The isoenzyme profiles (IP) of 33 strains of Entamoeba histolytica isolated from patients and carriers of two regions in Brazil (Amazonia and Southeast) were determined. The enzymes phosphoglucomutase, glucose-phosphate isomerase, hexokinase and malic enzyme were considered. IP of the strains was correlated with culture conditions, time of maintenance in laboratory and clinical history of patients. The strains were maintained under polyxenic, monoxenic and axenic culture conditions: 27 polyxenic, 1 polyxenic and monoxenic, 1 polyxenic, monoxenic and axenic and 4 axenic only. The patients were symptomatic and asymptomatic. The symptomatic patients presented either non dysenteric (NDC) or dysenteric colitis (DC), associated or not with hepatic abscess (HA). One patient presented anal amoeboma (AM). The analysis of IP for isolates maintained in polyxenic culture showed non pathogenic IP (I) for strains from carriers and patients with NDC, while the strains isolated from patients presenting DC, HA and AM resulted in isolates II or XIX pathogenic IP. This parameter was not able to differentiate strains from carriers from symptomatic patients when these strains were found in axenic or monoxenic culture. All these strains displayed pathogenic IP (II), demonstrating the inability of this parameter to classifying for virulence since it showed identical IP for strains isolated from carriers or symptomatic patients.

This paper presents the first phase of the redevelopment of the Electric Vehicle Scenario Simulator (EVeSSi) tool. A new methodology to generate traffic demand scenarios for the Simulation of Urban MObility (SUMO) tool for urban traffic simulation is described. This methodology is based on a Portugal census database to generate a synthetic population for a given area under study. A realistic case study of a Portuguese city, Vila Real, is assessed. For this area the road network was created along with a synthetic population and public transport. The traffic results were obtained and an electric buses fleet was evaluated assuming that the actual fleet would be replaced in a near future. The energy requirements to charge the electric fleet overnight were estimated in order to evaluate the impacts that it would cause in the local electricity network.

Pretende-se com este trabalho fazer um estudo detalhado de um troço da Estrada Municipal 607, situado no concelho de Valongo, caracterizando o seu estado de degradação e definindo qual ou quais as soluções mais convenientes de reabilitação a adotar. O pavimento alvo deste trabalho é abrangido por três ruas pertencentes à EM 607, Rua de Vilar, Rua de Cabeda e Rua D. Pedro IV e apresenta uma extensão total de 4,8 km. Este trabalho pode ser dividido em duas partes distintas: uma primeira parte, mais do âmbito teórico, onde é feita uma abordagem histórica desde o desenvolvimento dos pavimentos rodoviários até aos métodos e pavimentos existentes na atualidade e uma segunda parte, que diz respeito ao desenvolvimento prático do trabalho, onde é feito um estudo do pavimento e são apresentadas soluções de reabilitação e respetivos dimensionamentos. Iniciou-se o estudo do pavimento com uma análise geral das patologias existentes, definindo o estado de degradação em que este se encontra. Concluiu-se que se estava na presença de um pavimento muito degradado, havendo troços com um nível de degradação muito elevado, o que proporciona desconforto na circulação dos condutores. Foi efetuado um estudo de tráfego de pesados, onde se percebeu que se tratava de um pavimento com intenso tráfego de pesados e que talvez este pavimento não esteja apto para o suportar. A retirada de carotes veio confirmar este pressuposto, pois as espessuras das camadas betuminosas mostraram ser insuficientes para o atual volume de tráfego. As soluções de reabilitação foram ao encontro de várias soluções para diferentes situações, dando aos responsáveis da Câmara Municipal de Valongo a possibilidade de optarem por aquela que melhor se ajuste às suas necessidades, bem como às suas condições financeiras. Assim, foram propostas três soluções para três capacidades de resistência e períodos de vida úteis distintos. A primeira solução engloba a remoção do pavimento existente e colocação de uma estrutura completamente nova, com três camadas betuminosas e dimensionada para o volume de tráfego atual, apresentando um período de vida útil de 20 anos, mas que é mais dispendiosa. A segunda solução apresenta apenas duas camadas betuminosas, onde seriam removidas apenas as camadas betuminosas, aproveitando a camada de sub-base. Esta solução tem um período de vida útil estimado de cerca de 8 anos. A terceira solução, com um período de vida útil de 5 anos, baseia-se na remoção parcial de uma largura da faixa de rodagem, aproveitando a restante. Na largura removida seria colocada uma estrutura de pavimento nova e dimensionada na segunda solução. Por último, foi dimensionado o sistema de drenagem transversal e longitudinal, sendo caracterizadas as linhas de águas e as respetivas bacias hidrográficas. As valetas e passagens hidráulicas existentes não estão dimensionadas para o atual escoamento de águas pluviais a que o pavimento está sujeito. As valetas são muito reduzidas, apresentando-se em alguns casos degradadas ou a necessitarem de limpeza urgente.

The determination of the rabies neutralizing antibody (VNA) response after immunization against rabies is an acceptable index of the efficacy of a vaccine and a successful treatment. Several tests have been developed in attempt to improve the assessment of VNA, from mice inoculation to cell-culture fluorescence inhibition tests. All of them, however, present special difficulties in terms of reading or accuracy. The present study describes a neutralization test performed in cell-culture appraised by flow cytometry (FC). Serial dilutions of the serum samples were mixed in vitro with rabies virus before the addition of BHK-21 cells. After 24h-incubation, cells were released by trypsin treatment, fixed and permeabilized with a p-formaldehyde solution and stained with a rabies virus nucleocapsid protein-specific antibody conjugate. The percentage of virus infection inhibition caused by specific antibodies present in the serum were evaluated in a Beckton & Dickinson FACSCalibur® flow cytometer. A correlation curve between the IU/ml content and the percentage of infective inhibition was built with a reference serum and the VNA titers of serum samples were obtained by extrapolation. Titers obtained by FC and standard test showed an effective pairing results (p < 0.01), with a correlation coefficient (r) = 0.7. These results permit to envisage the FC as a suitable technique to evaluate VNA in sera from immunized animals and likely in human serum samples. Nevertheless, new studies comparing FC to gold-standard techniques are required for determining the FC values of Sensibility and Specificity .

A turvação é uma das principais preocupações das empresas de rede de distribuição de água potável. Para se saber a sua origem existem diversos mecanismos, sugeridos em bibliografia, que explicam a formação de depósitos de partículas nas tubagens, que se acumulam em camadas sobrepostas, criando entre si forças de coesão. A turvação pode surgir se o caudal ou a velocidade na tubagem aumentar, por alguma razão, pois um aumento da tensão de corte sobre o depósito, pode superar as forças de coesão levando à sua ressuspensão. Esta explicação é traduzida matematicamente pelo modelo PODDS (Prediction of Discolouration in Drinking Water Distribtuion System). O modelo PODDS foi compilado em EPANET recentemente por uma equipa de investigadores da Universidade de Sheffield e foi utilizado para fazer o estudo num ramal da empresa AdDP - Águas do Douro e Paiva, SA. Os resultados das simulações permitirão prever o caudal mínimo teórico a utilizar no ramal que origina o valor limite de turvação. Foram realizados ensaios de variação do caudal no ramal de Jovim-Ramalde para a calibração do modelo. Os parâmetros obtidos foram: Cmáx igual a 100 NTUm, b e n igual a 1, k é igual -1 NTUm3/N, e por fim, P igual a 0,0001 NTUm3/N.s. Após calibração do modelo realizaram-se simulações com atualização das forças de coesão para conhecer o caudal que provocaria a mobilização das partículas até uma turvação de 4 NTU e concluiu-se que o valor seria de 4,32 m3/s. O tempo necessário para regressar a valores inferiores a 1 NTU também foi determinado e 4 horas e 8 minutos foi o valor alcançado. Por fim realizou-se um estudo da influência da temperatura neste modelo e o resultado mostrou que este parâmetro é pouco influente para a gama de temperaturas considerada.

Sulfadiazine is an antibiotic of the sulfonamide group and is used as a veterinary drug in fish farming. Monitoring it in the tanks is fundamental to control the applied doses and avoid environmental dissemination. Pursuing this goal, we included a novel potentiometric design in a flow-injection assembly. The electrode body was a stainless steel needle veterinary syringe of 0.8-mm inner diameter. A selective membrane of PVC acted as a sensory surface. Its composition, the length of the electrode, and other flow variables were optimized. The best performance was obtained for sensors of 1.5-cm length and a membrane composition of 33% PVC, 66% onitrophenyloctyl ether, 1% ion exchanger, and a small amount of a cationic additive. It exhibited Nernstian slopes of 61.0 mV decade-1 down to 1.0×10-5 mol L-1, with a limit of detection of 3.1×10-6 mol L-1 in flowing media. All necessary pH/ionic strength adjustments were performed online by merging the sample plug with a buffer carrier of 4-(2-hydroxyethyl)-1-piperazineethanesulfonic acid, pH 4.9. The sensor exhibited the advantages of a fast response time (less than 15 s), long operational lifetime (60 days), and good selectivity for chloride, nitrite, acetate, tartrate, citrate, and ascorbate. The flow setup was successfully applied to the analysis of aquaculture waters. The analytical results were validated against those obtained with liquid chromatography–tandem mass spectrometry procedures. The sampling rate was about 84 samples per hour and recoveries ranged from 95.9 to 106.9%.

The local fractional Burgers’ equation (LFBE) is investigated from the point of view of local fractional conservation laws envisaging a nonlinear local fractional transport equation with a linear non-differentiable diffusion term. The local fractional derivative transformations and the LFBE conversion to a linear local fractional diffusion equation are analyzed.

Fractional Calculus (FC) goes back to the beginning of the theory of differential calculus. Nevertheless, the application of FC just emerged in the last two decades, due to the progress in the area of chaos that revealed subtle relationships with the FC concepts. In the field of dynamical systems theory some work has been carried out but the proposed models and algorithms are still in a preliminary stage of establishment. Having these ideas in mind, the paper discusses a FC perspective in the study of the dynamics and control of some distributed parameter systems.

This paper presents the most recent developments of the Simulator of Intelligent Transportation Systems (SITS). The SITS is based on a microscopic simulation approach to reproduce real traffic conditions in an urban or non-urban network. In order to analyse the quality of the microscopic traffic simulator SITS a benchmark test was performed. A dynamical analysis of several traffic phenomena, applying a new modelling formalism based on the embedding of statistics and Laplace transform, is then addressed. The paper presents also a new traffic control concept applied to a freeway traffic system.

Abstract: Preferential flow and transport through macropores affect plant water use efficiency and enhance leaching of agrochemicals and the transport of colloids, thereby increasing the risk for contamination of groundwater resources. The effects of soil compaction, expressed in terms of bulk density (BD), and organic carbon (OC) content on preferential flow and transport were investigated using 150 undisturbed soil cores sampled from 15 × 15–m grids on two field sites. Both fields had loamy textures, but one site had significantly higher OC content. Leaching experiments were conducted in each core by applying a constant irrigation rate of 10 mm h−1 with a pulse application of tritium tracer. Five percent tritium mass arrival times and apparent dispersivities were derived from each of the tracer breakthrough curves and correlated with texture, OC content, and BD to assess the spatial distribution of preferential flow and transport across the investigated fields. Soils from both fields showed strong positive correlations between BD and preferential flow. Interestingly, the relationships between BD and tracer transport characteristics were markedly different for the two fields, although the relationship between BD and macroporosity was nearly identical. The difference was likely caused by the higher contents of fines and OC at one of the fields leading to stronger aggregation, smaller matrix permeability, and a more pronounced pipe-like pore system with well-aligned macropores.

INTRODUCTION: A growing body of evidence shows the prognostic value of oxygen uptake efficiency slope (OUES), a cardiopulmonary exercise test (CPET) parameter derived from the logarithmic relationship between O(2) consumption (VO(2)) and minute ventilation (VE) in patients with chronic heart failure (CHF). OBJECTIVE: To evaluate the prognostic value of a new CPET parameter - peak oxygen uptake efficiency (POUE) - and to compare it with OUES in patients with CHF. METHODS: We prospectively studied 206 consecutive patients with stable CHF due to dilated cardiomyopathy - 153 male, aged 53.3±13.0 years, 35.4% of ischemic etiology, left ventricular ejection fraction 27.7±8.0%, 81.1% in sinus rhythm, 97.1% receiving ACE-Is or ARBs, 78.2% beta-blockers and 60.2% spironolactone - who performed a first maximal symptom-limited treadmill CPET, using the modified Bruce protocol. In 33% of patients an cardioverter-defibrillator (ICD) or cardiac resynchronization therapy device (CRT-D) was implanted during follow-up. Peak VO(2), percentage of predicted peak VO(2), VE/VCO(2) slope, OUES and POUE were analyzed. OUES was calculated using the formula VO(2) (l/min) = OUES (log(10)VE) + b. POUE was calculated as pVO(2) (l/min) / log(10)peakVE (l/min). Correlation coefficients between the studied parameters were obtained. The prognosis of each variable adjusted for age was evaluated through Cox proportional hazard models and R2 percent (R2%) and V index (V6) were used as measures of the predictive accuracy of events of each of these variables. Receiver operating characteristic (ROC) curves from logistic regression models were used to determine the cut-offs for OUES and POUE. RESULTS: pVO(2): 20.5±5.9; percentage of predicted peak VO(2): 68.6±18.2; VE/VCO(2) slope: 30.6±8.3; OUES: 1.85±0.61; POUE: 0.88±0.27. During a mean follow-up of 33.1±14.8 months, 45 (21.8%) patients died, 10 (4.9%) underwent urgent heart transplantation and in three patients (1.5%) a left ventricular assist device was implanted. All variables proved to be independent predictors of this combined event; however, VE/VCO2 slope was most strongly associated with events (HR 11.14). In this population, POUE was associated with a higher risk of events than OUES (HR 9.61 vs. 7.01), and was also a better predictor of events (R2: 28.91 vs. 22.37). CONCLUSION: POUE was more strongly associated with death, urgent heart transplantation and implantation of a left ventricular assist device and proved to be a better predictor of events than OUES. These results suggest that this new parameter can increase the prognostic value of CPET in patients with CHF.

Bipedal gaits have been classified on the basis of the group symmetry of the minimal network of identical differential equations (alias cells) required to model them. Primary bipedal gaits (e.g., walk, run) are characterized by dihedral symmetry, whereas secondary bipedal gaits (e.g., gallop-walk, gallop- run) are characterized by a lower, cyclic symmetry. This fact has been used in tests of human odometry (e.g., Turvey et al. in P Roy Soc Lond B Biol 276:4309–4314, 2009, J Exp Psychol Hum Percept Perform 38:1014–1025, 2012). Results suggest that when distance is measured and reported by gaits from the same symmetry class, primary and secondary gaits are comparable. Switching symmetry classes at report compresses (primary to secondary) or inflates (secondary to primary) measured distance, with the compression and inflation equal in magnitude. The present research (a) extends these findings from overground locomotion to treadmill locomotion and (b) assesses a dynamics of sequentially coupled measure and report phases, with relative velocity as an order parameter, or equilibrium state, and difference in symmetry class as an imperfection parameter, or detuning, of those dynamics. The results suggest that the symmetries and dynamics of distance measurement by the human odometer are the same whether the odometer is in motion relative to a stationary ground or stationary relative to a moving ground.

The objective of the thesis is to analyze the behaviour of the wind flow when it is passing beside the forest. To complete this analysis, a parametric study was done based upon generalized situations. Some abacus have been made, which are related to forest and wind characteristics. The abacus were compared with a particular real case, namely Alexandrovo (Bulgaria), where it was concluded that the applicability of the abacus in projects with complex terrain is low and they must be used, from a quantitative point of view, for flat terrain, being hc the most important parameter.
