Purpose: Mossy fiber sprouting (MFS) is a frequent finding following status epilepticus (SE). The present study aimed to test the feasibility of using manganese-enhanced magnetic resonance imaging (MEMRI) to detect MFS in the chronic phase of the well-established pilocarpine (Pilo) rat model of temporal lobe epilepsy (TLE). Methods: To modulate MFS, cycloheximide (CHX), a protein synthesis inhibitor, was coadministered with Pilo in a subgroup of animals. In vivo MEMRI was performed 3 months after induction of SE and compared to the neo-Timm histologic labeling of zinc mossy fiber terminals in the dentate gyrus (DG). Key Findings: Chronically epileptic rats displaying MFS as detected by neo-Timm histology had a hyperintense MEMRI signal in the DG, whereas chronically epileptic animals that did not display MFS had minimal MEMRI signal enhancement compared to nonepileptic control animals. A strong correlation (r = 0.81, p < 0.001) was found between MEMRI signal enhancement and MFS. Significance: This study shows that MEMRI is an attractive noninvasive method for detection of mossy fiber sprouting in vivo and can be used as an evaluation tool in testing therapeutic approaches to manage chronic epilepsy.

Background: Neutrophils have an impressive array of microbicidal weapons, and in the presence of a pathogen, progress from a quiescent state in the bloodstream to a completely activated state. Failure to regulate this activation, for example, when the blood is flooded with cytokines after severe trauma, causes inappropriate neutrophil activation that paradoxically, is associated with tissue and organ damage. Acidic proteomic maps of quiescent human neutrophils were analyzed and compared to those of activated neutrophils from severe trauma patients. The analysis revealed 114 spots whose measured volumes differed between activated and quiescent neutrophils, with 27 upregulated and 87 downregulated in trauma conditions. Among the identified proteins, grancalcin, S100-A9 and CACNB2 reinforce observed correlations between motility and ion flux, ANXA3, SNAP, FGD1 and Zfyve19 are involved in vesicular transport and exocytosis, and GSTP1, HSPA1 HSPA1L, MAOB, UCH-L5, and PPA1 presented evidence that activated neutrophils may have diminished protection against oxidative damage and are prone to apoptosis. These are discussed, along with proteins involved in cytoskeleton reorganization, reactive oxygen species production, and ion flux. Proteins such as Zfyve19, MAOB and albumin-like protein were described for the first time in the neutrophil. In this work we achieved the identification of several proteins potentially involved in inflammatory signaling after trauma, as well as proteins described for the first time in neutrophils.

Um questionamento muito frequente: qual o tempo que se deve esperar para movimentar um dente submetido a tratamento endodôntico, inclusive os de perfuração radicular? A extrapolação dos fenômenos observados em outras regiões da raiz e a fundamentação experimental com base em situações correlatas permitem afirmar que 30 dias correspondem a um período mais do que razoável para o reparo periapical estar em fase avançada de maturação e síntese. As forças ortodônticas são muito leves e dissipantes - muito mais do que o traumatismo dentário, o trauma oclusal e as forças mastigatórias normais -, e não devem interferir na patogenicidade e virulência das microbiotas envolvidas nas necroses e lesões periapicais crônicas, assim como não devem interferir nos fenômenos celulares e teciduais durante a reorganização dos tecidos apicais e periapicais.
Regulation of corticosterone function during early weaning and effects on gastric cell proliferation

Objectives: The development of the gastrointestinal tract depends on many elements, including glucocorticoids. In the current study, we evaluated the effects of early weaning on corticosterone function and the growth of rat gastric mucosa. Methods: By using Wistar rats submitted to early weaning at 15 d, we analyzed plasma corticosterone, corticosteroid-binding globulin (CBG), and glucocorticoid receptor (GR) distribution in the gastric epithelium. Results: With the use of radioimmunoassay, we found that early weaning increased corticosterone concentration at day 16 and 17 in test subjects as compared with controls, whereas it was equivalent between groups at day 18. CBG binding capacity decreased during treatment, and it was significantly lower at day 18. At this age, GR levels and distribution in the gastric mucosa were also reduced as compared with suckling counterparts. To reduce corticosterone activity during early weaning and to explore cell proliferation responses, we administered RU486 to 15-d-old pups. We found that cytoplasmic GR reached a peak after 48 h, whereas nuclear levels remained constant, thereby confirming the inhibition of receptor function. Next, by checking gastric proliferative responses, we observed that RU486 induced higher DNA synthesis and mitotic indices in test subjects as compared with control groups. Conclusions: We demonstrated that early weaning changed corticosterone activity by increasing hormone levels, reducing CBG binding capacity, and decreasing GR distribution in the gastric epithelium. These modifications seem to be important to the reorganization of gastric growth after the abrupt interruption of suckling.

Large scale wireless adhoc networks of computers, sensors, PDAs etc. (i.e. nodes) are revolutionizing connectivity and leading to a paradigm shift from centralized systems to highly distributed and dynamic environments. An example of adhoc networks are sensor networks, which are usually composed by small units able to sense and transmit to a sink elementary data which are successively processed by an external machine. Recent improvements in the memory and computational power of sensors, together with the reduction of energy consumptions, are rapidly changing the potential of such systems, moving the attention towards datacentric sensor networks. A plethora of routing and data management algorithms have been proposed for the network path discovery ranging from broadcasting/floodingbased approaches to those using global positioning systems (GPS). We studied WGrid, a novel decentralized infrastructure that organizes wireless devices in an adhoc manner, where each node has one or more virtual coordinates through which both message routing and data management occur without reliance on either flooding/broadcasting operations or GPS. The resulting adhoc network does not suffer from the deadend problem, which happens in geographicbased routing when a node is unable to locate a neighbor closer to the destination than itself. WGrid allow multidimensional data management capability since nodes' virtual coordinates can act as a distributed database without needing neither special implementation or reorganization. Any kind of data (both single and multidimensional) can be distributed, stored and managed. We will show how a location service can be easily implemented so that any search is reduced to a simple query, like for any other data type. WGrid has then been extended by adopting a replication methodology. We called the resulting algorithm WRGrid. Just like WGrid, WRGrid acts as a distributed database without needing neither special implementation nor reorganization and any kind of data can be distributed, stored and managed. We have evaluated the benefits of replication on data management, finding out, from experimental results, that it can halve the average number of hops in the network. The direct consequence of this fact are a significant improvement on energy consumption and a workload balancing among sensors (number of messages routed by each node). Finally, thanks to the replications, whose number can be arbitrarily chosen, the resulting sensor network can face sensors disconnections/connections, due to failures of sensors, without data loss. Another extension to {WGrid} is {W*Grid} which extends it by strongly improving network recovery performance from link and/or device failures that may happen due to crashes or battery exhaustion of devices or to temporary obstacles. W*Grid guarantees, by construction, at least two disjoint paths between each couple of nodes. This implies that the recovery in W*Grid occurs without broadcasting transmissions and guaranteeing robustness while drastically reducing the energy consumption. An extensive number of simulations shows the efficiency, robustness and traffic road of resulting networks under several scenarios of device density and of number of coordinates. Performance analysis have been compared to existent algorithms in order to validate the results.

Self-incompatibility (SI) systems have evolved in many flowering plants to prevent self-fertilization and thus promote outbreeding. Pear and apple, as many of the species belonging to the Rosaceae, exhibit RNase-mediated gametophytic self-incompatibility, a widespread system carried also by the Solanaceae and Plantaginaceae. Pear orchards must for this reason contain at least two different cultivars that pollenize each other; to guarantee an efficient cross-pollination, they should have overlapping flowering periods and must be genetically compatible. This compatibility is determined by the S-locus, containing at least two genes encoding for a female (pistil) and a male (pollen) determinant. The female determinant in the Rosaceae, Solanaceae and Plantaginaceae system is a stylar glycoprotein with ribonuclease activity (S-RNase), that acts as a specific cytotoxin in incompatible pollen tubes degrading cellular RNAs. Since its identification, the S-RNase gene has been intensively studied and the sequences of a large number of alleles are available in online databases. On the contrary, the male determinant has been only recently identified as a pollen-expressed protein containing a F-box motif, called S-Locus F-box (abbreviated SLF or SFB). Since F-box proteins are best known for their participation to the SCF (Skp1 - Cullin - F-box) E3 ubiquitine ligase enzymatic complex, that is involved in protein degradation through the 26S proteasome pathway, the male determinant is supposed to act mediating the ubiquitination of the S-RNases, targeting them for the degradation in compatible pollen tubes. Attempts to clone SLF/SFB genes in the Pyrinae produced no results until very recently; in apple, the use of genomic libraries allowed the detection of two F-box genes linked to each S haplotype, called SFBB (S-locus F-Box Brothers). In Japanese pear, three SFBB genes linked to each haplotype were cloned from pollen cDNA. The SFBB genes exhibit S haplotype-specific sequence divergence and pollen-specific expression; their multiplicity is a feature whose interpretation is unclear: it has been hypothesized that all of them participate in the S-specific interaction with the RNase, but it is also possible that only one of them is involved in this function. Moreover, even if the S locus male and female determinants are the only responsible for the specificity of the pollen-pistil recognition, many other factors are supposed to play a role in GSI; these are not linked to the S locus and act in a S-haplotype independent manner. They can have a function in regulating the expression of S determinants (group 1 factors), modulating their activity (group 2) or acting downstream, in the accomplishment of the reaction of acceptance or rejection of the pollen tube (group 3). This study was aimed to the elucidation of the molecular mechanism of GSI in European pear (Pyrus communis) as well as in the other Pyrinae; it was divided in two parts, the first focusing on the characterization of male determinants, and the second on factors external to the S locus. The research of S locus F-box genes was primarily aimed to the identification of such genes in European pear, for which sequence data are still not available; moreover, it allowed also to investigate about the S locus structure in the Pyrinae. The analysis was carried out on a pool of varieties of the three species Pyrus communis (European pear), Pyrus pyrifolia (Japanese pear), and Malus × domestica (apple); varieties carrying S haplotypes whose RNases are highly similar were chosen, in order to check whether or not the same level of similarity is maintained also between the male determinants. A total of 82 sequences was obtained, 47 of which represent the first S-locus F-box genes sequenced from European pear. The sequence data strongly support the hypothesis that the S locus structure is conserved among the three species, and presumably among all the Pyrinae; at least five genes have homologs in the analysed S haplotypes, but the number of F-box genes surrounding the S-RNase could be even greater. The high level of sequence divergence and the similarity between alleles linked to highly conserved RNases, suggest a shared ancestral polymorphism also for the F-box genes. The F-box genes identified in European pear were mapped on a segregating population of 91 individuals from the cross 'Abbé Fétel' × 'Max Red Bartlett'. All the genes were placed on the linkage group 17, where the S locus has been placed both in pear and apple maps, and resulted strongly associated to the S-RNase gene. The linkage with the RNase was perfect for some of the F-box genes, while for others very rare single recombination events were identified. The second part of this study was focused on the research of other genes involved in the SI response in pear; it was aimed on one side to the identification of genes differentially expressed in compatible and incompatible crosses, and on the other to the cloning and characterization of the transglutaminase (TGase) gene, whose role may be crucial in pollen rejection. For the identification of differentially expressed genes, controlled pollinations were carried out in four combinations (self pollination, incompatible, half-compatible and fully compatible cross-pollination); expression profiles were compared through cDNA-AFLP. 28 fragments displaying an expression pattern related to compatibility or incompatibility were identified, cloned and sequenced; the sequence analysis allowed to assign a putative annotation to a part of them. The identified genes are involved in very different cellular processes or in defense mechanisms, suggesting a very complex change in gene expression following the pollen/pistil recognition. The pool of genes identified with this technique offers a good basis for further study toward a better understanding of how the SI response is carried out. Among the factors involved in SI response, moreover, an important role may be played by transglutaminase (TGase), an enzyme involved both in post-translational protein modification and in protein cross-linking. The TGase activity detected in pear styles was significantly higher when pollinated in incompatible combinations than in compatible ones, suggesting a role of this enzyme in the abnormal cytoskeletal reorganization observed during pollen rejection reaction. The aim of this part of the work was thus to identify and clone the pear TGase gene; the PCR amplification of fragments of this gene was achieved using primers realized on the alignment between the Arabidopsis TGase gene sequence and several apple EST fragments; the full-length coding sequence of the pear TGase gene was then cloned from cDNA, and provided a precious tool for further study of the in vitro and in vivo action of this enzyme.

Die Kapsidproteine L1 und L2 von humanen Papillomviren (HPV) werden im Cytoplasma infizierter Keratinocyten synthetisiert und gelangen unabhängig voneinander in den Kern (Florin et al. 2002b). L2 lokalisiert in speziellen Kerndomänen, sog. ND10, und induziert die Reorganisation dieser Kernstrukturen: L2-abhängig akkumuliert der transkriptionelle Modulator Daxx verstärkt in ND10 und außerdem kommt es zum Ausschluss des transkriptionellen Aktivators Sp100 aus diesen Domänen (Florin et al. 2002a). Im Anschluss an diese Umorganisation im Kern induziert L2 die Lokalisation des Kapsidproteins L1 in ND10 (Florin et al. 2002b). Da auch die Replikation und Transkription von Papillomviren in oder in unmittelbarer Nähe von ND10 stattfinden, werden ND10 als Orte der Papillomvirus-Morphogenese diskutiert (Swindle et al. 1999). Innerhalb dieser Arbeit konnte gezeigt werden, dass L1 und L2 im Cytoplasma der Zellen mit Chaperonen interagieren, und dass der Kerntransport von L2 von der L2/Hsc70-Assoziation abhängig ist. Hsc70, das mit dem C-Terminus von L2 assoziiert ist, wird in virusähnliche Partikel (VLPs) eingebaut. Erst durch die Verpackung von DNA in die Kapside kommt es zum Ausschluss von Hsc70 aus dem Papillomvirus-Kapsid. Ergebnisse dieser Arbeit lassen zudem vermuten, dass L2 über seinen C-Terminus mit Mikrotubuli interagieren kann, falls diese Aminosäure-Region in L2 nicht durch das Chaperon maskiert wird. Mit Hilfe dieser Erkenntnisse wurde eine Modellvorstellung für die Rolle von L2 während der Infektion und der Morphogenese von HPV entwickelt. Die ND10-Lokalisationsdomäne (NDLD) in L2 konnte wie bei keinem Protein zuvor auf eine sehr kurze Sequenz von 22 Aminosäuren eingeengt werden. Welcher Mechanismus für die ND10-Lokalisation verantwortlich ist, muss dagegen noch geklärt werden. Alle L2-Mutanten, die ND10-Lokalisation zeigen, induzieren auch die Reorganisation dieser Domänen. Dies spricht dafür, dass L2 direkt in ND10 die Veränderungen hervorruft und wahrscheinlich keine zusätzlichen Domänen in L2 daran beteiligt sind. Es konnten zwei L1-Interaktionsdomänen in L2 kartiert werden. Diese beiden Regionen in L2 konnten nicht genauer lokalisiert werden und umfassen möglicherweise mehrere L1-Interaktionsdomänen. Der Einbau von L2 in die Kapside kann nur im Kern infizierter Zellen stattfinden. Hierfür ist die Lokalisation der Kapsidproteine in ND10 nicht notwendig. Weiterführende Versuche müssen jedoch noch klären, inwieweit ND10 trotzdem unerlässlich für eine produktive Morphogenese sind. Zudem wurde klar, dass die ersten 150 Aminosäuren im L2-Protein für das L1/L2-Verhältnis in Kapsiden verantwortlich sind. In Virionen beträgt dieses Verhältnis 30:1, d. h. zwölf L2-Moleküle werden in die Partikel aus 360 L1-Molekülen eingebaut. Bei der Verwendung der Deletionsmutante L2-150/467 beträgt dieses Verhältnis 5:1. Weitere Analysen, welche Regionen von L1 und L2 miteinander interagieren und wodurch die Beschränkung des L1/L2-Verhältnisses in Papillomviren zustande kommt, können genauere Einblicke in den Aufbau der Kapside und speziell die Lage von L2 im Kapsid liefern.

Die Ränder des Labrador Meeres wurden während des späten Neoproterozoikums intensiv von karbonatreichen silikatischen Schmelzen durchsetzt. Diese Schmelzen bildeted sich bei Drucken zwischen ca. 4-6 GPa (ca. 120-180 km Tiefe) an der Basis der kontinentalen Mantel-Lithosphäre. Diese Magmengenerierung steht in zeitlichem und räumlichem Zusammenhang mit kontinentalen Extensionsprozessen, welche zu beiden Seiten des sich öffnenden Iapetus-Ozeans auftraten.

Grundlage für die hier gezeigte Arbeit stellt die Eigenschaft von amphiphilen Blockcopolymeren dar immer den Block mit der niedrigsten Grenzflächenenergie zum angrenzenden Medium an die Oberfläche zu bringen. Durch einen Austausch des Mediums an der Grenzfläche zum Blockcopolymer kann eine Reorientierung erzwungen werden, wenn die Grenzflächenenergie des anderen Blocks nun die niedrigere Grenzflächenenergie besitzt. Dieses Verhalten von dünnen amphiphilen Blockcopolymerfilmen wurde zur Strukturierung von Oberflächen ausgenutzt und in nachfolgenden Synthesen weiter verstärkt. Um dies zu erreichen wurde das zur Strukturierung erforderliche Poly(4-Octylstyrol)block(4-hydroxystyrol) durch kontrollierte radikalische Polymerisationsmethode mit dem Tempo Unimer (2,2,6,6-Tetramethyl-1-1(1-phenyl-ethoxy)-piperidin) synthetisiert. Für die geplanten Reorientierungen und Modifizierungen von Oberflächen wurden dünne Filme durch Schleuderbeschichtung auf verschiedenen Substraten (Siliziumwafern, Glassubstraten und Goldoberflächen) hergestellt. Das Verhalten der Oberflächen von diesen Filmen wurde durch Kontaktwinkelmessungen untersucht. Auf diese Weise konnte gezeigt werden, dass die Oberfläche von Polymerfilmen nach der Präparation aus dem hydrophoben Block des Polymers gebildet wird. Durch Kontakt des Polymerfilms mit Wasser kann dieser zur Reorientierung gebracht werden, so dass der hydrophile Block des Polymers an der Oberfläche erscheint. Dieses Verhalten wurde zur Strukturierung mit softlithographischen Techniken genutzt. Dazu wurden hydrophil/hydrophob strukturierte Oberflächen durch Aufsetzen von hydrophoben PDMS-Stempeln, die Teile der Oberfläche selektiv abdeckten, und Einbringen von Wasser in die dabei entstehenden Kapillaren hergestellt. Dies ermöglichte es die Oberfläche selektiv im Größenbereich von 500nm bis zu 50µm zu strukturieren und an den reaktiven Bereichen Materialien, wie z.B. Kupfer, Titandioxid, Polyelektrolyte, photonische Kristalle und angegraftete Polymere, mit verschiedenen Methoden selektiv auf die Oberfläche aufzubringen. Um den Reorganisationsprozess der Oberfläche genauer zu studieren, wurde ein für diese Aufgabe besser geeignetes Polymer (Poly(Styrol)-block-poly(essigsäure-2-(2-(4-vinyl-phenoxy)-ethoxy)ethylester)) synthetisiert. Aus diesem Blockcopolymer wurden wieder dünne Filme durch Spincoaten hergestellt. Die Reorientierung dieses Polymers in 70°C warmen Wasser konnte durch Kontaktwinkelmessungen und NEXAFS Spektroskopie nachgewiesen werden. Mit Hilfe der NEXAFS Spektroskopie konnte festgestellt werden, dass die Geschwindigkeit der Reorientierung durch eine exponentielle Funktion beschrieben werden kann. Eine Auswertung der Geschwindigkeitskonstante für die Reorientierung einer hydrophilen zu einer hydrophoben Oberfläche des Polymers bei 60°C führt zu =75min. Aufgrund des exponentiellen Charakters der Reorientierung macht es den Anschein, dass die Reorientierung bei verschiedenen Reorientierungstemperaturen bis zu einem gewissen Grad erfolgt und dann stoppt. Eine weitere Reorientierung scheint erst wieder bei einer Temperaturerhöhung zu beginnen. Aus AFM Messungen ist ein Beginnen der Reorientierung durch Bildung kleiner Löcher in der Polymeroberfläche zu erkennen, die sich zu runden Erhöhungen und Vertiefungen vergrößern, um letztendlich in ein spinodales Entmischungsmuster über zu gehen. Dieses heilt dann im Laufe der Zeit langsam durch Verschwinden der hydrophilen Bereiche langsam aus. Der Beginn des zuvor beschriebenen Reorientierungsprozesses einer hydrophilen Oberfläche in eine hydrophobe konnte sowohl in den AFM, als auch in den NEXAFS-Messungen zu ca. 50°C bestimmt werden.

In this thesis, atomistic simulations are performed to investigate hydrophobic solvation and hydrophobic interactions in cosolvent/water binary mixtures. Many cosolvent/water binary mixtures exhibit non-ideal behavior caused by aggregation at the molecular scale level although they are stable and homogenous at the macroscopic scale. Force-field based atomistic simulations provide routes to relate atomistic-scale structure and interactions to thermodynamic solution properties. The predicted solution properties are however sensitive to the parameters used to describe the molecular interactions. In this thesis, a force field for tertiary butanol (TBA) and water mixtures is parameterized by making use of the Kirkwood-Buff theory of solution. The new force field is capable of describing the alcohol-alcohol, water-water and alcohol-water clustering in the solution as well as the solution components’ chemical potential derivatives in agreement with experimental data. With the new force field, the preferential solvation and the solvation thermodynamics of a hydrophobic solute in TBA/water mixtures have been studied. First, methane solvation at various TBA/water concentrations is discussed in terms of solvation free energy-, enthalpy- and entropy- changes, which have been compared to experimental data. We observed that the methane solvation free energy varies smoothly with the alcohol/water composition while the solvation enthalpies and entropies vary nonmonotonically. The latter occurs due to structural solvent reorganization contributions which are not present in the free energy change due to exact enthalpy-entropy compensation. It is therefore concluded that the enthalpy and entropy of solvation provide more detailed information on the reorganization of solvent molecules around the inserted solute. Hydrophobic interactions in binary urea/water mixtures are next discussed. This system is particularly relevant in biology (protein folding/unfolding), however, changes in the hydrophobic interaction induced by urea molecules are not well understood. In this thesis, this interaction has been studied by calculating the free energy (potential of mean force), enthalpy and entropy changes as a function of the solute-solute distance in water and in aqueous urea (6.9 M) solution. In chapter 5, the potential of mean force in both solution systems is analyzed in terms of its enthalpic and entropic contributions. In particular, contributions of solvent reorganization in the enthalpy and entropy changes are studied separately to better understand what are the changes in interactions in the system that contribute to the free energy of association of the nonpolar solutes. We observe that in aqueous urea the association between nonpolar solutes remains thermodynamically favorable (i.e., as it is the case in pure water). This observation contrasts a long-standing belief that clusters of nonpolar molecules dissolve completely in the presence of urea molecules. The consequences of our observations for the stability of proteins in concentrated urea solutions are discussed in the chapter 6 of the thesis.

Con la prima parte, si intende fornire un quadro pressoché esaustivo delle principali disposizioni in materia di società a partecipazione pubblica regionale e locale operanti nel campo dei servizi pubblici locali e della loro interpretazione giurisprudenziale e dottrinale, prendendo le mosse dagli ultimi interventi legislativo. Nella seconda parte, si affronta, invece, il tema dei limiti legislativi alla capacità di azione delle società a partecipazione pubblica e dei connessi dubbi interpretativi, anche alla luce degli orientamenti giurisprudenziali e dottrinali. In particolare, l’analisi riguarda l’art. 13 del decreto”Bersani” e il comma 9 dell’art. 23 bis (ora pedissequamente trasfuso nel comma 33 dell’art. 4 del d.l. n. 138/2011), ossia le principali disposizioni che definiscono, rispettivamente, la capacità di azione delle società (a partecipazione pubblica) strumentali e di quelle operanti nel campo dei servizi pubblici locali titolari di affidamenti diretti (assentiti con modalità diverse dall’evidenza pubblica). Vengono forniti cenni di inquadramento in relazione al cd. procedimento di riordino delle partecipazioni societarie pubbliche previsto dalla legge finanziaria del 2008 (art. 3, commi 27 – 32). Dal combinato disposto delle suddette norme, così come interpretate dalla giurisprudenza costituzionale ed amministrativa, si ricavano, poi, utili indicazioni in ordine alla possibilità, per gli enti pubblici territoriali, di costituire società con scopo meramente lucrativo (ossia, soggetti societari privi del rapporto di strumentalità con gli enti costituenti o partecipanti, chiamati ad operare, in regime di concorrenza, in settori completamente liberalizzati) e società cd. multiutilities (aventi oggetto sociale complesso, la cui attività si estrinseca tanto nel campo dei servizi strumentali, quanto in quello dei servizi pubblici locali), nonché in relazione alla disciplina applicabile all’attività di detti soggetti societari. La finalità ultima del contributo consiste nell'individuazione delle linee guida finalizzate alla classificazione delle società pubbliche in funzione della loro attività.

Covalent grafting mesogenic groups to the coordination cores of the parent mononuclear low-spin and spin-crossover compounds afforded metallomesogenic complexes of iron(II). In comparison with the parent complexes the spin-crossover properties of the alkylated derivatives are substantially modified. The type of the modification was found to be dependent on the properties of the parent system and the nature of the used anion, however, the general tendency is the destabilization of the low-spin state at the favor of spin-crossover or high-spin behavior below 400 K. The structural insight revealed the micro-segregated layered organization. The effect of the alkylation of the parent compounds consists first of all in the change of the lattice to a two-dimensional lamellar one retaining significant intermolecular contacts only within the ionic bilayers. The comprehensive analysis of the structural and thermodynamic data in the homologous series pointed at the mechanism of the interplay between the structural modification on melting and the induced anomalous change of the magnetic properties. A family of one-dimensional spin-crossover polymers was synthesized and characterized using a series of spectroscopic methods, X-ray powder diffraction, magnetic susceptibility measurements and differential scanning calorimetry. The copper analogue of was also synthesized and its crystal structure solved. In comparison with the mononuclear systems, the polymeric mesogens of iron(II) are less sensitive to the glass transition, which was attributed to the moderate concomitant variation of the structure. Nevertheless, the observed increase of the magnetic hysteresis with lengthening of the alkyl substituents was ascribed to the interplay of the structural reorganization of the coordination core due to spin-crossover with the structural delay in the spatial reorganization of the mesogenic substituents. The classification of mononuclear and polymeric metallomesogens according to the interactions between the structural- and the spin-transition and analysis of the data on the reported spin-crossover metallomesogens led to the separation of three types, namely: Type i: systems with coupling between the electronic structure of the iron(II) ions and the mesomorphic behavior of the substance; Type ii: systems where both transitions coexist in the same temperature region but are not coupled due to competition with the dehydration or due to negligible structural transformation; Type iii: systems where both transitions occur in different temperature regions and therefore are uncoupled. Fine-tuning, in particular regarding the temperature at which the spin-transition occurs with hysteresis properties responsible for the memory effect, are still a major challenge towards practical implementation of spin-crossover materials. A possible answer to the problem could be materials in which the spin-crossover transition is coupled with another transition easily controllable by external stimuli. In the present thesis we have shown the viability of the approach realized in the mesogenic systems with coupled phase- and spin-transitions.

Neurosteroide können langsame genomische und schnelle nicht-genomische Effekte zeigen. Die Synthese und der Metabolismus von Neurosteroiden werden entwicklungsbedingt reguliert. In den letzten Jahren sind immer mehr schnelle Steroideffekte bekannt geworden, die sowohl über klassische als auch über nicht-klassische Rezeptoren laufen. Zum heutigen Stand der Forschung sind die morphologischen Effekte von Neurosteroiden auf das neuronale Cytoskelett und die involvierten Signalkaskaden noch weitgehend unerforscht. In diesem Zusammenhang stellen sich auch die Fragen nach den verantwortlichen Rezeptoren und dem Transportmechanismus sowie der subzellulären Lokalisation der Steroide. Die im Rahmen meiner Promotion erhaltenen Ergebnisse zeigen, dass die Steroide DHEA und Testosteron eine Reorganisation des Aktincytoskeletts in neuronalen Zellen induzieren und dass diese Effekte diesen Steroiden und nicht ihren Folgemetaboliten zuzuordnen sind. DHEA bewirkt die Kontraktion der Zellen, eine erhöhte Ausbildung von Stressfasern und fokalen Adhäsionskomplexen sowie die Bildung von Filopodien. Der diesen Effekten zu Grunde liegende Signalweg konnte eindeutig identifiziert werden. DHEA induziert in neuronalen Zellen die Aktivierung des Rho-Signalwegs. Diese Aktivierung führt zu einem erhöhten Phosphorylierungsstatus der regulatorischen leichten Kette von Myosin II (MRLC) an Serin 19 und der damit verbundenen erhöhten Myosin-Aktin-Interaktion. Die Ausbildung von Filopodien wird vermutlich über eine Aktivierung der GTPase Cdc42 vermittelt. Testosteron induziert das Auswachsen langer Neuriten sowie eine Verminderung von Stressfasern in neuronalen Zellen. Diese Effekte sind abhängig von der Aktivität der PI3-Kinase. Die im Rahmen dieser Arbeit gewonnenen Erkenntnisse deuten darauf hin, dass Testosteron über die PI3-Kinase und FAK den Rac-Signalweg induziert, da es zu einer Inhibierung des Rho-Signalwegs kommt. Zahlreiche Erkenntnisse weisen darauf hin, dass DHEA und Testosteron die Aktivierung der beteiligten Signalwege über einen G-Protein gekoppelten Rezeptor induzieren. DHEA und Testosteron beeinflussen auch die Expression und die Lokalisation der regulatorischen leichten Ketten von Myosin II. Im Gegensatz zu DHEA (Lokalisation der MRLC in der kortikalen Region der Zelle), induziert Testosteron eine Umlokalisation der MRLC in den Zellkern. Daher ist es denkbar, dass die MRLCs, wie auch Aktin, als Transkriptionsfaktoren wirken können. Die Synthese eines funktionalen, fluoreszierenden DHEA-Derivats (DHEA-Bodipy) ermöglichte erstmals, den Transport und die subzelluläre Lokalisation von DHEA in neuronalen Zellen zu beobachten. DHEA-Bodipy wird in neuronalen Zellen in den Mitochondrien lokalisiert. Diese Lokalisation ergibt völlig neue Ansätze im Verständnis zellulärer Wirkungsorte von Steroiden und beteiligter Rezeptoren. Das in meiner Arbeit vorgestellte Verfahren zur Fluoreszenzmarkierung von Steroiden bietet vielfältige Möglichkeiten im Einsatz zellbiologischer Methoden. Nach diesem Verfahren hergestellte, fluoreszierende Steroide eignen sich aufgrund ihrer Stabilität sehr gut für die Untersuchung des Transports und der subzellulären Lokalisation von Steroiden an fixierten und lebenden Zellen sowie für Colokalisationsexperimente. Diese Methode grenzt somit auch die Anzahl möglicher molekularer Interaktionspartner ein. Für Testosteron konnte ebenfalls ein fluoreszierendes Testosteron-Derivat (Testosteron-Bodipy) synthetisiert werden. Die Aufklärung der Effekte von Steroiden auf das neuronale Cytoskelett und der beteiligten Signalkaskaden sowie die Identifizierung der zellulären Wirkungsorte ermöglichen therapeutische Ansätze zur Behandlung neurodegenerativer Erkrankungen, deren Ursachen in Abnormitäten des Cytoskeletts oder fehlregulierter Neurosteroidogenese zu begründen sind.

L’autore ricostruisce ed esamina la storia dei rapporti italo-tedeschi negli anni immediatamente precedenti la riapertura ufficiale delle relazioni diplomatiche tra Italia e Repubblica federale tedesca. Un riavvicinamento economico e politico progressivo, ma che suscitò forti contrasti tra i principali attori della diplomazia italiana. Il saggio si basa su una documentazione conservata presso l’Archivio storico del ministero degli Esteri, l’Archivio centrale dello Stato, l’Archivio storico della Banca d’Italia e l’Archivio Politico del ministero degli Esteri della Repubblica federale. L’autore sostiene che le relazioni economiche italo-tedesche assunsero un ruolo centrale nel processo di elaborazione della politica estera italiana sulla questione tedesca nel corso di questi anni. Prima dell’istituzione della Repubblica federale tedesca, l’Italia divenne un partner economico fondamentale per la Germania occidentale. Tra il 1945 e il 1949 l'Italia fu il primo paese europeo favorevole alla rinascita di un nuovo stato tedesco non sottoposto alla diretta influenza dell’Unione Sovietica. Il presidente del Consiglio De Gasperi e il ministro degli Esteri Sforza per sostenere la nuova Germania attuarono una precisa azione diplomatica di riavvicinamento politico, promuovendo diversi scambi di visite e di incontri con i rappresentanti tedeschi.
Electrostatic supramolecular assembly of charged dendritic polymers and their biological application

The aim of this study was the development of functional multilayer films through electrostatic layer by layer (LbL) assembly of dendritic macromolecules, the investigation of the fundamental properties of these multilalyered films and the study of their biological applications. rnThe synthesis of the anionic hyperbranched polyglycerols (hbPG) and the preparation of multilayers made of hbPG/phosphorus dendrimer as well as the influences of deposition conditions on multilayers were reported. The thicknesses of multilayer films increase with a decrease of molecular weight of anionic hbPGs. The multilayer films fabricated by low molecular weight hbPGs grow less regularly due to the less charged carboxylic acid groups providing the relative weaker electrostatic forces for the deposition. The thicknesses of multilayer films are reduced with increasing pH values and decreasing the concentration of NaCl. The observed changes of multilayer thickness and surface morphology could be interpreted with the aid of theories regarding the charge density and conformation of the anionic hbPG chains in solution. rnBesides the study of fundamental properties of hbPG/phosphorus multilayer films, antifouling thin films derived from hbPG layers were developed. The antifouling properties of hbPG layers were found to correlate with factors of the molecular weight of anionic hbPG and the film thickness. It was demonstrated that anionic hbPG single layer with highest molecular weight can reduce non specific protein adsorption more efficiently than single layer with lower molecular weight and all the hbPG bilayers possessed excellent property of antifouling. rnPhosphorus dendrimer multilayers were successfully prepared as the platforms to detect DNA immobilization and hybridization. The effect of NaCl concentration on the multilayer film thickness was evaluated to obtain the optimized film thickness. Making use of the multilayer deposited at the optimized condition as a substrate, a high loading of DNA probes was achieved through covalent coupling of probe DNA with the as-formed multilayer films. The hybridization of target DNA with immobilized probe DNA was then carried out and studied by SPFS. The limit of detection upon hybridization was estimated on various dendrimer multilayer platforms. The minimum detection concentration for DNA hybridization is in the same order of magnitude compared with other neutral phosphorus dendrimer systems. Furthermore, the LbL deposition of phosphorus dendrimer multilayers provided a mild and simple way to prepare platforms as DNA microarrays. rnBased on the phosphorus dendrimer multilayer systems, dendritic star polymers were employed which have more reactive groups than that phosphorus dendrimers. The as-assembled dendritic star polymer multilayer films exhibited such distinct morphology characteristics that they underwent extensive structural reorganization upon post-treatment under different pH conditions. Kinetic binding of probe DNA molecules on the outermost negatively charged dendritic surface was studied by SPR as well. The binding capacities of probe DNA on the multilayer surfaces fabricated from the first-generation and the second-generation of dendritic star polymers were compared. The improved binding capacity was achieved from the second-generation of dendritic star polymer multilayer films due to their more reactive groups. DNA hybridization reaction on dendritic multilayer films was investigated by SPFS. The similar hybridization behaviors were found on both multilayer surfaces. Meanwhile, the hybridization kinetic affinities were compared with that of phosphorus dendrimer multilayer surfaces and showed improved detection sensitivity than phosphorus dendrimer multilayer films.rn
