição à fibra em detergente neutro (FDN) não promoveu diferenças nas frações de carboidratos B1 e C, mas influenciou as frações A e B2. Como os valores da fração B1, obtidos pelo modelo CNCPS foram menores que os teores de amido e pectina determinados em laboratório, supõe-se que a pectina e outros oligossacarÃdeos da parede celular, solubilizados pela solução de detergente neutro (fibra solúvel), nunca fizeram parte da fração B1, e sim da fração A. Apesar de os carboidratos da fibra solúvel apresentarem elevadas taxas de degradação, não parece adequada a caracterização da fibra solúvel na fração A. Parece mais adequado que a fibra solúvel (que inclui a pectina) seja alocada a uma fração exclusivamente sua, que pode ser a fração B2, e que seja criada uma nova fração, a B3, para os carboidratos digerÃveis da parede celular. Assim, a fração B1 seria composta apenas de amido. A equação da fração C, que estima os carboidratos indigerÃveis da parede celular, pode ser simplificada, relacionando a fração indigerÃvel ao teor de lignina na matéria seca, e não à FDN isenta de cinzas e proteÃna, como atualmente utilizado. Esta proposta tem implicações práticas, uma vez que a fração indigerÃvel da parede celular tem sido expressa em relação à FDN, e não na MS, com base no fato de que os efeitos inibitórios da lignina ocorrem sobre os componentes fibrosos da parede celular vegetal, e não sobre o conteúdo celular.
