Dengue congenital disease was not confirmed in 10 children whose mothers had the infection during pregnancy. The fetal sera presented anti-dengue IgG antibodies which progressively declined, and disappeared after 8 months. IgM antibodies to dengue were not observed in the sera. Other normal data suggesting the healthy state of the children included: absence of malformations, pregnancy time, Apgar index, weight, and placenta aspect

A necessidade de utilizar métodos de ligação entre componentes de forma mais rápida, eficaz e com melhores resultados, tem causado a crescente utilização das juntas adesivas, em detrimento dos métodos tradicionais de ligação tais como a soldadura, brasagem, ligações aparafusadas e rebitadas. A utilização das juntas adesivas tem vindo a aumentar em diversas aplicações industriais por estas apresentarem vantagens das quais se destacam a redução de peso, redução de concentrações de tensões e facilidade de fabrico. No entanto, também apresentam desvantagens, como a necessidade de preparação das juntas e o descentramento da carga aplicada que provoca efeitos de flexão, os quais dão origem a tensões normais na direcção da espessura do adesivo (tensões de arrancamento), afectando assim a resistência da junta. A combinação da ligação adesiva com a soldadura por pontos permite algumas vantagens em comparação com as juntas adesivas tradicionais como a maior resistência, aumento da rigidez, melhor resistência ao corte e arrancamento e também à fadiga. Neste trabalho é apresentado um estudo experimental e numérico de juntas de sobreposição simples adesivas e híbridas (adesivas-soldadas). Os adesivos utilizados são o Araldite AV138®, apresentado como sendo frágil, e os adesivos Araldite 2015® e Sikaforce® 7752, intitulados como adesivos dúcteis. Foram considerados substratos de aço (C45E) em juntas com diferentes comprimentos de sobreposição ( ), que foram sujeitas a esforços de tracção. Foi realizada uma análise dos valores experimentais e efectuada uma comparação destes valores com os resultados obtidos por Elementos Finitos (EF) no software ABAQUS®, que incluiu uma análise de tensões na camada de adesivo e previsão do comportamento das juntas por Modelos de Dano Coesivo (MDC). A análise por MDC permitiu obter os modos de rotura, as curvas força-deslocamento e a resistência das juntas com bastante precisão, com excepção das juntas coladas com o adesivo Sikaforce® 7752. Estes resultados permitiram validar a técnica de modelação proposta para as juntas coladas e híbridas, o que representa uma base para posterior aplicação desta técnica em projecto, com as vantagens decorrentes da redução do tempo de projecto e maior facilidade de optimização.

Fungi have been considered a potential source of natural anticancer drugs. However, studies on these organisms have mainly focused on compounds present in the sporocarp and mycelium. The aim of this study was to assess the anticancer potential of fungal spores using a bioassay-guided fractionation with cancer and normal cell lines. Crude extracts from spores of the basidiomycetous fungus Pisolithus tinctorius were prepared using five solvents/solvent mixtures in order to select the most effective crude extraction procedure. A dichloromethane/methanol (DCM/MeOH) mixture was found to produce the highest extraction yield, and this extract was fractionated into 11 fractions. Crude extracts and fractions were assayed for cytotoxicity in the human osteocarcinoma cell line MG63, the human breast carcinoma cell line T47D, the human colon adenocarcinoma cell line RKO, and the normal human brain capillary endothelial cell line hCMEC/D3. Cytotoxicity was assessed by the 3-(4,5-dimethyl-2-thiazolyl)-2,5-diphenyl-2H-tetrazolium bromide (MTT) reduction assay. The results showed a reduction in cancer cell viability of approximately 95% with 4 of 11 fractions without a significant reduction in viability of hCMEC/D3 cells. Data demonstrated that spores of P. tinctorius might serve as an interesting source of compounds with potential anticancer properties.

Hepatitis C virus infection evolves progressively persisting in the majority of patients (85%). Most patients have high ALT (alanine aminotransferase) levels and approximately 25% normal ALT. The latter are usually female and there is no association between genotype and severity of hepatic lesion. Histologic analysis usually shows small lesion and absence or low amount of fibrosis, despite cirrhosis having been reported. Aiming at assessing prevalence, demographic, genotypical and anatomopathological characteristics in patients with normal ALT levels, we have carried out a study of 68 chronic hepatitis C patients between January 1997 and April 2000. There was a prevalence of 13.8% chronic hepatitis C patients with normal ALT levels, 45.6% of which were male and 54.4% female, the mean age being 38 +/- 13 years. We found a predominance of genotype 1 in 84.7% of the patients, genotype 2 in 6.8% and genotype 3 in 10.7%. In 52.9% of the cases liver biopsies revealed liver reaction, periportal activity score 0-1 was observed in 85.3% of the patients and score 2-4 was seen in 14.7%. Structural activity score 0-1 was seen in 73.5% of the patients and score 2-4 in 26.5% of them. Periportal activity > 2 and structural activity > 1 was seen in 29%, but steatosis was not seen in 73.5%. Our results suggest the need to revisit for liver biopsy practice in patients with Chronic Hepatitis C and normal transaminases.

O método de união com ligações adesivas está cada vez mais a ser utilizado na conceção de estruturas mecânicas, por causa das vantagens significativas desta técnica em comparação com as ligações tradicionais. De facto, as juntas com ligação adesiva estão sob investigação intensa há bastante tempo. Entre as vantagens, destaca-se a redução de peso e possibilidade de unir diferentes materiais, incluindo compósitos, sem danificar as estruturas a ligar. Os adesivos comerciais variam desde resistentes e frágeis (por exemplo, Araldite® AV138) a menos resistentes e dúcteis (por exemplo, Araldite® 2015). Uma nova família de adesivos de poliuretano combina elevada resistência e ductilidade (por exemplo, Sikaforce® 7888). Este trabalho compara o desempenho à tração dos três adesivos supracitados, em juntas de alumínio (Al6082-T651) de sobreposição simples e dupla, com variação dos valores de comprimento de sobreposição (LO). A análise numérica de modelos de dano coesivo (MDC) foi realizada para analisar as tensões de arrancamento (oy) e as de corte (txy) na camada adesiva, para estudar a variável de dano do MDC durante o processo de rotura e para avaliar a capacidade MDC na previsão da resistência da junta. A análise da resistência e da variável de dano ajudou na compreensão das diferenças entre os adesivos no que se refere ao processo de rotura e resistência da junta. Observou-se que as juntas de sobreposição dupla apresentam uma distribuição de tensões bastante mais favorável relativamente às juntas de sobreposição simples, principalmente devido à eliminação da flexão do substrato interior. Como resultado, a resistência destas juntas foi tipicamente superior ao dobro da observada para as juntas de sobreposição simples, com exceção de algumas configurações de junta em que houve plastificação extensa ou mesmo rotura dos substratos por tração. O trabalho proposto permitiu também concluir que as previsões MDC são tipicamente precisas, e qual a família de adesivos é mais adequada para cada configuração de junta, com a clara vantagem para o Sikaforce® 7888. Como resultado deste trabalho, foram propostas diretrizes de conceção para juntas adesivas.

Decreased responses to hepatitis B vaccine have been associated with some host conditions including obesity. Susceptible non-responders to a primary three-dose vaccine series should be revaccinated. Those who maintain a non-responder condition after revaccination with three vaccine doses are unlikely to develop protection using more doses. This is a description of an obese woman who received six doses of hepatitis B vaccine and persisted as a non-responder. She was submitted to a vertical banded gastroplasty Roux-en-Y gastric bypass Capellas's technique. After weight reduction, she received three additional doses of vaccine and seroconverted. Further studies should help clarify the need to evaluate antibody levels and eventually revaccinate the increasing population of individuals who undergo weight reduction.

PURPOSE: 1. Identify differences in optic nerve sheath diameter (ONSD) as an indirect measure of intracranial pressure (ICP) in glaucoma patients and a healthy population. 2. Identify variables that may correlate with ONSD in primary open-angle glaucoma (POAG) and normal tension glaucoma (NTG) patients. METHODS: Patients with NTG (n = 46) and POAG (n = 61), and healthy controls (n = 42) underwent B-scan ultrasound measurement of ONSD by an observer masked to the patient diagnosis. Intraocular pressure (IOP) was measured in all groups, with additional central corneal thickness (CCT) and visual field defect measurements in glaucomatous patients. Only one eye per patient was selected. Kruskal-Wallis or Mann-Whitney were used to compare the different variables between the diagnostic groups. Spearman correlations were used to explore relationships among these variables. RESULTS: ONSD was not significantly different between healthy, NTG and POAG patients (6.09 ± 0.78, 6.03 ± 0.69, and 5.71 ± 0.83 respectively; p = 0.08). Visual field damage and CCT were not correlated with ONSD in either of the glaucoma groups (POAG, p = 0.31 and 0.44; NTG, p = 0.48 and 0.90 respectively). However, ONSD did correlate with IOP in NTG patients (r = 0.53, p < 0.001), while it did not in POAG patients and healthy controls (p = 0.86, p = 0.46 respectively). Patient's age did not relate to ONSD in any of the groups (p > 0.25 in all groups). CONCLUSIONS: Indirect measurements of ICP by ultrasound assessment of the ONSD may provide further insights into the retrolaminar pressure component in glaucoma. The correlation of ONSD with IOP solely in NTG patients suggests that the translaminar pressure gradient may be of particular importance in this type of glaucoma.

Biomechanical gait parameters—ground reaction forces (GRFs) and plantar pressures—during load carriage of young adults were compared at a low gait cadence and a high gait cadence. Differences between load carriage and normal walking during both gait cadences were also assessed. A force plate and an in-shoe plantar pressure system were used to assess 60 adults while they were walking either normally (unloaded condition) or wearing a backpack (loaded condition) at low (70 steps per minute) and high gait cadences (120 steps per minute). GRF and plantar pressure peaks were scaled to body weight (or body weight plus backpack weight). With medium to high effect sizes we found greater anterior-posterior and vertical GRFs and greater plantar pressure peaks in the rearfoot, forefoot and hallux when the participants walked carrying a backpack at high gait cadences compared to walking at low gait cadences. Differences between loaded and unloaded conditions in both gait cadences were also observed.

Objective To examine the combined effects of physical activity and weight status on blood pressure (BP) in preschool-aged children. Study design The sample included 733 preschool-aged children (49% female). Physical activity was objectively assessed on 7 consecutive days by accelerometry. Children were categorized as sufficiently active if they met the recommendation of at least 60 minutes daily of moderate-to-vigorous physical activity (MVPA). Body mass index was used to categorize children as nonoverweight or overweight/obese, according to the International Obesity Task Force benchmarks. BP was measured using an automated BP monitor and categorized as elevated or normal using BP percentile-based cut-points for age, sex, and height. Results The prevalence of elevated systolic BP (SBP) and diastolic BP was 7.7% and 3.0%, respectively. The prevalence of overweight/obese was 32%, and about 15% of children did not accomplish the recommended 60 minutes of daily MVPA. After controlling for age and sex, overweight/obese children who did not meet the daily MVPA recommendation were 3 times more likely (OR 3.8; CI 1.6-8.6) to have elevated SBP than nonoverweight children who met the daily MVPA recommendation. Conclusions Overweight or obese preschool-aged children with insufficient levels of MVPA are at significantly greater risk for elevated SBP than their non overweight and sufficiently active counterparts. The aims of this study were as follows: (1) to analyze differences in cardiorespiratory fitness (CRF), parents' body mass index (BMI) and birth weight (BW) between non-overweight (NOW) and overweight/obese (OV/OB) adolescents, and (2) to investigate the association of those variables with the risk of their biological offspring being OV/OB. This study comprised 788 adolescents (477 girls and 311 boys), aged between 12 and 18 years. CRF was predicted by maximal multistage 20-m shuttle-run test according to the procedures described in FITNESSGRAM. Children's BMI was classified according to the International Obesity Task Force. Adolescents' BW was assessed from each child's pediatric record at birth. Parents' OV/OB status was defined and classified according to the World Health Organization. Socioeconomic status was defined by parental education. The prevalence OV/OB was 21.4 and 5.3%, respectively, and there were no gender differences. SUMMARY The clinical outcome of infection with Leishmania species of the subgenus Viannia in hamster model (Mesocricetus auratus) has shown to be different depending on experimental protocol. Body weight has been a relevant determinant of the clinical outcome of the infection in hamsters with visceral leishmaniasis but its importance as a clinical parameter in hamsters with cutaneous leishmaniasis is not known. In this study, the clinical evolution of infection with L. (V) panamensis was evaluated in juvenile and adult male hamsters during 11 weeks by comparing clinical parameters such as attitude, temperature, respiratory rate, appearance of the stool, and body weight between infected and non-infected groups. Results showed that body weight decreased in adult hamsters after infection by L. (V) panamensis; this observation supports the use of body weight as an additional parameter to define the management or treatment of cutaneous leishmaniasis in infected adult hamsters used as an animal experimental model for leishmaniasis.

A presente dissertação apresenta uma abordagem ao tema Fluência, com um desenvolvimento geral para quatro materiais: madeira, alumínio, betão e aço. No entanto, particularizou-se este estudo apenas para dois destes materiais, a madeira e o alumínio. A madeira é um material viscoelástico, logo fortemente influenciado quando submetido a uma ação constante (Fluência) sendo agravada com alterações do teor em água. Iniciou-se o estudo, com uma introdução aos objetivos principais e a revisão teórica do conhecimento da propriedade mecânica (Fluência). Efetuou-se o estudo inicial para os quatro materiais indicados. Posteriormente, fez-se avaliação do efeito mecânico sortivo durante 60 dias com sete ciclos, em provetes de dimensões 20 x 20 x 400 mm3 (escala 1:10) de madeira de Eucalyptus globulus Labill. Recorrendo ao levantamento de um conjunto de modelos numéricos, procedeu-se ao ajuste e extrapolação do comportamento, em Fluência, para distintos períodos de tempo (1, 10 e 50 anos). Os resultados obtidos demonstraram que a madeira de Eucalipto não apresenta um limite no seu comportamento em Fluência, logo instável para os 60 dias de duração de ensaio. Os diferentes modelos de Fluência (x6) apresentaram uma variabilidade crescente de resultados, de acordo com o aumento de extrapolação dos resultados. Para a espécie de madeira de Eucalipto constatou-se ainda, que o ajuste e extrapolação de deformação para 50 anos ultrapassaram os valores sugeridos pelo Eurocódigo 5. A última parte deste trabalho, incidiu sobre o desenvolvimento da metodologia do ensaio em flexão de 3 provetes de alumínio similares, cujas dimensões eram de 20 x 20 x 400mm3. O objetivo desta metodologia foi avaliar o seu comportamento em Fluência. Numa primeira fase, foi aplicada uma carga contante de 160 kN. E numa segunda fase, foi aplicada o dobro dessa carga sujeita adicionalmente a uma temperatura bastante superior à temperatura ambiente (a rondar os 50 oC). Conclusivamente verificou-se que o alumínio não apresenta deformação por Fluência, para as condições de ensaio apresentadas.

SUMMARY Introduction: We present a fatal case of disseminated cryptococcosis in a young man whose diagnosis of HIV infection was made at the time of admission to the emergency room. Case report: The patient was a twenty-three-year-old man, with a history of daily fever during one month associated with diarrhea, weight loss, headache, vomiting and generalized seizures. He also had a history of diabetes mellitus, alcoholism and drug addiction. Upon physical examination the patient was pale, disoriented and had periods of agitation. White blood cells count was 3,440/mm3 (5% lymphocytes), hemoglobin was 10g/dL, platelets were 83,000/ mm3. Creatinine was 0.7 mg/dL; urea 19 mg/dL; Na, K, and liver enzymes were within normal limits. Lactic dehydrogenase was 494 IU/L. Cerebrospinal fluid (CSF) analysis revealed 10 white blood cells/mm3 (58% neutrophils, 31% lymphocytes, 11% monocytes) and 2 red blood cells/mm3. India ink test revealed six Cryptococcus yeasts/mm3. CSF glucose was 122 mg/dL and protein was 36 mg/ dL. VDRL test was negative and anti-HIV test was positive. Intravenous hydration, insulin, phenytoin, fluconazole, pyrimethamine, sulfadiazine, folinic acid, and amphotericin B were started. The patient did not improve and became obtunded and hypotensive. He was intubated and put on mechanical respiration. He received vasoactive drugs and died less than 24 hours after admission. A postmortem examination was performed and revealed disseminated cryptococcosis, with severe involvement of the kidneys. Conclusion: Cryptococcosis, as a rule, is a systemic disease that affects mostly immunocompromised individuals, especially patients with AIDS. When diagnosed late in its course it has a very high mortality.
