The objective of this work was to evaluate the root system distribution and the yield of 'Conilon' coffee (Coffea canephora) propagated by seeds or cuttings. The experiment was carried out with 2x1 m spacing, in an Oxisol with sandy clay loam texture. A randomized complete block design was used, following a 2x9x6 factorial arrangement, with two propagation methods (seeds and cuttings), nine sampling spacings (0.15, 0.30, 0.45, 0.60, 0.75, and 0.90 m between rows, and 0.15, 0.30, and 0.45 between plants within rows), six soil depths (0.10-0.20, 0.20-0.30, 0.30-0.40, 0.40-0.50, and 0.50-0.60 m), and six replicates. Soil cores (27 cm3) with roots were taken from 12 experimental units, 146 months after planting. The surface area of the root system and root diameter, length, and volume were assessed for 13 years and, then, correlated with grain yield. The highest fine root concentration occurred at the superficial soil layers. The variables used to characterize the root system did not differ between propagation methods. Moreover, no differences were observed for net photosynthetic CO2 assimilation rate, stomatal conductance, internal CO2 concentrations, and instantaneous water-use efficiency in the leaves. Cutting-propagated plants were more productive than seed-propagated ones.

PURPOSE: To investigate the mechanism(s) of resistance to the RAF-inhibitor vemurafenib, we conducted a comprehensive analysis of the genetic alterations occurring in metastatic lesions from a patient with a BRAF(V600E)-mutant cutaneous melanoma who, after a first response, underwent subsequent rechallenge with this drug. EXPERIMENTAL DESIGN: We obtained blood and tissue samples from a patient diagnosed with a BRAF(V600E)-mutant cutaneous melanoma that was treated with vemurafenib and achieved a near-complete response. At progression, he received additional lines of chemo/immunotherapy and was successfully rechallenged with vemurafenib. Exome and RNA sequencing were conducted on a pretreatment tumor and two subcutaneous resistant metastases, one that was present at baseline and previously responded to vemurafenib (PV1) and one that occurred de novo after reintroduction of the drug (PV2). A culture established from PV1 was also analyzed. RESULTS: We identified two NRAS-activating somatic mutations, Q61R and Q61K, affecting two main subpopulations in the metastasis PV1 and a BRAF alternative splicing, involving exons 4-10, in the metastasis PV2. These alterations, known to confer resistance to RAF inhibitors, were tumor-specific, mutually exclusive, and were not detected in pretreatment tumor samples. In addition, the oncogenic PIK3CA(H1047R) mutation was detected in a subpopulation of PV1, but this mutation did not seem to play a major role in vemurafenib resistance in this metastasis. CONCLUSIONS: This work describes the coexistence within the same patient of different molecular mechanisms of resistance to vemurafenib affecting different metastatic sites. The objective of this work was to evaluate the morphological diversity of oil palm seeds and to cluster the accessions according to their morphological characteristics. Forty-one accessions from the oil palm germplasm bank of Embrapa Amazônia Ocidental were evaluated - 18 of Elaeis oleifera and 23 of E. guineensis. The groups were formed based on morphological characteristics, by principal component analysis. In E. oleifera, four groups were formed, tied to their region of origin, but with significant morphological differences between accessions from the same population. For tenera-type E. guineensis seeds, three widely divergent groups were formed, especially as to external parameters, which differentiated them from the other ones. The parameter endocarp thickness stood out in intra- and inter-population differentiation. For dura-type E. guineensis, three groups were formed, with larger seeds and thicker endocarps, which differed from all the other ones. The variability observed for seed characteristics in the analyzed accessions allows the establishment of different groups, to define strategies for genetic improvement.

In an open trial 11 in-patients with a major depressive episode (ICD-10), extensive metabolizers of mephenytoin (CYP2C19) and dextromethorphan (CYP2D6) and who were non-responders to a 3-wk pretreatment with 40 mg/d citalopram (Cit), were co-medicated for 7 wk (days 0-49) with fluoxetine (Fluox) (10 mg/d). Plasma concentrations of S-Cit and R-Cit significantly increased from day 0 (means+/-S.D.: 28+/-9 and 47+/-11 &mgr;g/l, respectively) to day 49 (58+/-12 and 72+/-21 &mgr;g/l, respectively) (p & 0.01 for each comparison), and the S-Cit/R-Cit ratio increased from 0.61+/-0.16 to 0.82+/-0.12 (p & 0.01). Therefore, Fluox increases the pharmacologically more active S-Cit (in comparison with R-Cit) with some stereoselectivity, most probably by inhibition of CYP2D6 and CYP3A4. Eight of the 11 patients showed clinical improvement (reduction > 50% of the MADRS score) and the combined treatment was generally well tolerated.

In this study, we used faecal analysis to determine the diet of the alpine mouse Apodemus alpicola in the field for the first time. This species consumes mainly insects in spring and fruits and seeds of small herbaceous dicotyledons in summer and autumn. Compared to the two congeneric species A. flavicollis and A. sylvaticus, which also occur in the Alps, the diet of A. alpicola is rich in contrasts, with a very pronounced difference between spring and summer. The absence of tree seeds also suggests a mostly terrestrial behaviour. We explain these facts by the specificity of the habitat of A. alpicola: the extreme weather conditions produce great seasonal changes in the vegetation, and the rocky ground favours the adaptation to rock climbing rather than to arboreal Living. The canistel is a fruit originated in Mexico and Central America, being introduced in Brazil in 1986. The plants present medium size, however it can reach up to 15 meters of height; the leaves measure about 10 to 25 cm; the flowers are complete and small and the fruit presents yellow coloration when ripe, with whitish pulp and sweet flavor. The propagation can be realized by seed or grafting. In view of almost total absence of information about the culture and the possibility to have a commercial cultivation, the present work, was live in which the effect of the temperature was evaluated in the percentage of germination of the seeds. It was checked that the best averages were obtained in temperature of 30ºC and the minor in 15ºC, 20ºC and 40ºC could be considerate the worst of them.

Continuous positive airway pressure, aimed at preventing pulmonary atelectasis, has been used for decades to reduce lung injury in critically ill patients. In neonatal practice, it is increasingly used worldwide as a primary form of respiratory support due to its low cost and because it reduces the need for endotracheal intubation and conventional mechanical ventilation. We studied the anesthetized in vivo rat and determined the optimal circuit design for delivery of continuous positive airway pressure. We investigated the effects of continuous positive airway pressure following lipopolysaccharide administration in the anesthetized rat. Whereas neither continuous positive airway pressure nor lipopolysaccharide alone caused lung injury, continuous positive airway pressure applied following intravenous lipopolysaccharide resulted in increased microvascular permeability, elevated cytokine protein and mRNA production, and impaired static compliance. A dose-response relationship was demonstrated whereby higher levels of continuous positive airway pressure (up to 6 cmH(2)O) caused greater lung injury. Lung injury was attenuated by pretreatment with dexamethasone. These data demonstrate that despite optimal circuit design, continuous positive airway pressure causes significant lung injury (proportional to the airway pressure) in the setting of circulating lipopolysaccharide. Although we would currently avoid direct extrapolation of these findings to clinical practice, we believe that in the context of increasing clinical use, these data are grounds for concern and warrant further investigation.

Spondias mombin L. shoot cultures were initiated from nodal explants taken from plants propagated by seeds. Explants coming from 4-6 months old plants, previously disinfected, were cultivated on WPM medium supplemented with a wide range of concentrations of BAP (0.0, 0.22, 0.44, 2.22 and 4.44 muM) and NAA (0.0, 0.27 and 2.70 muM). After four weeks, the responses obtained were axillary shoot and root formation. The first response were preferentially induced with the medium containing only BAP, regardless of the BAP concentration. The addition of NAA on medium reduced significantly axillary shoot formation and induced rhizogenesis. Roots were formed on nodal explant basis, preferentially on medium supplemented with 4.44 muM NAA. The medium supplemented with BAP reduced significantly root formation.

In this thesis fouling of conventional filter fabrics and membranes was studied. In the beginning of the thesis fouling and how it can be measured and predicted is reviewed. Information on different methods on how fouling canbe decreased or cleaned away is also given. The experimental part is divided into two sections; fabric filtration and membrane filtration. Fouling of the filter fabrics was studied with silica or cupper slurries and fouling of the membranes was studied with pulp and paper mill waters. The fouled filter materials were characterised according to many different methods. The most useful way to observe fouling is to measure the changes in the permeate flux. Fouling can also be seen visually e.g. with scanning electron microscopy. Consequently, also the reason for the fouling in question might be found. Different filtration characteristics affect fouling e.g. as the filtration pressure was increased it did not have much influence on the permeate flux, but the pressure caused the membrane to get fouled faster. Also, an increase of shear rate on the membrane surface decreased fouling. Different pretreatment methods for the effluent were tested in membrane filtration to decrease fouling. The tested methods; biological treatment, ozonation, enzymatic treatment and flocculation, did not have a clear influence on the fouling of the membrane, but e.g. a biological treatment combined with ultrafiltration made the tested groundwood mill circulation water purer than ultrafiltration alone.

Field poppy, Papaver rhoeas L., is a very common weed in winter cereals in North-Eastern Spain. Its control is becoming difficult due to expanding herbicide resistance. To control field poppies there are alternative strategies such as non-chemical control that take into account the weed emergence period. However, there is a lack of knowledge of P. rhoeas emergence patterns in semi-arid conditions. Thus, here we conducted pot experiments on the emergence of P. rhoeas. We aimed to describe the emergence period and to quantify the emergence of a susceptible and of a herbicide-resistant P. rhoeas population at two locations in Catalonia, Spain, from 1998 to 2001 and until 2004 at one of them. Therefore, pots containing seeds of both populations were established at the two locations and emergence was recorded monthly. We studied the origin of the population, the sowing location, the effect of cultivation and the sowing year. First, we found that the main emergence peaks in our experiments occurred in autumn, accounting for between 65.7 and 98.5% of the annual emergence from October to December, and only little emergence was recorded in spring. This emergence pattern is different from those found in the literature corresponding to Northern European countries, where in some cases main flushes occur only in autumn, in spring and winter or only in spring. The emergence was mainly affected by cultivation, but the effect of light stimulus was observed several months later. As a consequence, cultivation should be done in early autumn, promoting emergence during the whole autumn and winter so that emerged seedlings can be controlled before sowing a spring crop. Second, most experiments showed that the emergence was significantly higher in the first autumn than in the following seasons, e.g. 4.1% emergence in the first year and only 2.1, 2.3, 0.5 and 0.6% new emergence at one of the locations for the second, third, fourth and fifth years. Thus, after having a severe P. rhoeas infestation causing a big seed rain, emergence should be stimulated by autumn cultivation in the following season and seedlings controlled by trying to deplete the soil seed bank as much as possible. Despite the fact that emergence will be staggered throughout several years and that there was a significant relationship between rainfall and emergence, so that dry years will cause a smaller emergence rate of the weed, these findings define a cultural management strategy to reduce P. rhoeas infestations and to contribute to integrated weed management strategies combining it with other tools.

The experiment was carried out at the Embrapa Semi-Árido, Petrolina-PE, Brazil, in order to study the physiological responses of umbu plants propagated by seeds and by stem cuttings under water stress conditions, based on leaf water potential and gas exchange measurements. Data were collected in one-year plants established in pots containing 30 kg of a sandy soil and submitted to twenty-day progressive soil water deficit. The evaluations were based on leaf water potential and gas exchange data collection using psychrometric chambers and a portable infra-red gas analyzer, respectively. Plants propagated by seeds maintained a significantly higher water potential, stomatal conductance, transpiration and photosynthesis under decreasing soil water availability. However, plants propagated by stem cuttings were unable to maintain a favorable internal water balance, reflecting negatively on stomatal conductance and leaf gas exchange. This fact is probably because umbu plants propagated by stem cuttings are not prone to formation of root tubers which are reservoirs for water and solutes. Thus, the establishing of umbu plants propagated by stem cuttings must be avoided in areas subjected to soil water deficit.

Apparently, there are no custard apple cultivars defined for the northeastern region of Brazil. The establishment of breeding programs aimed at the selection of types from productive locations for later cloning is desirable. This work's objective was to evaluate the yield (during the first three crops) and quality (first crop) of fruits from 20 half-sibling custard apple tree progenies, selected from home orchards. An additional objective was to estimate genetic parameters for the traits evaluated. A micro sprinkling-irrigated experiment was conducted in Mossoró-RN, Brazil, as random blocks with five replications. In characteristics evaluated for periods longer than a year (diameter, height and mean weight of fruits, number of fruits ha-1 and fruit yield (kg ha-1), and a split-plot design was adopted, with progenies considered as plots and annual cropping seasons as subplots. The best progenies in terms of fruit yield (A3 and A4) are not necessarily the best for fruit dimensions and fruit mean weight (A2, FE4, JG1, JG2, SM1, SM7, and SM8). These progenies show great potential to be used in future studies on crosses or on vegetative propagation. In this regard, progeny JG2 should be highlighted as promising in terms of yield and fruit size. The progenies are not different with regard to percentages (in relation to mean fruit mass) of pericarp, endocarp, seeds, and receptacle, in the fruit, and fruit volume, number of seeds/fruit, and total soluble solids content in the fruit pulp, but progeny FE4 presents higher total titratable acidity in the fruit pulp. Narrow-sense heritability estimates were relatively high for all characteristics in which there was variability between progenies, with higher values for number of fruits ha-1 (80 %) and fruit yield (78 %). Relatively high coefficients of genotypic variation (around 20%) were observed for number of fruits ha-1 and fruit yield, with lower values for the other characteristics. There were positive genotypic and phenotypic correlations between fruit diameter (FD) and fruit height, FD and mean fruit weight, and number of fruits ha-1 and fruit yield.

The aim of this work was to determine the sugar, organic acid and mineral compositions of the whole fruit and fractions (skin, pulp and seed) of the Paulista (Plinia cauliflora) and Sabará (Plinia jaboticaba) jabuticaba tree genotypes, as well as the oil compositions of their skin and seeds. High levels of sugar, especially fructose, followed by glucose and sucrose, were encountered in the fruit. In the Paulista genotype, higher levels of total and reducing sugars were found in the pulp and skin, which was not observed when comparing the whole fruit of both genotypes. Five organic acids were found in the whole fruit and in the fractions of the two jabuticaba genotypes in quantitative order: citric acid > succinic acid > malic acid > oxalic acid > acetic acid. Potassium was the most abundant mineral found. This fruit was also shown to be rich in magnesium, phosphorus, calcium and copper. The seed oil had nearly the same constitution as the oil extracted from the skin in both genotypes and the major compounds were an unidentified phytosterol, palmitic, linoleic and oleic acids, and squalene.

Different solvents were evaluated for the extraction of jabuticaba anthocyanin pigments, identifying, quantifying and verifying the stability of the anthocyanins, as well as the conduction of three antioxidant activity assays and determination of the vitamin C levels. The maceration with ethanol acidified with HCl 1.5 mol L-1 (85:15) provides better pigment extraction and stability. The skin is anthocyanin rich, presenting 1.59 and 2.06 g 100 g-1 of dry matter in the Paulista and Sabará varieties, respectively. Cyanidin 3-glucoside is the majority pigment of the skins, followed by delphinidin 3-glucoside. The highest level of vitamin C was found in the skins and seeds of both varieties. It was verified that the skins, presented more antioxidant activity, in free radical capture, as well as in retarding the lipid oxidation process.

The citrus nursery tree is produced through the bud grafting process, in which rootstock is usually grown from seed germination. The objective of this research was to evaluate, in two dissimilar environmental conditions, the viability and polyembryony expression of five citrus rootstocks seeds stored in different periods under refrigeration. The rootstock varieties evaluated were: Rangpur lime (Citrus limonia Osb. cv. Limeira), Trifoliate orange (Poncirus trifoliata Raf. cv. Limeira), Citrumelo (P. trifoliata x C. paradisi Macf. cv. Swingle), Sunki mandarin (C. sunki Hort. ex Tanaka) and Volkamer lemon (C. volkameriana Ten. & Pasq. cv. Catania 2). The experimental design was the randomized blocks in a 11 x 5 x 2 factorial scheme, evaluating from time zero to the tenth month of storage, the five varieties of rootstock in two environments: germination and growth B.O.D type chamber (Biological Oxygen Demand - Eletrolab Brand Model FC 122) at 25 °C; and greenhouse seedbed with partial temperature control (22 °C to 36 °C) and humidity control (75-85%). The plot had 24 seeds in four replicates, using trays with substrate in greenhouse and Petri dishes with filter paper in B.O.D. chamber. The seed germination rate and polyembryony expression were evaluated monthly. It was concluded that Trifoliate and Citrumelo Swingle seeds can be stored for up to seven months, while Volkamer lemon, Rangpur lime and Sunki seeds can be stored for up to ten months. The polyembryony expression rate was slightly higher when measured in greenhouse than in B.O.D. chamber and remained stable in both environments until the seventh month, from which dropped sharply. Citrumelo Swingle seeds expressed the highest polyembryony rate (18.8%), followed by Rangpur lime and Volkamer lemon (average value of 13.7%), Sunki (9.4%) and Trifoliate (3.2%). Despite some differences among varieties, the viability of rootstock stored seeds can be monitored either in the greenhouse or in B.O.D. germination chamber, the latter being the faster and more suitable method.
