Fifty years have passed since Cyert and March’s 1963 A Behavioral Theory of the Firm. During this time, BTOF has been adopted across different research domains to investigate how organizations set goals, how they determine aspirations and how they finally react to performance aspiration discrepancies. Cyert and March’s framework has also recently emerged as one of the dominant paradigms to understand the ways in which family business organizations make decisions. In this chapter, I review the theoretical development and empirical results of BTOF and its application in the family business field of study in order to identify theoretical and empirical gaps and propose suggestions for future research. The conclusions suggest that BTOF is both a theoretically and empirically valid perspective in family business research, particularly when combined with other theoretical frameworks. The principal recommendation is to apply behavioral theory to enhance scholarly understanding of how family organisations define their aspiration levels and respond to organizational problems.

L’objectif de cet essai repose sur l’élaboration d’un plan de gestion des matières résiduelles pour la communauté algonquine de Long Point First Nation, située dans la Municipalité régionale de comté de Témiscamingue. La Politique québécoise de gestion des matières résiduelles n’établit pas clairement à qui revient la responsabilité d’une saine gestion dans le cas des communautés autochtones. Le flou législatif derrière cet enjeu ne fait qu’accentuer l’isolement de ces communautés dans leur aspiration à développer des actions précises et à les concrétiser. Pourtant, le milieu autochtone se doit de poursuivre la voie de l’amélioration continue de son administration à ce sujet puisque celui-ci participe également au bilan provincial. Cet essai s’inscrit dans le but précis d’accompagner Long Point First Nation dans son cheminement vers une gestion éclairée de ses matières résiduelles. La communauté de Long Point First Nation a introduit la collecte sélective depuis quelques années déjà et en assure elle-même sa gestion. Néanmoins, elle est aux prises avec des situations problématiques limitant la gestion adéquate de ses matières résiduelles. L’éloignement géographique, les partenariats laborieux, les coûts élevés reliés à la gestion et l’instabilité au niveau de la collecte hebdomadaire freinent sa facilité administrative et son déploiement. L’usage des dépôts sauvages pour éliminer les matières non collectées amplifie les problèmes environnementaux et de santé publique. La désuétude de certaines installations et l’absence de campagnes usuelles d’information, de sensibilisation et d’éducation n’assurent pas une gestion optimale ni sa pérennité. Un plan d’action a été réalisé dans le but ultime de contrer ces enjeux. Ce plan priorise deux objectifs fondamentaux, celui d’instaurer un système de récupération des matières organiques au niveau résidentiel et celui de développer des infrastructures pouvant accueillir les matières résiduelles non collectées par la collecte sélective actuellement en place. En plus de ces priorités, cinq autres objectifs sont mis de l’avant, soit le regroupement d’informations concernant la gestion des matières résiduelles et son suivi, la qualité de gestion de la collecte municipale, l’assurance de la qualité environnementale de son territoire, l’affinement du tri des matières recyclables et l’amélioration de la transmission d’informations auprès des résidents. Le tout représente un essai se voulant pragmatique colligeant objectifs et mesures afin d’élaborer un plan d’action réaliste à caractère environnemental.

Idiopathic pulmonary fibrosis (IPF) is a chronic progressive disease with no curative pharmacological treatment. Animal models play an essential role in revealing molecular mechanisms involved in the pathogenesis of the disease. Bleomycin (BLM)-induced lung fibrosis is the most widely used and characterized model for anti-fibrotic drugs screening. However, several issues have been reported, such as the identification of an optimal BLM dose and administration scheme as well as gender-specificity. Moreover, the balance between disease resolution, an appropriate time window for therapeutic intervention and animal welfare remains critical aspects yet to be fully elucidated. In this thesis, Micro CT imaging has been used as a tool to identify the ideal BLM dose regimen to induce sustained lung fibrosis in mice as well as to assess the anti-fibrotic effect of Nintedanib (NINT) treatment upon this BLM administration regimen. In order to select the optimal BLM dose scheme, C57bl/6 male mice were treated with BLM via oropharyngeal aspiration (OA), following either double or triple BLM administration. The triple BLM administration resulted in the most promising scheme, able to balance disease resolution, appropriate time-window for therapeutic intervention and animal welfare. The fibrosis progression was longitudinally assessed by micro-CT every 7 days for 5 weeks after BLM administration and 5 animals were sacrificed at each timepoint for the BALF and histological evaluation. The antifibrotic effect of NINT was assessed following different treatment regimens in this model. Herein, we have developed an optimized mouse model of pulmonary fibrosis, enabling three weeks of the therapeutic window to screen putative anti-fibrotic drugs. micro-CT scanning, allowed us to monitor the progression of lung fibrosis and the therapeutical response longitudinally in the same subject, drastically reducing the number of animals involved in the experiment.

Biomarkers are biological indicators of human health conditions. Their ultra-sensitive quantification is of cardinal importance in clinical monitoring and early disease diagnosis. Biosensors are some worldwide simple and easy-to-use analytical devices as a matter of fact, biosensors using electrochemiluminescence (ECL) are one of the most promising biosensors that needs an ever-increasing sensitivity for improving its clinical effectiveness. The principal aspiration of this project is the investigation of the ECL generation mechanisms for enhancing the ECL intensity and the development of an ultrasensitive sensor, the use of metal-oxide materials (Mox) and the substitution of metal-free dyes. Novel dyes such as BODIPY, TADF are used to improve the sensitivity of ECL techniques thanks to their advantageous and tunable properties, enhancing the signal and also the ECL efficiency. Additionally, the use of Mox could be beneficial for the investigation of two different ECL mechanisms, which occur simultaneously. In this thesis, the investigation of size and distance effects on electrochemical (EC) mechanisms was carried out through the innovative combination of a standard detection system using different size of micromagnetic beads (MBs). That allowed the discovery of an unexpected and highly efficient mechanistic path for electrochemical generation at small distances from the electrode’s surface. The smallest MBs (0.1μm) demostrate an enhancement of electrochemical signal than the bigger one (2.8μm) until 4 times of magnitude. Finally, a novel ultrasensitive sensor, based on the coreactant-luminophores mechanism, was developed for the determination of whole viral genome specific for cardiac HBV and COVID-19 virus. In conclusion, the ECL and the use of EC techniques (such as amperometry), improved the understanding of mechanisms responsible for the ECL/EC signal led to a great enhancement in the signal.

Il Mieloma Multiplo (MM) è una patologia neoplastica delle cellule B caratterizzata dalla proliferazione di più cloni di plasmacellule portatrici di diverse anomalie genomiche. Il MM presenta tipicamente un’eterogeneità genomica spaziale e intraclonale, che rende l’aspirato midollare "a singolo sito", attualmente utilizzato per la valutazione della malattia residua (MRD) dopo trattamento, non realmente informativo sulla taglia di malattia e sul panorama genomico della malattia. In considerazione della crescente importanza che sta assumendo la valutazione della MRD, i test per monitorarla dovrebbero essere non invasivi, affidabili e in grado di rappresentare le eterogeneità che caratterizzano il MM. Il presente studio ha permesso di dimostrare la possibilità di utilizzare la biopsia liquida, una metodica innovativa e non invasiva, per caratterizzare i pazienti con MM attivo o con MM smoldering ad alto rischio di evoluzione (HR-SMM) e per determinale l’MRD nei pazienti sottoposti a terapia di prima linea, integrando le metodiche attualmente validate. Nei pazienti arruolati nel presente studio è stato possibile identificare la frazione tumorale di DNA libero circolante (cfDNA-TF) nel sangue periferico, ed è stato possibile caratterizzare la malattia da un punto di vista qualitativo, dimostrando un’elevata concordanza del profilo genomico tra DNA libero circolante e DNA midollare (100% nei pazienti con HR-SMM e 86% nei pazienti con MM attivo). L’esecuzione seriata di biopsie liquide in corso di terapia, con un follow-up mediano di 24 mesi, ha mostrato una rapida e netta riduzione della cfDNA-TF xdalle prime fasi di terapia, con una tendenza a mantenersi mediamente sotto la soglia di sensibilità della metodica anche nelle fasi successive, indipendentemente dall’eventuale persistenza di MRD individuabile a livello midollare o mediante PET-CT. Con un follow-up più lungo probabilmente sarà possibile valutare meglio la capacità di questa metodica di affiancare o eventualmente sostituire l’aspirato midollare.
