Embora a doença de Chagas seja amplamente estudada, ainda não se conhece todos os interferentes que modulam a relação parasitária Trypanosoma cruzi- vetor- hospedeiro vertebrado. Sabe-se que as aves são refratárias ao T. cruzi, ou seja, o parasito não consegue estabelecer parasitismo nas mesmas, enquanto que nos camundongos a infecção ocorre. Essa refratariedade representa um exemplo de resistência natural do hospedeiro a uma infecção parasitária. Originalmente, acreditava-se que isso ocorria devido à presença de um anticorpo natural contra o T. cruzi no sangue das aves, porém estudos atuais atribuem tal atividade ao sistema complemento presente no sangue das mesmas. Pretendeu-se avaliar neste projeto se a referida característica peculiar do sangue das aves interfere no processo e no nível de infecção dos triatomíneos, em especial de Rhodnius neglectus, espécie de comprovada importância epidemiológica. Para tanto, utilizou-se a cepa Y de T. cruzi e ninfas de 4º estádio de R. neglectus. Avaliou-se também a capacidade lítica do soro de galinha em formas epimastigotas e tripomastigotas em meio de cultura LIT, utilizando a cepa Y. Referente à influência de diferentes fontes alimentares na infecção de R. neglectus por T. cruzi, observou-se um decréscimo no número de exemplares infectados após alimentação em patos , apresentando Índice de Infecção médio de 19,74%. Por outro lado, exemplares infectados após alimentação em camundongos não apresentaram diferença significante na parasitemia total. Os testes in vitro demonstraram que a adição de soro de galinha em meio de cultura LIT, com formas epimastigotas e tripomastigotas, foi capaz de diminuir notoriamente a contagem de parasitos móveis até a contagem zero, uma hora após a adição. O conjunto desses resultados sugere que a refratariedade das aves ao T. cruzi influencia a infecção de R. neglectus pelo flagelado e foi demonstrada, in vitro, pela ...

Introduction. American trypanosomiasis, also known as Chagas disease, is a zoonosis caused by Trypanosoma cruzi (T. cruzi). Dogs and cats participate actively in this parasite's transmission cycle. This study aimed at evaluating the occurrence of T. cruzi in dogs and cats from Botucatu, SP, Brazil, as well as at evaluating the technique of hemoculture in LIT (liver infusion tryptose) medium by polymerase chain reaction (PCR). Methods. Blood samples were collected from 50 dogs and 50 cats in Botucatu-SP, Brazil. For hemoculture, the samples were inoculated in LIT medium, and readings were performed for four months. Upon completion of such period, all the hemocultures were processed for parasitic DNA extraction. The PCR reactions were performed by using primers TCZ1/TCZ2. Results. Ten dogs and ten cats (20%) were positive to PCR, and four dogs and three cats (7%) were positive to hemoculture. Only in a one cat sample (1%) there was confirmation of positive hemoculture by PCR for T. cruzi. Conclusions. Results showed that PCR was a suitable tool for the confirmation of the parasite detection in hemoculture samples, and that dogs and cats from Botucatu, SP, Brazil, are maintaining the role of household reservoirs of T. cruzi, which reinforces the need for constant epidemiologic surveillance for this zoonosis.

Ischaemia modified albumin (IMA) is considered a biomarker of myocardial ischaemia in humans in contrast to other biomarkers released when cardiac necrosis occurs. Little is known about release conditions of IMA in exercise and this is the first report in equine species. For this purpose, ten clinically healthy untrained horses were submitted to a high intensity test (HIT) followed by a low intensity test (LIT) seven days later. Blood samples were taken before, during and immediately after exercise, and 15 min and 30 min thereafter. Serum IMA, lactate and albumin, and plasma malondialdehyde (MDA) were determined. There were no significant changes in IMA concentration in any of the exercise tests. There was also a negative correlation between IMA and albumin levels in both tests, and between IMA and lactate levels in LIT, suggesting possible assay interferences. It was concluded that HIT and LIT did not promote significant changes in IMA concentration in horses under these conditions.

A doença de Chagas, causada pelo protozoário flagelado Trypanosoma cruzi, foi descrita pelo pesquisador brasileiro Carlos Chagas em 1909. É transmitida ao homem por insetos hemípteros conhecidos como barbeiros dos quais os gêneros mais importantes são Panstrongylus, Rhodnius e Triatoma. Essa zoonose representa um risco para aproximadamente 20 milhões de pessoas em todo o mundo, principalmente na América Latina. Para tentar explicar as diferentes manifestações observadas na doença de Chagas, vários estudos foram realizados com intuito de averiguar as possíveis correlações entre as formas clínicas com a variabilidade genética do parasito. Algumas hipóteses estão relacionadas provavelmente ao fato de a doença ser um processo multifatorial, em que tanto aspectos do parasito como do hospedeiro estão inter-relacionados ou ainda a escolha inadequada de alvos como marcadores de patogenicidade na tentativa de estabelecer a correlação entre as formas clínicas e a variabilidade genética do parasito. Com o intuito de contribuir para ampliar o conhecimento sobre as populações de T. cruzi, foi realizada a cinética de crescimento em meio LIT e o estudo genotípico de seis cepas de T. cruzi isoladas de exemplares de R. montenegrensis, T. rubrovaria e T. sordida por meio de marcadores genotípicos utilizando as sequência dos genes 24Sα do DNA ribossomal, HSP60 e GPI.

OBJECTIVE: To investigate a possible direct, growth hormone-releasing, hormone-independent action of a growth hormone secretagogue, GHRP-2, in pituitary somatotroph cells in the presence of inactive growth hormone-releasing hormone receptors. MATERIALS AND METHODS: The responses of serum growth hormone to acutely injected growth hormone-releasing P-2 in lit/litmice, which represent a model of GH deficiency arising frommutated growth hormone-releasing hormone-receptors, were compared to those observed in the heterozygous (lit/+) littermates and wild-type (+/+) C57BL/6J mice. RESULTS: After the administration of 10 mcg of growth hormone-releasing P-2 to lit/lit mice, a growth hormone release of 9.3 +/- 1.5 ng/ml was observed compared with 1.04 +/- 1.15 ng/ml in controls (p<0.001). In comparison, an intermediate growth hormone release of 34.5 +/- 9.7 ng/ml and a higher growth hormone release of 163 +/- 46 ng/ml were induced in the lit/+ mice and wild-type mice, respectively. Thus, GHRP-2 stimulated growth hormone in the lit/lit mice, and the release of growth hormone in vivo may be only partially dependent on growth hormone-releasing hormone. Additionally, the plasma leptin and ghrelin levels were evaluated in the lit/lit mice under basal and stimulated conditions. CONCLUSIONS: Here, we have demonstrated that lit/lit mice, which harbor a germline mutation in the Growth hormone-releasing hormone gene, maintain a limited but statistically significant growth hormone elevation after exogenous stimulation with GHRP-2. The present data probably reflect a direct, growth hormone-independent effect on Growth hormone S (ghrelin) stimulation in the remaining pituitary somatotrophs of little mice that is mediated by growth hormone S-R 1a.

The applicability of laboratory bioassays to diagnose ivermectin (IVM) resistance in Rhipicephalus microplus was evaluated. Adult immersion tests (AITs), larval immersion tests (LITs) and larval packet tests (LPTs) were performed to characterise the effects of ivermectin toxicity on adults and larvae of a susceptible reference strain. The AIT was determined to be a reasonable assay but requires a large number of individuals to attain interpretable results. The LIT and LPT were validated with an IVM resistant strain, revealing resistance ratios (RRs) of 6.73 and 1.49, respectively. In a field survey, nine different populations of cattle tick from the states of Sao Paulo and Mato Grosso do Sul, Brazil, were analysed with the LIT. Populations without previous exposure to ivermectin exhibited RRs between 0.87 and 1.01. Populations previously exposed to IVM showed RRs between 1.83 and 4.62. The LIT was more effective at discriminating between resistant and susceptible populations than the LPT. The use of the LIT is recommended for the diagnosis of ivermectin resistance in R microplus. The recently described genus Philcoxia comprises three species restricted to well lit and low-nutrient soils in the Brazilian Cerrado. The morphological and habitat similarities of Philcoxia to those of some carnivorous plants, along with recent observations of nematodes over its subterranean leaves, prompted the suggestion that the genus is carnivorous. Here we report compelling evidence of carnivory in Philcoxia of the Plantaginaceae, a family in which no carnivorous members are otherwise known. We also document both a unique capturing strategy for carnivorous plants and a case of a plant that traps and digests nematodes with underground adhesive leaves. Our findings illustrate how much can still be discovered about the origin, distribution, and frequency of the carnivorous syndrome in angiosperms and, more generally, about the diversity of nutrient-acquisition mechanisms that have evolved in plants growing in severely nutrient-impoverished environments such as the Brazilian Cerrado, one of the world's 34 biodiversity hotspots.

In dieser Arbeit aus dem Bereich der Wenig-Nukleonen-Physik wird die neu entwickelte Methode der Lorentz Integral Transformation (LIT) auf die Untersuchung von Kernphotoabsorption und Elektronenstreuung an leichten Kernen angewendet. Die LIT-Methode ermoeglicht exakte Rechnungen durchzufuehren, ohne explizite Bestimmung der Endzustaende im Kontinuum. Das Problem wird auf die Loesung einer bindungzustandsaehnlichen Gleichung reduziert, bei der die Endzustandswechselwirkung vollstaendig beruecksichtigt wird. Die Loesung der LIT-Gleichung wird mit Hilfe einer Entwicklung nach hypersphaerischen harmonischen Funktionen durchgefuehrt, deren Konvergenz durch Anwendung einer effektiven Wechselwirkung im Rahmem des hypersphaerischen Formalismus (EIHH) beschleunigt wird. In dieser Arbeit wird die erste mikroskopische Berechnung des totalen Wirkungsquerschnittes fuer Photoabsorption unterhalb der Pionproduktionsschwelle an 6Li, 6He und 7Li vorgestellt. Die Rechnungen werden mit zentralen semirealistischen NN-Wechselwirkungen durchgefuehrt, die die Tensor Kraft teilweise simulieren, da die Bindungsenergien von Deuteron und von Drei-Teilchen-Kernen richtig reproduziert werden. Der Wirkungsquerschnitt fur Photoabsorption an 6Li zeigt nur eine Dipol-Riesenresonanz, waehrend 6He zwei unterschiedliche Piks aufweist, die dem Aufbruch vom Halo und vom Alpha-Core entsprechen. Der Vergleich mit experimentellen Daten zeigt, dass die Addition einer P-Wellen-Wechselwirkung die Uebereinstimmung wesentlich verbessert. Bei 7Li wird nur eine Dipol-Riesenresonanz gefunden, die gut mit den verfuegbaren experimentellen Daten uebereinstimmt. Bezueglich der Elektronenstreuung wird die Berechnung der longitudinalen und transversalen Antwortfunktionen von 4He im quasi-elastischen Bereich fuer mittlere Werte des Impulsuebertrages dargestellt. Fuer die Ladungs- und Stromoperatoren wird ein nichtrelativistisches Modell verwendet. Die Rechnungen sind mit semirealistischen Wechselwirkungen durchgefuert und ein eichinvarianter Strom wird durch die Einfuehrung eines Mesonaustauschstroms gewonnen. Die Wirkung des Zweiteilchenstroms auf die transversalen Antwortfunktionen wird untersucht. Vorlaeufige Ergebnisse werden gezeigt und mit den verfuegbaren experimentellen Daten verglichen.

L’un des buts de ce mémoire est de définir le concept de « mise en abyme » appliqué au court-métrage Mouettes, réalisé en 2013 par le cinéaste belge Zeno Graton. Un autre but est celui de présenter une version sous-titrée en langue italienne de ce même court-métrage tout en explorant le monde de la traduction audiovisuelle et notamment du sous-titrage. Ce mémoire est divisé en trois chapitres. Le premier chapitre porte principalement sur la mise en abyme, sur les différents types de mise en abyme, à savoir pictural, littéraire et filmique, sur les études réalisées et les théories conçues à propos de ce procédé, à partir d’André Gide jusqu’à Lucien Dällenbach en passant par Jean Ricardou. Ce chapitre se penche également sur les différences entre la mise en abyme filmique et d’autres modes de réflexivité cinématographique, pour ensuite se concentrer sur le court-métrage Mouettes, décrire le procédé de la mise en abyme hétérogène utilisée dans Mouettes et présenter les thèmes abordés dans le court-métrage. Parmi les thèmes exposés, l’un des plus évocateurs est celui de la figure de l’acteur qui ne sait plus distinguer, parfois, ses passions et ses sentiments réels de ceux ressentis dans le rôle du personnage qu’il doit jouer. Ensuite, le deuxième chapitre est consacré aux différents procédés utilisés dans la traduction audiovisuelle, du doublage, au voice-over au respeaking, en analysant surtout la technique du sous-titrage, la théorie et les problèmes liés. Enfin, le dernier chapitre propose mon travail de sous-titrage à travers un tableau avec le time code, le texte de départ transcrit, la traduction et, si nécessaire, l’adaptation. Dans le troisième chapitre, il y a également un commentaire sur les difficultés rencontrées pendant le travail de sous-titrage, dans la traduction ainsi que dans l’utilisation du logiciel Jubler.
