Ground-penetrating radar (GPR) has the potential to provide valuable information on hydrological properties of the vadose zone because of their strong sensitivity to soil water content. In particular, recent evidence has suggested that the stochastic inversion of crosshole GPR data within a coupled geophysical-hydrological framework may allow for effective estimation of subsurface van-Genuchten-Mualem (VGM) parameters and their corresponding uncertainties. An important and still unresolved issue, however, is how to best integrate GPR data into a stochastic inversion in order to estimate the VGM parameters and their uncertainties, thus improving hydrological predictions. Recognizing the importance of this issue, the aim of the research presented in this thesis was to first introduce a fully Bayesian inversion called Markov-chain-Monte-carlo (MCMC) strategy to perform the stochastic inversion of steady-state GPR data to estimate the VGM parameters and their uncertainties. Within this study, the choice of the prior parameter probability distributions from which potential model configurations are drawn and tested against observed data was also investigated. Analysis of both synthetic and field data collected at the Eggborough (UK) site indicates that the geophysical data alone contain valuable information regarding the VGM parameters. However, significantly better results are obtained when these data are combined with a realistic, informative prior. A subsequent study explore in detail the dynamic infiltration case, specifically to what extent time-lapse ZOP GPR data, collected during a forced infiltration experiment at the Arrenaes field site (Denmark), can help to quantify VGM parameters and their uncertainties using the MCMC inversion strategy. The findings indicate that the stochastic inversion of time-lapse GPR data does indeed allow for a substantial refinement in the inferred posterior VGM parameter distributions. In turn, this significantly improves knowledge of the hydraulic properties, which are required to predict hydraulic behaviour. Finally, another aspect that needed to be addressed involved the comparison of time-lapse GPR data collected under different infiltration conditions (i.e., natural loading and forced infiltration conditions) to estimate the VGM parameters using the MCMC inversion strategy. The results show that for the synthetic example, considering data collected during a forced infiltration test helps to better refine soil hydraulic properties compared to data collected under natural infiltration conditions. When investigating data collected at the Arrenaes field site, further complications arised due to model error and showed the importance of also including a rigorous analysis of the propagation of model error with time and depth when considering time-lapse data. Although the efforts in this thesis were focused on GPR data, the corresponding findings are likely to have general applicability to other types of geophysical data and field environments. Moreover, the obtained results allow to have confidence for future developments in integration of geophysical data with stochastic inversions to improve the characterization of the unsaturated zone but also reveal important issues linked with stochastic inversions, namely model errors, that should definitely be addressed in future research.

Aquest projecte tracta sobre la implantació d'un sistema ERP per tal de gestionar una empresa dedicada a la producció i distribució de material odontològic. El que durem a terme en aquesta aplicació serà importar les dades de l'aplicació VolkSoft a la nova aplicació. La idea principal es buscar una aplicació que satisfaci les necessitats de l'aplicació VolkSoft i a continuació, importar les dades d'una aplicació a l'altra.

Este estudo teve como objetivo desenvolver estratégias facilitadoras no processo de ensino e aprendizagem, utilizando-se de materiais recicláveis na construção de estruturas e órgãos anatômicos. Foi desenvolvido na disciplina Enfermagem Médico-Cirúrgica, a 31 alunos no curso de nível médio para formação do Auxiliar de Enfermagem. Após leituras de referências bibliográficas relacionadas à Anatomia, Fisiologia e Patologias mais comuns dos diversos sistemas do corpo humano, os alunos selecionaram os materiais recicláveis disponíveis para a confecção das principais estruturas anatômicas, o que possibilitou a visualização das alterações morfológicas decorrentes das afecções, facilitando assim a compreensão das patologias. As estratégias utilizadas favoreceram a aquisição de conhecimentos ao se projetarem em algo concreto, resultado da troca de sugestões e experiências entre os estudantes, favorecido pelo trabalho em grupo.

O estudo teve com objetivo caracterizar o processo de inserção nos Centros de Material e Esterilização - CME, de trabalhadores que não possuem formação específica na área de enfermagem. Fizeram parte do estudo 75 trabalhadores de dois hospitais públicos. Após observação dos aspectos éticos legais, os dados foram obtidos por meio de questionário previamente validado e testado. Foram encontrados 15 trabalhadores sem formação específica, provenientes de serviços diversos intra e extra hospitalar, os quais não foram submetidos a processos de seleção ou treinamentos, tendo aprendido a rotina do serviço com colegas de trabalho. Tais aspectos são preocupantes, considerando-se o risco ocupacional do trabalhador, o comprometimento da qualidade do reprocessamento de artigos hospitalares e o cumprimento da legislação do exercício profissional.

Estudo sobre o trabalho do enfermeiro na central de material e esterilização (CME), com a finalidade de analisar sua realidade social e seu papel transformador no cuidar em saúde e pela enfermagem. Tratou-se de pesquisa estratégica, com análise de discurso de enfermeiras da CME. Obteve-se que a gerência é sua principal atividade, em um processo estruturado que confirma a prática tradicional da enfermagem, tendo como objeto a coordenação do processamento de materiais médico-hospitalares utilizados em atos cuidadores. Tal prática caracteriza-se pelo cuidado indireto, que instrumentaliza não apenas o trabalho da enfermagem, mas os de outros profissionais. Em conseqüência, não se caracteriza como cuidado específico da enfermagem e do enfermeiro. Seu potencial transformador nas relações e novos modos de produção do cuidar em saúde e pelo enfermeiro deverá ocorrer antes pela competência do conhecimento específico em CME do que administrativo.

O estudo identifica parâmetros de produtividade no Centro de Material e Esterilização, com base em documentos, observação e cronometragem do processamento de duas amostras: caixa de herniorrafia e pacote de curativo. A produção média mensal foi de 30.466,42 artigos, a capacidade de produção, por hora, foi de 10,3 artigos por funcionário. O tempo de processamento do curativo do expurgo ao armazenamento foi, em média, de 295 minutos; o processo manual utilizou 46 minutos e o automatizado, 88 minutos. O tempo da caixa de herniorrafia foi de 329 minutos; o processo manual utilizou 60 minutos e o automatizado, 98 minutos. O processamento manual do curativo utilizou 59,23% horas produtivas no expurgo; 26,31% guarda e distribuição; 10,94% esterilização e 3,28% preparo. Quanto à caixa de herniorrafia: no preparo 16,20%; no expurgo 11,84%; na guarda e distribuição 7,47% e na esterilização 1,89%. Este estudo possibilitou aferir resultados e analisar o processo de trabalho no CME.

Este estudo buscou compreender a experiência dos trabalhadores de enfermagem da Unidade de Centro de Material (UCM) de um hospital de Londrina, utilizando a pesquisa qualitativa femomenológica. Foram realizadas dez entrevistas. A análise dos discursos permitiu construir quatro categorias: 1. Falando do ingresso na UCM - situa as diferentes formas de ingresso nesta unidade; 2. Falando sobre o trabalho desenvolvido na UCM - discorre sobre a rotatividade, o trabalho concomitante e a predileção por determinadas áreas; o ambiente fechado; a evolução tecnológica e o relacionamento interpessoal; 3. Vivenciando as dificuldades - descreve a rotina diária; o trabalho repetitivo e monótono, o cansaço físico, a falta de materiais e equipamentos; o período de adaptação, o desconhecimento, a desvalorização e a discriminação vivenciada; 4. Superando os obstáculos - revela a tomada de consciência do seu papel na unidade reconhecendo a sua importância.

The expression of Ia-associated human Invariant (In) chain glycoproteins was studied in the Raji B cells as well as in their RJ 2.2.5 Ia-negative derived variant cells by using a specific rabbit anti-human In chain antiserum. Two-dimensional gel electrophoresis of immunoprecipitates from either biosynthetically labeled or surface labeled cells were analyzed. In addition, flow microfluorometric analysis of stained cells was performed. The results indicate that the In chain is constitutively produced in the Ia-negative B cell variant. Moreover, it appears that several forms of In chain-related molecules, with different charges and distinct m.w. are equally expressed in Ia-positive and Ia-negative B cells. Finally, no evidence could be obtained that the In molecular family was expressed on the cell surface of Ia-positive Raji and Ia-negative RJ 2.2.5 cells.

O estudo tem como objetivo identificar a produção científica sobre o acidente de trabalho do profissional de enfermagem com material biológico contaminado por HIV e caracterizar os fatores predisponentes aos acidentes de trabalho, assim como os procedimentos realizados pós-exposição ao material perfurocortante potencialmente contaminado pelo HIV. É uma revisão de literatura, cuja busca bibliográfica por palavras-chave foi realizada por meio da base de dados LILACS a partir de 2000. Constatou-se que os fatores predisponentes à ocorrência dos acidentes de trabalho com perfurocortantes estão relacionados tanto às condições de trabalho, como as condições individuais. Frente a estes acidentes, os trabalhadores de enfermagem têm necessidade de conhecer as condutas pós-exposição ao material perfurocortante potencialmente contaminado pelo HIV. Concluímos que a adoção das precauções-padrão no trabalho em saúde é condição fundamental para a segurança dos trabalhadores, independente da área de atuação, pois o número de casos de HIV é crescente.

The Computational Biophysics Group at the Universitat Pompeu Fabra (GRIB-UPF) hosts two unique computational resources dedicated to the execution of large scale molecular dynamics (MD) simulations: (a) the ACMD molecular-dynamics software, used on standard personal computers with graphical processing units (GPUs); and (b) the GPUGRID. net computing network, supported by users distributed worldwide that volunteer GPUs for biomedical research. We leveraged these resources and developed studies, protocols and open-source software to elucidate energetics and pathways of a number of biomolecular systems, with a special focus on flexible proteins with many degrees of freedom. First, we characterized ion permeation through the bactericidal model protein Gramicidin A conducting one of the largest studies to date with the steered MD biasing methodology. Next, we addressed an open problem in structural biology, the determination of drug-protein association kinetics; we reconstructed the binding free energy, association, and dissaciociation rates of a drug like model system through a spatial decomposition and a Makov-chain analysis. The work was published in the Proceedings of the National Academy of Sciences and become one of the few landmark papers elucidating a ligand-binding pathway. Furthermore, we investigated the unstructured Kinase Inducible Domain (KID), a 28-peptide central to signalling and transcriptional response; the kinetics of this challenging system was modelled with a Markovian approach in collaboration with Frank Noe’s group at the Freie University of Berlin. The impact of the funding includes three peer-reviewed publication on high-impact journals; three more papers under review; four MD analysis components, released as open-source software; MD protocols; didactic material, and code for the hosting group.

Using a direct binding assay based on photoaffinity labeling, we studied the interaction of T cell receptor (TCR) with a Kd-bound photoreactive peptide derivative on living cells. The Kd-restricted Plasmodium berghei circumsporozoite (PbCS) peptide 253-260 (YIPSAEKI) was reacted NH2-terminally with biotin and at the TCR contact residue Lys259 with photoreactive iodo, 4-azido salicylic acid (IASA) to make biotin-YIPSAEK(IASA)I. Cytotoxic T lymphocyte (CTL) clones derived from mice immunized with this derivative recognized this conjugate, but not a related one lacking the IASA group nor the parental PbCS peptide. The clones were Kd restricted. Recognition experiments with variant conjugates, lacking substituents from IASA, revealed a diverse fine specificity pattern and indicated that this group interacted directly with the TCR. The TCR of four clones could be photoaffinity labeled by biotin-YIPSAEK(125IASA)I. This labeling was dependent on the conjugates binding to the Kd molecule and was selective for the TCR alpha (2 clones) or beta chain (1 clone), or was common for both chains (1 clone). TCR sequence analysis showed a preferential usage of J alpha TA28 containing alpha chains that were paired with V beta 1 expressing beta chains. The TCR that were photoaffinity labeled at the alpha chain expressed these J alpha and V beta segments. The tryptophan encoded by the J alpha TA28 segment is rarely found in other J alpha segments. Moreover, we show that the IASA group interacts preferentially with tryptophan in aqueous solution. We thus propose that for these CTL clones, labeling of the alpha chain occurs via the J alpha-encoded tryptophan residue.

Estudo transversal, tipo survey, realizado com a equipe multiprofissional de Atendimento Pré-hospitalar (APh) de Belo Horizonte, entre junho e dezembro de 2006. Objetivou-se determinar a incidência dos acidentes ocupacionais por exposição a material biológico, condutas pós-acidente, e fatores demográficos determinantes. Utilizou-se questionário estruturado, análise descritiva, cálculo de incidências e regressão logística. A incidência de acidentes com material biológico foi de 20,6%: 40,8% por pérfuro-cortantes e 49,0% por fluidos corporais; 35,3% entre médicos e 24,0% entre enfermeiros. Condutas pós-acidente: sem avaliação médica, 63,3%; subnotificação, 81,6%; nenhuma conduta, 55,0%; e, sem acompanhamento sorológico, 61,2%. Estiveram associados ao acidente: tempo na instituição (Odds ratio-OR 2,84; Intervalo de confiança-IC 95% 1,22-6,62), lotação na Unidade de Suporte Avançado (OR 4,18; IC 95% 1,64-10,64); interação: tempo na instituição e lotação na Unidade de Suporte Básico (OR 0,27; IC 95% 0,07-1,00). Sugere-se a implantação de protocolos pós-acidentes, visando a sua redução; a subnotificação e o aumento do acompanhamento pós-acidente.

OBJECTIVE: To identify which physician and patient characteristics are associated with physicians' estimation of their patient social status.DESIGN: Cross-sectional ulticentric survey. SETTING: Fourty-seven primary care private offices in Western Switzerland. PARTICIPANTS: Random sample of 2030 patients ≥ 16, who encountered a general practitioner (GP) between September 2010 and February 2011. MAIN MEASURES: PRIMARY OUTCOME: patient social status perceived by GPs, using the MacArthur Scale of Subjective Social Status, ranging from the bottom (0) to the top (10) of the social scale.Secondary outcome: Difference between GP's evaluation and patient's own evaluation of their social status. Potential patient correlates: material and social deprivation using the DiPCare-Q, health status using the EQ-5D, sources of income, and level of education. GP characteristics: opinion regarding patients' deprivation and its influence on health and care. RESULTS: To evaluate patient social status, GPs considered the material, social, and health aspects of deprivation, along with education level, and amount and type of income. GPs declaring a frequent reflexive consideration of their own prejudice towards deprived patients, gave a higher estimation of patients' social status (+1.0, p = 0.002). Choosing a less costly treatment for deprived patients was associated with a lower estimation (-0.7, p = 0.002). GP's evaluation of patient social status was 0.5 point higher than the patient's own estimate (p<0.0001). CONCLUSIONS: GPs can perceive the various dimensions of patient social status, although heterogeneously, according partly to their own characteristics. Compared to patients' own evaluation, GPs overestimate patient social status.

In order to characterize the gene encoding the ligand binding (1(st); alpha) chain of the human IFN-gamma receptor, two overlapping cosmid clones were analyzed. The gene spans over 25 kilobases (kb) of the genomic DNA and has seven exons. The extracellular domain is encoded by exons 1 to 5 and by part of exon 6. The transmembrane region is also encoded by exon 6. Exon 7 encodes the intracellular domain and the 3' untranslated portion. The gene was located on chromosome 6q23.1, as determined by in situ hybridization. The 4 kb region upstream (5') of the gene was sequenced and analyzed for promoter activity. No consensus-matching TATA or CAAT boxes in the 5' region were found. Potential binding sites for Sp1, AP-1, AP-2, and CREB nuclear factors were identified. Compatible with the presence of the Sp1/AP-2 sites and the lack of TATA box, S1-nuclease mapping experiments showed multiple transcription initiation sites. Promoter activity of the 5' flanking region was analyzed with two different reporter genes: the Escherichia coli chloramphenicol acetyltransferase and human growth hormone. The smallest 5' region of the gene that still had full promoter activity was 692 base pairs in length. In addition, we found sequences belonging to the oldest family of Alu repeats, 2 - 3 kb upstream of the gene, which could be useful for genetic studies.

The fungus Aspergillus nidulans contains both a mitochondrial and peroxisomal ß-oxidation pathway. This work was aimed at studying the influence of mutations in the foxA gene, encoding a peroxisomal multifunctional protein, or in the scdA/echA genes, encoding a mitochondrial short-chain dehydrogenase and an enoyl-CoA hydratase, respectively, on the carbon flux to the peroxisomal ß-oxidation pathway. A. nidulans transformed with a peroxisomal polyhydroxyalkanoate (PHA) synthase produced PHA from the polymerization of 3-hydroxyacyl-CoA intermediates derived from the peroxisomal ß-oxidation of external fatty acids. PHA produced from erucic acid or heptadecanoic acid contained a broad spectrum of monomers, ranging from 5 to 14 carbons, revealing that the peroxisomal ß-oxidation cycle can handle both long and short-chain intermediates. While the ∆foxA mutant grown on erucic acid or oleic acid synthesized 10-fold less PHA compared to wild type, the same mutant grown on octanoic acid or heptanoic acid produced 3- to 6-fold more PHA. Thus, while FoxA has an important contribution to the degradation of long-chain fatty acids, the flux of short-chain fatty acids to peroxisomal ß-oxidation is actually enhanced in its absence. While no change in PHA was observed in the ∆scdA∆echA mutant grown on erucic acid or oleic acid compared to wild type, there was a 2- to 4-fold increased synthesis of PHA in ∆scdA∆echA cells grown in octanoic acid or heptanoic acid. These results reveal that a compensatory mechanism exists in A. nidulans that increases the flux of short-chain fatty acids towards the peroxisomal ß-oxidation cycle when the mitochondrial ß-oxidation pathway is defective.
