BACKGROUND Recently, histopathological tumour regression, prevalence of signet ring cells, and localisation were reported as prognostic factors in neoadjuvantly treated oesophagogastric (junctional and gastric) cancer. This exploratory retrospective study analyses independent prognostic factors within a large patient cohort after preoperative chemotherapy including clinical and histopathological factors. METHODS In all, 850 patients presenting with oesophagogastric cancer staged cT3/4 Nany cM0/x were treated with neoadjuvant chemotherapy followed by resection in two academic centres. Patient data were documented in a prospective database and retrospectively analysed. RESULTS Of all factors prognostic on univariate analysis, only clinical response, complications, ypTNM stage, and R category were independently prognostic (P<0.01) on multivariate analysis. Tumour localisation and signet ring cells were independently prognostic only when investigator-dependent clinical response evaluation was excluded from the multivariate model. Histopathological tumour regression correlates with tumour grading, Laurén classification, clinical response, ypT, ypN, and R categories but was not identified as an independent prognostic factor. Within R0-resected patients only surgical complications and ypTNM stage were independent prognostic factors. CONCLUSIONS Only established prognostic factors like ypTNM stage, R category, and complications were identified as independent prognostic factors in resected patients after neoadjuvant chemotherapy. In contrast, histopathological tumour regression was not found as an independent prognostic marker.

Recent studies have demonstrated that the improved prognosis derived from resection of gliomas largely depends on the extent and quality of the resection, making maximum but safe resection the ultimate goal. Simultaneously, technical innovations and refined neurosurgical methods have rapidly improved efficacy and safety. Because gliomas derive from intrinsic brain cells, they often cannot be visually distinguished from the surrounding brain tissue during surgery. In order to appreciate the full extent of their solid compartment, various technologies have recently been introduced. However, radical resection of infiltrative glioma puts neurological function at risk, with potential detrimental consequences for patients' survival and quality of life. The allocation of various neurological functions within the brain varies in each patient and may undergo additional changes in the presence of a tumour (brain plasticity), making intra-operative localisation of eloquent areas mandatory for preservation of essential brain functions. Combining methods that visually distinguish tumour tissue and detect tissues responsible for critical functions now enables resection of tumours in brain regions that were previously considered off-limits, and benefits patients by enabling a more radical resection, while simultaneously lowering the risk of neurological deficits. Here we review recent and expected developments in microsurgery for glioma and their respective benefits.

A compact and planar donor–acceptor molecule 1 comprising tetrathiafulvalene (TTF) and benzothiadiazole (BTD) units has been synthesised and experimentally characterised by structural, optical, and electrochemical methods. Solution-processed and thermally evaporated thin films of 1 have also been explored as active materials in organic field-effect transistors (OFETs). For these devices, hole field-effect mobilities of μFE=(1.3±0.5)×10−3 and (2.7±0.4)×10−3 cm2 V s−1 were determined for the solution-processed and thermally evaporated thin films, respectively. An intense intramolecular charge-transfer (ICT) transition at around 495 nm dominates the optical absorption spectrum of the neutral dyad, which also shows a weak emission from its ICT state. The iodine-induced oxidation of 1 leads to a partially oxidised crystalline charge-transfer (CT) salt {(1)2I3}, and eventually also to a fully oxidised compound {1I3}⋅1/2I2. Single crystals of the former CT compound, exhibiting a highly symmetrical crystal structure, reveal a fairly good room temperature electrical conductivity of the order of 2 S cm−1. The one-dimensional spin system bears compactly bonded BTD acceptors (spatial localisation of the LUMO) along its ridge.

Le relief accidenté, le climat agressif favorisé par l’Alizé, ainsi que l’étroite dépendance de la population de l’exploitation forestière et de l’exploitation agricole sans dispositifs de protection antiérosifs, sont les contextes qui permettent de dire que la région de Mandraka est sujette au problème d’érosion. L’étude a pour objectif de déterminer les relations entre les caractéristiques du sol, son mode d’occupation, les précipitations ainsi que l’érosion dans la région de Mandraka et ce en vue d’un aménagement rationnel. Des dispositifs pour la mesure de pertes en terre et de ruissellement ont été matérialisés sur des parcelles d’expérimentation. A cet effet, le choix de ces parcelles a porté sur le mode d’occupation du sol (sous terrains aménagés, sous « tavy », sous « savoka », et sous forêt naturelle), sur la similitude des pentes et sur leur localisation de manière à ce qu’elles soient situées sur les mi-versants. Les résultats ont montré que le sol dans cette région contient un taux de matière organique satisfaisant à élevé (2,41 à 3,74% de MO), lui permettant une bonne stabilité structurale (Is = 0,85 et K = 0,08). Les pertes en terre (0,13 à 20,93t/ha/an) et les ruissellements (0,3 à 5%) obtenus indiquent que l’érosion est faible à accélérée suivant le mode d’occupation du sol. En deux ans d’aménagement, les terrasses se sont stabilisées et les racines de Vetiveria zizanoides peuvent remplacer les fascines initialement installées pour tenir les talus. Toutefois, l’exposition du sol à la battance de la pluie et la mauvaise infiltration d’eau de ruissellement dans le sol accélèrent le phénomène d’érosion. Par ailleurs, il y a l’efficacité de la SCV sans brûlis bien qu’il s’agit de la première année d’expérience. Dans ces conditions, un plan d’aménagement simplifié a été proposé en vue (i) de protéger le sol contre la battance de la pluie, (ii) de réduire les transports solides par l’érosion et (iii) d’améliorer l’infiltration d’eau du sol.

STUDY DESIGN Retrospective data analysis. OBJECTIVES To document fracture characteristics, management and related complications in individuals with traumatic spinal cord injury (SCI). SETTING Rehabilitation centre for SCI individuals. METHOD Patients' records were reviewed. Patients with traumatic SCI and extremity fractures that had occurred after SCI were included. Patient characteristics, fractured bone, fracture localisation, severity and management (operative/conservative), and fracture-related complications were extracted. RESULTS A total of 156 long-bone fractures in 107 SCI patients (34 women and 73 men) were identified. The majority of patients were paraplegics (77.6%) and classified as American Spinal Injury Association Impairment Scale A (86.0%). Only the lower extremities were affected, whereby the femur (60.9% of all fractures) was fractured more frequently than the lower leg (39.1%). A total of 70 patients (65.4%) had one fracture, whereas 37 patients (34.6%) had two or more fractures. Simple or extraarticular fractures were most common (75.0%). Overall, 130 (83.3%) fractures were managed operatively. Approximately half of the femur fractures (48.2%) were treated with locking compression plates. In the lower leg, fractures were mainly managed with external fixation (48.8%). Conservative fracture management was applied in 16.7% of the cases and consisted of braces or a well-padded soft cast. Fracture-associated complications were present in 13.5% of the cases but did not differ significantly between operative (13.1%) and conservative (15.4%) fracture management. CONCLUSION SCI was associated with simple or extraarticular fractures of the distal femur and the lower leg. Fractures were mainly managed operatively with a low complication rate.

Periazetabuläre Frakturen bei Hüftprothesen nehmen aufgrund der Überalterung und der zunehmenden Aktivität alter Menschen zu. Die periprothetischen Azetabulumfrakturen werden anhand der Einteilung von Letournel klassifiziert. Wenn beide Azetabulumpfeiler bei Hüftprothese betroffen sind, wird auch von einer Beckendiskontinuität gesprochen. Durch eine laterale Kompression können auch periazetabuläre Schambeinastfrakturen und/oder transiliakale Frakturen auftreten. Für die Therapieentscheidung (konservativ, alleinige Osteosynthese, Revisionshüfttotalprothese mit oder ohne zusätzliche Osteosynthese des Vorder- und/oder Hinterpfeilers) und die Zugangswahl bei operativer Versorgung werden patientenspezifische (Alter, Morbidität, Osteoporose, Aktivitätslevel des Patienten), frakturspezifische (Frakturtyp, Dislokationsausmaß, Impression des Doms oder der Hinterwand) und auch prothesenspezifische Faktoren (Art der implantierten Prothese [Hemiprothese vs. Totalprothese], Pfannenstabilität, Zeichen eines Prothesenabriebs, Ausmaß und Lokalisation einer azetabulären Lyse, Stabilität und Lysezeichen des Prothesenschafts) berücksichtigt. Bei akuten Beckendiskontinuitäten werden neben einer Osteosynthese des dorsalen Pfeilers zunehmend eine schnell ossär integrierbare Pfanne (Tantalum [„Trabecular Metal“: TM]) mit oder ohne Augment und/oder Allograft und allenfalls in einer sog. „Cup-Cage“-Technik (TM-Pfanne mit einem abstützenden Revisionsring [Burch-Schneider-Ring] analog zur Therapie von chronischen Beckendiskontinuitäten empfohlen. Bei großen Lysezonen und starken Dislokationen des vorderen Pfeilers und der quadrilateralen Fläche können intrapelvine Zugänge (modifizierter Stoppa- oder Pararectus-Zugang nach Keel) zur zusätzlichen Zuggurtungsosteosynthese des vorderen Pfeilers und Abstützung der quadrilateralen Fläche gewählt werden.

The coding sequence of the wild-type, cys-sensitive, cysE gene from Escherichia coli, which encodes an enzyme of the cysteine biosynthetic pathway, namely serine acetyltransferase (SAT, EC 2.3.1.30), was introduced into the genome of potato plants under the control of the cauliflower mosaic virus 35S promoter. In order to target the protein into the chloroplast, cysE was translationally fused to the 5′-signal sequence of rbcS from Arabidopsis thaliana. Transgenic plants showed a high accumulation of the cysE mRNA. The chloroplastic localisation of the E. coli SAT protein was demonstrated by determination of enzymatic activities in enriched organelle fractions. Crude leaf extracts of these plants exhibited up to 20-fold higher SAT activity than those prepared from wild-type plants. The transgenic potato plants expressing the E. coli gene showed not only increased levels of enzyme activity but also exhibited elevated levels of cysteine and glutathione in leaves. Both were up to twofold higher than in control plants. However, the thiol content in tubers of transgenic lines was unaffected. The alterations observed in leaf tissue had no effect on the expression of O-acetylserine(thiol)-lyase, the enzyme which converts O-acetylserine, the product of SAT, to cysteine. Only a minor effect on its enzymatic activity was observed. In conclusion, the results presented here demonstrate the importance of SAT in plant cysteine biosynthesis and show that production of cysteine and related sulfur-containing compounds can be enhanced by metabolic engineering.

Protozoan parasites which reside inside a host cell avoid direct destruction by the immune system of the host. The infected cell, however, still has the capacity to counteract the invasive pathogen by initiating its own death, a process which is called programmed cell death or apoptosis. Apoptotic cells are recognised and phagocytosed by macrophages and the parasite is potentially eliminated together with the infected cell. This potent defence mechanism of the host cell puts strong selective pressure on the parasites which have, in turn, evolved strategies to modulate the apoptotic program of the host cell to their favour. Within the last decade, the existence of cellular signalling pathways which inhibit the apoptotic machinery has been demonstrated. It is not surprising that intracellular pathogens subvert these pathways to ensure their own survival in the infected cell. Molecular mechanisms which interfere with apoptotic pathways have been studied extensively for viruses and parasitic bacteria, but protozoan parasites have come into focus only recently. Intracellular protozoan parasites which have been reported to inhibit the apoptotic program of the host cell, are Toxoplasma gondii, Trypanosoma cruzi, Leishmania sp., Theileria sp., Cryptosporidium parvum, and the microsporidian Nosema algerae. Although these parasites differ in their mechanism of host cell entry and in their final intracellular localisation, they might activate similar pathways in their host cells to inhibit apoptosis. In this respect, two families of molecules, which are known for their capacity to interrupt the apoptotic program, are currently discussed in the literature. First, the expression of heat shock proteins is often induced upon parasite infection and can directly interfere with molecules of the cellular death machinery. Secondly, a more indirect effect is attributed to the parasite-dependent activation of NF-kappaB, a transcription factor that regulates the transcription of anti-apoptotic molecules.

They are used to measure the impact of biomass removal by fisheries and to evaluate the models skills, while the use of standard dataset facilitates models inter-comparison. North Atlantic albacore tuna is exploited all year round by longline and in summer and autumn by surface fisheries and fishery statistics compiled by the International Commission for the Conservation of Atlantic Tunas (ICCAT). Catch and effort with geographical coordinates at monthly spatial resolution of 1° or 5° squares were extracted for this species with a careful definition of fisheries and data screening. In total, thirteen fisheries were defined for the period 1956-2010, with fishing gears longline, troll, mid-water trawl and bait fishing. However, the spatialized catch effort data available in ICCAT database represent a fraction of the entire total catch. Length frequencies of catch were also extracted according to the definition of fisheries above for the period 1956-2010 with a quarterly temporal resolution and spatial resolutions varying from 1°x 1° to 10°x 20°. The resolution used to measure the fish also varies with size-bins of 1, 2 or 5 cm (Fork Length). The screening of data allowed detecting inconsistencies with a relatively large number of samples larger than 150 cm while all studies on the growth of albacore suggest that fish rarely grow up over 130 cm. Therefore, a threshold value of 130 cm has been arbitrarily fixed and all length frequency data above this value removed from the original data set.

North Atlantic albacore tuna is exploited all year round by longline and in summer and autumn by surface fisheries and fishery statistics compiled by the International Commission for the Conservation of Atlantic Tunas (ICCAT). Catch and effort with geographical coordinates at monthly spatial resolution of 1° or 5° squares were extracted for this species with a careful definition of fisheries and data screening. Length frequencies of catch were also extracted according to the definition of fisheries for the period 1956-2010. Using these data, an application of the spatial ecosystem and population dynamics model (SEAPODYM) was developed for the North Atlantic albacore population and fisheries and provided the first spatially explicit estimate of albacore density in the North Atlantic by life stage. These densities by life stage (larval recruits, young immature fish adult mature fish and total biomass) are provided in gridded file (Netcdf) at resolution of 2° x 2° x month.
