Notre intention est montrer comment les conceptions mdicales de Descartes, disséminées dans ses oeuvres et dans sa correspondance, se sont constituées a partir de deux questions fondamentales posées par le philosophe: 1 - comment fonctionne-t-il notre organisme; 2 - quel est-t-il la nature du corps animal et, en particulier, du corps humain.

We consider a scalar field theory on AdS in both minimally and non-minimally coupled cases. We show that there exist constraints which arise in the quantization of the scalar field theory on AdS which cannot be reproduced through the usual AdS/CFT prescription. We argue that the usual energy, defined through the stress-energy tensor, is not the natural one to be considered in the context of the AdS/CFT correspondence. We analyze a new definition of the energy which makes use of the Noether current corresponding to time displacements in global coordinates. We compute the new energy for Dirichlet, Neumann and mixed boundary conditions on the scalar field and for both the minimally and non-minimally coupled cases. Then, we perform the quantization of the scalar field theory on AdS showing that, for 'regular' and 'irregular' modes, the new energy is conserved, positive and finite. We show that the quantization gives rise, in a natural way, to a generalized AdS/CFT prescription which maps to the boundary all the information contained in the bulk. In particular, we show that the divergent local terms of the on-shell action contain information about the Legendre transformed generating functional, and that the new constraints for which the irregular modes propagate in the bulk are the same constraints for which such divergent local terms cancel out. In this situation, the addition of counterterms is not required. We also show that there exist particular cases for which the unitarity bound is reached, and the conformai dimension becomes independent of the effective mass. This phenomenon has no bulk counterpart.

A Tese elege por objeto de investigaão os auto-biografismos na temtica da Formação de Professores. Como material significante, aborda um conjunto de textos de características autobiográficas coligidos a partir dos Anais do Congresso Internacional sobre Pesquisa (Auto)biográfica – CIPA, correspondentes às edições dos anos de 2006, 2008 e 2010. O estudo se orienta, sobretudo a partir da noção de biografema, pela problematizaão da Escrita da Vida em textos autobiográficos de docentes da Educaão Superior, apresentados nas edições do CIPA na forma de Comunicaão Oral. A colocaão da Escrita da Vida (em sua forma autobiográfica) como problema, possibilita vislumbrar as permeabilidades cambiantes (caracterizadas por encontros, desencontros, bifurcaões, liames, contiguidades, sinuosidades, fragmentaões) de uma vida. Uma vida escrita tomada não como uma correspondência direta e verídica com A Vida, mas como possibilidade de criaão, de invenção de uma escrita de vida que contagia... Pormenores de uma vida, traos, espaos vazios, fragmentos que encantam, tocam, saem do texto e entram em nossas vidas; produzem uma co-existência: felicidade de escrita: imanente artifício de uma vida. O estudo, como exercício de crítica, não se propõe a analisar textos autobiográficos submetendo-os a uma crítica judicativa ou veridicativa do caráter de seu objeto de escrita ou mesmo de sua forma de escrita, mas se orienta por recolher fragmentos, linhas, frases de prazer que permitam flagrar liames de escrita e vida, uma vida atravessando linhas de escrita, uma vida como fenda de linhas de escrita e a afirmação, como ideia força desta tese-experimento, da impossibilidade de “aprisionamento” de uma vida em uma auto-biografia-destino e a proposição de uma auto-biografia-deriva.

Esse artigo tem como objetivo apresentar uma leitura da correspondência de Guy de Maupassant (1850-1893). Trata-se de descrever e analisar a prática epistolar no conjunto das cartas por ele escritas a Émile Zola, entre 1875 e 1890, de seguir os passos dos dois escritores e de observar que, paralelamente a uma relaão mtua de amizade e de respeito, surge uma crítica aos princípios estéticos e às obras do mestre do naturalismo. Assim, é possível entender a amizade e a experiência estética que se constroem ao longo desses quinze anos.

This article analyzes the data resultant of the accomplishment, between 2008 and 2009, of an extension and research project on the international exchange of letters between teachers and students from elementary schools in the towns of Garça and Marília, São Paulo State, Brazil, with a school in the city of Azaruja, Portugal and another in the city of Luanda, Angola. It was developed by a team of students and teachers, UNESP, Marília, in order to promote the exchange via personal letters for that students, between 8 and 9 years old, could take possession of the written language, in this case Portuguese, and understand it as an instrument steeped in culture. The choice of this kind in the world of the epistolary genre was made because it promotes the participation of the Other in the configuration of dialogic relations in the elaboration of the statements, taken as a reference in a Bakhtinian conception of language. It aimed to (1) point, in the letters exchanged between teachers and students between the Portuguese, Brazilians and Angolans in the early years, the evidence of authorship linguistic behavior that could signal the beginning of autonomy in the use of the written modality of language, or signs of submissive behavior; (2) reveal the cultural content of language tools used to construct indicators listed on the customs of each people, including those constituting school doings. For the generation of data were used principles of action research, which enabled direct action, along with Brazilian teachers, and indirectly, in the case of the foreign teachers. From the analysis of the corpus of research - letters exchanged between teachers and students - it is clear that the appropriation of language as a speech act realized in human relations makes the old personal correspondence a powerful instrument of development in the area of the written language.

The DOK1 gene is a putative tumour suppressor gene located on the human chromosome 2p13 which is frequently rearranged in leukaemia and other human tumours. We previously reported that the DOK1 gene can be mutated and its expression down-regulated in human malignancies. However, the mechanism underlying DOK1 silencing remains largely unknown. We show here that unscheduled silencing of DOK1 expression through aberrant hypermethylation is a frequent event in a variety of human malignancies. DOK1 was found to be silenced in nine head and neck cancer (HNC) cell lines studied and DOK1 CpG hypermethylation correlated with loss of gene expression in these cells. DOK1 expression could be restored via demethylating treatment using 5-aza-2'deoxycytidine. In addition, transduction of cancer cell lines with DOK1 impaired their proliferation, consistent with the critical role of epigenetic silencing of DOK1 in the development and maintenance of malignant cells. We further observed that DOK1 hypermethylation occurs frequently in a variety of primary human neoplasm including solid tumours (93% in HNC, 81% in lung cancer) and haematopoietic malignancy (64% in Burkitt's lymphoma). Control blood samples and exfoliated mouth epithelial cells from healthy individuals showed a low level of DOK1 methylation, suggesting that DOK1 hypermethylation is a tumour specific event. Finally, an inverse correlation was observed between the level of DOK1 gene methylation and its expression in tumour and adjacent non tumour tissues. Thus, hypermethylation of DOK1 is a potentially critical event in human carcinogenesis, and may be a potential cancer biomarker and an attractive target for epigenetic-based therapy.

The growth and the metabolism of Bifidobacterium adolescentis MB 239 fermenting GOS, lactose, galactose, and glucose were investigated. An unstructerd unsegregated model for growth of B. adolescentis MB 239 in batch cultures was developed and kinetic parameters were calculated with a Matlab algorithm. Galactose was the best carbon source; lactose and GOS led to lower growth rate and cellular yield, but glucose was the poorest carbon source. Lactate, acetate and ethanol yields allowed calculation of the carbon fluxes toward fermentation products. Similar distribution between 3- and 2-carbon products was observed on all the carbohydrates (45 and 55%, respectively), but ethanol production was higher on glucose than on GOS, lactose and galactose, in decreasing order. Based on the stoichiometry of the fructose 6-phosphate shunt and on the carbon distribution among the products, ATP yield was calculated on the different carbohydrates. ATP yield was the highest on galactose, while it was 5, 8, and 25% lower on lactose, GOS, and glucose, respectively. Therefore, a correspondance among ethanol production, low ATP yields, and low biomass production was established demonstrating that carbohydrate preferences may result from different sorting of carbon fluxes through the fermentative pathway. During GOS fermentation, stringent selectivity based on the degree of polymerization was exhibited, since lactose and the trisaccharide were first to be consumed, and a delay was observed until longer oligosaccharides were utilized. Throughout the growth on both lactose and GOS, galactose accumulated in the cultural broth, suggesting that β-(1-4) galactosides can be hydrolysed before they are taken up. The physiology of Bifidobacterium adolescentis MB 239 toward xylooligosaccharides (XOS) was also studied and our attention was focused on an extracellular glycosyl-hydrolase (β-Xylosidase) expressed by a culture of B. adolescentis grown on XOS as sole carbon source. The extracellular enzyme was purified from the the supernatant, which was dialyzed and concentrated by ultrafiltration. A two steps purification protocol was developed: the sample was loaded on a Mono-Q anion exchange chromatography and then, the active fractions were pooled and β-Xylosidase was purified by gel filtration chromatography on a Superdex-75. The enzyme was characterized in many aspects. β- Xylosidase was an homo-tetramer of 160 kDa as native molecular mass; it was a termostable enzyme with an optimum of temperature at 53 °C and an optimum of pH of 6.0. The kinetics parameter were calculated: km = 4.36 mM, Vmax = 0.93 mM/min. The substrate specificity with different di-, oligo- and polysaccharides was tested. The reactions were carried out overnight at pH 7 and at the optimum of temperature and the carbohydrates hydrolysis were analyzed by thin layer chromatography (TLC). Only glycosyl-hydrolase activities on XOS and on xylan were detected, whereas sucrose, lactose, cellobiose, maltose and raffinose were not hydrolyzed. It’s clearly shown that β-Xylosidase activity was higher than the Xylanase one. These studies on the carbohydrate preference of a strain of Bifidobacterium underlined the importance of the affinity between probiotics and prebiotics. On the basis of this concept, together with Barilla G&R f.lli SpA, we studied the possibility to develop a functional food containing a synbiotic. Three probiotic strains Lactobacillus plantarum BAR 10, Streptococcus thermophilus BAR 20, and Bifidobacterium lactis BAR 30 were studied to assess their suitability for utilization in synbiotic products on the basis of antioxidative activity, glutathione production, acid and bile tolerance, carbohydrates fermentation and viability in food matrices. Bile and human gastric juice resistance was tested in vitro to estimate the transit tolerance in the upper gastrointestinal tract. B. lactis and L. plantarum were more acid tolerant than S. thermophilus. All the strains resisted to bile. The growth kinetics on 13 prebiotic carbohydrates were determined. Galactooligosaccharides and fructo-oligosaccharides were successfully utilized by all the strains and could be considered the most appropriate prebiotics to be used in effective synbiotic formulations. The vitality of the three strains inoculated in different food matrices and maintained at room temperature was studied. The best survival of Lactobacillus plantarum BAR 10, Streptococcus thermophilus BAR 20, and Bifidobacterium lactis BAR 30 was found in food chocolate matrices. Then an in vivo clinical trial was carried out for 20 healthy volunteers. The increase in faecal bifidobacteria and lactobacilli populations and the efficacy of the pre-prototype was promising for the future develop of potential commercial products.

Quelle est la différence entre un flibustier et un pirate ? Ce projet de recherche est basé sur cette question enfantine. En fait, les historiens contemporains, qui ont consacré de nombreuses pages à la « piraterie » et à la « flibuste » de l’âge moderne, n’ont pas réussi à répondre à cette – apparemment – simple question. Les reconstructions et les théories développées, en fait, ont assimilé ces deux phénomnes, en traitant les deux termes comme de simples synonymes. Mais, si cela peut être considéré comme vrai aujourd’hui, il n'était pas au cours des XVIIème et XVIIIème siècles. À l'époque, pirate était équivalent de « hostis humani generis », et comme tel craint et persécuté. Les flibustiers, au contraire, ont été considérés comme l’un des groupes les plus importants dans les premiers établissements des îles de la Mer des Carabes. En outre, le terme flibustier, dans la correspondance des gouverneurs français, se réfère à un élément considéré comme essentiel pour la réussite du processus de construction de la colonie, et également cruciale pour la consolidation des établissements américains. C’est donc la relation spéciale entre les flibustiers et le Nouveau Monde est l’objet de cette thèse. En utilisant une approche historico-culturel, on a essayé de contextualiser le phénomne de la flibuste dans le « processus d’américanisation » des premières communautés européennes en Amérique. La relation étroite entre les colons des établissements français de Saint-Domingue (aujourd’hui Hati) et les nombreux espaces - économique, militaire, diplomatique, social, humain - des Carabes a fait des flibustiers un élément fortement « américanisé ». Grâce à la lecture des documents d'archives et les mmoires des années entre 1684 et 1727, on a reconstruit la dynamique de la relation entre ce groupe et le contexte américain, avec une attention particulière à la dynamique culturelle et sociale.

Bioconjugation of peptides and asymmetric synthesis of gem-difluoromethylene compounds are areas of the modern organic chemistry for which mild and selective methods continue to be developed. This thesis reports new methodologies for these two areas based on the use of stabilized carbenium ions. The reaction that makes the bioconjugation of peptides possible takes place via the direct nucleophilic substitution of alcohols and is driven by the spontaneous formation of stabilized carbenium ions in water. By reacting with the thiol group of cysteine in very mild conditions and with a high selectivity, these carbenium ions allow the site-specific ligation of polypeptides containing cysteine and their covalent derivatization with functionalized probes. The ligation of the indole ring of tryptophan, an emerging target in bioconjugation, is also shown and takes place in the same conditions. The second area investigated is the challenging access to optically active gem-difluoromethylene compounds. We describe a methodology relying on the synthesis of enantioenriched 1,3-benzodithioles intermediates that are shown to be precursors of the corresponding gem-difluoromethylene analogues by oxidative desulfurization-fluorination. This synthesis takes advantage of the highly enantioselective organocatalytic α-alkylation of aldehydes with the benzodithiolylium ion and of the wide possibilities of synthetic transformations offered by the 1,3-benzodithiole group. This approach allows the asymmetric access to complex gem-difluoromethylene compounds through a late-stage fluorination step, thus avoiding the use of fluorinated building blocks.

In vitro engineered tissues which recapitulate functional and morphological properties of bone marrow and bone tissue will be desirable to study bone regeneration under fully controlled conditions. Among the key players in the initial phase of bone regeneration are mesenchymal stem cells (MSCs) and endothelial cells (ECs) that are in close contact in many tissues. Additionally, the generation of tissue constructs for in vivo transplantations has included the use of ECs since insufficient vascularization is one of the bottlenecks in (bone) tissue engineering. Here, 3D cocultures of human bone marrow derived MSCs (hBM-MSCs) and human umbilical vein endothelial cells (HUVECs) in synthetic biomimetic poly(ethylene glycol) (PEG)-based matrices are directed toward vascularized bone mimicking tissue constructs. In this environment, bone morphogenetic protein-2 (BMP-2) or fibroblast growth factor-2 (FGF-2) promotes the formation of vascular networks. However, while osteogenic differentiation is achieved with BMP-2, the treatment with FGF-2 suppressed osteogenic differentiation. Thus, this study shows that cocultures of hBM-MSCs and HUVECs in biological inert PEG matrices can be directed toward bone and bone marrow-like 3D tissue constructs.

Ce numro de la RSH revient sur ces inédits à travers un dossier critique (contributions de C. Roullier, J. Kaempfer, M. Mgevand & P. Suter), tout en en proposant d’autres : Le Couloir, adapté à partir de 3 manuscrits de Pinget par L. Koechlin ; la correspondance Pinget – Robbe-Grillet, ainsi que de nombreux documents inédits).
