La presente tesis doctoral aborda el estudio de un nuevo material mineral, compuesto principalmente por una matriz de yeso (proveniente de un conglomerante industrial basado en sulfato de calcio multifase) y partículas de aerogel de sílice hidrófugo mesoporoso, compatibilizadas mediante un surfactante polimérico, debido a su alto carácter hidrófugo. La investigación se centra en conocer los factores que influyen en las propiedades mecánicas y conductividad térmica del material compuesto generado. Este estudio pretende contribuir al conocimiento sobre el desarrollo de nuevos morteros de elevado aislamiento térmico que puedan ser utilizados en la rehabilitación energética de edificios de viviendas existentes, debido a que estos representan gran parte del consumo energético del parque de viviendas de España, aunque también a nivel internacional. De los materiales utilizados para desarrollar los morteros estudiados, el yeso, además de ser un material muy abundante, especialmente en España, requiere una menor cantidad de energía para la fabricación de un conglomerante (debido a una menor temperatura de fabricación), en comparación con el cemento o la cal, por lo que presenta una menor huella de carbono que estos últimos. Por otro lado, el aerogel de sílice hidrófugo mesoporoso es, de acuerdo con la documentación disponible, el material que posee actualmente la mayor capacidad de aislamiento térmico en el mercado. El desarrollo de nuevos morteros minerales con una capacidad de aislamiento térmico mayor que los materiales aislantes utilizados tradicionalmente, tiene una aplicación relevante en los casos de rehabilitación energética de edificios históricos y patrimoniales, en los que se requiere la aplicación del aislamiento por el interior de la fachada, ya que este tipo de soluciones tienen el inconveniente de reducir el espacio habitable de las áreas involucradas, especialmente en zonas climáticas en las que el aislamiento térmico puede suponer un espesor considerable, por lo que es ideal utilizar materiales de altas prestaciones de aislamiento térmico capaces de aportar el mismo nivel de aislamiento (o incluso mayor), pero en un espesor considerablemente menor. La investigación se desarrolla en tres etapas: bibliográfica, experimental y de simulación. La primera etapa, parte del estudio de la bibliografía existente, relacionada con materiales aislantes, incluyendo soluciones basadas, tanto en morteros aislantes, como en paneles de aislamiento térmico. La segunda, de carácter experimental, se centra en estudiar la influencia de la microestrucrura y macroestructura, del nuevo material mineral, en las propiedades físicas elementales, mecánicas y conductividad térmica del compuesto. La tercera etapa, mediante una simulación del consumo energético, consiste en cuantificar teóricamente el potencial ahorro energético que puede aportar este material en un caso de rehabilitación energética en particular. La investigación experimental se centró principalmente en conocer los factores principales que influyen en las propiedades mecánicas y conductividad térmica de los materiales compuestos minerales desarrollados en esta tesis. Para ello, se llevó a cabo una caracterización de los materiales de estudio, así como el desarrollo de distintas muestras de ensayo, de tal forma que se pudo estudiar, tanto la hidratación del yeso en los compuestos, como su posterior microestructura y macroestructura, aspectos fundamentales para el entendimiento de las propiedades mecánicas y conductividad térmica del compuesto aislante. De este modo, se pudieron conocer y cuantificar, los factores que influyen en las propiedades estudiadas, aportando una base de conocimiento y entendimiento de este tipo de compuestos minerales con aerogel de sílice hidrófugo, no existiendo estudios publicados hasta el momento de finalización de esta tesis, con la aproximación al material propuesta en este estudio, ni con yeso (basado en sulfato de calcio multifase), ni con otro tipo de conglomerantes. Particularmente, se determinó la influencia que tiene la incorporación de partículas de aerogel de sílice hidrófugo, en grandes proporciones en volumen, en un compuesto mineral basado en distintas fases de sulfato de calcio. No obstante, para llevar a cabo las mezclas, fue necesario utilizar un surfactante para compatibilizar este tipo de partículas, con el conglomerante basado en agua. El uso de este tipo de aditivos tiene una influencia, no solo en el aerogel, sino en las propiedades del compuesto en general, dependiendo de su concentración, por lo que se establecieron dos porcentajes de adición: la primera, determinada a partir de la cantidad mínima necesaria para compatibilizar las mezclas (0,1% del agua de amasado), y la segunda, como límite superior, la concentración utilizada habitualmente a nivel industrial para estabilizar burbujas de aire en hormigones espumados (5%). El surfactante utilizado mostró la capacidad de modificar la superficie del aerogel, cambiando el comportamiento de las partículas frente al agua, permitiendo una invasión parcial de su estructura porosa, por parte del agua de amasado. Este comportamiento supone un aumento muy importante en la relación agua/yeso, afectando el hábito cristalino e influenciando negativamente las propiedades mecánicas de la matriz de yeso, presentando un efecto aún notable a mayor concentración de surfactante (5%). En cuanto a las propiedades finales alcanzadas, fue posible lograr un compuesto mineral ultraligero (200 kg/m3), con alrededor de un 60% de aerogel en volumen y de alta capacidad aislante (0,028 W/m•K), presentando una conductividad térmica notablemente menor que los morteros aislantes del mercado, e incluso también menor que la de los aislantes tradicionales basado en las lanas minerales o EPS; no obstante, con la limitante de presentar bajas propiedades mecánicas, condicionando su posible aplicación futura. Entre los factores principales relacionados con las propiedades mecánicas, se encontró que estas dependen exponencialmente del volumen de yeso en el compuesto; no obstante, factores de segundo orden, como el grado de hidratación, o una mejor distribución del conglomerante entre las partículas de aerogel, debido al aumento de la superficie específica del polvo mineral, pueden aumentar las propiedades mecánicas entre el doble y el triple, dependiendo del volumen de aerogel en cuestión. Además, se encontró que el aerogel, en conjunto con el surfactante, es capaz de introducir una gran cantidad de aire (0,70 m3 por cada m3 de aerogel), que unido al agua evaporada (no consumida por el conglomerante durante la hidratación), el volumen de aire total alcanza, generalmente, un 40%, independientemente de la cantidad de aerogel en la mezcla. De este modo, el aire introducido en la matriz desplaza las proporciones en volumen del aerogel y del yeso, disminuyendo, tanto las propiedades mecánicas, como la capacidad aislante de compuesto mineral. Por otro lado, la conductividad térmica mostró tener una dependencia directa de la contribución de las tres fases principales en el compuesto: yeso, aerogel y aire ocluido. De este modo, se pudo desarrollar un modelo matemático, adaptado de uno existente, capaz de calcular, con bastante precisión, la relación de los tres componentes mencionados, en la conductividad térmica de los compuestos, para el rango de volúmenes y materiales utilizados en esta tesis. Finalmente, la simulación del consumo energético realizada a una vivienda típica de España, de los años 1900 a 1959 (basada en muros de ladrillo macizo), para las zonas climáticas estudiadas (A, D y E), permitió observar el potencial ahorro energético que puede aportar este material, dependiendo de su espesor, como aislamiento interior de los muros de fachada. Particularmente, para la zona A, se determinó un espesor óptimo de 1 cm, mientras que para la zona D y E, 3,5 y 3,9 cm respectivamente. En este sentido, el nuevo material estudiado es capaz de disminuir, entre un 35% y un 80%, el espesor de la capa aislante, en comparación con paneles de lana de roca o los morteros minerales de mayor capacidad aislante del mercado español respectivamente. ABSTRACT The present doctoral thesis studies a new mineral-based composite material, composed by a gypsum matrix (based on an industrial multiphase gypsum binder) and mesoporous hydrophobic silica aerogel particles, compatibilized with a polymeric surfactant due to the high hydrophobic character of the insulating particles. This study pretends to contribute to the development of new composite insulating materials that could be used in energy renovation of existing dwellings, in order to reduce their high energy consumption, as they represent a great part of the total energy consumed in Spain, but also internationally. Between the materials used to develop de studied insulating mortars, gypsum, besides being an abundant material, especially in Spain, requires less energy for the manufacture of a mineral binder (due to lower manufacturing temperatures), compared to lime or cement, thus presenting lower carbon footprint. In other hand, the hydrophobic mesoporous silica aerogel, is, according to the existing references, the material with the highest know insulating capacity in the market. The development of new mineral mortars with higher thermal insulation capacity than traditional insulating materials, presents a relevant application in energy retrofitting of historic and cultural heritage buildings, in which implies that the insulating material should be installed as an internal layer, rather than as an external insulating system. This type of solution involves a reduced internal useful area, especially in climatic zones where the demand for thermal insulation is higher, and so the insulating layer thickness, being idealistic to use materials with very high insulating properties, in order to reach same insulating level (or higher), but in lower thickness than the provided by traditional insulating materials. This research is developed in three main stages: bibliographic, experimental and simulation. The first stage starts by studying the existing references regarding thermally insulating materials, including existing insulating mortars and insulating panels. The second stage, mainly experimental, is centered in the study of the the influence of the microstructure and macrostructure in the physical and mechanical properties, and also in the thermal conductivity of the new mineral-based material. The thirds stage, through energy simulation, consists in theoretically quantifying the energy savings potential that can provide this type of insulating material, in a particular energy retrofitting case study. The experimental research is mainly focused in the study of the factors that influence the mechanical properties and the thermal conductivity of the thermal insulating mineral composites developed in this thesis. For this, the characterization of the studied materials has been performed, as well as the development of several experimental samples, in order to study the hydration of the mineral binder within the composites, but also the final microstructure and macrostructure, fundamental aspects for the understanding of the composite’s mechanical and insulating properties. Thus, is was possible to determine and quantify the factors that influence the studied material properties, providing a knowledge base and understanding of mineral composites that comprises mesoporous hydrophobic silica aerogel particles, being the first study up to date regarding the specific approach of the present study, regarding not just multiphase calcium sulfate plaster, but also other mineral binders. Particularly, the influence of the incorporation of hydrophobic silica aerogel particles, in high volume ratios into a mineral compound, based on different phases of calcium sulfate has been determined. However, to perform mixing, it is necessary to use a surfactant in order to compatibilize these particles with the water-based mineral binder. The use of such additives has an influence, not only in the aerogel, but the overall properties of the compound, so two different surfactant concentration has been studied: the first, the minimum amount of surfactant (used in this thesis) in order to develop the slurries (0.1% concentration of the mixing water), and the second, as the upper limit, the concentration usually used industrially to stabilize air bubbles in foamed concrete (5%). One of the side effects of using such additive, was the modification of the aerogel particles, by changing their behavior in respect to water, generating a partial invasion of the aerogel’s porous structure, by the mixing water. This behavior produces a very important increase in water/binder ratios, affecting the crystal habit and negatively influencing the mechanical properties of the gypsum matrix. This effect further increased when a higher concentration of surfactant (5%) is used. Regarding final materials properties, it was possible to achieve an ultra-lightweight mineral composite (200 kg/m3), with around 60% by volume of aerogel, presenting a very high insulating capacity (0.028 W/m•K), a noticeable lower thermal conductivity compared to the insulating mortars and traditional thermal insulating panels on the market, such as mineral wool or EPS; however, the limiting factor for future’s material application in buildings, is related to the very low mechanical properties achieved. Among the main factors related to the mechanical properties, it has been found an exponential correlation to the volume of gypsum in the composite. However, second-order factors such as the degree of hydration, or a better distribution of the binder between the aerogel particles, due to the increased surface area of the mineral powder, can increase the mechanical properties between two to three times, depending aerogel volume involved. In addition, it was found that the aerogel, together with the surfactant, is able to entrain a large amount of air volume (around 0.70 m3 per m3 of aerogel), which together with the evaporated water (not consumed by the binder during hydration), can reach generally around 40% of entrained air within the gypsum matrix, regardless of the amount of aerogel in the mixture. Thus, the entrained air into the matrix displaces the volume proportions of the aerogel and gypsum, reducing both mechanical and insulating properties of the mineral composite. On the other hand, it has been observed a direct contribution of three main phases into the thermal conductivity of the composite: gypsum, aerogel and entrained air. Thus, it was possible to develop a mathematical model (adapted from an existing one), capable of calculating quite accurate the thermal conductivity of such mineral composites, from the ratio these three components and for the range of volumes and materials used in this thesis. Finally, the energy simulation performed to a typical Spanish dwelling, from the years 1900 to 1959 (mainly constructed with massive clay bricks), within three climatic zones of Spain (A, D and E), showed the energy savings potential that can provide this type of insulating material, depending on the thickness of the applied layer. Particularly, for the climatic A zone, it has been found an optimal layer thickness of 1 cm, while for zone D and E, 3.5 and 3.9 cm respectively. In this manner, the new studied materials is capable of decreasing the thickness of the insulating layer by 35% and 80%, compared with rock wool panels or mineral mortars with the highest insulating performance of the Spanish market respectively.

A finales del siglo XIX y principios del XX, la aparición de nuevos materiales, como el acero y el hormigón armado, y la experimentación en procedimientos industriales provocan un cambio en el concepto de cerramiento y en la forma de construir. La fachada se libera y se independiza de la estructura principal, y el nuevo cerramiento debe responder a los principios arquitectónicos y constructivos de este momento. Se busca, por tanto, un cerramiento nuevo. Un cerramiento ligero, de poco peso, de poco espesor, autoportante, multicapa, montado en seco, de grandes dimensiones y que cumpla las exigencias de todo cerramiento. Se puede afirmar que, hasta que Jean Prouvé experimenta con distintos materiales y sistemas de fabricación, la técnica de los cerramientos ligeros no se desarrolla por completo. En sus trabajos se pueden encontrar aplicaciones de los nuevos materiales y nuevas técnicas, e investigaciones sobre prefabricación ligera en acero y aluminio, en un intento de aplicar la producción industrial y en serie a la construcción. Esta Tesis realiza un análisis en profundidad, tanto gráfico como escrito, de los cerramientos verticales desarrollados por Jean Prouvé, sin tratarlos como objetos aislados, entendiendo que forman parte de una obra arquitectónica concreta y completa. Dicho análisis sirve para clasificarlos según las funciones esenciales que debe garantizar un cerramiento: aislar, iluminar, ventilar y proteger, y para comprender cuáles son las claves, los recursos e intenciones, utilizadas por el autor para conseguir este propósito. El resultado de la investigación se plasma de dos formas diferentes. En la primera, se realizan reflexiones críticas para extraer los temas importantes de los elementos analizados, lo que posibilita el acercamiento a otros arquitectos y ampliar el campo de visión. En la segunda, de tipo gráfico, se elabora un atlas de los distintos tipos de cerramientos verticales desarrollados por Jean Prouvé. ABSTRACT In the late nineteenth and the early twentieth century, the appearance of new materials, like steel or reinforced concrete, and the experimentation in industrial procedures cause a change in the concept of façade and in the way of build. The façade is released and become independent of the main structural frame, and the new building enclosure must answer the architectural and construction principles of that moment. A new façade is therefore looked for. A light, thin, self supported, multi layer, dry mounted and big dimensions façade that meet the exigencies of all building enclosure. You can ensure that until Jean Prouvé experiment with several materials and fabrication systems, the light façade technic does not develop completely. In his work we can find new materials applications and new technics and studies about light prefabrication with steel and aluminium, in an attempt of apply the mass production to construction. This Thesis carries out a deep analysis, graphic and written, of the vertical enclosure panels of Jean Prouvé’s work. This is made without studying them like isolated objects, but understanding that they are part of a particular architectural work, as a whole. The analysis is used for classify the panels according to main functions that a façade must satisfy: isolate, light up, ventilate and protect. And also to understand which are the keys, the resources and intentions used by Prouvé to achieve this goal. The result of the research is presented in two different ways. In the first one, a critical reflection is made in order to extract the important issues of the analyzed elements. That makes possible the approach to other architects and gives us a bigger range of vision. In the second, graphic, an atlas of the different types of vertical façade panels of Jean Prouvé is made.

Inbred 129 strain mice are predisposed to developing male germ cell tumors (GCTs) of the testes. The inherent genetic defects that underlie male GCT susceptibility in the 129 mouse strain are unknown. GCT incidence is increased in 129 strain males that lack functional p53 protein, and we have used this finding to facilitate the generation of panels of GCT-bearing intercross and backcross mice for genetic mapping analysis. A 129 strain locus, designated pgct1, that segregates with the male GCT phenotype has been identified on chromosome 13 near D13Mit188. This region of murine chromosome 13 may be syntenic to a portion of human chromosome 5q that is implicated in male GCT susceptibility in humans.

A large library of phage-displayed human single-chain Fv antibodies (scFv), containing 6.7 × 109 members, was generated by improving the steps of library construction. Fourteen different protein antigens were used to affinity select antibodies from this library. A panel of specific antibodies was isolated with each antigen, and each panel contained an average of 8.7 different scFv. Measurements of antibody–antigen interactions revealed several affinities below 1 nM, comparable to affinities observed during the secondary murine immune response. In particular, four different scFv recognizing the ErbB2 protein had affinities ranging from 220 pM to 4 nM. Antibodies derived from the library proved to be useful reagents for immunoassays. For example, antibodies generated to the Chlamydia trachomatis elementary bodies stained Chlamydia-infected cells, but not uninfected cells. These results demonstrate that phage antibody libraries are ideally suited for the rapid production of panels of high-affinity mAbs to a wide variety of protein antigens. Such libraries should prove especially useful for generating reagents to study the function of gene products identified by genome projects.

Here we present the successful application of the microarray technology platform to the analysis of DNA polymorphisms. Using the rice genome as a model, we demonstrate the potential of a high-throughput genome analysis method called Diversity Array Technology, DArT‘. In the format presented here the technology is assaying for the presence (or amount) of a specific DNA fragment in a representation derived from the total genomic DNA of an organism or a population of organisms. Two different approaches are presented: the first involves contrasting two representations on a single array while the second involves contrasting a representation with a reference DNA fragment common to all elements of the array. The Diversity Panels created using this method allow genetic fingerprinting of any organism or group of organisms belonging to the gene pool from which the panel was developed. Diversity Arrays enable rapid and economical application of a highly parallel, solid-state genotyping technology to any genome or complex genomic mixtures.

The Zebrafish Information Network, ZFIN, is a WWW community resource of zebrafish genetic, genomic and developmental research information (http://zfin.org). ZFIN provides an anatomical atlas and dictionary, developmental staging criteria, research methods, pathology information and a link to the ZFIN relational database (http://zfin.org/ZFIN/). The database, built on a relational, object-oriented model, provides integrated information about mutants, genes, genetic markers, mapping panels, publications and contact information for the zebrafish research community. The database is populated with curated published data, user submitted data and large dataset uploads. A broad range of data types including text, images, graphical representations and genetic maps supports the data. ZFIN incorporates links to other genomic resources that provide sequence and ortholog data. Zebrafish nomenclature guidelines and an automated registration mechanism for new names are provided. Extensive usability testing has resulted in an easy to learn and use forms interface with complex searching capabilities.

Early detection is an effective means of reducing cancer mortality. Here, we describe a highly sensitive high-throughput screen that can identify panels of markers for the early detection of solid tumor cells disseminated in peripheral blood. The method is a two-step combination of differential display and high-sensitivity cDNA arrays. In a primary screen, differential display identified 170 candidate marker genes differentially expressed between breast tumor cells and normal breast epithelial cells. In a secondary screen, high-sensitivity arrays assessed expression levels of these genes in 48 blood samples, 22 from healthy volunteers and 26 from breast cancer patients. Cluster analysis identified a group of 12 genes that were elevated in the blood of cancer patients. Permutation analysis of individual genes defined five core genes (P ≤ 0.05, permax test). As a group, the 12 genes generally distinguished accurately between healthy volunteers and patients with breast cancer. Mean expression levels of the 12 genes were elevated in 77% (10 of 13) untreated invasive cancer patients, whereas cluster analysis correctly classified volunteers and patients (P = 0.0022, Fisher's exact test). Quantitative real-time PCR confirmed array results and indicated that the sensitivity of the assay (1:2 × 108 transcripts) was sufficient to detect disseminated solid tumor cells in blood. Expression-based blood assays developed with the screening approach described here have the potential to detect and classify solid tumor cells originating from virtually any primary site in the body.

The ability to isolate fetal nucleated red blood cells (NRBCs) from the maternal circulation makes possible prenatal genetic analysis without the need for diagnostic procedures that are invasive for the fetus. Such isolation requires antibodies specific to fetal NRBCs. To generate a panel of antibodies to antigens present on fetal NRBCs, a new type of nonimmune phage antibody library was generated in which multiple copies of antibody fragments are displayed on each phage. Antibody fragments specific for fetal NRBCs were isolated by extensive predepletion of the phage library on adult RBCs and white blood cells (WBCs) followed by positive selection and amplification on fetal liver erythroid cells. After two rounds of selection, 44% of the antibodies analyzed bound fetal NRBCs, with two-thirds of these showing no binding of WBCs. DNA fingerprint analysis revealed the presence of at least 16 unique antibodies. Antibody specificity was confirmed by flow cytometry, immunohistochemistry, and immunofluorescence of total fetal liver and adult RBCs and WBCs. Antibody profiling suggested the generation of antibodies to previously unknown fetal RBC antigens. We conclude that multivalent display of antibodies on phage leads to efficient selection of panels of specific antibodies to cell surface antigens. The antibodies generated to fetal RBC antigens may have clinical utility for isolating fetal NRBCs from maternal circulation for noninvasive prenatal genetic diagnosis. Some of the antibodies may also have possible therapeutic utility for erythroleukemia.

The incubation period (IP) and the neuropathology of transmissible spongiform encephalopathies (TSEs) have been extensively used to distinguish prion isolates (or strains) inoculated into panels of inbred mouse strains. Such studies have shown that the bovine spongiform encephalopathy (BSE) agent is indistinguishable from the agent causing variant Creutzfeldt–Jakob disease (vCJD), but differs from isolates of sporadic CJD, reinforcing the idea that the vCJD epidemic in Britain results from consumption of contaminated beef products. We present a mouse model for genetic and environmental factors that modify the incubation period of BSE cross-species transmission. We have used two mouse strains that carry the same prion protein (PrP) allele, but display a 100-day difference in their mean IP following intracerebral inoculation with primary BSE isolate. We report genetic effects on IP that map to four chromosomal regions, and in addition we find significant factors of host environment, namely the age of the host's mother, the age of the host at infection, and an X-cytoplasm interaction in the host.

Clinical evidence of hematopoietic restoration with placental/umbilical cord blood (PCB) grafts indicates that PCB can be a useful source of hematopoietic stem cells for routine bone marrow reconstitution. In the unrelated setting, human leukocyte antigen (HLA)-matched donors must be obtained for candidate patients and, hence, large panels of frozen HLA-typed PCB units must be established. The large volume of unprocessed units, consisting mostly of red blood cells, plasma, and cryopreservation medium, poses a serious difficulty in this effort because storage space in liquid nitrogen is limited and costly. We report here that almost all the hematopoietic colony-forming cells present in PCB units can be recovered in a uniform volume of 20 ml by using rouleaux formation induced by hydroxyethyl starch and centrifugation to reduce the bulk of erythrocytes and plasma and, thus, concentrate leukocytes. This method multiples the number of units that can be stored in the same freezer space as much as 10-fold depending on the format of the storage system. We have also investigated the proportion of functional stem/progenitor cells initially present that are actually available to the recipient when thawed cryopreserved PCB units are infused. Progenitor cell viability is measurably decreased when thawed cells, still suspended in hypertonic cryopreservative solutions, are rapidly mixed with large volumes of isotonic solutions or plasma. The osmotic damage inflicted by the severe solute concentration gradient, however, can be averted by a simple 2-fold dilution after thawing, providing almost total recovery of viable hematopoietic progenitor cells.

The mutagen-sensitive CHO line irs1SF was previously isolated on the basis of hypersensitivity to ionizing radiation and was found to be chromosomally unstable as well as cross-sensitive to diverse kinds of DNA-damaging agents. The analysis of somatic cell hybrids formed between irs1SF and human lymphocytes implicated a human gene (defined as XRCC3; x-ray repair cross-complementing), which partially restored mitomycin C resistance to the mutant. A functional cDNA that confers mitomycin C resistance was transferred to irs1SF cells by transforming them with an expression cDNA library and obtaining primary and secondary transformants. Functional cDNA clones were recovered from a cosmid library prepared from a secondary transformant. Transformants also showed partial correction of sensitivity to cisplatin and gamma-rays, efficient correction of chromosomal instability, and substantially improved plating efficiency and growth rate. The XRCC3 cDNA insert is approximately 2.5 kb and detects an approximately 3.0-kb mRNA on Northern blots. The cDNA was mapped by fluorescence in situ hybridization to human chromosome 14q32.3, which was consistent with the chromosome concordance data of two independent hybrid clone panels.

Human monoclonal antibodies have considerable potential in the prophylaxis and treatment of viral disease. However, only a few such antibodies suitable for clinical use have been produced to date. We have previously shown that large panels of human recombinant monoclonal antibodies against a plethora of infectious agents, including herpes simplex virus types 1 and 2, can be established from phage display libraries. Here we demonstrate that facile cloning of recombinant Fab fragments against specific viral proteins in their native conformation can be accomplished by panning phage display libraries against viral glycoproteins "captured" from infected cell extracts by specific monoclonal antibodies immobilized on ELISA plates. We have tested this strategy by isolating six neutralizing recombinant antibodies specific for herpes simplex glycoprotein gD or gB, some of which are against conformationally sensitive epitopes. By using defined monoclonal antibodies for the antigen-capture step, this method can be used for the isolation of antibodies to specific regions and epitopes within the target viral protein. For instance, monoclonal antibodies to a nonneutralizing epitope can be used in the capture step to clone antibodies to neutralizing epitopes, or antibodies to a neutralizing epitope can be used to clone antibodies to a different neutralizing epitope. Furthermore, by using capturing antibodies to more immunodominant epitopes, one can direct the cloning to less immunogenic ones. This method should be of value in generating antibodies to be used both in the prophylaxis and treatment of viral infections and in the characterization of the mechanisms of antibody protective actions at the molecular level.

Steroidogenic acute regulatory protein (StAR) appears to mediate the rapid increase in pregnenolone synthesis stimulated by tropic hormones. cDNAs encoding StAR were isolated from a human adrenal cortex library. Human StAR, coexpressed in COS-1 cells with cytochrome P450scc and adrenodoxin, increased pregnenolone synthesis > 4-fold. A major StAR transcript of 1.6 kb and less abundant transcripts of 4.4 and 7.5 kb were detected in ovary and testis. Kidney had a lower amount of the 1.6-kb message. StAR mRNA was not detected in other tissues including placenta. Treatment of granulosa cells with 8-bromo-adenosine 3',5'-cyclic monophosphate for 24 hr increased StAR mRNA 3-fold or more. The structural gene encoding StAR was mapped using somatic cell hybrid mapping panels to chromosome 8p. Fluorescence in situ hybridization placed the StAR locus in the region 8p11.2. A StAR pseudogene was mapped to chromosome 13. We conclude that StAR expression is restricted to tissues that carry out mitochondrial sterol oxidations subject to acute regulation by cAMP and that StAR mRNA levels are regulated by cAMP.

Elicitins are a family of small proteins secreted by species of Phytophthora. They are thought to be major determinants of the resistance response of tobacco against these oomycetes, since purified elicitins, alone and at low concentrations, can induce vigorous defense responses in tobacco (i.e., hypersensitive cell death and resistance against subsequent pathogen attack), and in vitro elicitin production by Phytophthora isolates is strongly negatively correlated with their pathogenicity on tobacco plants. A number of elicitins have been purified and their amino acid sequences have been determined and found to be conserved. A three-dimensional structure for elicitin is emerging from nuclear magnetic resonance studies. Two structural classes, alpha and beta, are distinguished by their biological effects when applied to decapitated stems or petioles; the beta class causes more necrosis on leaves and provides better subsequent protection against pathogen attack. However, both these classes of elicitins will similarly cause necrosis when each is, instead, directly infiltrated into tobacco leaf panels. Effects of elicitins on tobacco cells include rapid electrolyte leakage, changes in protein phosphorylation and amounts of active oxygen species, and later production of ethylene and capsidiol. The sites of initial interaction with tobacco cells are unknown, but the interaction appears to induce general defense-related responses.

No presente trabalho, considerou o estudo do potencial de utilização de sacos de cimento e embalagens de longa vida com o intuito de agregar valor a esses resíduos, mediante a fabricação de painéis de partículas para uso como forro em protótipos de galpões avícolas. Para fabricação desse material foi utilizado sacos de cimento descartados nas obras civis, embalagens longa vida residuais e resina poliuretana bicomponente à base de óleo de mamona. O estudo foi dividido em quatro etapas: 1) Caracterização da matéria-prima (sacos de cimento); 2) Efeito da densidade e teor de resina nas propriedades físicas, mecânicas e térmicas de painéis de partículas de saco de cimento; 3) Avaliação do desempenho de embalagens longa vida e verniz como revestimentos dos painéis selecionados na etapa anterior; 4) Determinação do desempenho térmico de protótipos de aviários executados em escala reduzida e distorcida com forro de painel de saco de cimento e embalagem longa vida. Os resultados obtidos indicaram: Em razão do painel com 0,6 g.cm-3 e 12% de resina ter apresentado melhor desempenho nas propriedades investigadas, essa combinação foi selecionada para avaliar o desempenho do material (físicas, mecânicas e térmicas) quando revestido com embalagens longa vida, adotando a testemunha e o verniz como um revestimento comparativo. Dentre os revestimentos avaliados, os painéis com embalagens de longa vida, foram superiores aos revestidos com verniz, quando comparados as suas propriedades físicas, mecânicas e térmicas. Dessa maneira, painéis com 0,6 g.cm-3 e 12% de resina revestidos embalagens longa vida, foram testados como forro quanto ao desempenho térmico (primavera, verão, outono e inverno) em protótipos em escala reduzida e distorcida, mediante a determinação de índices de conforto térmico (Entalpia e IAPfc) para aves de corte. A associação do forro sob o protótipo permitiu reduzir a temperatura interna do ar e dos índices de conforto térmico na primavera, verão e outono, enquanto que no inverno não foi constatado eficácia do material. Dentre as estações estudadas, o verão e a primavera, demonstraram serem as épocas mais críticas para criação de aves corte em instalações avícolas que apresentem características semelhantes aos protótipos experimentais.
