The bacterial GatCAB operon for tRNA-dependent amidotransferase (AdT) catalyzes the transamidation of mischarged glutamyl-tRNA(Gln) to glutaminyl-tRNA(Gln). Here we describe the phenotype of temperature-sensitive (ts) mutants of GTF1, a gene proposed to code for subunit F of mitochondrial AdT in Saccharomyces cerevisiae. The ts gtf1 mutants accumulate an electrophoretic variant of the mitochondrially encoded Cox2p subunit of cytochrome oxidase and an unstable form of the Atp8p subunit of the F(1)-F(0) ATP synthase that is degraded, thereby preventing assembly of the F(0) sector. Allotopic expression of recoded ATP8 and COX2 did not significantly improve growth of gtf1 mutants on respiratory substrates. However, ts gft1 mutants are partially rescued by overexpression of PET112 and HER2 that code for the yeast homologues of the catalytic subunits of bacterial AdT. Additionally, B66, a her2 point mutant has a phenotype similar to that of gtf1 mutants. These results provide genetic support for the essentiality, in vivo, of the GatF subunit of the heterotrimeric AdT that catalyzes formation of glutaminyl-tRNA(Gln) (Frechin, M., Senger, B., Braye, M., Kern, D., Martin, R. P., and Becker, H. D. (2009) Genes Dev. 23, 1119-1130).

Salmonella flagellin, the flagellum structural subunit, has received particular interest as a vaccine adjuvant conferring enhanced immunogenity to soluble proteins or peptides, both for activation of antibody and cellular immune responses. In the present study, we evaluated the Salmonella enterica FliCd flagellin as a T cell vaccine adjuvant using as model the 9-mer (SYVPSAEQI) synthetic H2(d)-restricted CD8(+) T cell-specific epitope (CS(280-288)) derived from the Plasmodium yoelii circumsporozoite (G) protein. The FliCd adjuvant effects were determined under two different conditions: (i) as recombinant flagella, expressed by orally delivered live S. Dublin vaccine strains expressing the target CS(280-288) peptide fused at the central hypervariable domain, and (ii) as purified protein in acellular vaccines in which flagellin was administered to mice either as a recombinant protein fused or admixed with the target CS(280-288) peptide. The results showed that CS(280-288)-specific cytotoxic CD8(+) T cells were primed when BALB/c mice were orally inoculated with the expressing the CS280-288 epitope S. Dublin vaccine strain. In contrast, mice immunized with purified FliCd admixed with the CS280-288 peptide and, to a lesser extent, fused with the target peptide developed specific cytotoxic CD8(+) T cell responses without the need of a heterologous booster immunization. The CD8(+) T cell adjuvant effects of flagellin, either fused or not with the target peptide, correlated with the in vivo activation of CD11c(+) dendritic cells. Taken together, the present results demonstrate that Salmonella flagellins are flexible adjuvant and induce adaptative immune responses when administered by different routes or vaccine formulations. We sequenced the small subunit (SSU) rRNA and glycosomal glyceraldehyde-3-phosphate dehydrogenase (gGAPDH) genes of two trypanosomes isolated from the Brazilian snakes Pseudoboa nigra and Crotalus durissus terrificus. Trypanosomes were cultured and their morphometrical and ultrastructural features were characterized by light microscopy and scanning and transmission electron microscopy. Phylogenetic trees inferred using independent or combined SSU rRNA and gGAPDH data sets always clustered the snake trypanosomes together in a clade closest to lizard trypanosomes, forming a strongly supported monophyletic assemblage (i.e. lizard-snake clade). The positioning in the phylogenetic trees and the barcoding based on the variable V7-V8 region of the SSU rRNA, which showed high sequence divergences, allowed us to classify the isolates from distinct snake species as separate species. The isolate from P. nigra is described as a new species, Trypanosoma serpentis n. sp., whereas the isolate from C. d. terrificus is redescribed here as Trypanosoma cascavelli.

Patients and methods: Clinical data from all patients admitted with acute respiratory failure due to novel viral H1N1 infection were reviewed. Lung tissue was submitted for viral and bacteriological analyses by real-time RT-PCR, and autopsy was conducted on all patients who died. Results: Eight patients were admitted, with ages ranging from 55 to 65 years old. There were five patients with solid organ tumors (62.5%) and three with hematological malignancies (37.5%). Five patients required mechanical ventilation and all died. Four patients had bacterial bronchopneumonia. All deaths occurred due to multiple organ failure. A milder form of lung disease was present in the three cases who survived. Lung tissue analysis was performed in all patients and showed diffuse alveolar damage in most patients. Other lung findings were necrotizing bronchiolitis or extensive hemorrhage. Conclusions: H1N1 viral infection in patients with cancer can cause severe illness, resulting in acute respiratory distress syndrome and death. More data are needed to identify predictors of unfavorable evolution in these patients.

Introduction: This study prospectively accessed the immune response to the inactivated influenza vaccine in renal transplant recipients receiving either azathioprine or mycophenolate mofetil (MMF). Side effects were investigated. Methods: Sixty-nine patients received one dose of inactivated trivalent influenza vaccine. Antihemagglutinin (HI) antibody response against each strain was measured before and one to six months after vaccination. Results: Geometric mean HI antibody titers for H1N1 and H3N2 strains increased from 2.57 and 2.44 to 13.45 (p = 0.001) and 7.20 (p < 0.001), respectively. Pre- and post-vaccination protection rates for H1N1 and H3N2 increased from 8.7% to 49.3% (p < 0.001); and 36.3% (p < 0.001) and seroconversion rates were 36% and 25.3%, respectively. There was no response to influenza B. The use of MMF reduced the H1N1 and H3N2 protection rates and the seroconversion rate for the H1N1 strain when compared with the use of azathioprine, and subjects transplanted less than 87 months also had inferior antibody response. Adverse events were mild and there were no change on renal function post-vaccination. Conclusion: Renal transplant patients vaccinated against influenza responded with antibody production for in. uenza A virus strains, but not for in. uenza B. Use of MMF and shorter time from transplantation decreased the immune response to the vaccine.

In trypanosomatids the involvement of mitochondrial complex I in NADH oxidation has long been debated. Here, we took advantage of natural Trypanosoma cruzi mutants which present conspicuous deletions in ND4, ND5 and ND7 genes coding for complex I subunits to further investigate its functionality. Mitochondrial bioenergetics of wild type and complex I mutants showed no significant differences in oxygen consumption or respiratory control ratios in the presence of NADH-linked substrates or FADH(2)-generating succinate. No correlation could be established between mitochondrial membrane potentials and ND deletions. Since release of reactive oxygen species occurs at complex I, we measured mitochondrial H(2)O(2) formation induced by different substrates. Significant differences not associated to ND deletions were observed among the parasite isolates, demonstrating that these mutations are not important for the control of oxidant production. Our data support the notion that complex I has a limited function in T. cruzi.

Trata-se de um estudo sobre o impacto da introdução da vacina contra influenza no perfil de morbimortalidade por pneumonias na população acima de 65 anos, em Porto Alegre, no período de 1995 a 2001. Associado ao estudo da série histórica, investigamos os motivos de adesão ou recusa à prática de vacinação contra influenza entre 138 idosos. Aprofundamos a análise das concepções do processo saúde-doença e das práticas preventivas em saúde com um grupo de 30 idosos usuários do sistema público de saúde. A metodologia utilizada é de caráter epidemiológico do tipo série temporal, combinando entrevistas com idosos e categorização temática das informações obtidas. Os dados de morbidade foram obtidos pela pesquisa documental estatística, a partir dos dados de internação hospitalar do Sistema Único de Saúde (Tabwin) e os dados de mortalidade foram obtidos a partir do Sistema de Informação em Mortalidade (SIM). Os resultados apontam um comportamento de tendência à queda na morbidade, através das internações hospitalares e na mortalidade por pneumonias, após a introdução regular da vacina contra influenza em nosso meio no ano de 1999. Particularizando a adesão ou recusa à prática de vacinação, os idosos demonstram que fatores culturais e sociais influenciaram suas decisões. Os idosos que participaram deste estudo também revelaram que é fundamental a manutenção de uma atividade física, intelectual ou laboral para um envelhecimento saudável. Os resultados deste estudo, acredita-se, contribuem para o aperfeiçoamento das práticas de promoção em saúde através da educação em saúde e da adoção de medidas de proteção específica como a vacinação com eficácia e credibilidade junto à população.

As infecções respiratórias de etiologia viral constituem um problema alarmante de Saúde Pública, sendo responsáveis pelo elevado e constante aumento dos índices de morbimortalidade registados no Mundo associados ao vírus influenza. O presente estudo teve como objetivo avaliar a prevalência dos anticorpos IgG e IgM em soros de utentes com requisições para análises serológicas ao vírus influenza A e B. Os utentes foram atendidos entre 1 de Abril de 2009 e 30 de Abril de 2011. Outro objectivo foi determinar a epidemiologia do vírus pandémico A (H1N1) 2009 nos indivíduos com sintomatologia de gripe durante o período entre Julho de 2009 e Julho de 2010 utilizando a técnica de RT-PCR em amostras de exsudado (ou aspirado) nasofaríngeo. Tendo por base o universo de amostragem de 981 indivíduos, constatou-se que 10,7 e 8,2% da população analisada apresenta valores positivos de anticorpos IgM e IgG indicativos de infecção pelo vírus influenza A e B, respectivamente. Constatou-se, também, que entre os 1934 indivíduos submetidos a procedimentos de diagnóstico laboratorial para a detecção de infecção pelo vírus pandémico A (H1N1) 2009, cerca de 747 (38,6%) estavam infectados. Verificou-se que, a população mais jovem foi mais susceptível à infecção pelo vírus influenza A (H1N1) 2009. Isto difere da típica época de gripe sazonal, na qual as pessoas mais idosas estão mais propensas a tornarem-se infectadas e a desenvolver doença grave por influenza A e/ou B. A prevalência de gripe na RAM é reduzida – um dos aspectos plausíveis que justifiquem esta afirmação poderá dever-se às características genéticas da população da RAM estudada. Embora seja de elevada relevância salientar que o Programa Regional de Vacinação (PRV) da RAM tem alcançado reconhecimento nacional e internacional devido às excelentes taxas de cobertura vacinal, fruto da atitude entre cidadãos e profissionais de saúde.

The transmission of influenza in health care settings is a major threat to patients, especially those with severe diseases. The attitude of health care workers (HCWs) may influence the transmission of countless infections. The current study aimed to quantify knowledge and identify attitudes of HCWs involved in intensive care units (ICUs) regarding the risk of nosocomial influenza transmission. A questionnaire was applied through interviews to HCWs who worked in one of the five ICUs from a teaching hospital. Questions about influenza were deliberately dispersed among others that assessed several infectious agents. Forty-two HCWs were interviewed: nine physicians, ten nurses and 23 nursing technicians or auxiliaries. Among the 42 HCWs, 98% were aware of the potential transmission of influenza virus in the ICUs, but only 31% would indicate droplet precautions for patients with suspected infection. Moreover, only 31% of them had been vaccinated against influenza in the last campaign (2008). Nursing technicians or auxiliaries were more likely to have been vaccinated, both by univariate and multivariable analysis. When asked about absenteeism, only 10% of the study subjects stated that they would not go to work if they had an influenza-like illness. Those findings suggest that, in non-pandemic periods, influenza control in hospitals requires strategies that combine continuous education with changes in organizational culture.
