O presente trabalho foi elaborado no âmbito da Dissertação Final de Mestrado do curso de Engenharia Mecânica – Gestão Industrial do Instituto Superior de Engenharia do Porto. Este foi realizado numa empresa da indústria de pneus, a Continental Mabor S.A.. Nos dias atuais, a indústria esta cada vez mais competitiva, os custos e os prazos da entrega são cada vez mais reduzidos e a qualidade cada vez mais exigente, assim sendo, é imprescindível uma constante melhoria do sistema produtivo. Este fato fez com que o presente trabalho tivesse como principal objetivo determinar o estado atual e delinear um plano de melhoria para um equipamento (Extrusora de pisos nº6), recentemente instalado na fábrica mas oriundo de outra fábrica do grupo, recorrendo para tal ao Lean Manufacturing e a ferramentas que lhe estão associadas. Inicialmente realizou-se uma análise e diagnóstico ao processo de extrusão de pisos na Extrusora nº6, com o intuito de avaliar todas as suas ineficiências e ainda formular um plano de melhorias para a ineficiência de maior impacto no sistema produtivo. Esta análise foi realizada em diferentes turnos e diferentes equipas de trabalho de forma a se obter uma amostra mais representativa da realidade global. Após esta análise verificou-se que as principais ineficiências eram Setup, conformidade do material, dimensões e encravamentos, entre outros. Sendo que os Setup provocam um tempo de paragem de 101 minutos por turno, escolheu-se esta perturbação como o foco do plano de melhorias a realizar posteriormente. De forma a reduzir os tempos de mudança (Setup), o autor no presente trabalho utilizou ferramentas Lean Manufacturing, principalmente o SMED. Conjuntamente com o SMED ainda foram utilizadas outras ferramentas Lean Manufacturing tais como: 5S, Gestão Visual, Problem Solving e Normalização do Método de Trabalho. Após a implementação de todas estas ferramentas obteve-se uma redução de tempos de mudança de 43% com 1 operador e 71% com 2 operadores, ou seja, reduziu-se de 40,5 minutos gastos por turno para a mudança de fieira para 23,13 min e 11,79 min respetivamente, o que corresponde a um ganho monetário anual de 63.621€ ou 105.045€, respetivamente. Com este trabalho conclui-se que a utilização de ferramentas Lean Manufacturing contribuem para a redução dos desperdícios do processo produtivo. Por isso, espera-se que este estudo seja aplicado na Extrusora nº6 e nas restantes Extrusoras de pisos existentes na fábrica, e num futuro próximo que se realize estudos semelhantes em máquinas com diferentes funções.

The thesis is divided into two parts corresponding to structural studies on two different proteins. The first part concerns the study of two UDP-glucose dehydrogenases (UGDs) from Sphingomonas elodea ATCC 31461 and Burkholderia cepacia IST 408, both involved in exopolysaccharide production. Their relevance arises because some of these bacterial exopolysaccharides are valuable as established biotechnological products, the former case, whilst others are highly problematic, when used by pathogens in biofilm formation over biological surfaces, as the latter case, namely in the human lungs. The goal of these studies is to increase our knowledge regarding UGDs structural properties, which can potentiate either the design of activity enhancers to respond to the increased demand of useful biofilms, or the design of inhibitors of biofilm production, in order to fight invading pathogens present in several infections. The thesis reports the production and crystallisation of both proteins, the determination of initial phases by single-wavelength anomalous dispersion (SAD) in S. elodea crystals using a seleno-methionine isoform, and phasing of B. cepacia crystals by molecular replacement (MR) using the S. elodea model, as well as the refinement, structural analysis and comparison between the several UGDs structures available during this work.(...)

With the emergence of low-power wireless hardware new ways of communication were needed. In order to standardize the communication between these low powered devices the Internet Engineering Task Force (IETF) released the 6LoWPAN stand- ard that acts as an additional layer for making the IPv6 link layer suitable for the lower-power and lossy networks. In the same way, IPv6 Routing Protocol for Low- Power and Lossy Networks (RPL) has been proposed by the IETF Routing Over Low power and Lossy networks (ROLL) Working Group as a standard routing protocol for IPv6 routing in low-power wireless sensor networks. The research performed in this thesis uses these technologies to implement a mobility process. Mobility management is a fundamental yet challenging area in low-power wireless networks. There are applications that require mobile nodes to exchange data with a xed infrastructure with quality-of-service guarantees. A prime example of these applications is the monitoring of patients in real-time. In these scenarios, broadcast- ing data to all access points (APs) within range may not be a valid option due to the energy consumption, data storage and complexity requirements. An alternative and e cient option is to allow mobile nodes to perform hand-o s. Hand-o mechanisms have been well studied in cellular and ad-hoc networks. However, low-power wireless networks pose a new set of challenges. On one hand, simpler radios and constrained resources ask for simpler hand-o schemes. On the other hand, the shorter coverage and higher variability of low-power links require a careful tuning of the hand-o parameters. In this work, we tackle the problem of integrating smart-HOP within a standard protocol, speci cally RPL. The simulation results in Cooja indicate that the pro- posed scheme minimizes the hand-o delay and the total network overhead. The standard RPL protocol is simply unable to provide a reliable mobility support sim- ilar to other COTS technologies. Instead, they support joining and leaving of nodes, with very low responsiveness in the existence of physical mobility.

Eighteen Cebus apella monkeys, (juvenile and adult of both sexes) were inoculated five years ago, with three Trypanosoma cruzi strains (CA1, n = 10; Colombian, n=4 and Tulahuen, n=4), either by conjunctival or intraperitoneal route, once or repeatedly. Parasitological, hematological, serological, enzymatic, radiographic, electro and echocardiographic findings have been peviously published15 and they are similar to those observed in human pathology. The most frequent electrocardiographic alteration was right branch bundle block. Six animals, chosen at random, were sacrificed. Those sacrificed 20 to 25 months post-first inoculation showed focal accumuli of leukocytes with myocytolysis. Foci of diffuse interstitial fibrosis with mild infiltrate of leukocytes among fibers were observed in the animals sacrificed 36 to 47 months post-inoculation. No parasites were seen. The lesions were more prominent in the ventricular walls and the septum. The fact that the infiltrates were predominant in the animals sacrificed at a shorter time after first inoculation and that fibrosis was more severe in those sacrificed at a longer time suggests that there is a progression of the infiltrative lesions to fibrosis, with a leukocytic activity indicative of a chronic phase. These lesions are similar to those described in human chronic Chagas' disease. This would demonstrate that this model is useful in evaluating a progress in the knowledge of the pathogenesis which is still a controversial issue, immunology, immunogenesis and chemotherapeutic agents of the chronic and indeterminate phases of this disease.

In an attempt to be as close as possible to the infected and treated patients of the endemic areas of schistosomiasis (S. mansoni) and in order to achieve a long period of follow-up, mice were repeatedly infected with a low number of cercariae. Survival data and histological variables such as schistosomal granuloma, portal changes, hepatocellular necrosis, hepatocellular regeneration, schistosomotic pigment, periductal fibrosis and chiefly bile ducts changes were analysed in the infected treated and non treated mice. Oxamniquine chemotherapy in repeatedly infected mice prolonged survival significantly when compared to non-treated animals (chi-square 9.24, p = 0.0024), thus confirming previous results with a similar experimental model but with a shorter term follow-up. Furthermore, mortality decreased rapidly after treatment suggesting an abrupt reduction in the severity of hepatic lesions. A morphological and immunohistochemical study of the liver was carried out. Portal fibrosis, with a pattern resembling human Symmers fibrosis was present at a late phase in the infected animals. Bile duct lesions were quite close to those described in human Mansonian schistosomiasis. Schistosomal antigen was observed in one isolated altered bile duct cell. The pathogenesis of the bile duct changes and its relation to the parasite infection and/or their antigens are discussed.

A double pi'npin heterostructure based on amorphous SiC has a non linear spectral gain which is a function of the signal wavelength that impinges on its front or back surface. An impulse of a configurable length and amplitude is applied to a 390 nm LED which illuminates one of the sensor surfaces, followed by a time period without any illumination after which an input signal with a different wavelength is impinged upon the front surface. Results show that the intensity and duration of the impulse illumination of the surfaces influences the sensor's response with different output for the same input signal. This paper studies this effect and proposes an application as a short term light memory. (C) 2015 Elsevier B.V. All rights reserved.

Microwave assisted synthesis of the Cu(I) compound [Cu(µ4-4-ptz)]n [1, 4-ptz = 5-(4-pyridyl)tetrazolate] has been performed by employing a relatively easy method and within a shorter period of time compared to its sister compounds. The syntheses of the Cu(II) compounds [Cu3(µ3-4-ptz)4(µ2-N3)2(DMF)2]n∙(DMF)2n (2) and [Cu(µ2-4-ptz)2(H2O)2]n (3) using a similar method were reported previously by us. MOFs 1-3 revealed high catalytic activity toward oxidation of cyclic alkanes (cyclopentane, -hexane and -octane) with aqueous hydrogen peroxide, under very mild conditions (at room temperature), without any added solvent or additive. The most efficient system (2/H2O2) showed, for the oxidation of cyclohexane, a turnover number (TON) of 396 (TOF of 40 h−1), with an overall product yield (cyclohexanol and cyclohexanone) of 40% relative to the substrate. Moreover, the heterogeneous catalytic systems 1–3 allowed an easy catalyst recovery and reuse, at least for four consecutive cycles, maintaining ca. 90% of the initial high activity and concomitant high selectivity.

We present results, obtained by means of an analytic study and a numerical simulation, about the resonant condition necessary to produce a Localized Surface Plasmonic Resonance (LSPR) effect at the surface of metal nanospheres embedded in an amorphous silicon matrix. The study is based on a Lorentz dispersive model for a-Si:H permittivity and a Drude model for the metals. Considering the absorption spectra of a-Si:H, the best choice for the metal nanoparticles appears to be aluminium, indium or magnesium. No difference has been observed when considering a-SiC:H. Finite-difference time-domain (FDTD) simulation of an Al nanosphere embedded into an amorphous silicon matrix shows an increased scattering radius and the presence of LSPR induced by the metal/semiconductor interaction under green light (560 nm) illumination. Further results include the effect of the nanoparticles shape (nano-ellipsoids) in controlling the wavelength suitable to produce LSPR. It has been shown that is possible to produce LSPR in the red part of the visible spectrum (the most critical for a-Si:H solar cells applications in terms of light absorption enhancement) with aluminium nano-ellipsoids. As an additional results we may conclude that the double Lorentz-Lorenz model for the optical functions of a-Si:H is numerically stable in 3D simulations and can be used safely in the FDTD algorithm. A further simulation study is directed to determine an optimal spatial distribution of Al nanoparticles, with variable shapes, capable to enhance light absorption in the red part of the visible spectrum, exploiting light trapping and plasmonic effects. © 2015 WILEY-VCH Verlag GmbH & Co. Hyperspectral imaging has become one of the main topics in remote sensing applications, which comprise hundreds of spectral bands at different (almost contiguous) wavelength channels over the same area generating large data volumes comprising several GBs per flight. This high spectral resolution can be used for object detection and for discriminate between different objects based on their spectral characteristics. One of the main problems involved in hyperspectral analysis is the presence of mixed pixels, which arise when the spacial resolution of the sensor is not able to separate spectrally distinct materials. Spectral unmixing is one of the most important task for hyperspectral data exploitation. However, the unmixing algorithms can be computationally very expensive, and even high power consuming, which compromises the use in applications under on-board constraints. In recent years, graphics processing units (GPUs) have evolved into highly parallel and programmable systems. Specifically, several hyperspectral imaging algorithms have shown to be able to benefit from this hardware taking advantage of the extremely high floating-point processing performance, compact size, huge memory bandwidth, and relatively low cost of these units, which make them appealing for onboard data processing. In this paper, we propose a parallel implementation of an augmented Lagragian based method for unsupervised hyperspectral linear unmixing on GPUs using CUDA. The method called simplex identification via split augmented Lagrangian (SISAL) aims to identify the endmembers of a scene, i.e., is able to unmix hyperspectral data sets in which the pure pixel assumption is violated. The efficient implementation of SISAL method presented in this work exploits the GPU architecture at low level, using shared memory and coalesced accesses to memory.
