Elimination of paternal mitochondrial DNA in intraspecific crosses during early mouse embryogenesis.
Resumo:
To examine whether mtDNA is uni- or biparentally transmitted in mice, we developed an assay that can detect sperm mtDNA in a single mouse embryo. In intraspecific hybrids of Mus musculus, paternal mtDNA was detected only through the early pronucleus stage, and its disappearance co-incided with loss of membrane potential in sperm-derived mitochondria. By contrast, in interspecific hybrids between M. musculus and Mus spretus, paternal mtDNA was detected throughout development from pronucleus stage to neonates. We propose that oocyte cytoplasm has a species-specific mechanism that recognizes and eliminates sperm mitochondria and mtDNA. This mechanism must recognize nuclearly encoded proteins in the sperm midpiece, and not the mtDNA or the proteins it encodes, because sperm mitochondria from the congenic strain B6.mtspr, which carries M. spretus mtDNA on background of M. musculus (B6) nuclear genes, were eliminated early by B6 oocytes as in intraspecific crosses. We conclude that cytoplasmic genomes are transmitted uniparentally in intraspecific crosses in mammals as in Chlamydomonas and that leakage of parental mtDNA is limited to interspecific crosses, which rarely occur in nature.

Sandinista ideology and its political culture are born in 1927, with the refusal of the Pacto del Espino Negro by Augusto César Sandino, while its disappearance can be placed in 1999, date of the signature of the Liberal-Sandinista pact with which –effectively- the two main protagonist of nicaraguan politics at that time, Daniel Ortega and Arnoldo Alemán, halt the democratization process of Nicaragua, so putting on ice also its political development. Meanwhile, in the lapse of time between these two pacts, the most intense, feverish, dramatic and participated period of political history of the Central American country develops: an anti-imperialist guerrilla warfare ended in a bloodbath; a dynastic dictatorship of predatory authoritarianism for more than 40 years; a popular revolution that throws down the dictatorship; a decade of revolutionary government attacked by a counter-revolutionary war; an electoral defeat that will lead to a season of “pactismo” that will end the Sandinista anomaly and that will give an opening to something that we could consider –not with a certain difficulty- its pretence. The aim of this essay consists in analyzing how it has been possible that a political experience like the Sandinista Front , created not only for gaining power and for revolutionizing politically, socially and economically Nicaragua, but also for changing radically cultural, ethic and moral perspective of the country and its people, arrived being the contrary of what had been posed as the horizon to aspire...

In Europe, Dupont’s Lark Chersophilus duponti is a threatened open-habitat bird. Prescribed burning has sometimes been proposed for its conservation, but without evidence of its effectiveness. To evaluate the short-term effects of a summer wildfire on this species, we performed several transect counts in the burnt and unburnt parts of a shrubsteppe in central Spain. The same transects were counted within a three-year interval prior to the fire and were repeated during the first two springs after the fire. We also measured the vegetation during the first two springs after the fire. In the burnt area, we observed a decrease of about 85–100% in Dupont’s Lark abundance, and about 7–15% in the control area. The disappearance of the scrub cover after fire and its slow regeneration, as well as the large increase in grass cover during the second year, may explain the decrease in this habitat-specialist bird species. Fire should be avoided in areas occupied by the Dupont’s Lark, as its negative effects in the short-term may cause local extinctions. However, prescribed burning may be used in neighboring areas to create new open habitats that may be subsequently colonized by this species.

The distribution and composition of Amphipoda assemblages were analysed off the coasts of Alicante (Spain, Western Mediterranean), a disturbed area affected by several co-occurring anthropogenic impacts. Although differences among sampled stations were mainly related to natural parameters, anthropogenic activities were linked with changes in amphipod assemblages. Expansion of the Port of Alicante, a sewage outfall and a high salinity brine discharge could be causing the disappearance of amphipods at stations closer to these disturbances. However, the completion of port enlargement works and mitigatory dilution of the brine discharge has led to the recovery of the amphipod assemblage. Among the natural parameters, depth determines the distribution of some of the species. While Siphonoecetes sabatieri was abundant at shallow stations, Ampelisca spp., Photis longipes, Pseudolirius kroyeri, Apherusa chiereghinii and Phtisica marina were more abundant at deeper stations. Grain size and percentage of organic matter also influenced amphipod distribution, resulting in changes in species composition and in the relative percentages of different trophic groups. Species such as Ampelisca brevicornis, Perioculodes longimanus, Urothoe hesperiae and Urothoe elegans were more abundant at stations with a high content of fine sand. Carnivorous species, mainly of the Oedicerotidae family, were more abundant at those stations with a low organic matter content, while detritivorous species were more abundant at stations with a higher mud content. Among 62 identified species, three were reported for the first time from the Spanish Mediterranean coast, two species were recorded for the second time and a new species of Siphonoecetes was found, Siphonoecetes (Centraloecetes) bulborostrum. These results confirm the need for further data on amphipods from the Mediterranean Spanish coast.

Peroxide-mediated reactive extrusion of linear isotactic polypropylene (L-PP) was conducted in the presence of trimethylolpropane trimethacrylate (TMPTMA) and triallyl trimesate (TAM) coagents, using a twin screw extruder. The resulting coagent-modified polypropylenes (CM-PP) had higher viscosities and elasticities, as well as increased crystallization temperature compared to PP reacted only with peroxide (DCP-PP). Additionally, deviations from terminal flow, and strain hardening were observed in PP modified with TAM, signifying the presence of long chain branching (LCB). The CM-PP formulations retained the modulus and tensile strength of the parent L-PP, in spite of their lower molar mass and viscosities, whereas their elongation at break and the impact strength were better. This was attributed to the finer spherulitic structure of these materials, and to the disappearance of the skin-core layer in the injection molded specimens.

Despite accounting for a significant share of global trade and the resulting interdependencies from it, energy governance remains largely fragmented and there is no global framework or agreement defining the rules of energy trade. This paper, after presenting the main global and regional energy market developments, discusses the opportunities to ‘energise the TTIP’, i.e. to include a chapter dedicated to trade and cooperation in the sphere of energy. The shale revolution in the US, the ever-rising interconnectedness of energy markets (recently proven by the disappearance of the ‘Asian gas premium’) and the EU’s quest to diversify its energy supplies generally sets favourable conditions to reinforce energy relations between the EU and the US. The question, as is often the case, is whether there is sufficient political will to tighten relations in a strategic sphere with connotations for national security and sovereignty.

T2K reports its first measurements of the parameters governing the disappearance of νµ in an off-axis beam due to flavor change induced by neutrino oscillations. The quasimonochromatic νµ beam, produced with a peak energy of 0.6 GeV at J-PARC, is observed at the far detector SuperKamiokande, 295 km away, where the νµ survival probability is expected to be minimal. Using a dataset corresponding to 4.01×10²⁰ protons on target, 34 fully contained µ-like events were observed. The best-fit oscillation parameters are sin²(θ₂₃) = 0.45 and |∆m^2_32| = 2.51 × 10⁻³ eV² with 68% confidence intervals of 0.38 - 0.64 and 2.26 - 2.80 ×10⁻³ eV² respectively. These results are in agreement with existing antineutrino parameter measurements and also with the νµ disappearance parameters measured by T2K.

The steep environmental gradients of mountain ecosystems over short distances reflect large gradients of several climatic parameters and hence provide excellent possibilities for ecological research on the effects of environmental change. To gain a better understanding of the dynamics of abiotic and biotic parameters of mountain ecosystems, long-term records are required since permanent plots in mountain regions cover in the best case about 50 - 70 years. In order to extend investigations of ecological dynamics beyond these temporal limitations of permanent plots, paleoecological approaches can be used if the sampling resolution can be adapted to ecological research questions, e.g. a sample every 10 years. Paleoecological studies in mountain ecosystems can provide new ecological insights through the combination of different spatial and temporal scales. [f we thus improve our understanding of processes across both steep environmental gradients and different time scales, we may be able to better estimate ecosystem responses to current and future environmental change (Ammann et al. 1993; Lotter et al. 1997). The complexity of ecological interactions in mountain regions forces us to concentrate on a number of sub-systems - without losing sight of the wider context. Here, we summarize a few case studies on the effects of Holocene climate change and disturbance on the vegetation of the Western Alps. To categorize the main response modes of vegetation to climatic change and disturbance in the Alps we use three classes of ecological behaviour: "resilience", "adjustment", and "vulnerability", We assume a resilient (or elastic) behaviour if vegetation is able to recover to its former state, regaining important ecosystem characteristics, such as floristic composition, biodiversity, species abundances, and biomass (e.g. Küttel 1990; Aber and Melillo 199 1). Conversely, vegetation displacements may occur in response to climatic change and/or disturbance. In some cases, this may culminate in irreversible large-scale processes such as species and/or community extinctions. Such drastic developments indicate high ecosystem vulnerability (or inelasticity or instability, for detailed definitions see Küttel 1990; Aber and Melillo 199 1) to climatic change and/or disturbance. In this sense, the "vulnerability" (or instability) of an ecosystem is expressed by the degree of failure to recover to the original state before disturbance and/or climatic change. Between these two extremes (resilience vs. vulnerability), ecosystem adjustments to climatic change and/or disturbance may occur, including the appearance of new and/or the disappearance of old species. The term "adjustment" is hence used to indicate the response of vegetational communities, which adapted to new environmental conditions without losing their main character. For forest ecosystems, we assume vegetational adjustments (rather than vulnerability) if the dominant (or co-dominant) tree species are not outnumbered or replaced by formerly unimportant plant species or new invaders. Adaptation as a genetic process is not discussed here and will require additional pbylogeographical studies (that incorporate the analysis of ancient DNA) in order to fully understand the distributions of ecotypes.

New palaeoecological investigations (pollen, macrofossil, and charcoal analyses) provide important evidence on the fire history and the long-term fire ecology of different regions of Switzerland. The results from the Swiss plateau, the Northern and Central Alps and Southern Switzerland suggest that fire played a different role for the long-term vegetational development in the different regions. In the Northern Alps and Southern Switzerland anthropogenic fires led to the disappearance of entire forest communities. These fires especially affected the fire-sensitive species Abies alba. On the Swiss Plateau fire frequencies were markedly lower than in the Southern Alps. Nevertheless, fires probably led to a decline in the occurrence of fire-sensitive taxa such as Ulmus, Fraxinus excelsior or Tilia at lower altitudes (Fagus silvatica-Quercus belt). First evidences from the Central Alps suggest that forest fires were naturally more frequent in this continental region and that the vegetation might be better fire-adapted than the original (partly or completely vanished) plant communities of the Swiss Plateau, the Northern Alps and Southern Switzerland.

Hole 633A was drilled in the southern part of Exuma Sound on the toe-of-slope of the southeastern part of Great Bahama Bank during ODP Leg 101. The top 55 m, collected as a suite of six approximately 9.5-m-long hydraulic piston cores, represents a Pliocene-Pleistocene sequence of periplatform carbonate ooze, a mixture of pelagic calcite (foraminifer and coccolith tests), some pelagic aragonite (pteropod tests), and bank-derived fine aragonite and magnesian calcite. A 1.6-m.y.-long hiatus was identified at 43.75 mbsf using calcareous nannofossil biostratigraphy and magnetostratigraphy. The 43.75-m-thick periplatform sequence above the hiatus is a complete late Pliocene-Quaternary record of the past 2.15 m.y. The d18O curve, primarily based on Globigerinoides sacculifera, clearly displays high-frequency/low-amplitude cycles during the early Pleistocene and low-frequency/high-amplitude cycles during the middle and late Pleistocene. Variations in aragonite content in the fine fraction of the periplatform ooze show a cyclic pattern throughout the Pleistocene, as previously observed in piston cores of the upper Pleistocene. These variations correlate well with the d18O record: high aragonite corresponds to light interglacial d18O values, and vice versa. Comparison of the d18O record and the aragonite curve helps to identify 23 interglacial and glacial oxygen-isotope stages, corresponding to 10.5 aragonite cycles (labeled A to K) commonly established during the middle and late Pleistocene (0.9 Ma-present). Strictly based on the aragonite curve, another 11 aragonite cycles, labeled L to V, were identified for the early Pleistocene (0.9 to 1.6 Ma). Mismatches between the d18O record and the aragonite curve occur mainly at some of the glacial-to-interglacial transitions, where aragonite increases usually lag behind d18O depletion. When one visually connects the minima on the Pleistocene aragonite curve, low-frequency (0.4 to 0.5 m.y.) supercycles seem to be superimposed on the high-frequency cycles. The timing of this supercycle roughly matches the timing of the Pleistocene carbonate preservation supercycles described in the Pacific, Indian, and Atlantic oceans. Mismatches between aragonite and d18O cycles are even more obvious for the late Pliocene (1.6 to 2.15 Ma). Irregular aragonite variations are observed for the late Pliocene, although after the onset of late Pleistocene-like glaciations in the North Atlantic Ocean 2.4 m.y. ago the d18O record has shown a mode of high-frequency/low-amplitude cycles. Initiation of climatically induced aragonite cycles occurs only at the Pliocene-Pleistocene transition, 1.6 m.y. ago. After that time, aragonite cycles are fully developed throughout the Quaternary. The 11-m-thick periplatform sequence below the hiatus represents a lower Pliocene interval between 3.75 and 4.45 Ma. The bottom half (4.25-4.45 Ma) has a fairly constant, high aragonite content (averaging 60%) and high sedimentation rates (28 m/m.y.) and corresponds to the end of the prolonged early Pliocene interglacial interval (4.1-5.0 Ma), established as a worldwide high sea-level stand. The second half (3.75-4.25 Ma), in which aragonite content decreases by successive steps, paralleled by a gradual 5180 enrichment in Globigerinoides sacculifera and low sedimentation rates (10 m/m.y), corresponds to the climatic deterioration established worldwide between 4.1 and 3.8 Ma, to a decrease of carbonate preservation observed in the equatorial Pacific Ocean, and to a global sea-level decline. Dolomite, a ubiquitous secondary component in the lower Pliocene, is interpreted as being authigenic and possibly related to diagenetic transformation of primary bank-derived fine magnesian calcite. Transformation of the primary mineralogical composition of the periplatform ooze was evidently minor, as the sediments have retained a detailed record of the Pliocene-Pleistocene climatic evolution. Clear evidence of diagenetic transformations in the periplatform ooze includes (1) the disappearance of magnesian calcite in the upper 20 m of Hole 633A, (2) the occurrence of calcite overgrowths on foraminiferal tests and microclasts at intermittent chalky core levels, and (3) the ubiquitous presence of authigenic dolomite in the lower Pliocene.

Pliocene and Pleistocene planktonic foraminiferal biogeography and paleoceanography have been examined in Deep Sea Drilling Project (DSDP) sites of the Panama Basin (Pacific Ocean) and Colombian and Venezuelan Basins (Atlantic Ocean) to determine the timing of the isolation of Atlantic and Pacific tropical planktonic faunas resulting from the development of the Central American isthmus. Previous studies have suggested a late Miocene to middle Pliocene occurrence of this event. The Panama Basin (DSDP site 157) and the Colombian Basin (DSDP site 154A) share two early Pliocene biogeographic events: (1) great abundance of sinistral coiling Neogloboquadrina pachyderma at 4.3 m.y. ago at site 157 and 0.7 m.y. later at site 154A, and (2) a sinistral-to-dextral change in the coiling-direction preference in Pulleniatina 3.5 m.y. ago at both locations. Identification of these events farther to the east in the Venezuelan Basin (DSDP site 148) is complicated by insufficient lower Pliocene core recovery, but abundant sinistral N. pachydcrma appear to have extended far to the east in the Caribbean 3.6 m.y. ago; perhaps the early Pliocene abundance of this form is not indicative of cool water. The coiling-direction history and stratigraphic ranges of Pulleniatina became different in the Atlantic and Pacific Oceans during the early Pliocene; this is inferred to result from geographic isolation of the assemblages. Saito (1976) used the temporary disappearance of this genus from Atlantic waters at 3.5 m.y. ago to mark the closure of the Isthmus of Panama, but I show that in the Colombian Basin (site 154A) its disappearance was closer to 3.1 m.y. ago. This suggests the possibility of surface-water communication between the Atlantic and Pacific until that time.
