O objetivo deste trabalho foi avaliar o perfil de ácidos graxos da carne de novilhos mestiços alimentados, em confinamento, com dietas contendo níveis crescentes de grão de milheto moído em substituição ao grão de milho moído. Foram utilizados 24 tourinhos mestiços europeus e 24 mestiços zebuínos, abatidos aos 24 meses de idade, após 96 dias de confinamento. Utilizou-se o delineamento experimental inteiramente casualizado, com os tratamentos em arranjo fatorial 4x2 (quatro dietas com 0, 33, 66 e 100% de milheto no concetrado e dois grupos genéticos) com seis repetições. O aumento dos níveis de grão de milheto na dieta elevou linearmente a participação dos ácidos graxos, araquídico (C20:0), heneicosanoico (C21:0), α-linolênico (C18:3 n-3) e dihomo-γ-linolênico (C20:3 n-6). Tourinhos europeus apresentaram carne com menor teor dos ácidos graxos, mirístico (C14:0), heneicosanoico (C21:0) e γ-linolênico (C18:3 n-6) do que tourinhos zebuínos. A concentração total de ácidos graxos saturados (45,2%), monoinsaturados (41,2%) e poli-insaturados (8,7%), e a relação monoinsaturados/saturados (1,09) e poli-insaturados/saturados (0,18) não foi alterada pelos grupos genéticos e pelas dietas. O aumento da percentagem de grão de milheto na dieta de tourinhos europeus ou zebuínos melhora a relação entre os ácidos graxos ω-6/ω-3.

BACKGROUND: Neurospheres (NS) are colonies of neural stem and precursor cells capable of differentiating into the central nervous system (CNS) cell lineages upon appropriate culture conditions: neurons, and glial cells. NS were originally derived from the embryonic and adult mouse striatum subventricular zone. More recently, experimental evidence substantiated the isolation of NS from almost any region of the CNS, including the hypothalamus. METHODOLOGY/FINDINGS: Here we report a protocol that enables to generate large quantities of NS from both fetal and adult rat hypothalami. We found that either FGF-2 or EGF were capable of inducing NS formation from fetal hypothalamic cultures, but that only FGF-2 is effective in the adult cultures. The hypothalamic-derived NS are capable of differentiating into neurons and glial cells and most notably, as demonstrated by immunocytochemical detection with a specific anti-GnRH antibody, the fetal cultures contain cells that exhibit a GnRH phenotype upon differentiation. CONCLUSIONS/SIGNIFICANCE: This in vitro model should be useful to study the molecular mechanisms involved in GnRH neuronal differentiation.

O objetivo deste trabalho foi avaliar o efeito de dietas com diferentes concentrações de fósforo disponível sobre o perfil metabólico e o peso corporal de leitões em fase inicial de crescimento, mantidos em ambientes térmicos distintos. Foram utilizados 120 leitões machos castrados, de linhagem comercial e com peso corporal médio de 15 kg. Os tratamentos consistiram de dietas com 0,107, 0,214, 0,321, 0,428 e 0,535% de fósforo disponível, e de dois ambientes: termoneutro e quente. O peso corporal foi determinado no início e no final da fase experimental, e amostras de sangue foram coletadas para a medição de eletrólitos, albumina, proteína total, substâncias nitrogenadas, fosfatase alcalina total e óssea, hormônios tireoidianos, glicose e lactato. Tanto as dietas quanto os ambientes produziram efeito sobre o ganho de peso corporal e sobre os valores de sódio, potássio, cálcio total, magnésio, fósforo, fosfatase alcalina total e óssea, T4 livre e lactato. O aumento nos teores de fósforo na dieta tem efeito positivo sobre o peso corporal, nos ambientes estudados, mas desequilibra as relações sanguíneas cálcio:fósforo. O estresse por calor diminui as concentrações de glicose, fosfatase alcalina total e T4 livre no sangue, e aumenta as de lactato. Parâmetros bioquímicos sanguíneos retratam o estado metabólico dos animais e podem ser usados para avaliação do balanço nutricional e para detecção precoce de alterações metabólicas.

In vivo fetal magnetic resonance imaging provides aunique approach for the study of early human braindevelopment [1]. In utero cerebral morphometry couldpotentially be used as a marker of the cerebralmaturation and help to distinguish between normal andabnormal development in ambiguous situations. However,this quantitative approach is a major challenge becauseof the movement of the fetus inside the amniotic cavity,the poor spatial resolution provided by very fast MRIsequences and the partial volume effect. Extensiveefforts are made to deal with the reconstruction ofhigh-resolution 3D fetal volumes based on severalacquisitions with lower resolution [2,3,4]. Frameworkswere developed for the segmentation of specific regionsof the fetal brain such as posterior fossa, brainstem orgerminal matrix [5,6], or for the entire brain tissue[7,8], applying the Expectation-Maximization MarkovRandom Field (EM-MRF) framework. However, many of theseprevious works focused on the young fetus (i.e. before 24weeks) and use anatomical atlas priors to segment thedifferent tissue or regions. As most of the gyraldevelopment takes place after the 24th week, acomprehensive and clinically meaningful study of thefetal brain should not dismiss the third trimester ofgestation. To cope with the rapidly changing appearanceof the developing brain, some authors proposed a dynamicatlas [8]. To our opinion, this approach however faces arisk of circularity: each brain will be analyzed /deformed using the template of its biological age,potentially biasing the effective developmental delay.Here, we expand our previous work [9] to proposepost-processing pipeline without prior that allow acomprehensive set of morphometric measurement devoted toclinical application. Data set & Methods: Prenatal MRimaging was performed with a 1-T system (GE MedicalSystems, Milwaukee) using single shot fast spin echo(ssFSE) sequences (TR 7000 ms, TE 180 ms, FOV 40 x 40 cm,slice thickness 5.4mm, in plane spatial resolution1.09mm). For each fetus, 6 axial volumes shifted by 1 mmwere acquired under motherâeuro?s sedation (about 1min pervolume). First, each volume is segmentedsemi-automatically using region-growing algorithms toextract fetal brain from surrounding maternal tissues.Inhomogeneity intensity correction [10] and linearintensity normalization are then performed. Brain tissues(CSF, GM and WM) are then segmented based on thelow-resolution volumes as presented in [9]. Ahigh-resolution image with isotropic voxel size of 1.09mm is created as proposed in [2] and using B-splines forthe scattered data interpolation [11]. Basal gangliasegmentation is performed using a levet setimplementation on the high-resolution volume [12]. Theresulting white matter image is then binarized and givenas an input in FreeSurfer software(http://surfer.nmr.mgh.harvard.edu) to providetopologically accurate three-dimensional reconstructionsof the fetal brain according to the local intensitygradient. Ultrasound scans in the mid trimester of pregnancy are now a routine part of antenatal care in most European countries. With the assistance of Registries of Congenital Anomalies a study was undertaken in Europe. The objective of the study was to evaluate prenatal detection of congenital heart defects (CHD) by routine ultrasonographic examination of the fetus. All congenital malformations suspected prenatally and all congenital malformations, including chromosome anomalies, confirmed at birth were identified from the Congenital Malformation Registers, including 20 registers from the following European countries: Austria, Croatia, Denmark, France, Germany, Italy, Lithuania, Spain, Switzerland, The Netherlands, UK and Ukrainia. These registries follow the same methodology. The study period was 1996-1998, 709 030 births were covered, and 8126 cases with congenital malformations were registered. If more than one cardiac malformation was present the case was coded as complex cardiac malformation. CHD were subdivided into 'isolated' when only a cardiac malformation was present and 'associated' when at least one other major extra cardiac malformation was present. The associated CHD were subdivided into chromosomal, syndromic non-chromosomal and multiple. The study comprised 761 associated CHD including 282 cases with multiple malformations, 375 cases with chromosomal anomalies and 104 cases with non-chromosomal syndromes. The proportion of prenatal diagnosis of associated CHD varied in relation to the ultrasound screening policies from 17.9% in countries without routine screening (The Netherlands and Denmark) to 46.0% in countries with only one routine fetal scan and 55.6% in countries with two or three routine fetal scans. The prenatal detection rate of chromosomal anomalies was 40.3% (151/375 cases). This rate for recognized syndromes and multiply malformed with CHD was 51.9% (54/104 cases) and 48.6% (137/282 cases), respectively; 150/229 Down syndrome (65.8%) were livebirths. Concerning the syndromic cases, the detection rate of deletion 22q11, situs anomalies and VATER association was 44.4%, 64.7% and 46.6%, respectively. In conclusion, the present study shows large regional variations in the prenatal detection rate of CHD with the highest rates in European regions with three screening scans. Prenatal diagnosis of CHD is significantly higher if associated malformations are present. Cardiac defects affecting the size of the ventricles have the highest detection rate. Mean gestational age at discovery was 20-24 weeks for the majority of associated cardiac defects.

Resumo:O objetivo deste trabalho foi determinar a variação sazonal dos componentes do balanço de energia e avaliar o controle biofísico da evapotranspiração, em área de Caatinga preservada, em condições de seca intensa. O experimento foi conduzido em 2012, tendo-se utilizado o sistema "eddy covariance", instalado a 16,9 m acima da superfície do solo. Além disso, foram realizadas medidas de temperatura, umidade, radiação solar, saldo de radiação, fluxo de calor no solo e temperatura do solo. Os dados dos fluxos de calor sensível e latente foram processados com o programa Alteddy e analisados em escalas diária e sazonal. Com os dados do fluxo de calor latente, foram determinados os valores da evapotranspiração, bem como os indicadores da sua sensibilidade às condições ambientais: fator de desacoplamento, e resistências aerodinâmica e da superfície. O saldo de radiação apresentou forte sazonalidade, com comportamento dependente da radiação solar. A partição dos componentes do balanço de energia revelou que a ocorrência de seca intensa maximiza a destinação da energia disponível ao fluxo de calor sensível, e que a evapotranspiração nessa condição é especialmente dependente do deficit de pressão de vapor e da resistência da superfície.

El Trastorn Específic del Llenguatge (TEL) és un dètficit lingüístic que afecta el desenvolupament normal del llenguatge, en qui es poden veure implicades tant la comprensió com l'expressió, i en què es mostra un rendiment cognitiu superior al linguístic. L'objectiu d'aquest treball és estudiar el perfil psicolingüístic de nens amb TEL. S'hi estudien setze nens, vuit amb TEL i vuit que segueixen la pauta estàndard de desenvolupament del llenguatge (NP); tots els nens tenen entre 5 i 9;4 anys d'edat. Les diferències són notables entre els dos grups de nens estudiats, tant quantitativament com qualitativament. En l'estudi es pot observar que la comprensió es relaciona directament amb els errors gramaticals i sintàctics en el nens amb TEL. També es destaquen les relacions existents entre errors de formulació i errors gramaticals i sintàctics, d'una banda, i entre aquests dos últims, de ['altra. Les característiques que mostren els nens amb TEL comprometen el seu procés d'escolarització i la seva integració social.

Triiodothyronine (30 nM) added to serum-free cultures of mechanically dissociated re-aggregating fetal (15-16 days gestation) rat brain cells greatly increased the enzymatic activity of choline acetyltransferase and acetylcholinesterase throughout the entire culture period (33 days), and markedly accelerated the developmental rise of glutamic acid decarboxylase specific activity. The enhancement of choline acetyltransferase and acetylcholinesterase specific activities in the presence of triiodothyronine was even more pronouned in cultures of telencephalic cells. If triiodothyronine treatment was restricted to the first 17 culture days, the level of choline acetyltransferase specific activity at day 33 was 84% of that in chronically treated cultures and 270% of that in cultures receiving triiodothyronine between days 17 and 33, indicating that relatively undifferentiated cells were more responsive to the hormone. Triiodothyronine had no apparent effect on the incorporation of [3H]thymidine at day 5 or on the total DNA content of cultures, suggesting that cellular differentiation, rather than proliferation was affected by the hormone. Our findings in vitro are in good agreement with many observations in vivo, suggesting that rotation-mediated aggregating cell cultures of fetal rat brain provide a useful model to study thyroid hormone action in the developing brain.

Insults during the fetal period predispose the offspring to systemic cardiovascular disease, but little is known about the pulmonary circulation and the underlying mechanisms. Maternal undernutrition during pregnancy may represent a model to investigate underlying mechanisms, because it is associated with systemic vascular dysfunction in the offspring in animals and humans. In rats, restrictive diet during pregnancy (RDP) increases oxidative stress in the placenta. Oxygen species are known to induce epigenetic alterations and may cross the placental barrier. We hypothesized that RDP in mice induces pulmonary vascular dysfunction in the offspring that is related to an epigenetic mechanism. To test this hypothesis, we assessed pulmonary vascular function and lung DNA methylation in offspring of RDP and in control mice at the end of a 2-wk exposure to hypoxia. We found that endothelium-dependent pulmonary artery vasodilation in vitro was impaired and hypoxia-induced pulmonary hypertension and right ventricular hypertrophy in vivo were exaggerated in offspring of RDP. This pulmonary vascular dysfunction was associated with altered lung DNA methylation. Administration of the histone deacetylase inhibitors butyrate and trichostatin A to offspring of RDP normalized pulmonary DNA methylation and vascular function. Finally, administration of the nitroxide Tempol to the mother during RDP prevented vascular dysfunction and dysmethylation in the offspring. These findings demonstrate that in mice undernutrition during gestation induces pulmonary vascular dysfunction in the offspring by an epigenetic mechanism. A similar mechanism may be involved in the fetal programming of vascular dysfunction in humans.

This paper presents the evaluation results of the methods submitted to Challenge US: Biometric Measurements from Fetal Ultrasound Images, a segmentation challenge held at the IEEE International Symposium on Biomedical Imaging 2012. The challenge was set to compare and evaluate current fetal ultrasound image segmentation methods. It consisted of automatically segmenting fetal anatomical structures to measure standard obstetric biometric parameters, from 2D fetal ultrasound images taken on fetuses at different gestational ages (21 weeks, 28 weeks, and 33 weeks) and with varying image quality to reflect data encountered in real clinical environments. Four independent sub-challenges were proposed, according to the objects of interest measured in clinical practice: abdomen, head, femur, and whole fetus. Five teams participated in the head sub-challenge and two teams in the femur sub-challenge, including one team who tackled both. Nobody attempted the abdomen and whole fetus sub-challenges. The challenge goals were two-fold and the participants were asked to submit the segmentation results as well as the measurements derived from the segmented objects. Extensive quantitative (region-based, distance-based, and Bland-Altman measurements) and qualitative evaluation was performed to compare the results from a representative selection of current methods submitted to the challenge. Several experts (three for the head sub-challenge and two for the femur sub-challenge), with different degrees of expertise, manually delineated the objects of interest to define the ground truth used within the evaluation framework. For the head sub-challenge, several groups produced results that could be potentially used in clinical settings, with comparable performance to manual delineations. The femur sub-challenge had inferior performance to the head sub-challenge due to the fact that it is a harder segmentation problem and that the techniques presented relied more on the femur's appearance.

Este estudo analisou os principais fatores envolvidos na comercialização do mamão em 27 supermercados e 27 varejões do Distrito Federal (Brasília e cidades satélites), no período de janeiro a dezembro de 2001. Entrevistaram-se 162 consumidores. Na tabulação e análise dos questionários, obteve-se: o principal fornecedor de mamão para o varejo foi a Ceasa-DF; a presença de danos físicos foi o principal aspecto observado pelos responsáveis pela compra; os frutos são expostos em prateleiras sem refrigeração, e o responsável pelo prejuízo ocasionado pelas perdas foi o próprio estabelecimento varejista. Os estabelecimentos varejistas diferenciaram-se em alguns tópicos, tais como: responsável pela compra, existência de contrato; forma de exposição dos frutos; grau de maturação dos frutos adquiridos, e acompanhamento das perdas. Os consumidores preferem frutos nos estádios 3 a 5 (acima de 25% da superfície amarela); escolhem o estabelecimento para compra de mamão orientados pela qualidade e preço; não pagariam mais por um mamão embalado e classificado, preferindo mamão a granel pela possibilidade de escolher-se o fruto que mais lhe agrade no momento da compra, e elegeram, como principal problema, a presença de frutos com danos físicos.

NK1.1+TCR alpha beta+ (NK1+) T cells are an unusual subset of mouse TCR alpha beta+ cells found primarily in adult thymus and liver. In contrast to conventional TCR alpha beta+ cells, NK1+ T cells have a TCR repertoire that is highly skewed to V alpha14 and to Vbeta8, -7, and -2. The developmental origin and ligand specificity of NK1+ T cells are controversial. We show here that NK1+ T cells with a typically biased V alpha and V beta repertoire develop in cytokine-supplemented suspension cultures of fetal liver established from either normal or athymic mice. Furthermore, NK1+ T cell development in fetal liver cultures is abrogated in beta2m-deficient mice (which lack MHC class I and other related molecules) and can be partially inhibited by the presence of anti-CD1 mAbs. Moreover, mixing experiments indicate that recombination-deficient SCID fetal liver cells can reconstitute NK1+ T cell development in beta2m-deficient fetal liver cultures. Collectively, our data demonstrate that NK1+ T cells can develop extrathymically from fetal liver precursors and that a beta2m-associated ligand (putatively CD1) present on nonlymphoid cells is essential for their positive selection and/or expansion.
