Foram avaliados os aspectos biológicos dos estádios imaturos de Chrysoperla externa (Hagen) e Ceraeochrysa cincta (Schneider), alimentados com ovos e ninfas de Bemisia tabaci (Genn.) biótipo B. Ovos dos predadores foram individualizados em recipientes de vidro de fundo chato com 2,5 cm de diâmetro e 8,5 cm de altura que foram mantidos à temperatura de 25±2ºC, UR de 70±10% e fotofase de 14h. Após a eclosão das larvas, discos de folhas de tomateiro e de couve com 2 cm de diâmetro, contendo ovos e ninfas de B. tabaci, respectivamente, foram colocados nos recipientes. A duração e a viabilidade de cada ínstar e da fase pupal de ambas as espécies predadoras alimentadas com diferentes estágios da presa foram avaliados. Larvas de primeiro ínstar de C. externa apresentaram desenvolvimento mais lento quando alimentadas com ovos de B. tabaci biotipo B monstrando influência do estágio da presa fornecida no período de desenvolvimento do predador. O mesmo não ocorreu para a espécie C. cincta. O desenvolvimento de larvas de primeiro ínstar de ambas as espécies predadoras foi semelhante quando foram alimentadas com ovos de B. tabaci; porém C. cincta apresentou período de desenvolvimento mais longo (5,4 dias) comparada a C. externa (4,0 dias) quando alimentada com ninfas da presa. Para larvas de segundo e terceiro ínstares e para a duração da fase larval não houve influência do estágio da presa fornecida (ovo ou ninfa) no período de desenvolvimento. A duração do segundo ínstar foi maior para C. cincta do que para C. externa (4,8 e 4,1 dias, respectivamente). Porém a duração do terceiro instar foi maior e da fase larval foi menor para C. externa (6,6 e 15,3 dias, respectivamente) do que para C. cincta (6,4 e 16,7 dias, respectivamente). A viabilidade dos ínstares foi acima de 90% para ambas as espécies. No entanto, menor porcentagem de viabilidade da fase de pupa foi registrada para C. cincta, quando alimentada com ovos (30%) ou ninfas (55,6%).

Avaliou-se o comportamento de genótipos de feijoeiro sob a infestação de Bemisia tabaci (Genn.) biótipo B e Caliothrips phaseoli (Hood) em condições de campo, nas épocas de cultivo de inverno, das águas e da seca. Adotou-se o delineamento de blocos casualizados em esquema de parcelas sub-subdividida (19x3x6), com 19 genótipos (IAC Tybatã, IAC Una, FT Nobre, Pérola, Gen 96A98-15-3-32-1, Gen 96A45-3-51-52-1, IAC Alvorada, IAC Diplomata, Gen 96A3-P1-1-1, LP 98-122, LP 02-130, LP 01-38, LP 9979, BRS-Pontal, BRS-Requinte, BRS-Triunfo, BRS-Grafite, CV-48 e Z-28), três épocas de cultivos (inverno, águas e seca) e seis períodos de avaliação. As avaliações foram realizadas semanalmente, contando-se o número de ovos e ninfas de B. tabaci e ninfas de C. phaseoli em dez folíolos por parcela. Os genótipos IAC Una, Pérola, Gen 96A45-3-51-52-1, Gen 96A98-15-32-1, FT Nobre, IAC Tybatã, IAC Alvorada, LP 02-130, LP 01-38, LP 98-122, IAC Diplomata e Gen 96A3P1-1-1 foram menos ovipositados por B. tabaci no cultivo das águas. Os genótipos Pérola, Gen 96A45-3-51-52-1, IAC Una, Gen 96A98-15-32-1, IAC Tybatã e FT Nobre foram menos infestados por C. phaseoli. A maior incidência de ninfas de mosca-branca ocorreu no fim de janeiro no cultivo das águas e início de maio no cultivo da seca; para tripés, foi em junho durante o cultivo de inverno. A maior incidência da população de B. tabaci e C. phaseoli, ocorreu dos 46 aos 60 dias após a emergência das plantas.

Os objetivos deste trabalho foram avaliar o desempenho germinativo de sementes de palmeira-australiana em diferentes substratos e estabelecer a granulometria e intensidade de umedecimento adequado. A semeadura foi realizada com quatro repetições de 25 sementes em solo, areia, rolo de papel e em vermiculita de diferentes granulometrias: mícron (0,15-0,20 mm), superfina (0,21-0,30 mm), fina (0,30-0,50 mm) e média (1,19-0,50 mm), umedecidas com 0,5; 1,0; 1,5; e 2,0 vezes o seu peso em água. O teste de germinação foi conduzido a 20-30 °C, avaliando-se a primeira contagem do teste aos sete dias após a semeadura e, semanalmente, a germinação (plântulas normais) até os 35 dias, quando foram contabilizadas, também, as plântulas anormais e as sementes mortas. Calcularam-se o tempo médio e frequência relativa de germinação. A vermiculita mícron umedecida com uma vez o seu peso em água apresentou o melhor desempenho como substrato para o teste de germinação de sementes de palmeira-real-australiana, por possibilitar a máxima germinação das sementes (90%) e velocidade de germinação, demandando tempo médio de 15,3 dias nesse processo.

Venous ulcers (VU) is a chronic injury of the lower extremities and because of its high incidence and recurrence implies long and complex treatments, damaging the quality of life (QOL) and self-esteem (SE) of the people. This study aimed to analyze the association between self-esteem with the quality of life of people with venous ulcers treated in primary care. Cross sectional analytical study with a quantitative approach conducted with 44 people met with VU at 13 primary care units 2 and Mixed units in Natal/RN. The study was approved by the Ethics Committee in Research of the Federal University of Rio Grande do Norte (UFRN), CAAE: 07556312.0.0000.5537. Held data collection from February to April 2014 and used three instruments: a structured form covering sociodemographic, health care and clinical variables, the Rosenberg Self-Esteem Scale and the SF-36. The collected data were entered into a database and processed on computerized software for descriptive and inferential analyzes. The results showed a predominance of people with UV females (65,9%), with more than 60 years (59,1%), married or in a stable relationship (52,3%), low education (86,4%) without occupation (68,2%) and less than one minimum wage income (81,8%). Regarding assistance characteristics was observed that most patients performed the dressing with appropriate material (72,7%), professional or trained caregiver (61,4%) did not use compression therapy (81,8%), treating the injury for more than 6 months (77,3%), lack of guidelines for the use of compression therapy, elevation of legs, and regular exercise (77,3%) and consulting the angiologist last year (52,3% ). Regarding clinical features of the lesion was found that most of the recurrent lesions are (77,3%), over one year of current lesion (52,3%) medium to large lesions (54,8%), without signs of infection (61,3%) and pain (79,5%). The mean SE of respondents was 9,3 (± 5,1). The relations between the SE and the sociodemographic variables, health care and clinics showed that individuals without a partner (a) (p = 0,01), who did not wear compression therapy (p = 0,04), with more 6 months of treatment (p = 0,01) and larger lesions (p = 0,01) had a lower SE. The mean domain and the dimensions of the SF-36 were lower emphasizing the functional capacity 36.5 (± 27,6) and the physical aspects of 15.3 (± 30,6). There were significant correlations between AE people with VU and the domains and dimensions of the SF-36: physical functioning (r = -0,432), general health (r = -0,415), vitality (r = -0,573), aspects social (r = -0,517), mental health (r = -0,612) and mental health dimensions (r = -0,612) and physical health (r = -0,473). Based on these results it is concluded by rejecting the null hypothesis and accept the alternative proposed in the study in which it was found that there is a negative correlation between the SE and the QOL of people with venous ulcers

A resistência mecânica do solo à penetração exerce grande influência sobre o desenvolvimento vegetal, uma vez que o crescimento das raízes e a produtividade das culturas variam de forma inversamente proporcional ao seu valor. O objetivo do trabalho foi estudar a variabilidade espacial e a correlação entre a produtividade do milho (PG) e a resistência mecânica do solo à penetração (RP) de um Latossolo Vermelho sob preparo convencional, em área pertencente à Fazenda Experimental de Ensino e Pesquisa da Faculdade de Engenharia - FE/UNESP - Câmpus de Ilha Solteira. Foram definidos 133 pontos de amostragem no campo, segundo malha com espaçamento regular de 10,0 m, nos quais foram medidas a RP, a umidade gravimétrica (UG), nas camadas de 0-0,10; 0,10-0,20; 0,20-0,30 e 0,30-0,40 m, e a PG. O modelo exponencial mostrou-se adequado para descrever todos os semivariogramas experimentais construídos. A RP e a UG apresentaram moderada dependência espacial, com alcance variando entre 20,2 e 63,0 m. A PG apresentou forte dependência espacial com alcance de 15,3 m. A RP não apresentou correlação espacial com a PG. Valores de RP variando entre 0,9 e 2,0 MPa não restringiram a PG.

A análise fatorial de componentes principais (CP) foi usada no exame do relacionamento entre variáveis de um banco de dados da Empresa Brasileira de Pesquisa Agropecuária (EMBRAPA) Gado de Leite. As variáveis disponíveis foram relacionadas às vacas (dias em lactação, teores lácteos de gordura e extrato seco total, produção de leite, ordem de lactação, peso vivo e grau de sangue), ao manejo (dia de pastejo, disponibilidade e períodos de descanso da pastagem), ao ambiente (estação do ano, precipitação pluviométrica) e ao alimento (consumo de nutrientes do concentrado e da cana × uréia, consumo de MS de pastagem de capim-elefante, composição química e digestibilidade in vitro da pastagem e concentração fecal de PB, FDN e FDA). O primeiro CP (33,7% da inércia dos dados) representou o uso da suplementação volumosa (cana × uréia) da pastagem em resposta à redução sazonal da disponibilidade e do consumo de capim-elefante. O segundo CP (15,3% da inércia) foi relacionado ao consumo de nutrientes do concentrado. O terceiro CP (8,5% da inércia) representou efeitos do manejo sobre a composição química da pastagem. A interpretação gráfica dos resultados favoreceu a percepção mais dinâmica da intensidade da associação e do antagonismo entre as variáveis contextualizadas no estudo.

Morbidly obese patients present an increase in heart rate, blood pressure and perceived exertion besides lower walking ability compared to normal weight people. However, little is known about how these variables are presented after bariatric surgery. Moreover, despite the distance walked during the six-minute walk (6MWT) improve after surgery is not well established if the level of physical activity influences this improvement. Objective: To evaluate cardiovascular performance, perceived effort, ability of walking and physical activity level of patients with morbid obesity before and after bariatric surgery. Methods: The cardiovascular performance, perception of effort, the ability to walk and level of physical activity were assessed in 22 patients before (BMI = 50.4 kg/m2) and after (BMI = 34.8 kg/m2) bariatric surgery through the 6MWT. The heart rate, blood pressure and perceived exertion were assessed at rest, at the end of the 6MWT and in the second minute post-test (HR recovery). The ability to walk was measured by total distance walked at the end of the test while the level of physical activity was estimated by applying the Baecke questionnaire, analyzing domains occupation, leisure and locomotion and leisure and physical activity. Results: The HR at rest and recovery decreased significantly (91.2 ± 15.8 bpm vs. 71.9 ± 9.8 bpm, 99.5 ± 15.3 bpm vs 82.5 ± 11.1 bpm, respectively), as well as all the arterial pressure and perceived exertion after surgery. The distance achieved by the patients increased by 58.4 m (p = 0.001) postoperatively. Time postoperatively had correlation with the percentage of excess weight lost (r = 0.48, p = 0.02), BMI (r =- 0.68, p = 0.001) and the Baecke (r = 0.52, p = 0.01) which did not happen with the distance walked (r = 0.37, p = 0.09). Despite weight loss, patients showed no difference in the level of physical activity in any of the areas before and after surgery. Conclusion: The cardiovascular performance, the perception of effort and ability to walk seem to improve after bariatric surgery. However, despite improvement in the ability to walk by the distance achieved in the 6MWT after weight loss, this is not reflected in an increase in physical activity level of obese patients after surgery

Recently, genetically encoded optical indicators have emerged as noninvasive tools of high spatial and temporal resolution utilized to monitor the activity of individual neurons and specific neuronal populations. The increasing number of new optogenetic indicators, together with the absence of comparisons under identical conditions, has generated difficulty in choosing the most appropriate protein, depending on the experimental design. Therefore, the purpose of our study was to compare three recently developed reporter proteins: the calcium indicators GCaMP3 and R-GECO1, and the voltage indicator VSFP butterfly1.2. These probes were expressed in hippocampal neurons in culture, which were subjected to patchclamp recordings and optical imaging. The three groups (each one expressing a protein) exhibited similar values of membrane potential (in mV, GCaMP3: -56 ±8.0, R-GECO1: -57 ±2.5; VSFP: -60 ±3.9, p = 0.86); however, the group of neurons expressing VSFP showed a lower average of input resistance than the other groups (in Mohms, GCaMP3: 161 ±18.3; GECO1-R: 128 ±15.3; VSFP: 94 ±14.0, p = 0.02). Each neuron was submitted to current injections at different frequencies (10 Hz, 5 Hz, 3 Hz, 1.5 Hz, and 0.7 Hz) and their fluorescence responses were recorded in time. In our study, only 26.7% (4/15) of the neurons expressing VSFP showed detectable fluorescence signal in response to action potentials (APs). The average signal-to-noise ratio (SNR) obtained in response to five spikes (at 10 Hz) was small (1.3 ± 0.21), however the rapid kinetics of the VSFP allowed discrimination of APs as individual peaks, with detection of 53% of the evoked APs. Frequencies below 5 Hz and subthreshold signals were undetectable due to high noise. On the other hand, calcium indicators showed the greatest change in fluorescence following the same protocol (five APs at 10 Hz). Among the GCaMP3 expressing neurons, 80% (8/10) exhibited signal, with an average SNR value of 21 ±6.69 (soma), while for the R-GECO1 neurons, 50% (2/4) of the neurons had signal, with a mean SNR value of 52 ±19.7 (soma). For protocols at 10 Hz, 54% of the evoked APs were detected with GCaMP3 and 85% with R-GECO1. APs were detectable in all the analyzed frequencies and fluorescence signals were detected from subthreshold depolarizations as well. Because GCaMP3 is the most likely to yield fluorescence signal and with high SNR, some experiments were performed only with this probe. We demonstrate that GCaMP3 is effective in detecting synaptic inputs (involving Ca2+ influx), with high spatial and temporal resolution. Differences were also observed between the SNR values resulting from evoked APs, compared to spontaneous APs. In recordings of groups of cells, GCaMP3 showed clear discrimination between activated and silent cells, and reveals itself as a potential tool in studies of neuronal synchronization. Thus, our results indicate that the presently available calcium indicators allow detailed studies on neuronal communication, ranging from individual dendritic spines to the investigation of events of synchrony in neuronal networks genetically defined. In contrast, studies employing VSFPs represent a promising technology for monitoring neural activity and, although still to be improved, they may become more appropriate than calcium indicators, since neurons work on a time scale faster than events of calcium may foresee

BackgroundDetection and quantification of hepatitis C virus (HCV) RNA is integral to diagnostic and therapeutic regimens. All molecular assays target the viral 5'-noncoding region (59-NCR), and all show genotype-dependent variation of sensitivities and viral load results. Non-western HCV genotypes have been under-represented in evaluation studies. An alternative diagnostic target region within the HCV genome could facilitate a new generation of assays.Methods and FindingsIn this study we determined by de novo sequencing that the 3'-X-tail element, characterized significantly later than the rest of the genome, is highly conserved across genotypes. To prove its clinical utility as a molecular diagnostic target, a prototype qualitative and quantitative test was developed and evaluated multicentrically on a large and complete panel of 725 clinical plasma samples, covering HCV genotypes 1-6, from four continents (Germany, UK, Brazil, South Africa, Singapore). To our knowledge, this is the most diversified and comprehensive panel of clinical and genotype specimens used in HCV nucleic acid testing (NAT) validation to date. The lower limit of detection (LOD) was 18.4 IU/ml (95% confidence interval, 15.3-24.1 IU/ml), suggesting applicability in donor blood screening. The upper LOD exceeded 10(-9) IU/ml, facilitating viral load monitoring within a wide dynamic range. In 598 genotyped samples, quantified by Bayer VERSANT 3.0 branched DNA (bDNA), X-tail-based viral loads were highly concordant with bDNA for all genotypes. Correlation coefficients between bDNA and X-tail NAT, for genotypes 1-6, were: 0.92, 0.85, 0.95, 0.91, 0.95, and 0.96, respectively; X-tail-based viral loads deviated by more than 0.5 log10 from 5'-NCR-based viral loads in only 12% of samples (maximum deviation, 0.85 log10). The successful introduction of X-tail NAT in a Brazilian laboratory confirmed the practical stability and robustness of the X-tail-based protocol. The assay was implemented at low reaction costs (US$8.70 per sample), short turnover times (2.5 h for up to 96 samples), and without technical difficulties.ConclusionThis study indicates a way to fundamentally improve HCV viral load monitoring and infection screening. Our prototype assay can serve as a template for a new generation of viral load assays. Additionally, to our knowledge this study provides the first open protocol to permit industry-grade HCV detection and quantification in resource-limited settings.

Background: The purpose of this study was to compare laboratory and clinical outcomes of intracytoplasmic morphologically selected sperm injection (IMSI) and conventional intracytoplasmic sperm injection (ICSI) in couples with repeated implantation failures.Methods: A total of 200 couples with at least two prior unsuccessful ICSI cycles were enrolled: 100 couples were submitted to IMSI and 100 were submitted to routine ICSI. For IMSI, spermatozoa were selected at 8400x magnification using an inverted microscope equipped with Nomarski (differential interference contrast) optics. For conventional ICSI, spermatozoa were selected at 400x magnification. Clinical outcomes were evaluated between the two groups.Results: Study patients were comparable in age, number of treatment failures, aetiology of infertility, percentage of normal form assessed by MSOME (motile sperm organelle morphology examination), semen parameters, total number of oocytes collected, number of mature oocytes collected, total number of embryos transferred and number of high-quality embryos transferred. No statistically significant differences between the two groups were observed with regard to rates of fertilisation, implantation and pregnancy/cycle. Although not statistically significant, rates of miscarriage (IMSI:15.3% vs ICSI:31.7%), ongoing pregnancy (IMSI:22% vs ICSI:13%) and live births (IMSI:21% vs ICSI:12%) showed a trend towards better outcomes in the IMSI group. In addition, analysis of subpopulations with or without male factor showed similar results.Conclusions: Our results suggest that IMSI does not provide a significant improvement in clinical outcome compared to ICSI, at least in couples with repeated implantation failures after conventional ICSI. However, it should be noted that there were clear trends for lower miscarriage rates (approximate to 50% reduced) and higher rates of ongoing pregnancy and live births (both nearly doubled) within the IMSI group. Further confirmation as well as randomized large-scale trials are needed to confirm the beneficial effects of IMSI in couples with poor reproductive prognoses.

Leptospirosis and toxoplasmosis are zoonoses with high importance because of the economic and public health impact. This study was aimed to determine the seroprevafence of leptospirosis and toxoplamosis in 714 serum samples of horses from different farms from Botucatu, São Paulo, Brazil. The samples were researched for Toxoplasma gondii antibodies by indirect immunofluorescence antibody test (IFAT) and for Leptospira spp. antibodies by microscopic agglutination test. of 714 serum samples, 128 (17.9%; 95% Cl: 15.3%-20.9%) were positive for one or more serovars of Leptospira spp., with icterohaemorrhagiae, canicola, and castellonis as the most prevalent serovars, whereas 42 (5.9%; 95% CI: 4.4%-7.9%) were positive for T gondii, of which 33 samples (78.57%; 95% CI: 64.0%-88.2%) presented a titer of 16, 7 (16.7%; 95% CI: 8.4%-30.7%) a titer of 64, and 1 (2.38%; 95% CI: 0.6%-12.3%) a titer of 256. No significant difference was found among the results obtained and the associated variables such as age and sex. (C) 2012 Elsevier B.V. All rights reserved.
