The effectiveness of fungicides in controlling white mold (Sclerotinia sclerotiorum) of dry beans (Phaseolus vulgaris) was evaluated when they were applied through irrigation water directly onto the plants or only to the soil. Two field trials were installed in April 1998 and April 1999 in Viçosa, MG. Trials were conducted as a (2 x 3) + 1 factorial: two fungicides x three application modes + one untreated control. The fungicides were benomyl (1.0 kg a.i. ha-1) and fluazinam (0.5 l a.i. ha-1). The three application modes were: (a) by backpack sprayer (667 l ha-1), (b) by garden watering-cans simulating sprinkler irrigation with 35,000 l ha-1 of water, and (c) by garden watering-cans applying water between the rows and near the soil surface in 35,000 l ha-1 of water. In 1998, fungicides were applied at 43 and 54 days after emergence (DAE); in 1999, at 47 and 61 DAE. Both fungicides were similarly effective on white mold control when applied by either chemigation or backpack sprayer, resulting in yields 21% higher than untreated control. Only fluazinam provided disease control when applications were made only in soil. Chemigation provided white mold control equivalent to that of backpack sprayer in terms of incidence, severity and number of diseased pods. Consequently, yield differences between these application methods were not significant.

In this work is proposed a solid phase preconcentration system of Co2+ ions and its posterior determination by GFAAS in which fractional factorial design and response surface methodology (RSM) were used for optimization of the variables associated with preconcentration system performance. The method is based on cobalt extraction as a complex Co2+-PAN (1:2) in a mini-column of polyurethane foam (PUF) impregnated with 1-(2-pyridylazo)-naphthol (PAN) followed by elution with HCl solution and its determination by GFAAS. The chemical and flow variables studied were pH, buffer concentration, eluent concentration and preconcentration and elution flow rates. Results obtained from fractional factorial design 2(5-1) showed that only the variables pH, buffer concentration and interaction (pH X buffer concentration) based on analysis of variance (ANOVA) were statistically significant at 95% confidence level. Under optimised conditions, the method provided an enrichment factor of 11.6 fold with limit of detection and quantification of 38 and 130 ng L-1, respectively, and linear range varying from 0.13 to 10 µg L-1. The precision (n = 9) assessed by relative standard deviation (RSD) was respectively 5.18 and 2.87% for 0.3 and 3.0 µg L-1 cobalt concentrations.

This paper describes a method for quantitative spot test analysis of hydrochlorothiazide using diffuse reflectance spectroscopy. The reflectance measurements were performed analyzing the colored compound (l = 585 nm) produced from the reaction between hydrochlorothiazide and p-dimethylaminocinnamaldehyde (PDAC) in acid medium. This reaction occurred on filter paper after heating to 80ºC for 8 minutes. Factorial designs allowed varying multiple reaction factors simultaneously in order to obtain the best reaction conditions. These factors included heating temperature, heating time, acid volume and PDAC volume. The linearity was studied in the range of 3.36x10-2 to 1.01x10-1 mol L-1 with a correlation coefficient of 0.998. The limit of detection was estimated to be 1.32x10² mol L-1. Commercial samples were analyzed using the proposed method and the results were favorably compared with those of the United States Pharmacopeia method, showing that quantitative spot test analysis by diffuse reflectance could be successfully used to determine hydrochlorothiazide in medicines.

A flow-injection system with sample and reagent addition by the synchronous merging zones approach for calcium determination in milk by flame AAS is proposed. Main parameters were optimized using a factorial design with central point. The optimum conditions were 2.5% (m/v) for La concentration, 8 mL min-1 for the carrier flow-rate, 20 cm for coiled reactor and 250 ìL for sample volume. Different sample preparation procedures were evaluated such as dilution in water or acid and microwave-assisted decomposition using concentrated or diluted acids. The optimized flow system was applied to determine Ca in eleven commercial milk samples and two standard reference materials diluted in water. Similar calcium levels were encountered comparing the results obtained by the proposed method (dilution in water) with those obtained using microwave-oven digestion. Results obtained in two standard reference materials were in agreement at 95% confidence level with those certified. Recoveries of spiked samples were in the 93% - 116% range. Relative standard deviation (n = 12) was < 5.4% and the sample throughput was 150 measurements per hour, corresponding to a consumption of 250 µL of sample and 6.25 mg La per determination.

The damage and the resistance levels of cultivars and accessions of common beans rescued in the South and mountain regions of Espírito Santo State, Brazil, to M. incognita race 3 and M. javanica parasitism were evaluated under a greenhouse. Four rescued bean genotypes ("FORT-10", "FORT-13", "FORT-16" and "FORT-19") and 2 commercial cultivars: "Pérola", and "Aporé", were tested. The cultivar "Rico-23" was included as standard of susceptibility to nematodes and non-inoculated plants constituted the control. Thus, the experiment was carried out in a completely randomized design in 3 (treatments considering nematodes) x 7 (genotypes and bean cultivars) factorial arrangement, with 7 replicates. Data were measured at 50 days after plant inoculation. For damage quantification, the following variables were evaluated: plant height (PHE), number of nodes (NNO), number of trifoliate leaves (NRT), fresh matter weight (FWE) and dry matter weight (DWE) of shoots, root weight (RWE), number of root nodules (NRO) and final population (FPO) of nematodes per root system. There were no significant differences between the effects caused by M. incognita and M. javanica, but both species showed inferior values of PHE, NNO, NRT, RWE, FWE and DWE compared to controls. Concerning the levels of resistance of bean plants to M. incognita, the genotypes "FORT-10", "FORT-13", "Aporé" and "FORT-16" behaved as moderately resistant, the cultivars "Rico 23" and "Pérola" low resistant, and the genotype "FORT-19" as highly susceptible. When parasitized by M. javanica, the beans "FORT-19", "Rico-23", "FORT-16" and "FORT-13" were low resistant, "Pérola" and "Aporé" susceptible and "FORT-10" highly susceptible.

Fungi require special substrates for their isolation, vegetative growth and sporulation. In experiments conducted in the laboratory, the influence of substrates, light, filter paper and pH on the sporulation of Cercospora sojina conidia, the causal agent of soybean frogeye leaf spot, was assessed. The media potato sucrose agar, V-8 agar, tomato extract agar, soybean leaf extract agar, soybean seed extract agar, soybean meal agar, soybean flour agar and wheat flour agar were tested, added on the surface, with and without filter paper and under two light regimes, with 12 h light at 25°± 2°C and in the dark. A triple factorial 8x2x2 (substrates x light/dark x with/without filter paper) design with four replicates was used. V-8 agar medium was employed and the pH was adjusted with HCl 0.1N or NaOH 0.1N before autoclaving to the values: 3, 4, 5, 6, 7 and 8, and the pH of V-8 agar medium is 6.7. The evaluation was done on the seventh day of incubation. Data underwent regression analysis. Sporulation was maximized on the agar media V-8, seed extract, oat flour, tomato extract, and potato sucrose in the presence of filter paper and 12h light. On V-8 medium, maximal sporulation was obtained with pH 6.7.

Although the functions of the element Boron (B) in plants have not been sufficiently clarified, several hypotheses have been raised. Some of the functions attributed to this element are synthesis and transport of carbohydrates. To evaluate the effect of B on the synthesis of some polyols and sugars in Eucalyptus grandis and hybrids "urograndis", these species were submitted to situations of supply and restriction of B. The experiment was carried out in a greenhouse, arranged in a randomized block design in a 2 x 2 factorial scheme, with 5 replicates of each genotype. The B levels were provided in the form of nutrient solution. No significant difference was observed in the content of mannitol, sorbitol, myo-inositol and scyllo-inositol and sugars α-glucose and β-glucose between E. grandis and hybrids. Arabinose was the only one to present a higher content in E. grandis on restriction of B. The effect of the presence of B was very expressive, but regarding B supply, the plants showed significant increase in the synthesis of the compounds evaluated.

The objective of this work was to evaluate the outplanting growth of Eucalyptus grandis e E. saligna clones, produced by cuttings in tubes (50cm³) and in pressed blocks (40x60x07cm) - 175 cm³/ seedlings, with different substrates (BT - sugarcane bagasse+sugarcane filter cake; AR - carbonized rice hull + eucalyptus bark; TF - peat). The experiment was arranged in a randomized block design, in a 2x7 factorial (2 clones and 7 treatments), with four replicates with 25 plants. Survival was evaluated two months later. Plant growth was monitored through height and ground level diameter at 20, 40, 60, 120 and 180 days after outplanting. To evaluate the effect of the containers on stem and root biomass in both clones, 180 days after outplanting, the cuttings grown in BT substrate with fertilizer were selected. One plant per plot of each clone, grown in tubes and in pressed blocks was selected. The E. grandis and E. saligna cuttings grown in pressed blocks with sugarcane bagasse+sugarcane filter cake presented greater height and diameter after out planting. Both clones presented larger root, bark, log and branch biomass production in plants produced in the block system. Cuttings of E. saligna grown in pressed blocks showed 80% most wood biomass 180 days after outplanting, compared to that grown in tubes. In E. grandis, the differences in diameter and height, in function of the cutting production system, decreased along time, while in E. saligna these differences increased along the evaluation period.

Termites are well known for their ability to damage wood and various types of wood-derived products. This study was performed in the municipality of Seropédica, Rio de Janeiro state, Brazil, with the objective of evaluating the susceptibility of the wood of five different forest species to the activity of the Asian subterranean termite, Coptotermes gestroi. Wood stakes from the following forest species were used as specimens: pine (Pinus sp., Pinaceae), Brazil nut (Bertholletia excelsa, Lecythidaceae), cabbage angelin (Andira inermis, Leguminosae-Papilionoideae), maçaranduba (Manilkara huberi, Sapotaceae) and pink ipê (Tabebuia avellanedae, Bignoniaceae). The stakes, with dimensions of 2 cm x 2 cm x 16 cm, were subjected to the activity of C. gestroi from September 2, 2006 to June 2, 2007. The experimental design consisted of randomized blocks with four replications in an 11 x 5 x 3 factorial arrangement. Twelve stakes of each of the five forest species were installed at each of eleven sites, totaling 660 stakes. Three evaluations were performed, at 90, 180 and 270 days after stake installation. The results indicated that the pine, Brazil nut and cabbage angelin stakes were more susceptible to C. gestroi attack, while the pink ipê and maçaranduba stakes suffered no significant damage from this termite. There was a significant negative correlation between the wood consumption rate and the wood density of the species studied. The woods most susceptible to C. gestroi attack presented moderate to low densities and therefore less resistance, as in the case of pine and cabbage angelin.

The Dimorphandra mollis Benth. - Caesalpiniaceae is a native forest species coming from the Cerrado and Caatinga due to its economical and ecological use, which justifies the studies on seed germination. In this work, germinative performance of D. mollis seeds were studied in different conditions of temperature regime and substrate. The experimental delineation used was completely randomized in factorial 4 x 4 (4 substrates -sand, coconut fiber, vermiculite and paper towel; and 4 temperatures: 25, 30, 35 and 20-30ºC), with four replications of 25 seeds each. The following parameters were evaluated: seed moisture content, final germination, first germination count, germination speed index, length and dry matter weight. The best germination and vigor is obtained at 30 and 35ºC. The substrates paper towel and vermiculite allow satisfactory germinative performance of seeds, being suitable to evaluate the physiological quality of D. mollis seeds.

Bauhinia x blakeana (B. purpurea x B. variegata) is a natural hybrid that has been cultivated in gardens, streets and parks. Due to its sterility, it must be vegetatively propagated. The objective of this work was to evaluate the viability of cuttings and grafting on its propagation. Semi-woody cuttings were collected during four seasons and treated with 0; 1,000; 2,000; and 3,000 mg L-1 of IBA. The experimental design was entirely randomized and the treatments were arranged in a 4x4 factorial scheme (four collecting times x four IBA concentrations) and five replications with 10 cuttings each, per collecting time and per IBA concentration. Characteristics of roots and shoots were evaluated after 90 days. The data means were compared by the Tukey test and submitted to the polynomial regression analysis. For the grafting experiment, B. variegata and B. variegata var. candida plants of six and 12 months were used as rootstocks and the splice graft and T-budding methods were tested. The experimental design was entirely randomized and the treatments were arranged in a 2x2x2 factorial scheme (two rootstock species x two grafting methods x two rootstock ages) and four replications with five plants each, per rootstock species, per grafting method and per rootstock age. Characteristics of shoots were evaluated after 90 days and the data means were compared by the Tukey test. B. x blakeana can be propagated by semi-woody cuttings collected in spring, without IBA application, or in summer, with the application of 3,000 mg L-1 of IBA. The tested grafting methods were not effective.

Planting trees is an important way to promote the recovery of degraded areas in the Caatinga region. Experiments (E1, E2, and E3) were conducted in a randomized blocks design, with three, three, and five replicates, respectively. The objectives were to evaluate biomass of the shoots of: a) gliricidia (G) and sabiá (S), as a response to planting density; b) G, S, and neem (N) in competition; c) G, and S in agroforestry. E1 was conducted in split-plots, and planting densities (400, 600, 800, 1000, and 1200 plants ha-1) as subplots. E2 consisted of a factorial comprising the following plots: GGG, NGN, SGS, NNN, GNG, SNS, SSS, GSG, NSN (each letter represents a row of plants). E3 was conducted with G and S in agroforestry experiment. The trees were harvested after 54, 42, and 27 months old, in E1, E2 and E3, respectively. In E1, G presented higher green biomass of the stems and leaf at smaller densities than S, but lower green biomass of branches at most densities. The species did not differ for mean stem dry biomass and leaf dry biomass, but G showed higher branch dry biomass at most densities. Higher planting densities increased green and dry biomass of stems, branches, and leaves in S, but decreased those characteristics in G, with the exception of leaf dry mass, which was not influenced by density. In E2, the behavior of each species was identical in plots containing the same or different species. Griricidia showed the highest green biomass of stems and branches, and the highest values for geren biomass of the leaf were observed for gliricidia and neem. The highest stem, branch, and leaf dry biomass values were obtained for G, S, and N, respectively. In E3, G was superior for stem and leaf green biomass, and for stem and branch dry biomass. There were no differences between species for the other biomass values.

The aim of this study was to identify, by multivariate statistical technique, the physic, chemical and biological variables that best characterize the quality of surface waters in two small rural catchments with different land uses (eucalyptus silviculture (SC) vs. pasture and extensive livestock (LC)) located in Rosário do Sul, RS - Brazil. Monitoring was conducted during the months of August 2011 to August 2012 and the following parameters were analyzed: Ca2+, Mg2+, K+, SO42-, Cl-, pH, electrical conductivity, turbidity, alkalinity, suspended and dissolved solids, biochemical oxygen demand , total coliforms, Escherichia coli and temperature, flow and rainfall. Through the use of FA/PCA, it was found that the model best fit to express water quality of in LC that was composed of five factors which account for 83.5% of the total variance, while for SC, four factors accounted for 85.12% of the variance. In LC, the five main factors were, respectively, soluble salts, diffuse pollution, solid, and both anthropogenic and organic factors. In SC, the four factors were namely: soluble salts, mineral, nutritional and diffuse pollution factors. The results of this study showed that by replacing the traditional soil usage (pasture and livestock) with planted forest, diffuse pollution was attenuated but, however, it did not result in major changes in the physical-chemical and biological characteristics of the water. Another point to note is that factorial analysis did not result in a large reduction in the number of variables, once the best model fit occurred with the addition of 15 of 18 analyzed variables (LC) and 17 of 18 analyzed variables (SC).

ABSTRACT The present study aimed to evaluate the growth and the levels of N, P, K, Ca and Mg in Australian cedar seedlings which had been inoculated with arbuscular mycorrhizal fungi (AMF) in different types of containers. The experiment was carried out in a greenhouse and the experimental design was that of randomized complete blocks (RCB), with a 4 x 4 factorial design consisting of four inoculation treatments with AMF (Rhizophagus clarum, Gigaspora margarita, a mixed inoculation (R. clarum + G. margarita) and the control (with no AMF inoculation); four types of containers (plastic bags measuring 250 cm3, tubes of 55 and 130 cm3 and pressed blocks 440 cm3. plant-1), with four repetitions. The height, the diameter of the stem base, the aerial part dry weight (APDW), the dry weight of the root (DWR) and the total plant dry weight (DW) were measured, along with the Dickson quality index, the percentage of mycorrhizal colonization and the levels of N, P, K, Ca and Mg in the aerial part dry weight. One hundred and thirty eight days (138) days after sowing, the greatest growth and/or the highest levels of P, K and Ca could be observed in the aerial part dry weight of the Australian cedar seedlings which had been planted in the pressed block container and inoculated with a mixture of the two AMF species (G. margarita + R. clarum) or with just R. clarum. Thus it can be seen that AMF can make a significant contribution to the production of Australian cedar seedlings.

This work objectified to evaluate the efficiency of two meter mechanism of corn seeds when submitted to different forward speed and soil management system during the non-tillage seeding. It was used a factorial design in randomized blocks. The factors whose effects were examined were related to the seeders with pneumatic and horizontal disk meter mechanisms for the distribution of the seeds, to the set tractor-seeder forward speeds (4.4; 8.0 and 9.8 km h-1), and to the soil management system considering the corn no-tillage seeding over minimum tillage with chisel plow and the no-tillage system for the seeding of oat culture (Avena strigosa Schreb). It was verified that the forward speed didn't influence the initial and final stands of plants but it interfered in the regularity of longitudinal distribution of plants. The smallest speed provided the largest percentile of normal spacing between plants. The pneumatic meter mechanism presented better performance than the horizontal disk perforated in the longitudinal distribution of plants. About corn productivity aspect it's indifferent the recommendation of use for pneumatic and perforated horizontal disk meter mechanism of seeds.
