Bacillus circulans D1 is a good producer of extracellular thermostable xylanase. Xylanase production in different carbon sources was evaluated and the enzyme synthesis was induced by various carbon sources. It was found that D-maltose is the best inducer of the enzyme synthesis ( 7.05 U/ mg dry biomass at 48 h), while D-glucose and D-arabinose lead to the production of basal levels of xylanase. The crude enzyme solution is free of cellulases, even when the microorganism was cultivated in a medium with D-cellobiose. When oat spelt xylan was supplemented with D-glucose, the repressive effect of this sugar on xylanase production was observed at 24 h, only when used at 5.0 g/ L, leading to a reduction of 60% on the enzyme production. on the other hand, when the xylan medium was supplemented with D- xylose ( 3.0 or 5.0 g/ L), this effect was more evident ( 80 and 90% of reduction on the enzyme production, respectively). Unlike that observed in the xylan medium, glucose repressed xylanase production in the maltose medium, leading to a reduction of 55% on the enzyme production at 24 h of cultivation. Xylose, at 1.0 g/ L, induced xylanase production on the maltose medium. on this medium, the repressive effect of xylose, at 3.0 or 5.0 g/ L, was less expressive when compared to its effect on the xylan medium.

Vegetated riparian buffer strips have been established in Southern Quebec (Canada) in order to intercept nutrients such as nitrate (NO(3)(-)) and protect water quality near agricultural fields. Buffer strips may also favour denitrification through a combination of high soil moisture, NO(3)(-) and carbon supply, which could lead to the production of nitrous oxide (N(2)O), a greenhouse gas. Denitrification could be further amplified by the presence of earthworms, or by plant species that promote earthworm and bacterial activity in soils. Soils from four farms, comprising maize fields and adjacent buffer strips, were sampled in the fall of 2008. A total of six earthworm species were found, but average earthworm biomass did not differ between buffer strips and maize agroecoecosystems. Nitrate concentrations and net nitrification rates were higher in the maize fields than in the buffer strips: there was no difference in N(2)O production in soils collected from the two sampling locations. Potential denitrification, measured by acetylene inhibition, varied by two orders of magnitude, depending on experimental conditions: when amended with H(2)O or with H(2)O + NO3-, potential denitrification was higher (P < 0.05) in soils from buffer strips than from maize fields. Potential denitrification was highest in soils amended with H(2)O+glucose, or with H(2)O+ NO(3)(-) + glucose. Using microcosms, we tested the effect of litter-soil mixtures on earthworm growth, and the effect of earthworm-litter-soil mixtures on potential denitrification. Based on four categories of chemical assays, litters of woody species (oak, apple, Rhododendron) were generally of lower nutritional quality than litter from agronomic species (alfalfa, switchgrass, corn stover). Alfalfa litter had the most positive effect, whereas apple litter had the most negative effect, on earthworm growth. Potential denitrification was 2-4 times higher in earthworm-litter-soil mixtures than in plain soil. Litter treatments that included corn stover had lower potential denitrification than those that included alfalfa or switchgrass, whereas litter treatments that included oak had lower potential denitrification than those that included apple or Rhododendron. Results suggest that potential N(2)O emissions may be higher in riparian buffer strips than in adjacent maize fields, that N(2)O emissions in buffer strips may be amplified by comminuting earthworms, and that plant litters that reduce earthworm growth may not be best at mitigating N(2)O emissions. This paper evaluates emissions to the atmosphere of biologically available nitrogen compounds in a region characterized by intensive sugar cane biofuel ethanol production. Large emissions of NH(3) and NO,, as well as particulate nitrate and ammonium, occur at the harvest when the crop is burned, with the amount of nitrogen released equivalent to similar to 35% of annual fertilizer-N application. Nitrogen oxides concentrations show a positive association with fire frequency, indicating that biomass burning is a major emission source, with mean concentrations of NO, doubling in the dry season relative to the wet season. During the dry season biomass burning is a source of NH3, with other sources (wastes, soil, biogenic) predominant during the wet season. Estimated NO(2)-N, NH(3)-N, NO(3)(-)-N and NH(4)(+)-N emission fluxes from sugar cane burning in a planted area,of ca. 2.2 x 10(6) ha are 11.0, 1.1, 0.2, and 1.2 Gg N yr(-1), respectively.

During the TROCCINOX field experiment in January and February 2005, the contribution of lightning-induced nitrogen oxides (LNOx) from tropical and subtropical thunderstorms in Southern Brazil was investigated. Airborne trace gas measurements (NO, NOy, CO and O-3) were performed up to 12.5 km with the German research aircraft Falcon. During anvil penetrations in selected tropical and subtropical thunderstorms of 4 and 18 February, NOx mixing ratios were on average enhanced by 0.7-1.2 and 0.2-0.8 nmol mol(-1) totally, respectively. The relative contributions of boundary layer NOx (BL-NOx) and LNOx to anvil-NOx were derived from the NOx-CO correlations. on average similar to 80-90% of the anvil-NOx was attributed to LNOx. A Lightning Location Network (LINET) was set up to monitor the local distribution of cloud-to-ground (CG) and intra-cloud (IC) radiation sources (here called 'strokes') and compared with lightning data from the operational Brazilian network RINDAT (Rede Integrada Nacional de Deteccao de Descargas Atmosfericas). The horizontal LNOx mass flux out of the anvil was determined from the mean LNOx mixing ratio, the horizontal outflow velocity and the size of the vertical cross-section of the anvil, and related to the number of strokes contributing to LNOx. The values of these parameters were derived from the airborne measurements, from lightning and radar observations, and from a trajectory analysis. The amount of LNOx produced per LINET stroke depending on measured peak current was determined. The results were scaled up with the Lightning Imaging Sensor (LIS) flash rate (44 flashes s(-1)) to obtain an estimate of the global LNOx production rate. The final results gave similar to 1 and similar to 2-3 kg(N) per LIS flash based on measurements in three tropical and one subtropical Brazilian thunderstorms, respectively, suggesting that tropical flashes may be less productive than subtropical ones. The equivalent mean annual global LNOx nitrogen mass production rate was estimated to be 1.6 and 3.1 Tg a(-1), respectively. By use of LINET observations in Germany in July 2005, a comparison with the lightning activity in mid-latitude thunderstorms was also performed. In general, the same frequency distribution of stroke peak currents as for tropical thunderstorms over Brazil was found. The different LNOx production rates per stroke in tropical thunderstorms compared with subtropical and mid-latitude thunderstorms seem to be related to the different stroke lengths (inferred from comparison with laboratory data and observed lengths). In comparison, the impact of other lightning parameters as stroke peak current and stroke release height was assessed to be minor. The results from TROCCINOX suggest that the different vertical wind shear may be responsible for the different stroke lengths.

Some factors influencing the growth and production of extracellular lipase by Rhizopus oligosporus were studied. Highest yields of enzyme were obtained when Tweens were the carbon source. Soybean meal extract supported good growth and enzyme production. Carbohydrates, vegetable oils, proteins or amino acids did not stimulate lipase production. The fungus grew well with carbohydrate- or protein-supplemented media but not with oils, unless emulsified with a non-metabolizable gum. The production of biomass in static cultures was maximum at 35-40°C after 4 d at pH 5.5. The yield of lipase was maximum at 25°C after 3 d at pH 6.5. Shaking cultures enhanced growth but decreased lipase production.

Cell cultures of Mikania glomerata Sprengel were established with leaf segments cultured on White medium supplemented with 1 mg/L BA and 3 mg/L NAA. Different types and concentrations of growth regulators were tested for callus maintenance. Determination of coumarin content was performed in HPLC using authentic coumarin standard. Growth regulator concentration affected biomass and coumarin accumulation. Cultures developed in semisolid medium containing both BA and NAA exhibited enhanced biomass production as well as coumarin accumulation. In the most favorable conditions tested, cells accumulated 25 μg/g of dry weight what is much inferior to the yield already reported in intact plants (5 mg/g of dry weight). However, results obtained so far suggest several alternatives for culture manipulation in order to optimize the productivity of coumarin by M. glomerata cultured cells.

Titanium and its alloys provide high strength-to-weight ratios, good fatigue strength and increased corrosion resistance compared with others materials. Its acceptance in aerospace has been limited by costs considerations such as high cost of raw material, high buy-to-fly ratios and expensive machining operations. Significant cost reductions can be obtained by vacuum sintering and powder metallurgy (P/M) techniques by producing near net shapes and consequently minimizing material waste and machining time. The Ti 35Nb alloy exhibit a low modulus of elasticity. Stemming from the unique combination of high strength, low modulus of elasticity and low density, this alloy is intrinsically more resistant to shock and explosion damages than most other engineering materials. Samples were produced by mixing of initial metallic powders followed by uniaxial and cold isostatic pressing with subsequent densification by sintering between 900 and 1600 °C, in vacuum. Sintering behavior was studied by means of dilatometry. Sintered samples were characterized for phase composition, microstructure and microhardness by X-ray diffraction, scanning electron microscopy and Vickers indentation, respectively. Fatty acid production by four strains of Mucor hiemalis grown in plant oil and soluble carbohydrates

Four Mucor hiemalis strains (M1, M2, M3 and M4), isolated from soil at a depth of 0 - 15 cm in the Juréia-Itatins Ecology Station (JIES), in the state of São Paulo, Brazil and were evaluated for the production of γ-linolenic (GLA) and other unsaturated fatty acids. Five growth variables (temperature, pH, carbon source, nitrogen source, and vegetable oils) were studied. Liquid media containing 2% vegetable oil (palm oil, canola oil, soybean oil, sesame oil, or sunflower oil) or 2% carbohydrate (fructose, galactose, glycerol, glucose, lactose, maltose, sucrose, sorbitol or xylose) and 1% yeast extract as a nitrogen source were used. The greatest biomass production was observed with M3 and M4 strains in palm oil (91.5 g l -1) and sunflower oil (68.3 g l -1) media, respectively. Strain M4 produced greater quantities of polyunsaturated acids in medium containing glucose. The GLA production in the M4 biomass was 1,132.2 mg l -1 in glucose medium. Plant oils were inhibitors of fatty acid production by these strains. © 2007 Academic Journals.

Due to the confirmation of its medicinal properties, the demand and the area planted with 'espinheira-santa' have been increasing. However, there are no sources of propagation material with known characteristics. Progenies of Maytenus ilicifolia Martius ex Reiss (espinheira-santa), originating from Southern Brazil have been analyzed, and currently comprise a germplasm inventory for the species. Studies included the assessment of in natura growth features (height and production of biomass canopy) and plant chemistry (lignin, phenols and mineral element contents), and targeted the creation of a data base, characterizing populations/progenies to be used in future selection and breeding programs of the species. Among the features assessed, were considered desirable the ones related to productivity (high foliar mass to branch mass ratio) and also the absence of thorns on the leaves. High total phenol and tannin content were observed in all the progenies assessed and these features is desirable as long the absence of toxic effects and preservation of the therapeutic quality of the plant medicinal elements produced can be proved.

The present study sought biotensoactive production from soybean oil fry waste using Pseudomonas aeruginosa ATCC 10145 and Pseudomonas aeruginosa isolated from the soil of a petroleum station having undergone gasoline and diesel oil spills. The results of the experiments were analyzed using a complete factorial experimental design, investigating the concentration of soybean oil waste, ammonia sulfate and residual brewery yeast. Assays were performed in 250-mL Erlenmeyer beakers containing 50 mL of production medium, maintained on a rotary shaker at 200 rpm and a temperature of 30±1 °C for a 48-hour fermentation period. Biosurfactant production was monitored through the determination of rhamnose, surface tension and emulsification activity. The Pseudomonas aeruginosa ATCC 10145 strain and isolated Pseudomonas aeruginosa were able to reduce the surface tension of the initial mexlium from 61 mN/m to 32.5 mN/m and 30.0 mN/m as well as produce rhamnose at concentrations of 1.96 and 2.89 g/L with emulsification indices of 96% and 100%, respectively.

Many plants utilized in the urban center shows substances considered toxic whose production could be influenced by some factors, like hydric stress, including ornamental Nerium oleander L., widely used in gardens in various parts of the world, which presents production of cardioative glucosides, considered toxic. This study had the objective to evaluate the effect of field capacity in the biomass and the level of cardioative glucosides in seedlings of Nerium oleander. The experiment was carried at UNIDERP, in Campo Grande City, Mato Grosso do Sul State, Brazil, at the University for Development of State and Pantanal Region, using the experimental delineation in randomized blocks. There were 4 treatments (25%; 50%; 75% and 100% of the field capacity), 5 replications and 4 plants by parcel, totalling 80 plants. The evaluations were realized 60 days after the seedlings were planted. The quantitative analysis of the cardioative glucosides was realized by gravimetric test, after selective extraction of the glucosides. Were conclude that increase of the quantity of water in the soil raised the biomass production until 75% of the field capacity and increased the level of cardioative glucosides, showing that water management is very important and should be provided only the necessary to development of the plant.

We present a search for associated production of Higgs and W bosons in pp̄ collisions at a center of mass energy of s=1.96 TeV in 5.3 fb-1 of integrated luminosity recorded by the D0 experiment. Multivariate analysis techniques are applied to events containing one lepton, an imbalance in transverse energy, and one or two b-tagged jets to discriminate a potential WH signal from Standard Model backgrounds. We observe good agreement between data and expected backgrounds, and set an upper limit of 4.5 (at 95% confidence level and for mH=115 GeV) on the ratio of the WH cross section multiplied by the branching fraction of H→bb̄ to its Standard Model prediction, which is consistent with an expected limit of 4.8. © 2011 Elsevier B.V.
